Electro-acupuncture inhibits C-fiber-evoked WDR neuronal activity of the trigeminocervical complex: Neurophysiological hypothesis of a complementary therapy for acute migraine modeled rats.

INTRODUCTION: Acupuncture has become a relevant complementary and alternative treatment for acute migraine; however, the neurophysiological mechanism (C-fibers) underlying this effect remains unclear. C-fibers play a crucial role for diffuse noxious inhibitory controls (DNIC) at wide dynamic range (WDR) neurons in the trigeminocervical complex (TCC) in migraine attacks, and we supposed that this may be the mechanism of acupuncture analgesia. This study aimed to examine the neurophysiology of acupuncture intervention in an acute migraine rat model. METHODS: Inflammatory soup (IS) or saline was

Characterising the Analgesic Effect of Different Targets for Deep Brain Stimulation in Trigeminal Anaesthesia Dolorosa.

BACKGROUND: Several deep brain stimulation (DBS) targets have been explored for the alleviation of trigeminal anaesthesia dolorosa. We aimed to characterise the analgesia produced from the periaqueductal grey (PAG) and centromedian-parafascicular (CmPf) nucleus using a within-subject design. METHOD: We report a case series of 3 subjects implanted with PAG and CmPf DBS systems for the treatment of anaesthesia dolorosa. At follow-up, testing of onset and offset times, magnitude, and thermal and mechanical sensitivity was performed. RESULTS: The mean pain score of the cohort was acutely reduced

Involvement of microglial activation in the brainstem in experimental dental injury and inflammation.

OBJECTIVE: The purpose of this study was to determine the effect of dental injury and inflammation on microglia in the trigeminal subnucleus caudalis (Vc). METHODS: Pulp exposure (PX) was performed on the first maxillary molar of 35 rats. Specimens were collected at 1, 3, 7, 14, 21 and 28 days after PX. Teeth were processed for H&E staining and immunohistochemical staining for OX-42, a marker of microgial activation, in the Vc. RESULTS: We observed that there was a progressive and persistent inflammation in the tooth. At 21-28 days after PX, the inflammation extended out into periodontal lig

GABA(A) receptor activation modulates corneal unit activity in rostral and caudal portions of trigeminal subnucleus caudalis.

Corneal nociceptors terminate at the trigeminal subnucleus interpolaris/caudalis (Vi/Vc) transition and subnucleus caudalis/upper cervical spinal cord (Vc/C1) junction regions of the lower brain stem. The aims of this study were to determine if local GABAA receptor activation modifies corneal input to second-order neurons at these regions and if GABAA receptor activation in one region affects corneal input to the other region. In barbiturate-anesthetized male rats, corneal nociceptors were excited by pulses of CO2 gas, and GABAA receptors were activated by microinjections of the selective agon

Masticatory movements and EMG activity following electrolytic lesions of the trigeminal motor nucleus in growing guinea pigs.

Unilateral electrolytic lesions were produced in the left trigeminal motor nucleus (TMN) of six guinea pigs at 49 days of age. Masticatory mandibular movement and EMG data were collected prior to lesioning and at 4 and 12 days postlesion. After the animals were killed 60 days postlesion, dissection and maceration revealed muscular atrophy and craniofacial asymmetries on the lesion side. Analyses of prelesion and postlesion mandibular movement and EMG data indicated significant (p less than 0.05; p less than 0.01) changes in chew cycle durations, dimensions, and EMG activity patterns. Shifts in

Sex-Specific Distribution of Glycine Transporter 2 in the Central Nervous System of Adult GlyT2-iCre-tdTomato Mouse Brain.

Glycine critically regulates locomotor and sensory processing, with its presynaptic glycine transporter 2 (GlyT2) serving as key therapeutic targets for modulating glycinergic neurotransmission. While GlyT2 inhibitors hold clinical promise, the specialization of spatially distributed GlyT2 neuronal populations across the brain remains incomplete. Here, we crossed GlyT2-iCre with Ai9 to generate GlyT2-iCre-tdTomato mice, enabling anatomical mapping and functional interrogation of GlyT2 circuits. Here, we generated GlyT2-iCre-tdTomato mice through crossbreeding GlyT2-iCre and Ai9 lines, enabling

Estrogen receptors α, β and GPER in the CNS and trigeminal system - molecular and functional aspects.

