Donepezil improves learning and memory deficits in APP/PS1 mice by inhibition of microglial activation.

Donepezil, a cholinesterase inhibitor, is a representative symptomatic therapy for Alzheimer's disease (AD). Recent studies have reported the anti-inflammatory effects of donepezil. However, limited studies that investigate its anti-inflammatory effect in AD have been reported. Considering the role of proinflammatory molecules and microglial activation in the pathogenesis of AD, the current study aimed to elucidate the effects of donepezil on microglial activation induced by amyloid deposition in transgenic mice. Our results showed that chronic treatment with donepezil significantly improved t

Lack of the transcription factor C/EBPδ impairs the intrinsic capacity of peripheral neurons for regeneration.

Adult neurons of the peripheral nervous system (PNS), in contrast to those of the central nervous system, have a remarkable capacity to repair themselves after injury, yet the mechanisms underlying this regenerative propensity of peripheral neurons are far from completely understood. Here we show that the transcription factor CCAAT enhancer binding protein delta (C/EBPδ) is necessary for the efficient axonal regeneration of dorsal root ganglia (DRG) neurons after sciatic nerve crush injury. Loss of C/EBPδ substantially impairs axonal growth in dissociated cultured DRG neurons. In addition, lac

Early postnatal expression of L1 by retinal fibers in the optic tract and synaptic targets of the Syrian hamster.

Previous immunohistochemical studies in mouse, rat, and chick have reported that the expression of the glycoprotein and cell adhesion molecule L1, a member of the immunoglobulin superfamily, shows regulation during development of retina and optic nerve. To extend our understanding of the role of L1 in developing neural circuitry, we have examined L1 expression in the optic tract and thalamic and midbrain synaptic targets of retinal fibers in the early postnatal Syrian hamster, a well-characterized developmental model of the primary visual projection. Metabolic labeling studies reveal that a sy

The Role of Dendritic Spines in Water Exchange Measurements With Diffusion MRI: Time-Dependent Single Diffusion Encoding MRI.

Time-dependent diffusion MRI (dMRI) using single diffusion encoding (SDE) is sensitive to water dynamics in biological tissues, yet interpreting its signals requires careful consideration of underlying microstructure. While prior work has focused on restricted/hindered diffusion and membrane permeation, additional exchange mechanisms such as diffusion-mediated exchange between dendritic shaft and spines in gray matter (GM) remain understudied. Here, we hypothesize that water diffusion within impermeable spiny dendrites can produce time-dependent SDE signals indistinguishable from those arising

The characterization of Parkinson's disease related pain based on the classification system: The severity of neuropathic pain and the clinical profile of nociplastic pain.

INTRODUCTION: Pain significantly impairs quality of life in Parkinson's disease (PD). While the PD Pain Classification System (PD-PCS) categorizes PD-related pain into nociceptive, neuropathic, and nociplastic pain, the clinical and structural correlates of these subtypes, particularly nociplastic pain, remain poorly characterized. The objective of this study was to elucidate the distinct features of each PD-related pain subtype. METHODS: This single-center cross-sectional study involved a two-stage PD-PCS-based evaluation. First, 192 PD patients were assessed for the prevalence of pain subty

Association of Cortical Free Water With Brain Tau Tangle Load in the Alzheimer Disease Continuum.

BACKGROUND AND OBJECTIVES: Neurofibrillary tangles (NFTs) progressively damage gray matter in Alzheimer disease (AD). Resulting cortical microstructural alterations might not be detectable using macrostructural metrics but may be studied using isotropic water diffusion, as it reflects extracellular free water content. The aim of this study was to examine the effect of NFTs on cortical microstructure by investigating whether cortical free water increases as a function of tau load. We also investigated whether phosphorylated tau in blood plasma also indicated cortical microstructural abnormaliti

Early hypoxia treatment and oxygen-concentration alteration promote the retinal progenitor proliferation and ganglion cell maturation in human retinal organoid development.

BACKGROUND: Retinal organoids (ROs) derived from human embryonic stem cells (hESCs) hold immense potential for modeling retinal development and diseases. However, current differentiation protocols often overlook the physiological hypoxic microenvironment of early embryogenesis, potentially compromising developmental fidelity. In this study, we investigated how staged oxygen modulation-hypoxic priming followed by normoxic transition-optimizes RO development by mimicking in vivo oxygen dynamics. METHODS: The H9 hESC line was differentiated into ROs using a modified serum-free floating culture o

Mesenchymal stromal/stem cell-derived extracellular vesicles in brain disorders: mechanisms of repair and recovery.

