Can community-based integrated vector control hasten the process of LF elimination?

Community-based integrated vector control (IVC) using polystyrene beads (EPS) and pyrethroid impregnated curtains (PIC) as an adjunct to mass drug administration (MDA) was implemented for lymphatic filariasis elimination, in the filaria endemic villages of Tirukoilur, south India. In all the villages, MDA was carried out by the state health machinery, as part of the national filariasis elimination programme. Thirty-six difficult-to-control villages were grouped as, viz, MDA alone, MDA + EPS and MDA + EPS + PIC arms. Implementation and monitoring of IVC was carried out by the community. After 3

Combining the Sterile Insect Technique with Wolbachia-Based Approaches: II--A Safer Approach to Aedes albopictus Population Suppression Programmes, Designed to Minimize the Consequences of Inadvertent Female Release.

Due to the absence of a perfect method for mosquito sex separation, the combination of the sterile insect technique and the incompatible insect technique is now being considered as a potentially effective method to control Aedes albopictus. In this present study first we examine the minimum pupal irradiation dose required to induce complete sterility in Wolbachia triple-infected (HC), double-infected (GUA) and uninfected (GT) female Ae. albopictus. The HC line is a candidate for Ae. albopictus population suppression programmes, but due to the risk of population replacement which characterizes

[Malaria transmission in 1999 in the rice field area of the Kou Valley (Bama), (Burkina Faso)].

A longitudinal study based on mosquitoes sampled by larvae prospecting and adult catches on humans was carried out during 1999 in the rice field area of the Kou Valley (South-West Burkina Faso) to evaluate the malaria transmission level. Two sites were studied: VK5 located in the rice field centre and VK7 in the periphery. Irrigation is sub-permanent and two crops are grown each year: from February to June during the dry season and from July to November during the rainy season. A man sleeping in VK5 without any protection is exposed to more than 60,000 mosquito bites/year. Two majors vectors a

[Geographic approach in malaria control in the central highlands of Madagascar].

Following the severe malaria outbreak in the central highlands in Madagascar in 1986, a vector control program by use DDT pm 75 house-spraying has been implemented to operate in areas located at altitudes between 1000 and 1500 m. Early treatment with chloroquine has also been incorporated in the control program. To detect areas at particular high risk for malaria outbreak the Geographic Information System (GIS) has been applied and tested. The study has shown that the system can be used in malaria surveillance in order to identify areas in which an intense distribution of Anopheles funestus ca

Deep learning classification and motion-based analysis of Aedes aegypti Red-eye GSS larvae for SIT applications.

Aedes aegypti is a major vector of arboviral diseases, including dengue, Zika, and chikungunya, posing significant global public health challenges. Effective control strategies, such as the sterile insect technique (SIT), require an accurate and reliable sex sorting system to ensure male only releases. Here, we present a compact deep learning framework for image-based classification of sex-linked eye-color phenotypes (black-eye males and red-eye females) and instar stages of Aedes aegypti larvae, complemented by an independent optical flow analysis of larval motor activity. Video recordings of

Dengue vector niche modeling with future SSP climatic scenarios over India using the machine learning-based MaxEnt model.

Dengue remains a major mosquito-borne public-health threat, and climate change is expected to reshape the geographic suitability of its principal vectors. We developed India-wide species distribution models for Aedes aegypti and Aedes albopictus using a presence-background MaxEnt framework driven by high-resolution (~ 1 km) CHELSA bioclimatic predictors. Occurrence records were compiled from GBIF and subjected to transparent quality control and de-duplication on the modelling grid, yielding 301 training presences for Aedes aegypti (363 raw → 341 quality-filtered → 301 thinned) and 180 for Aede

Genetic variations in three insecticide targets in the disease vector Culex quinquefasciatus from Mianyang, China: simultaneous detection of novel mutations RDL A296G and VGSC A1007T.

Culex quinquefasciatus is a major vector of filariasis and Japanese encephalitis in China. The application of insecticides is a regular measure for prevention and control of such mosquito-borne diseases. However, insecticide resistance, often caused by target-site mutations, threatens control efforts. In Mianyang City, a region previously reporting high pyrethroid resistance, the current genetic basis of resistance remains unclear. To address this gap, resistance-related mutations in three key insecticide targets, including acetylcholinesterase (AChE, encoded by ace1, conferring organophosphat

In silico screening of flavonoid-based inhibitors targeting UDP-galactopyranose mutase of Brugia malayi for lymphatic filariasis drug design.

Lymphatic filariasis (LF) is a vector-borne disease caused by parasitic roundworms, such as Wuchereria bancrofti, Brugia malayi, and Brugia timori, transmitted through mosquito vectors. Drugs such as albendazole, ivermectin, and diethylcarbamazine are used to treat LF; however, they mainly act against the microfilarial stage and have limited effect on the adult worms. So, it is time to identify or design a more efficient drug against the LF. Targeting the UDP-galactopyranose mutase (UGM) enzyme offers an effective strategy for treating lymphatic filariasis, as this enzyme is absent in mammals.

Sensitive and modular amplicon sequencing of Plasmodium falciparum diversity and resistance for research and public health.

