Rat pups, seven days old, with right carotid artery ligations were exposed to an atmosphere of oxygen 8% remainder nitrogen for 2 hr. The animals that survived for three weeks after the hypoxic-ischemic episode had clusters of darkly stained (hematoxylin-eosin) neurons in the cortex and reduced uptake of dopamine (frontal cortex) and choline (frontal cortex, hippocampus and striatum) in preparations of synaptosomes. Treatment with GM1 ganglioside partially corrected the loss of uptake activity and increased the number of darkly stained neurons.

A new ligand (Ts kappa) of the apamin binding site on rat brain synaptosomes (K0.5 = 300 pM) was purified and characterized from the venom of Tityus serrulatus. It is a polypeptide toxin of 35 amino acid residues, with three disulfide bridges. Its cDNA was amplified from a venom gland cDNA library and the nucleotide sequence determined. A model of Ts kappa was constructed by amino acid replacement using charybdotoxin structure as determined by 1H nuclear magnetic resonance as starting model.

Inhibition of Na, K-ATPase in fractions of unpurified synaptosomes and of heavy microsomes from lumbar part of spinal cord was found in dynamics of botulinic intoxication of the C type in rats. Both competitive and noncompetitive types of inhibition of the enzymatic activity were found in kinetic studies. Inhibition of transport functions across biological membranes caused by botulinic toxin was shown to have a reversible character. The inhibition might be prevented within the early periods of botulinic intoxication by administration of serotonin and guanidine hydrochloride.

The activity of 0.01-1 microM of melatonin on depolarization-induced 45Ca2+ influx by hypothalamic synaptosomes of rats killed at late evening (2000 h) depended on melatonin preincubation time, exhibiting an early (10 min) stimulation of uptake and an inhibition after 30 min of incubation. Both stimulatory and inhibitory effects were maximal at 2000 h and tended to a minimum at 2400 h, exhibiting intermediate values at 400 h. At 1000 h, only inhibitory effects of melatonin (1 microM) on Ca2+ uptake were found.

Acute effects of ethanol on Na(+)-dependent transport of gamma-aminobutyric acid (GABA) and glutamic acid (GLU) were investigated in crude synaptosomal preparations from rat cerebral cortex.