Mesenchymal stem/stromal cell-derived small extracellular vesicles (MSC-sEVs) have emerged as promising cell-free therapeutics for central nervous system (CNS) disorders including stroke, traumatic brain injury (TBI), dementia, and multiple sclerosis (MS). MSC-sEVs offer advantages of low immunogenicity, ease of storage, and ability to cross the blood-brain barrier. This review provides a comprehensive analysis of the mechanisms by which MSC-sEVs have been reported to promote neural repair and recovery in preclinical models, through two convergent categories of action. First, MSC-sEVs exert di

Antigliotic guiding regenerative gel: a bioactive hydrogel platform for peripheral nerve and spinal cord repair.

Peripheral nerve injury (PNI) and spinal cord injury (SCI) impose a major burden of traumatic neurological disability worldwide. Autologous nerve grafting, the current gold standard, achieves meaningful recovery in only 81.6% of repairs (2023 meta-analysis, n = 1,559), while hollow nerve graft conduits achieve only 62.2% (p < 0.05), with performance declining sharply beyond 20-30 mm gaps. Passive scaffolds cannot counter the inhibitory post-injury microenvironment of reactive gliosis, chondroitin sulfate proteoglycan (CSPG) deposition, oxidative stress, and myelin-associated inhibitors, which

Association Between Alcohol Consumption, Cognitive Abilities, and Neuropathologic Changes: A Population-Based Autopsy Study.

BACKGROUND AND OBJECTIVES: Heavy alcohol consumption is a major global health concern linked to increased morbidity and mortality. However, the long-term impact of excessive alcohol consumption on cognitive abilities and dementia-related neuropathology is unclear. The aim of this study was to analyze the association between alcohol consumption and age-related neuropathologic outcomes in a population-based autopsy study. METHODS: This cross-sectional study used data from the Biobank for Aging Studies, classifying participants as never, moderate, heavy, or former drinkers. Alzheimer disease pat

Non-invasive identification of mesenchymal glioblastoma using quantitative radiomic features from advanced diffusion MRI: a preclinical-to-clinical transfer learning strategy.

BACKGROUND: Glioblastoma (GBM) is no longer regarded as a single disease, as distinct molecular subgroups exist, with the mesenchymal (MES) having the worst prognosis. As such, there is a critical need for noninvasive methods to determine GBM molecular status. Although conventional magnetic resonance imaging (MRI)-based radiomics showed promise for predicting GBM characteristics, few studies evaluated pipelines that leverage advanced diffusion MRI (dMRI) techniques, such as diffusion tensor imaging (DTI) and neurite orientation dispersion and density imaging (NODDI), enabling characterization

Heat stress-induced genomic instability in neural stem cells and its association with neuronal developmental deficits.

BACKGROUND: Maternal hyperthermia, an increasingly prevalent environmental stressor linked to global climate change, is epidemiologically associated with a higher risk of neurodevelopmental disorders, including autism and schizophrenia. However, the molecular mechanisms underlying this neurotoxicity, particularly those leading to long-term neurological deficits, remain poorly understood. This study tested the novel hypothesis that hyperthermia acts not only as an acute physiological stressor but also as a potent genotoxic agent, inducing persistent genomic alterations in neural progenitor cell

The Importance of Coating Surface and Composition for Attachment and Survival of Neuronal Cells Under Mechanical Stimulation.

Cell culture of neuronal cells places high demands on the surface for these cells to adhere to and grow on. Native extracellular matrix (ECM) proteins are often applied to the cell culture surface. The substrate is even more important when mechanical strain is applied to the cells in culture. These cells will easily detach and die, precluding the study of how mechanical factors affect these cells. Mechanical factors are, for example, important in the eye disorder glaucoma, which is characterized by the loss of the retinal ganglion cells (RGCs), the retinal neurons that transfer the visual info

Subvoxel quantitative susceptibility mapping of chronic white matter lesions: Enhancing lesion heterogeneity characterization and providing complementary biomarkers for myelin integrity in multiple sclerosis.

