[Changes in N-acetyl-beta-glucosaminidase (NAG) in prostatitis patients].

NAG was measured before and after treatment and compared in 11 acute prostatitis patients, 13 chronic prostatitis patients and 10 healthy volunteers. Urinary NAG before treatment was 34.1 +/- 18.4 U/l in the acute prostatitis patients and was significantly higher than that in the healthy volunteers, which was 5.1 +/- 1.2 U/l. It was 8.5 +/- 3.2 U/l in the chronic prostatitis patients. Urinary NAG after treatment decreased and was 7.6 +/- 4.4 U/l and 6.7 +/- 1.7 U/l in the acute and chronic prostatitis groups, respectively. These results suggest that urinary NAG is useful in evaluating the chan

Utilization of exogenously added acetyl coenzyme A by intact isolated lysosomes.

Acetyl coenzyme A has been shown to be required for heparan sulfate degradation for the acetylation of terminal alpha-linked glucosamine residues (Klein, U., Kresse, H., and von Figura, K. (1978) Proc. Natl. Acad. Sci. U.S.A. 75, 5185-5189). We can now demonstrate this requirement in intact isolated lysosomes. When lysosomes deficient in the enzyme alpha-N-acetylglucosaminidase were incubated at 37 degrees C with [3H]acetyl-CoA, tritium was incorporated primarily into glycosaminoglycans. By utilizing hydrolytic enzymes of known specificity, the labeled glycosaminoglycan was identified as hepar…

Assay of urinary N-acetyl-beta-glucosaminidase in a centrifugal analyzer.

We describe a kinetic, fluorometric assay for urinary N-acetyl-beta-glucosaminidase (EC 3.2.1.30) by measurement of the release of 4-methylumbelliferone (7-hydroxy-4-methylcoumarin). The method is simple, involving only a single reagent, and is applicable to the assay of other enzymes that hydrolyze similar fluorescent-labeled substrates. The enzyme distribution in human tissues and fluids is described. The enzyme is present in high concentrations in human kidney, liver, and lung. Its concentration in urine is shown to be a sensitive indicator of early renal damage, which precedes changes in s

Acetylation-like H2BK109Q attenuates acute brain injury and preserves axon-myelin integrity after intracerebral hemorrhage.

AIMS: Axon-myelin damage contributes to neurological dysfunction after intracerebral hemorrhage (ICH), but the upstream chromatin regulators remain unclear. We aimed to characterize the H2B acetylation response to ICH, test the functional significance of modeled H2BK109 states, and identify a downstream effector of H2BK109Q-mediated protection. METHODS: Pan-H2B acetylation was examined in mice with collagenase-induced ICH and in hemoglobin-treated HT22 cells, and the histone acetylome of the mouse brain was profiled. H2BK109Q and H2BK109R were used to model acetylation-mimetic and non-acetyla…

p300 Degradation by the p53-SIAH1 Axis Relieves TBK1 Acetylation to Enhance Innate Antiviral Immunity.

p300 is an acetyltransferase that regulates gene expression by acetylating histones and transactivating some transcription factors such as nuclear Factor Kappa B (NF-κB) and interferon regulatory factor 3 (IRF3). p53 is an interferon (IFN)-inducible tumor suppressor that enhances antiviral responses. How p300 and p53 precisely regulate innate antiviral immunity remains incompletely understood. Herein, we report that conditional p300 knockout in alveolar epithelial cells does not suppress but rather enhances antiviral responses in mice infected with vesicular stomatitis virus (VSV) and herpes s

Epigenetic activation of PDLIM7 via H3K27 acetylation mitigates neuroinflammation and neurodegeneration in parkinson's disease models.

This study investigates the role and regulatory mechanisms of PDZ and LIM domain protein 7 (PDLIM7) in the pathology of Parkinson’s disease (PD) and evaluates its potential as a therapeutic target, specifically focusing on epigenetic regulation through histone acetylation. We investigated PDLIM7 expression and regulatory mechanisms in PD using a multi-level strategy that combined bioinformatics analysis, postmortem human substantia nigra tissues, and 6-hydroxydopamine (6-OHDA)-induced PD models in mice and cell lines. Utilizing functional tests, such as immunohistochemistry (IHC), ubiquitinati…

RNA acetylation modification ac4C: An emerging regulatory hub of RNA metabolism disruption in Alzheimer's disease.

RNA metabolic dysregulation is a key pathological mechanism underlying the onset and progression of Alzheimer's disease (AD), involving multiple aspects such as abnormal RNA splicing, loss of function in RNA-binding proteins, dysregulation of non-coding RNAs, and impaired nuclear-cytoplasmic transport. In recent years, the emergence of epigenome research has revealed the critical role of RNA chemical modifications in regulating RNA metabolism at the post-transcriptional level. N4-acetylcytidine (ac4C) is the only known RNA acetylation modification in eukaryotes and is specifically catalyzed by…

KAT5-mediated acetylation enhances the deubiquitination of HASPIN by OTUB2 and promotes breast cancer progression.

Breast cancer (BC) remains the leading cause of global female cancer-related mortality, with poor survival in advanced stages driven largely … pecifically counteracting its K48-linked polyubiquitination and subsequent proteasomal degradation. Acetylation of HASPIN at lysine 751 by acetyltransferase lysine acetyltransferase 5 (KAT5) enhances its affinity for OTUB2, promoting HASPIN stability. Functionally, OTUB2 depletion reduces HASPIN protein levels, while OTUB2 overexpression-induced HASPIN upregu …

cKAT acetylation of the CO2 hydration protein ChpX regulates the CO2 concentrating mechanism in cyanobacteria.

