BACKGROUND: Methylphenidate is widely prescribed for the clinical management of pediatric patients with attention-deficit/hyperactivity disorder (ADHD), yet its safety profile remains a subject of ongoing concern. This study aims to characterize the real-world safety profile of methylphenidate in children and adolescents using the FDA Adverse Event Reporting System (FAERS) database.
METHODS: Adverse drug event (ADE) reports for individuals aged 6-17 years were extracted from the FAERS database via OpenVigil 2.1. Disproportionality analyses were performed to detect methylphenidate-associated s

BACKGROUND: The adrenal gland plays a pivotal role in the stress response via the hypothalamic-pituitary-adrenal (HPA) axis and the sympatho-adrenomedullary system (SAMS). Congenital adrenal hyperplasia (CAH) results from pathogenic variants in genes encoding adrenal steroidogenic enzymes. The most common form, 21-hydroxylase deficiency, impairs cortisol biosynthesis, leading to compensatory overproduction of adrenocorticotropic hormone (ACTH) and chronic adrenal hyperplasia.

BACKGROUND: Virilization in girls younger than 2 years should prompt careful evaluation for pathological androgen excess and should be interpreted as a practical clinical reminder rather than as a stand-alone diagnostic tool. Pediatric adrenocortical tumors (ACTs) are rare but are usually hormonally active, and their interpretation requires pediatric-specific clinicopathological criteria rather than direct application of adult adrenal carcinoma criteria.
CASE PRESENTATION: A 19-month-old girl presented with a 6-month history of progressive voice deepening, pubic hair growth, and clitoromegaly

Background/Objectives: The skeletal system reaches peak bone mass through modeling and remodeling processes, influenced by environmental, dietary, hormonal, and genetic factors. In children with endocrinopathies, disturbances in bone mass and mineralization may correlate with hormonal levels, but conditions like short stature or obesity can confound DXA results. This study aimed to assess the prevalence of decreased bone mineral density (BMD) in children with endocrine disorders and evaluate the impact of auxological and hormonal abnormalities on BMD. Methods: This study analyzed medical recor

BACKGROUND: Bisphenol A (BPA), a widely distributed endocrine-disrupting chemical (EDC), has been associated with altered pubertal timing in children, including precocious puberty (PP). However, the multi-target molecular mechanisms potentially linking BPA exposure to PP remain incompletely understood.
OBJECTIVE: To explore the potential multi-target mechanisms by which BPA may contribute to PP using an integrated computational framework.
METHODS: We combined network toxicology, protein-protein interaction (PPI) network analysis, molecular docking, and molecular dynamics (MD) simulations. Sh

BACKGROUND: Hepatocellular adenomas (HCAs) are rare benign tumors of the liver that occur predominantly in women taking oral contraceptives. In children, HCAs comprise < 5% of hepatic tumors. We report a case of HCAs in a 7-year-old girl with estrogen and glucose imbalance.
CASE PRESENTATION: A 7-year-old girl was presented to our hospital with bilateral breast enlargement for 2 months, polydipsia, polyuria, polyphagia, hyperglycemia, and significant weight gain. Computed tomography (CT) showed a 7.2 cm×6.9 cm×5.3 cm round-shaped mass in the left inner lobe of the liver, ovarian ultrasound sh

OBJECT: Endoscopic third ventriculostomy (ETV) is an established treatment for hydrocephalus. Most studies focus on success rate, and complications are insufficiently documented. The aim of this study was to perform a systematic review of ETV complications.
METHODS: A Medline search discovered 34 series of ETV with detailed complications reports (17 series involving exclusively pediatric patient populations, 6 series involving exclusively adults, and 11 series involving mixed adult and pediatric populations).
RESULTS: The analysis included 2985 ETVs performed in 2884 patients. The cause of h

OBJECTIVE: Drug-resistant epilepsy associated with hypothalamic hamartomas (HHs) can be cured by microsurgical resection of the lesions. Morbidity and mortality rates for microsurgery in this area are significant. Gamma knife surgery (GKS) is less invasive and seems to be well adapted for this indication.
METHODS: To evaluate the safety and efficacy of GKS to treat this uncommon pathological condition, we organized a multicenter retrospective study. Ten patients were treated in seven different centers. The follow-up periods were more than 12 months for eight patients, with a median follow-up

BACKGROUND: Puberty consists of 2 components: gonadarche and adrenarche. Both components have distinct endocrine changes. Adrenarche has virtually been ignored with respect to examining hormone-behavior relations.
OBJECTIVES: To provide descriptive biological and behavioral information on children with premature adrenarche (PA) and to examine differences in biological, psychological, and cognitive variables of children with PA and a healthy comparison group of children with on-time adrenarche.
DESIGN: Descriptive pilot study.
SETTING: A consecutive sample of patients was recruited from pedi

The new imaging techniques (ultrasound, CT, digital subtraction angiography) have greatly modified the diagnostic approach to the endocrinopathies in pediatric age, notably if of surgical interest. The main clinical indications to the ultrasound and/or CT study of hypophysis, thyroid, adrenals and female internal genital tract are synthesized in this paper. A diagnostic flow-chart is also suggested for each clinical entity.

Activating mutations of the Gsalpha gene are responsible for McCune-Albright syndrome and have also been identified in sporadic tumors of the pituitary and thyroid. When associated with malignancy, activating Gsalpha mutations are known as gsp-oncogenes. We hypothesized that similar activating mutations might also account for some cases of premature thelarche and/ or granulosa cell tumors. Polymerase chain reaction and DNA sequencing was used to screen for activating mutations of Gsalpha genes in children with premature thelarche and in pathologic specimens from juvenile and adult granulosa ce

We report one patient with 21-hydroxylase deficiency and associated bilateral macro-orchidism caused by nodular hyperplasia of testicular adrenal rests (TAR). The boy, referred to us when 10 years old, was born with bilateral cryptorchidism that was treated unsuccessfully with i.m. injections of human chorionic gonadotropin (hCG) and later on with orchidopexy. He was treated with oral dexamethasone (0.625 mg per day) for the following 13 years. After one year, there was a marked reduction in steroid hormone levels (17-hydroxyprogesterone (17-OH P) from 27.2 to 1.2 nmol/l, testosterone from >10