OBJECTIVES: Quantitative susceptibility mapping (QSM) has been used to classify chronic white matter lesions (WMLs) in multiple sclerosis (MS), which exhibit varying myelin and iron content. Compared to QSM, subvoxel-QSM can further differentiate susceptibility alterations arising from demyelination and iron accumulation. This study aims to evaluate the performance of subvoxel-QSM in classifying chronic WMLs in relapsing-remitting MS (RRMS) and explore their heterogeneity. METHODS: In 57 RRMS patients, WMLs segmented manually were classified into 5 QSM lesion types (isointense, hypointense, h

In vivo cortical diffusion imaging relates to Alzheimer's disease neuropathology.

BACKGROUND: There has been increasing interest in cortical microstructure as a complementary and earlier measure of neurodegeneration than macrostructural atrophy, but few papers have related cortical diffusion imaging to post-mortem neuropathology. This study aimed to characterise the associations between the main Alzheimer's disease (AD) neuropathological hallmarks and multiple cortical microstructural measures from in vivo diffusion MRI. Comorbidities and co-pathologies were also investigated. METHODS: Forty-three autopsy cases (8 cognitively normal, 9 mild cognitive impairment, 26 AD) fro

Promising application of a novel biomaterial, light chain of silk fibroin combined with NT3, in repairment of rat sciatic nerve defect injury.

Autologous nerve transplantation is the gold standard for treating peripheral nerve defects, but it is associated with defects such as insufficient donor and secondary injury. Artificial nerve guidance conduits (NGCs) are now considered promising alternatives for bridging long nerve gaps, although exploring new biomaterials to construct NGCs remains challenging. Silk fibroin (SF) has good biocompatibility and can self-assemble in aqueous solutions. However, the lack of proximal neurotrophic factors after nerve injury is a major concern, leading to incomplete nerve regeneration. In this study,

The prevalence, correlation, and co-occurrence of neuropathology in old age: harmonisation of 12 measures across six community-based autopsy studies of dementia.

BACKGROUND: Population-based autopsy studies provide valuable insights into the causes of dementia but are limited by sample size and restriction to specific populations. Harmonisation across studies increases statistical power and allows meaningful comparisons between studies. We aimed to harmonise neuropathology measures across studies and assess the prevalence, correlation, and co-occurrence of neuropathologies in the ageing population. METHODS: We combined data from six community-based autopsy cohorts in the US and the UK in a coordinated cross-sectional analysis. Among all decedents aged

Cancer diagnosis is associated with a lower burden of dementia and less Alzheimer's-type neuropathology.

Cancer and Alzheimer's disease are common diseases in ageing populations. Previous research has reported a lower incidence of Alzheimer's disease-type (amnestic) dementia among individuals with a diagnosis of cancer. Both cancer and amnestic dementia are prevalent and potentially lethal clinical syndromes. The current study was conducted to investigate the association of cancer diagnosis with neuropathological and cognitive features of dementia. Data were analysed from longitudinally evaluated participants in a community-based cohort study of brain ageing who came to autopsy at the University

Multicompartmental models and diffusion abnormalities in paediatric mild traumatic brain injury.

The underlying pathophysiology of paediatric mild traumatic brain injury and the time-course for biological recovery remains widely debated, with clinical care principally informed by subjective self-report. Similarly, clinical evidence indicates that adolescence is a risk factor for prolonged recovery, but the impact of age-at-injury on biomarkers has not been determined in large, homogeneous samples. The current study collected diffusion MRI data in consecutively recruited patients (n = 203; 8-18 years old) and age and sex-matched healthy controls (n = 170) in a prospective cohort design. Pa

Early structural connectivity within the sensorimotor network: Deviations related to prematurity and association to neurodevelopmental outcome.

Consisting of distributed and interconnected structures that interact through cortico-cortical connections and cortico-subcortical loops, the sensorimotor (SM) network undergoes rapid maturation during the perinatal period and is thus particularly vulnerable to preterm birth. However, the impact of prematurity on the development and integrity of the emerging SM connections and their relationship to later motor and global impairments are still poorly understood. In this study we aimed to explore to which extent the early microstructural maturation of SM white matter (WM) connections at term-equ