BACKGROUND: Migraine occurs 2-3 times more often in females than in males and is in many females associated with the onset of menstruation. The steroid hormone, 17β-estradiol (estrogen, E2), exerts its effects by binding and activating several estrogen receptors (ERs). Calcitonin gene-related peptide (CGRP) has a strong position in migraine pathophysiology, and interaction with CGRP has resulted in several successful drugs for acute and prophylactic treatment of migraine, effective in all age groups and in both sexes. METHODS: Immunohistochemistry was used for detection and localization of pr

Multiple vibrissae sensory regions in rat cerebellum: a (14C) 2-deoxyglucose study.

Repetitive tactile sensory stimulation of the right mystacial vibrissae (whiskers) was performed in awake, adult rats. Regions of increased (14C) 2-deoxyglucose (2DG) uptake were mapped autoradiographically in cerebellum. Predominantly ipsilateral activation of multiple discrete granule cell regions occurred in paramedian lobule, crus 2, crus 1, lobulus simplex, and anterior lobe hemisphere. Bilateral and contralateral activation of cerebellum did occur. Multiple small patches, as well as large granule cell regions, were activated. Mossy fiber afferents from the spinal trigeminal nuclei (parti

Microcephaly with altered cortical layering in GIT1 deficiency revealed by quantitative neuroimaging.

G Protein-Coupled Receptor Kinase-Interacting Protein-1 (GIT1) regulates neuronal functions, including cell and axon migration and synapse formation and maintenance, and GIT1 knockout (KO) mice exhibit learning and memory deficits. We noted that male and female GIT1-KO mice exhibit neuroimaging phenotypes including microcephaly, and altered cortical layering, with a decrease in neuron density in cortical layer V. Micro-CT and magnetic resonance microscopy (MRM) were used to identify morphometric phenotypes for the skulls and throughout the GIT1-KO brains. High field MRM of actively-stained mou

Computed tomography of the brain stem with intrathecal metrizamide. Part I: the normal brain stem.

In summary, the anatomic details of the brain stem are exquisitely demonstrated with metrizamide CT. The obex is the zone of transition from the closed medulla, which has a conical pear-shaped appearance to the quadriconcave open medulla. The pyramids and inferior olivary nuclei characterize the anterior surface of the open medulla, whereas the median sulcus and the cranial nerve nuclei constitute the floor of the fourth ventricle at the same level. The mushroomlike appearance of the pontomedullary junction is easily reproducible with the restiform bodies forming the posteriorly diverging bipe

Distinct expression patterns of two zebrafish homologues of the human APP gene during embryonic development.

The human amyloid protein precursor (APP) gene correlates with early onset of Alzheimer's disease in humans. We have identified two APP homologues in zebrafish, which we call appa and appb. They show a high degree of identity to human APP particularly in the beta APP42 and the transmembrane domain. Widespread expression of both appa and appb was detected from mid-gastrulation until the bud stage. During segmentation, the two genes diverged in their pattern of expression: at 14 h post-fertilisation (hpf) and 18 hpf both genes were expressed rostrally in the prospective CNS, but only appa was fo

Herpes simplex encephalitis. An immunohistological study of the distribution of viral antigen within the brain.

An immunoperoxidase technique was used to map the sites of herpes simplex virus antigen (VA) within the brain in 29 autopsied cases of herpes simplex encephalitis. Attention was directed particularly to those parts of the brain that are known from pathological studies to be involved in the disease. Material was studied from cases surviving for varying periods from a few days to a few years after the onset of neurological disease. VA was found within the brain in all cases dying within 3 weeks of onset, but in none dying thereafter. VA was already most abundant in patients dying within the firs

The Infratrigeminal Suprafloccular Approach to Intrapontine Lesions: An Anatomical Overview and Relevance for the Approach to Intrapontine Lesions.

