Prevalence of hemochromatosis-related symptoms among individuals with mutations in the HFE gene.

OBJECTIVE: To determine the prevalence of hemochromatosis-related symptoms in homozygotes for the HFE mutation C282Y compared with controls without HFE mutations identified through a large screening program of subjects attending a health appraisal center. SUBJECTS AND METHODS: Presence of symptoms commonly associated with clinical hemochromatosis was ascertained by self-report on a written questionnaire among C282Y homozygotes and HFE wild-type subjects of white or Hispanic ethnicity identified from screening 41,599 adult subjects between March 1999 and August 2001. A subset of C282Y homozygo

Impact factors and genetic characteristics of head lice infestation in schoolchildren: a cross-sectional study from 2018 to 2023 in central China.

BACKGROUND: Head lice (Pediculus humanus capitis) infestation is a worldwide public health concern, especially in school-aged children. However, its main impact factors and genetic characteristics remain poorly understood in China. Hence, the purpose of the study was to explore the precise association between multiple impact factors and head lice infestation, characterize the genetic variation of the head lice, and develop a sensitive and specific mitochondrial (mt) DNA duplex polymerase chain reaction (PCR) for accurately distinguishing clades A and B. METHODS: A cross-sectional study was co

Bifocal germinoma in a patient with 16p11.2 microdeletion syndrome.

Intracranial germinomas are rare tumors affecting mostly patients at young age. Therefore, molecular data on its etiopathogenesis are scarce. We present a clinical case of a male patient of 25 years with an intracranial germinoma and a 16p11.2 microdeletion. His initial complaints were related to obesity, loss of facial hair and polydipsia. He also had a history of social-interaction difficulties during childhood. His blood tests were consistent with hypogonadotropic hypogonadism and secondary adrenal insufficiency, and he had been previously diagnosed with hypothyroidism. He also presented wi

Scalp stratum corneum histamine levels: novel sampling method reveals association with itch resolution in dandruff/seborrhoeic dermatitis treatment.

Dandruff and seborrhoeic dermatitis are accompanied by bothersome itch. We have established a novel non-invasive methodology to sample histamine levels in the stratum corneum in order to facilitate an understanding of pruritogenesis in this condition. Histamine levels were assessed in two groups of subjects with dandruff before and after 3 weeks of treatment with a commercial potentiated zinc pyrithione shampoo. A comparative population without dandruff was also studied. Itch self-perception was quantified on a visual analogue scale. The histamine level in subjects with dandruff was more than

Molecular dissection of mesenchymal-epithelial interactions in the hair follicle.

De novo hair follicle formation in embryonic skin and new hair growth in adult skin are initiated when specialized mesenchymal dermal papilla (DP) cells send cues to multipotent epithelial stem cells. Subsequently, DP cells are enveloped by epithelial stem cell progeny and other cell types to form a niche orchestrating hair growth. Understanding the general biological principles that govern the mesenchymal-epithelial interactions within the DP niche, however, has been hampered so far by the lack of systematic approaches to dissect the complete molecular make-up of this complex tissue. Here, we

Null mutation of endothelin receptor type B gene in spotting lethal rats causes aganglionic megacolon and white coat color.

Mutations in the gene encoding the endothelin receptor type B (EDNRB) produce congenital aganglionic megacolon and pigment abnormalities in mice and humans. Here we report a naturally occurring null mutation of the EDNRB gene in spotting lethal (sl) rats, which exhibit aganglionic megacolon associated with white coat color. We found a 301-bp deletion spanning the exon 1-intron 1 junction of the EDNRB gene in sl rats. A restriction fragment length polymorphism caused by this deletion perfectly cosegregates with the sl phenotype. The deletion leads to production of an aberrantly spliced EDNRB mR

Psoriatic arthritis digital phenotyping and inflammation drivers (PDPID) study: protocol for an international multicentre prospective cohort.

INTRODUCTION: Changes in psoriatic arthritis (PsA) disease activity, particularly flares, are unpredictable and can impact patients' quality of life. Digital biomarkers derived from data collected via smart devices provide an opportunity for unobtrusive continuous monitoring of symptoms reflecting disease activity. Besides, our understanding of the mechanisms and triggers behind flares is limited. The primary objectives of the Psoriatic Arthritis Digital Phenotyping and Inflammation Drivers (PDPID) study are: (1) to develop digital biomarker capable of detecting PsA flare using data collected

Exploring the Patient Pathway to Focused Ultrasound Ablation for Essential Tremor: A Mixed-Method Approach.

INTRODUCTION: Focused ultrasound ablation (FUSA) is an emerging treatment for essential tremor (ET). While disparities in access to deep brain stimulation for ET have been identified, potential barriers to FUSA remains understudied. METHODS: A single tertiary-care center, mixed methods study was completed of medication-refractory ET patients presenting for FUSA consultation. Baseline demographics and tremor scores were obtained. A semi-structured questionnaire and focus group discussion was implemented to estimate delays in care, quality of life (QOL) differences, and treatment barriers. Quan

Engineering of functional auditory neurons from human induced pluripotent stem cells.

BACKGROUND: Spiral ganglion neurons (SGNs) relay auditory sensory information from the cochlea to the brain. Their loss results in permanent hearing impairment in humans due to their limited regenerative capacity. Progress in hearing restoration has been constrained by the inaccessibility of human inner ear tissue and challenges in generating functionally mature human SGN-like neurons from stem cells in vitro. METHODS: To generate human SGN-like neurons from human induced pluripotent stem cells (hiPSCs), we recapitulated key signaling pathways involved in human inner ear development. On day (

Enhancing prestin oligomerization for superior sonogenetics.

Background: Sonogenetics enables non-invasive control of cellular activity using ultrasound (US)-responsive proteins. Prestin, a motor protein from outer hair cells, can render cells responsive to US when ectopically expressed, offering potential therapeutic applications for neurodegenerative diseases and epilepsy. However, current prestin variants exhibit suboptimal oligomerization, reducing their efficacy in neuromodulation. This study aimed to enhance prestin sonogenetic performance by improving oligomerization. Methods: We screened four echolocating bat prestin orthologs and point mutants

A longitudinal study investigating the effects of noise exposure on behavioural, electrophysiological and self-report measures of hearing in musicians with normal audiometric thresholds.

Musicians are at risk of hearing loss and tinnitus due to regular exposure to high levels of noise. This level of risk may have been underestimated previously since damage to the auditory system, such as cochlear synaptopathy, may not be easily detectable using standard clinical measures. Most previous research investigating hearing loss in musicians has involved cross-sectional study designs that may capture only a snapshot of hearing health in relation to noise exposure. The aim of this study was to investigate the effects of cumulative noise exposure on behavioural, electrophysiological, an