Lysine acetylation (Kac) is a critical post-translational modification that regulates photosynthesis and carbon metabolism in cyanobacteria. However, the diversity and functional roles of lysine acetyltransferases (KATs) beyond the well-characterized cGNAT2 remain poorly defined. This study identifies and functionally characterizes the previously unannotated protein A0096, designated here as cKAT, demonstrating its capacity to catalyze Kac both in vivo and in vitro in Synechococcus sp. PCC 7002 (Synechococcus). Deletion of cKAT significantly impaired cellular growth and photosynthetic efficien…

JMJD3-mediated HADH demethylation orchestrates hepatocyte maturation through fatty acid β-oxidation-dependent histone acetylation.

Functionally mature hepatocyte generation requires coordinated genetic, metabolic, and epigenetic regulation. Although transcriptional regul … these maturation defects, supporting a functional link between FAO-dependent metabolism and histone acetylation. Collectively, our findings establish a JMJD3-HADH-FAO-acetyl-CoA regulatory axis that coordinates metabolic remodeling and epigenetic regulation during hepatocyte maturation.

Regulation of Chromatin Acetylation by Alcohol: Dependence on Sex, Brain Region, and Mode of Exposure.

Both genetic and epigenetic factors influence the development and pathology of alcohol use disorder (AUD), which is further associated with changes in learning, memory, and synaptic plasticity. Histone acetylation is an epigenetic mechanism that changes the chromatin architecture, influencing gene transcription, which may further impact neuronal signaling, cognition, and addiction-related behaviors. In this review, we summarize the existing literature on how alcohol exposure impacts histone acetylation and the expression and activity of histone acetyltransferases (HATs) and histone deacetylase…

ACLY-Driven Metabolic Reprogramming Promotes Histone Acetylation and Inflammation-Associated Fibrosis in Chronic Kidney Disease.

The mechanisms by which metabolic stress drives epigenetic dysregulation and fibrosis in chronic kidney disease (CKD) remain incompletely understood. Using quantitative histone proteomics in murine fibrosis models, we uncovered a selective increase in  histone H3 lysine 27 acetylation (H3K27ac) as a conserved epigenetic feature. Unbiased metabolomics revealed citrate accumulation, nominating ATP-citrate lyase (ACLY) as a driver of acetyl-CoA-dependent histone acetylation. In murine models of folic acid and unilateral ureteral obstruction, ACLY expression, acetyl-CoA levels, and H3K27ac were in…

Palmitate-induced mitochondrial damage restricts histone acetylation in CD8 T cells to impair antitumor immunity.

Lipid accumulation in the tumor microenvironment is a hallmark of solid tumors, with increased palmitate (PA) availability fostering tumor progression. Although PA's direct effects on cancer cells are well described, its impact on CD8 T cells [cytotoxic T lymphocytes (CTLs)] remains unclear. Here, we show that PA irreversibly impairs CTL mitochondrial metabolism, leading to the loss of effector functions and compromised antitumor immunity. PA-induced mitochondrial dysfunction reduced histone acetylation and chromatin accessibility, suppressing transcription of genes involved in T cell replicat…

Aspirin and fibrin clot modulation in vascular pharmacology: Fibrinogen acetylation, disease context, and translational uncertainty.

Fibrin clot architecture is increasingly recognized as a determinant of thrombotic phenotype and a potentially modifiable endpoint in vascular pharmacology. Aspirin is established as an antiplatelet agent through irreversible cyclooxygenase-1 acetylation and thromboxane A₂ suppression. Beyond this canonical pathway, aspirin has been proposed to modulate fibrin clot properties through nonenzymatic Nε-lysine acetylation of fibrinogen. Experimental studies in purified, recombinant, and selected ex vivo systems suggest effects on fibrin polymerization, factor XIIIa-dependent cross-linking, clot pe…

CPEB1 drives ferroptosis-neuroinflammation crosstalk in temporal lobe epilepsy via the SIRT1-NRF2 acetylation axis.

BACKGROUND: Temporal lobe epilepsy (TLE) is a common neurological disorder frequently resistant to pharmacological treatment, yet its molecu … bust neuroprotective effects. Mechanistically, CPEB1 suppressed SIRT1 activity, leading to enhanced acetylation-dependent destabilization of NRF2, impaired downstream SLC7A11/GPX4 antioxidant signaling, and excessive reactive oxygen species (ROS) accumulation, ultimately triggering ferroptotic neuronal death. Importantly, pharmacological inhibition of SIRT1 o …

Association of Glycoprotein Acetylation With Carotid-Femoral Pulse Wave Velocity in Patients Receiving Maintenance Hemodialysis.

BACKGROUND Patients undergoing long-term maintenance hemodialysis (HD) experience persistent low-grade inflammation and are at high risk of cardiovascular complications. Aortic stiffness, commonly assessed by carotid-femoral pulse wave velocity (cfPWV), is a well-established marker of cardiovascular risk. Glycoprotein acetylation (GlycA) reflects cumulative systemic inflammation; however, its association with aortic stiffness in patients undergoing HD remains unclear. MATERIAL AND METHODS In this cross-sectional study, 160 patients receiving maintenance HD were enrolled. Aortic stiffness was d…
