Building new hearts: a review of trends in cardiac tissue engineering.

Cardiovascular disease (CVD) is the number one cause of death in the United States. However, few treatments for CVD provide a means to regain full cardiac function with no long-term side effects. Novel tissue-engineered products may provide a way to overcome the limitations of current CVD therapies by replacing injured myocardium with functioning tissue or by inducing more constructive forms of endogenous repair. In this review, we discuss some of the factors that should be considered in the development of tissue-engineered products, and we review the methods currently being investigated to ge

Successful human long-term application of in situ bone tissue engineering.

Tissue Engineering (TE) and Regenerative Medicine (RM) have gained much popularity because of the tremendous prospects for the care of patients with tissue and organ defects. To overcome the common problem of donor-site morbidity of standard autologous bone grafts, we successfully combined tissue engineering techniques for the first time with the arteriovenous loop model to generate vascularized large bone grafts. We present two cases of large bone defects after debridement of an osteomyelitis. One of the defects was localized in the radius and one in the tibia. For osseus reconstruction, arte…

Construction of three-dimensional vascularized cardiac tissue with cell sheet engineering.

Construction of three-dimensional (3D) tissues with pre-isolated cells is a promising achievement for novel medicine and drug-discovery research. Our laboratory constructs 3D tissues with an innovative and unique method for layering multiple cell sheets. Cell sheets maintain a high-efficiently regenerating function, because of the higher cell density and higher transplantation efficiency, compared to other cell-delivery methods. Cell sheets have already been applied in clinical applications for regenerative medicine in treating patients with various diseases. Therefore, in our search to develo

Current concepts of bone tissue engineering for craniofacial bone defect repair.

Craniofacial fractures and bony defects are common causes of morbidity and contribute to increasing health care costs. Successful regeneration of bone requires the concomitant processes of osteogenesis and neovascularization. Current methods of repair and reconstruction include rigid fixation, grafting, and free tissue transfer. However, these methods carry innate complications, including plate extrusion, nonunion, graft/flap failure, and donor site morbidity. Recent research efforts have focused on using stem cells and synthetic scaffolds to heal critical-sized bone defects similar to those s

Utilization of stem cells in alginate for nucleus pulposus tissue engineering.

In a general view of anatomy, intervertebral disc is composed of three parts: annulus fibrosus (AF), nucleus pulposus (NP), and cartilage endplate (CEP). Recently, several types of stem cells were successfully isolated from these corresponding regions, but up to now, no research was performed about which kind of stem cells is the most efficient candidate for NP tissue engineering or for stem cell-based disc regeneration therapy. In this study, we compared the regenerative potentials of the above-mentioned three kinds of disc-derived stem cells with that of the classic bone marrow (BM)-mesenchy…

Icariin: does it have an osteoinductive potential for bone tissue engineering?

Traditional Chinese medicines have been recommended for bone regeneration and repair for thousands of years. Currently, the Herba Epimedii and its multi-component formulation are the attractive native herbs for the treatment of osteoporosis. Icariin, a typical flavonol glycoside, is considered to be the main active ingredient of the Herba Epimedii from which icariin has been successfully extracted. Most interestingly, it has been reported that icariin can be delivered locally by biomaterials and that it has an osteoinductive potential for bone tissue engineering. This review focuses on the per…

Stem cell and biomaterials research in dental tissue engineering and regeneration.

This review summarizes approaches used in tissue engineering and regenerative medicine, with a focus on dental applications. Dental caries and periodontal disease are the most common diseases resulting in tissue loss. To replace or regenerate new tissues, various sources of stem cells have been identified such as somatic stem cells from teeth and peridontium. Advances in biomaterial sciences including microfabrication, self-assembled biomimetic peptides, and 3-dimensional printing hold great promise for whole-organ or partial tissue regeneration to replace teeth and periodontium.

[Application of tissue engineering technology and stem cells in urethral reconstruction].

Urethral defects caused by congenital abnormality, injury, inflammation or tumor are common diseases in urology. Conventional surgery of urethral reconstruction may lead to many complications, and its application has been greatly limited by insufficient supply of donor materials. Tissue engineering technology and stem cells can be used to construct biological substitutes for maintaining and repairing damaged tissue and organ function, which offers a new prospect for the strategies of urethral reconstruction.

Stem cells combined with bone graft substitutes in skeletal tissue engineering.

