Atmospheric particulate matter (PM) is a heterogeneous carrier of toxic metals whose environmental fate and biological effects depend on particle size, source-related composition, chemical form, solubility, and bioaccessibility. Lead (Pb), cadmium (Cd), and nickel (Ni) are of particular concern because atmospheric transport and deposition connect air pollution with persistent contamination of soils, vegetation, waters, sediments, food, and feed, followed by human and animal exposure. This review integrates evidence across a source-to-effect continuum encompassing emission, atmospheric transpor

Titanium dioxide (TiO2), used as a food additive (labeled E171 in Europe), was withdrawn from the European market in 2022. The E171 toxicity mechanism involves its uptake, oxidative stress, DNA damage, and inflammation. It has been hypothesized that the TiO2 accumulation nanoparticles (NPs) or E171 triggers tissue damage, and some studies have quantified titanium (Ti) concentration in several organs. Still, the accumulation pattern and toxicokinetics remain unknown. We aimed to systematically review the Ti accumulation in the liver, kidney, spleen, intestine, and colon as these tissues have be

Zinc phosphide (ZnP) is a rodenticide commonly used in agriculture for pest control, but it can cause acute human poisoning, through ingestion in suicide attempts. The toxicokinetics of ZnP are not well understood, although it is believed that its primary mechanism of toxicity involves the inhibition of Complex IV of Cytochrome C Oxidase, similar to aluminum phosphide (ALP) poisoning. However, there are notable differences between the two. The rarity of acute ZnP toxicity and the misconception that it mirrors ALP poisoning have hindered the development of effective treatment strategies. This r

Nonoxidative ethanol metabolites have been reported as ethanol biomarkers in clinical and forensic cases. However, their forensic toxicokinetics are still unclear. The study aimed to simultaneously research the postmortem distribution and dynamic distribution of ethanol and its nonoxidative metabolites in 62 poisoned rabbits. Firstly, 32 rabbits were randomly divided into three groups and sacrificed after three doses ethanol, and their organs were collected to study the postmortem distribution of ethanol and its nonoxidative metabolites. The results showed that the distribution trends of ethan

As the United States and the European Union continue their steady march towards the acceptance of new approach methodologies (NAMs), we need to ensure that the available tools are fit for purpose. Critics will be well-positioned to caution against NAMs acceptance and adoption if the tools turn out to be inadequate. In this paper, we focus on Quantitative Structure Activity-Relationship (QSAR) models and highlight how the training database affects quality and performance of these models. Our analysis goes to the point of asking, "are the endpoints extracted from the experimental studies in the

Nonclinical studies involve in vitro, in silico, and in vivo experiments to assess the toxicokinetics, toxicology, and safety pharmacology of drugs according to regulatory requirements by a national or international authority. In this review, we summarize the potential effects of various underlying diseases governing the absorption, distribution, metabolism, and excretion (ADME) of drugs to consider the use of animal models of diseases in nonclinical trials. Obesity models showed alterations in hepatic metabolizing enzymes, transporters, and renal pathophysiology, which increase the risk of dr

BACKGROUND: Current US federal action levels for domoic acid (DA) in seafood are based on acute toxicity observed in exposed adult humans. Life course considerations have not been incorporated. The potential for developmental neurotoxicity (DNT) at permissible DA levels has previously been noted, but not methodically assessed.
METHODS: Studies of DNT following DA exposure in experimental and wild animals were identified through a comprehensive search strategy. Evidence from papers meeting inclusion criteria was evaluated for specific outcomes reported for doses at which adverse effects were o

Recent epidemiological studies have reported associations between colorectal cancer incidence and nitrates in drinking-water, but not from the diet. The toxicokinetics of nitrates were reviewed and exposure data from New Zealand were analysed. Dietary (including drinking-water) exposure of New Zealanders to nitrates was found to be very similar to most other countries and within internationally-established acceptable daily intakes. Less than 10% of nitrate exposure was from drinking-water, with little difference between adults and children. Approximately half of the total water-based exposure

The last decade witnessed extraordinary advances in "omics" methods, particularly transcriptomics, proteomics and metabolomics, enabling toxicologists to integrate toxicokinetics and toxicodynamics with mechanistic insights on the mode-of-action of noxious chemicals, single or combined. The toxicology of mixtures is, nonetheless, a most challenging enterprise, especially for environmental toxicologists and ecotoxicologists, who invariably deal with chemical mixtures, many of which contain unknowns. Despite costs and demanding computations, the systems toxicology framework, of which "omics" is

