Biomaterials-based engineering of the bone microenvironment for osteoporosis therapy.

Osteoporosis is a progressive skeletal disorder marked by an imbalance between bone resorption and formation, resulting in compromised microarchitecture and increased fracture risk. However, conventional pharmacological therapies have systemic side effects and limited targeting efficiency. Therefore, these limitations highlights the need for innovative strategies, and biomaterials have emerged as versatile tools, offering both structural support and the ability to modulate the osteoporotic bone microenvironment. This review outlines the key pathophysiological changes in osteoporosis including

Orodispersible tablets in psychiatric and mood disorder management: clinical value, pharmacokinetics, and patient-centric formulation strategies.

OBJECTIVE: To analyze the extensive literature underlying the concept of using and formulating Orodispersible tablets (ODTs) for managing psychiatric and mood disorders. SIGNIFICANCE: Psychiatric and mood disorders represent a major global health burden across all age groups, significantly impairing quality of life and daily functioning. Emotional distress associated with these conditions often leads to poor medication adherence and difficulty swallowing, particularly in vulnerable populations. Thus, an increased emergency intervention is required during acute panic or agitation states. ODTs

An innovative and stable mRNA-LNP microneedle vaccine elicits humoral and multifunctional cellular immune responses.

During the COVID-19 pandemic, the use of lipid nanoparticles (LNPs) augmented the development of mRNA vaccines. However, their ultralow-temperature storage and transportation requirements, as well as their heavy reliance on injection by professional medical staff, have limited large-scale vaccination in many developing countries. Herein, we developed a simple and widely deployable microneedle (MN) vaccine delivery system (mLNP-man-MN) for mannose-modified LNPs (mLNP-man) loaded with mRNA encoding the SARS-CoV-2 spike receptor-binding domain by utilizing three-dimensional printing and polydimet

Metacarpophalangeal joint reconstruction using a costal osteochondral graft: A case report.

RATIONALE: The conventional treatment of giant cell tumors is intralesional curettage with local adjuvant therapy. Because hand tumors have a high local recurrence, the primary goal for treating tumors of the hand is to eradicate the lesion. PATIENT CONCERNS: To preserve the metacarpophalangeal (MCP) joint function as well as avoid further recurrence after surgery. DIAGNOSES: The giant cell tumor invades the patient's MCP joint in an index proximal phalanx. INTERVENTIONS: Using computer-aided design and three-dimensional printing techniques, we reformed the original shapes of the MCP joint

Hand eczema symptoms, exposures and skin care in orthodontics : A national, cross-sectional questionnaire-based survey.

OBJECTIVES: Occupational hand eczema is a common inflammatory skin condition among healthcare professionals. Orthodontists are frequently exposed to a variety of irritating and allergenic substances, and therefore they belong to a predisposed group to develop hand eczema. However, current data on the prevalence and predisposing factors among orthodontists to provide adequate prophylaxis are lacking. METHODS: An anonymous online survey was conducted in Germany between January and February 2023 and distributed to 2402 orthodontists. The questionnaire addressed general information on current ski

Novel techniques and future developments in minimally invasive pectus repair.

After the paradigm shift in the treatment of excavated deformities caused by the introduction of the Nuss technique in 1998, several innovative technical modifications and new treatment modalities have radically changed the surgical approach of pectus excavatum in the last couple of years. These new trends attain different topics. On one hand, the use of three-dimensional (3D) printing and implant customization surge as a possibility for a wider audience as 3D printers become available with lower costs. They provide the surgeon with new elements that enable precise planning, simulation, and cu

Fracture Non-Union in Osteoporotic Bones: Current Practice and Future Directions.

Given the compromised bone quality and altered healing environment, fracture non-union in osteoporotic bones presents a complex challenge in orthopedics. As global populations age, the incidence of osteoporotic fractures rises, leading to increased delayed healing and non-union cases. The pathophysiology underlying non-union in osteoporotic patients involves impaired bone regeneration, reduced osteoblast function, and poor vascularity. Traditional management strategies - ranging from pharmacological interventions like bisphosphonates and teriparatides to surgical approaches such as bone grafti

The Progress in Reconstruction of Mandibular Defect Caused by Osteoradionecrosis.

Osteoradionecrosis (ORN) is described as a disease with exposed, nonviable bone that fails to heal spontaneously or by means of conservative treatment after radiotherapy in at least 3 months. Though traditional theories in the early stage including hypoxic-hypocellular-hypovascular and fibro-atrophic in addition to new findings such as ferroptosis were put forward to explain the mechanisms of the osteoradionecrosis, the etiology of ORN is still unclear. With the high rate of occurrence in the head and neck area, especially in the mandible, this disease can disrupt the shape and function of the

3D Printing of Collagen/Oligomeric Proanthocyanidin/Oxidized Hyaluronic Acid Composite Scaffolds for Articular Cartilage Repair.