BACKGROUND: Targeted amplicon sequencing is a powerful and efficient tool for interrogating the Plasmodium falciparum genome, generating actionable data from infections to complement traditional malaria epidemiology. For maximum impact, genomic tools should be multi-purpose, robust, sensitive, and reproducible. METHODS: We developed, characterized, and implemented MAD4HatTeR, an amplicon sequencing panel based on Multiplex Amplicons for Drug, Diagnostic, Diversity, and Differentiation Haplotypes using Targeted Resequencing, along with a bioinformatic pipeline for data analysis. Additionally,

Biology and epidemiology of Plasmodium falciparum and Plasmodium vivax gametocyte carriage: Implication for malaria control and elimination.

Malaria is among the leading public health problems worldwide. Female anopheles mosquito orchestrates the transmission of malaria by taking gametocytes and introducing sporozoite while taking blood meals. Interrupting transmission is the major strategy for malaria elimination. The gametocyte stage is essential for the onward transmission of malaria. Thus, understanding its basic biology and epidemiology is key to malaria control and elimination. Therefore, the current review focuses on revealing the biology, prevalence, and determinants of gametocyte carriage as well as its implication on miti

Biological evaluation of 1,3-benzodioxole acids points to 3,4-(methylenedioxy) cinnamic acid as a potential larvicide against Aedes aegypti (Diptera: Culicidae).

Aedes aegypti serves as the primary vector for viruses like dengue, Chikungunya, Zika, and yellow fever, posing a significant public health challenge in Brazil. Given the absence of approved vaccines for these diseases, effective mosquito control becomes paramount in preventing outbreaks. However, currently available chemical insecticides face issues related to toxicity and the emergence of resistance, necessitating the exploration of new active compounds. Drawing inspiration from natural products, we identified the 1,3-benzodioxole group as a key pharmacophore associated with insecticidal act…

Short Report: Adult Aedes abundance and risk of dengue transmission.

Dengue is transmitted mainly by the adult female Aedes aegypti mosquito. However, little is known about the impact of adult Aedes abundance on the risk of dengue transmission. Here we analysed nationally representative dengue case and vector surveillance data collected from Singapore, to determine the effect of adult Aedes abundance on the risk of dengue transmission. A case was an area with active dengue transmission as indicated by the presence of dengue cluster. A control was an area where no dengue cluster was reported. Using multivariate logistic regression, we analysed 88 cases and 602 c

Do-It-Yourself Tick Control: Granular Gamma-Cyhalothrin Reduces Ixodes scapularis (Acari: Ixodidae) Nymphs in Residential Backyards.

Lyme disease is the most common vector-borne disease in the United States with hotspots in the Northeast and Midwest. Integrated vector control for mosquito-borne disease prevention is often organized at the community level, but tick control is primarily coordinated at the household and individual level. Management of the blacklegged tick, Ixodes scapularis (Say), the vector of the causative agent of Lyme disease in the Midwest and eastern United States in peridomestic environments may be critical as many tick encounters are reported to occur in the yard. Therefore, we assessed the effectivene

Identification of sequence motifs involved in Dengue virus-host interactions.

Dengue fever is a rapidly spreading mosquito-borne virus infection, which remains a serious global public health problem. As there is no specific treatment or commercial vaccine available for effective control of the disease, the attempts on developing novel control strategies are underway. Viruses utilize the surface receptor proteins of host to enter into the cells. Though various proteins were said to be receptors of Dengue virus (DENV) using Virus Overlay Protein Binding Assay, the precise interaction between DENV and host is not explored. Understanding the structural features of domain II

Bionomics and vectorial capacity of Anopheles annularis with special reference to India: a review.

Anopheles annularis is widely distributed mosquito species all over the country. An. annularis has been incriminated as a malaria vector in India, Sri Lanka, Bangladesh, Myanmar, Indonesia, Malaysia and China. In India, it has been reported to play an important role in malaria transmission as a secondary vector in certain parts of Assam, West Bengal and U.P. In Odisha and some neighbouring countries such as Sri Lanka, Nepal and Myanmar it has been recognised as a primary vector of malaria. This is a species complex of two sibling species A and B but the role of these sibling species in malaria

Diurnally subperiodic filariasis in India-prospects of elimination: precept to action?

The elimination of lymphatic filariasis in the Andaman and Nicobar Islands provides unique opportunities and challenges at the same time. Since these islands are remote, are sparsely populated, and have poor transport networks, mass drug administration programs are likely to be difficult to implement. Diurnally subperiodic Wuchereria bancrofti vectored by Downsiomyia nivea was considered for the scope of vector control options. Considering the bioecology of this mosquito, vector control including personal protection measures may not be feasible. However, since these islands are covered by sepa

[Emergent viral threats in blood transfusion].

During the last 20 years, the safety of blood products increased dramatically with regard to the infectious risk and notably to that represented by retroviruses (HIV and HTLV) and hepatitis B and C viruses. The aim of this review is to identify the residual and emergent viral threats that could be responsible for the occurring of new contaminations in the receivers of blood products. Beside many other viruses (HHV-8, erythrovirus B19, hepatitis A and E viruses...), a special attention has been paid to emerging arbovirus diseases (West Nile virus infection, dengue, chikungunya) that threaten to