INTRODUCTION: Bone grafting is used to repair large bone defects and autograft is recognised as producing the best clinical outcome, which is partly due to its cellular component. When autograft is unavailable, allograft and bone graft substitutes can be used; however, they rely on the host bed to provide cellular osteogenic activity. AREAS COVERED: Bone graft substitutes have the potential to benefit from the addition of stem cells aimed at enhancing the rate and quality of defect repair. Mesenchymal stem cells (MSCs) can be isolated from bone marrow or periosteum and culture expanded. Other

Stem cells from umbilical cord and placenta for musculoskeletal tissue engineering.

Mesenchymal stem cells isolated from amnion/amniotic fluid, umbilical cord blood, placental tissue, umbilical cord vein and the Wharton's Jelly are promising candidates for musculoskeletal tissue engineering of bone and cartilage tissues. The extracorporeal nature of this source avoids the ethical concerns that plague the isolation of embryonic stem cells. Moreover, the harvesting does not require the invasive and discomfort extraction procedures as well as patient risks that attend adult stem cell isolation. Current preclinical studies support the application of these cell-based therapies for…

Porous keratin scaffold-promising biomaterial for tissue engineering and drug delivery.

A porous keratin scaffold, prepared from the reduced keratin solution, has shown good cell viability which makes it a potential candidate for cell seeding. An aqueous solution of reduced keratin was extracted from horn meal using a mixture of urea, sodium dodecyl sulfate, mercaptoethanol, and water at 60 degrees C. The molecular mass of the extracted keratin is found to be ranging between 225 and 150 KDa. The CD spectrum of aqueous solution of keratin shows the presence of infinity-helical structure with beta-turns as negative absorption band at 225 nm and as positive absorption band at 195 nm

Mesenchymal stem cells as a potential pool for cartilage tissue engineering.

Osteoarthritis (OA) resulting from trauma, degenerative or age-related disease presents a major clinical challenge due to the limited repair capacity of articular cartilage. This poor self-repair capacity of osteochondral defects has resulted in the development of a wide variety of new treatment approaches. Although the use of chondrocytes in applications of cartilage tissue engineering is still prevalent, concerns associated with donor-site morbidity, cell de-differentiation and the limited lifespan of these cells have brought the use of mesenchymal stem cells (MSCs) to the forefront of such …

Mesenchymal stem cell tissue engineering: techniques for isolation, expansion and application.

Mesenchymal stem cells (MSCs) are undifferentiated multipotent cells which reside in various human tissues and have the potential to differentiate into osteoblasts, chondrocytes, adipocytes, fibroblasts and other tissues of mesenchymal origin. In the human body they could be regarded as readily available reservoirs of reparative cells capable to mobilize, proliferate and differentiate to the appropriate cell type in response to certain signals. These properties have triggered a variety of MSC-based therapies for pathologies including nonunions, osteogenesis imperfecta, cartilage damage and myo

Muscle-based gene therapy and tissue engineering to improve bone healing.

Failed fracture healing is a significant problem in orthopaedics, often seen in patients with scaphoid fractures, high-energy injuries, and osteoporosis. Current treatments often result in poor outcomes and donor site morbidity. Gene therapy has been the focus of much recent research to improve bone healing. In the current review, the authors specifically evaluate the use of muscle-derived cells as a gene delivery vehicle and inducible osteoprogenitor cell that can enhance bone regeneration. Muscle-derived cells have been used to deliver bone morphogenetic protein-2 and produce ectopic bone. T

Functional tissue engineering: the role of biomechanics in articular cartilage repair.

Articular cartilage shows little or no intrinsic capacity for repair in response to injury or disease, and even minor lesions or injuries may lead to progressive damage and joint degeneration. Tissue engineering is a relatively new but rapidly growing field that has sought to use combinations of implanted cells, biomaterials, and biologically active molecules to repair or regenerate injured or diseased tissues. Despite many advances, tissue engineers have faced significant challenges in repairing or replacing tissues that serve a predominantly biomechanical function, such as articular cartilag…

Comparative evaluation of cell types for endothelialization in cardiovascular tissue engineering.

Tissue engineering represents a promising approach to overcome the limitations of current vascular grafts by promoting endothelialization. However, the successful fabrication of small-diameter (<6 mm) tissue-engineered vascular grafts (TEVGs) that maintain the desired mechanical strength, biocompatibility, and long-term patency of native vessels remains a significant challenge. The development of an ideal TEVG depends largely on achieving complete endothelial coverage using appropriate autologous cells that can mimic the functional properties of native endothelium. Several studies have highlig…