Background and objectives Brainstem lesions have long been considered complex pathologies that may lead to permanent deficits or life-threatening complications, posing significant challenges for surgical removal. Among these lesions, intrapontine lesions are particularly challenging in the field of neurosurgery. However, with advancements in microsurgical anatomy knowledge and technology, these lesions have become more amenable to surgical treatment. In this study, the authors examine an infratrigeminal suprafloccular approach, which has been shown to be a safe surgical route, resulting in few

Analgesic and Neuroprotective Effects of Electroacupuncture in a Dental Pulp Injury Model-A Basic Research.

Irreversible pulpitis is an extremely painful condition and its consequence in the central nervous system (CNS) remains unclear. A mouse model of dental pulp injury (DPI) resembles the irreversible pulpitis profile in humans. This study sought to determine whether pain induced by DPI activates microglia and astrocytes in the trigeminal subnucleus caudalis (Vc), as well as increases levels of proinflammatory cytokines, and whether electroacupuncture (EA) can be a potential analgesic and neuroprotective therapy following DPI. Pain behavior was measured via head-withdrawal threshold (HWT) and bur

Differential activation of mitogen-activated protein kinases and glial cells in the trigeminal sensory nuclear complex following lingual nerve injury.

Mitogen-activated protein kinases (MAPKs) play a pivotal role in the mediation of cellular responses to a variety of signaling molecules. The current study demonstrates phosphorylation of extracellular signal-regulated kinase (ERK) and p38 MAPK in each subdivision of the trigeminal sensory nuclear complex (TSNC) following lingual nerve injury. Immunohistochemical labeling for phosphorylated ERK (p-ERK) or phosphorylated p38 (p-p38) MAPK was performed in histological sections of the brainstem. A transient increase in the immunoreactivity for p-ERK was found in each subdivision of the TSNC follo

[Action of high and superhigh doses of fission neutrons and gamma quanta on the central noradrenergic formation of the brain--on the locus coeruleus (the blue spot)].

Most of the blue spot neurons of Wistar rats exhibited a pronounced central chromatolysis 24 h following irradiation with fission neutrons (above 100 Gy) and gamma-quanta (above 200 Gy). The changes in the mesencephalic nucleus of the trigeminal nerve were distinct on day 2 after 200 Gy gamma-irradiation. The number of the modified nervous cells in these regions was higher than that in other brain parts.

Substance P and the sensory innervation of intracranial and extracranial feline cephalic arteries. Implications for vascular pain mechanisms in man.

Substance P, a putative neurotransmitter peptide present in a subpopulation of small sensory neurons, was measured in the walls of feline cranial arteries and systemic veins and arteries using a sensitive and specific radioimmunoassay. Substance P immunoreactivity exhibited a retention time identical to that of synthetic substance P when vessel extracts were subjected to reverse phase high performance liquid chromatography. Levels in cephalic arteries (453-1083 fmol/mg protein) were at least twice as high as amounts in systemic arteries and veins, and were significantly higher than those measu

[Critical remarks on so-called cervicogenic vertigo].

A critical review of neurophysiological literature is given about the part played by the neck proprioceptors in the regulation of vestibular functions. In comparison with vestibular and optokinetic influences the neck proprioceptors are of secondary importance. A critical review of the publications by the supporters of a cervical genesis of vertigo leads to the conclusion that "cervical nystagmus" must be proven before the concept of neck-triggered vertigo can be accepted. In relation to all forms of vertigo, vertigo induced by cervical disorder represents a very low percentage only.

The reaction of primary sensory neurons to peripheral nerve injury with particular emphasis on transganglionic changes.

This paper reviews light- and electron microscopic, histochemical and physiological evidence which demonstrate that peripheral nerve injury in mammals is followed by profound structural and functional changes in the central terminals of the affected primary sensory neurons. Available evidence indicates that at least some of these so-called transganglionic changes are the result of ganglion cell degeneration and death, although other mechanisms are probably in effect as well. Existing data suggest that this ganglion cell death does not effect all types of ganglion cells equally, but do not perm