Chromium(VI) compounds are classified as carcinogenic to humans. Whereas chromium measurements in urine and whole blood (i.e., including plasma) are indicative of recent exposure, chromium in red blood cells (RBC) is attributable specifically to Cr(VI) exposure. Before recommending Cr in RBC as a biological indicator of Cr(VI) exposure, in-vitro studies must be undertaken to assess its reliability. The present study examines the relationship between the chromium added to a blood sample and that subsequently found in the RBC. After incubation of total blood with chromium, RBC were isolated, cou

PURPOSE: Monocarboxylate transporter (MCT) inhibition represents a potential treatment strategy for γ-hydroxybutyric acid (GHB) overdose by blocking its renal reabsorption in the kidney. This study further evaluated the effects of a novel, highly potent MCT inhibitor, AR-C155858, on GHB toxicokinetics/toxicodynamics (TK/TD).
METHODS: Rats were administered GHB (200, 600 or 1500 mg/kg i.v. or 1500 mg/kg po) with and without AR-C155858. Breathing frequency was continuously monitored using whole-body plethysmography. Plasma and urine samples were collected up to 8 h. The effect of AR-C155858 on

Many epoxides and their precursors are high production volume chemicals that have major uses in the polymer industry and as intermediates in the manufacture of other chemicals. Several of these chemicals were demonstrated to be carcinogenic in laboratory animal studies conducted by the Ramazzini Foundation (e.g., vinyl chloride, acrylonitrile, styrene, styrene oxide, and benzene) and by the National Toxicology Program (e.g., ethylene oxide, 1,3-butadiene, isoprene, chloroprene, acrylonitrile, glycidol, and benzene). The most common sites of tumor induction were lung, liver, harderian gland, an

The Canadian Environmental Protection Act (CEPA) authorizes the Ministers of the Environment and of Health in Canada to investigate a wide variety of substances that may contaminate the environment and cause adverse effects on the environment and/or on human health. Under the Act, assessments have been completed for 44 environmental contaminants on the first Priority Substances List, including four metals and their compounds. The principles developed for the assessment of risk to human health for priority substances under CEPA are outlined, with specific emphasis on the metals. These include g

The derivation of Chemical Specific Adjustment Factors (CSAFs) (IPCS, 2005; U.S. EPA, 2014) depends on the choice of appropriate dose metric. EPA and IPCS guidance was applied to derive a CSAF for developmental toxicity for procymidone (PCM). Although kinetic data were not available in humans at any dose, sufficient toxicokinetic data are available in a surrogate species, primates, and from chimeric mice with both rat and human liver cells to offer insights. Alternative approaches were explored in the derivation of the CSAG based on review of the available kinetic data. The most likely dosimet

Essentials Hemophilia patients on concizumab prophylaxis may need rFVIIa to treat breakthrough bleeds. Effect and safety of concizumab + rFVIIa were tested in vitro and in vivo. Concizumab + rFVIIa had no additive effects on bleeding in hemophilic rabbits. High steady-state levels of concizumab did not affect the safety of rFVIIa in cynomolgus monkeys. SUMMARY: Background Concizumab is a monoclonal antibody (mAb) against tissue factor pathway inhibitor (TFPI), currently in clinical development as a subcutaneous prophylactic therapy for hemophilia A/B with and without inhibitors. In patients wi

Leachables are chemicals that can potentially occur in a drug product; therefore, it is important for patient safety to assess potential toxicity risk. Many parenteral drug products are liquid formulations and have an increased likelihood for leachables compared to other dosage forms. One approach for the safety assessment of potential leachables is to derive a Permitted Daily Exposure (PDE) from toxicity data. However, there is often a lack of availability of parenteral repeat dose toxicity studies to facilitate the assessment. In lieu of parenteral data, a bioavailability adjustment factor (

Evaluating the oral acute toxicity (AT) of corrosive acids and bases remains a challenge in chemical safety assessment. In current regulatory practice, waiving experimental testing often results in assignment to the highest hazard category. This may overestimate systemic toxicity and influence the treatment prioritization in poisoning cases, where local corrosive injury is the dominant driver of clinical outcome. This study proposes a mechanistically grounded hypothesis that the systemic AT of corrosive acids and bases is primarily determined by their counterions. Based on this premise, a read

In the current research, we examined the effects of warfarin (W1 = 5 mg/L and W2 = 25 mg/L) and tinzaparin (T1 = 5 mg/L and T2 = 25 mg/L) on meiofauna using microcosms. These microcosms were intentionally contaminated with both anticoagulants for one month. The findings indicated that nematodes and amphipods demonstrated the greatest resistance to the two anticoagulants evaluated. Specifically, the number of amphipods increased after exposure to the treatment that included T2. Following the separate introduction of each drug, amphipods displayed a taxonomic restructuring, with a more significa