Articular cartilage defects affect millions of people worldwide, including children, adolescents, and adults. Progressive wear and tear of articular cartilage can lead to progressive tissue loss, further exposing the bony ends and leaving them unprotected, which may ultimately cause osteoarthritis (degenerative joint disease). Unlike other self-repairing tissues, cartilage has a low regenerative capacity; once injured, the cartilage is much more difficult to heal. Consequently, developing methods to repair this defect remains a challenge in clinical practice. In recent years, tissue engineerin

3D printing of microscopic bacterial communities.

Bacteria communicate via short-range physical and chemical signals, interactions known to mediate quorum sensing, sporulation, and other adaptive phenotypes. Although most in vitro studies examine bacterial properties averaged over large populations, the levels of key molecular determinants of bacterial fitness and pathogenicity (e.g., oxygen, quorum-sensing signals) may vary over micrometer scales within small, dense cellular aggregates believed to play key roles in disease transmission. A detailed understanding of how cell-cell interactions contribute to pathogenicity in natural, complex env

L1 vertebral body replacement using 3D-printed polylactic acid bioimplants: in vitro cellular evaluation, in vivo rat model assessment, and histological analysis of implant osseointegration.

BACKGROUND: The vertebral body plays a crucial role in supporting compressive loads and maintaining spinal biomechanics. An ideal biomaterial for total vertebral body replacement should combine biological and mechanical properties, yet no current material fulfills all criteria. This pilot study explores the use of a novel three-dimensional (3D)-printed porous polylactic acid (PLA) implant for total L1 vertebral body replacement. METHODS: This study had four stages: first, design, optimization, and 3D printing of the PLA device; second, in vitro evaluation of biocompatibility and cell growth u

Radiotherapy planning in a prostate cancer phantom model with intraprostatic dominant lesions using stereotactic body radiotherapy with volumetric modulated arcs and a simultaneous integrated boost.

AIM: In the treatment of prostate cancer with radiation therapy, the addition of a simultaneous integrated boost (SIB) to the dominant intraprostatic lesions (DIL) may improve local control. In this study, we aimed to determine the optimal radiation strategy in a phantom model of prostate cancer using volumetric modulated arc therapy for stereotactic body radiotherapy (SBRT-VMAT) with a SIB of 1-4 DILs. METHODS: We designed and printed a three-dimensional anthropomorphic phantom pelvis to simulate individual patient structures, including the prostate gland. A total of 36.25 Gy (SBRT) was deli

Evaluation of the Properties of 3D-Printed Ti Alloy Plates: In Vivo and In Vitro Comparative Experimental Study.

Titanium (Ti)-based implants play a significant role in rigid internal fixation in maxillofacial surgery. No study has reported that three-dimensional-printed Ti alloy plates (3D-Ti plates) have comprehensively excellent properties similar to standard plates (Matrix-MANDIBLE, SYNTHES, Switzerland) (Synthes-Ti plates). In this work, we manufactured 3D-Ti plates by selective laser melting with Ti6Al4V powder. The surface morphology, mechanical properties, and bone-plate contact rate of the 3D-Ti plates and the Synthes-Ti plates were characterized and compared via electron microscopy, atomic forc

Field Expansion with Multiplexing Prism Glasses Improves Pedestrian Detection for Acquired Monocular Vision.

PURPOSE: Patients with acquired monocular vision (AMV) lose vision in the temporal crescent on the side of the blind eye. This visual field loss affects patients' ability to detect potential hazards in the blind field. Mounting a base-in multiplexing prism (MxP) on the nasal side of the seeing eye can provide true field expansion and enable detection of potential collision hazards. We evaluated the efficacy of the MxP glasses in a virtual reality walking environment. METHODS: A three-dimensional printed clip-on MxP holder that can be adjusted for an individual user's facial parameters was dev

Next generation of ventricular catheters for hydrocephalus based on parametric designs.

BACKGROUND: The flow pattern of the cerebrospinal fluid is probably the most important factor related to obstruction of ventricular catheters during the normal treatment of hydrocephalus. To better comprehend the flow pattern, we have carried out a parametric study via numerical models of ventricular catheters. In previous studies, the flow was studied under steady and, recently, in pulsatile boundary conditions by means of computational fluid dynamics (CFD) in three-dimensional catheter models. OBJECTIVE: This study aimed to bring in prototype models of catheter CFD flow solutions as well to

Conjoint and extended neural networks for the computation of speech codes: the neural basis of selective impairment in reading words and pseudowords.

The computation of speech codes (i.e. phonology) is an important aspect of word reading. Understanding the neural systems and mech- anisms underlying phonological processes provides a foundation for the investigation of language in the brain. We used high-resolution three-dimensional positron emission tomography (PET) to investigate neural systems essential for phonological processes. The burden of neural activities on the computation of speech codes was maximized by three rhyming tasks (rhyming words, pseudowords and words printed in mixed letter cases). Brain activation patterns associated w
