This study comprehensively evaluated the in vitro and in vivo anthelmintic efficacy and pharmacokinetics of four benzimidazole compounds: febantel (FB), fenbendazole (FBZ), albendazole (ABZ), and mebendazole (MBZ) against the gill fluke Microcotyle sebastis in cultured black rockfish (Sebastes schlegelii). In vitro tests with isolated adult parasites demonstrated that ABZ exhibited the highest efficacy, followed by FBZ, while MBZ showed minimal activity and FB remained largely inactive at lower concentrations. At the highest concentration tested (200 mg/L), FB showed only weak, nonspecific act

Anthelmintics are drugs used for controlling pathogenic helminths in animals and plants. The natural compound betaine and the recently developed synthetic compound monepantel are both anthelmintics that target the acetylcholine receptor ACR-23 and its homologs in nematodes. Here, we present cryo-electron microscopy structures of ACR-23 in apo, betaine-bound, and betaine- and monepantel-bound states. We show that ACR-23 forms a homo-pentameric channel, similar to some other pentameric ligand-gated ion channels (pLGICs). While betaine molecules are bound to the classical neurotransmitter sites i…

Zebrafish in research facilities are frequently infected with capillarids. Since the health status (as a nonprotocol source of variation) of zebrafish can affect the validity of experiments, it is important to develop therapies for common zebrafish diseases. Regarding the likelihood of (1) the development of drug resistance and (2) the loss of the efficacy of a drug for laboratory zebrafish, the availability of alternatives for treatment is of direct importance. The efficacy of five dewormers from the same or different therapeutic groups was assessed in the current study. The exposure to each

Fatty acid and retinol binding proteins (FARs) are nematode-specific proteins that orchestrate lipid metabolism, development, and host immune response. Here, the antagonists of Nippostrongylus brasiliensis FAR-1 (NbFAR-1) were identified through integrating virtual screening, fluorescent ligand binding assay, and in vitro egg hatching assays. An in vivo mouse model was employed to evaluate anthelmintic efficacy against intestine-parasitized N. brasiliensis and brain-parasitized Angiostrongylus cantonensis. Forty-eight candidates were selected by virtual screening, six of them showed more than

In vivo anthelmintic assay models are essential for evaluating candidate compounds in target species. Several rodent-helminth systems have been developed, with the Cobb-Wistar rat-T. colubriformis model emerging as promising for early-stage screening. Although this model has demonstrated utility, the development of an alternative model using a more widely accessible rat strain represents a valuable opportunity for advancing in vivo screening of anthelmintics. This study evaluated different immunosuppressants and regimens to identify treatments yielding the highest and most consistent worm burd…

Anthelmintic resistance in gastrointestinal nematodes (GIN) of cattle is of increasing concern, but the number of studies monitoring anthelmintic efficacy is insufficient to provide a representative picture. In this study, resistance to macrocyclic lactones (ML) and benzimidazoles (BZ) in GINs from 14 Scottish dairy farms (four non-organic, 10 organic) was evaluated using multiple approaches. The faecal egg count reduction test (FECRT) remains the primary tool for evaluating anthelmintic resistance in the field and was used on the four non-organic farms. Differing methodologies and recent guid

In this study, the anthelmintic potential of the ethanolic and aqueous extracts of Moringa oleifera leaves and Embelia ribes seeds was evaluated against Haemonchus contortus by both in vitro (egg hatch assay, EHA; larval development assay, LDA; and adult mortality assay, AMA) and in vivo assays (faecal egg count reduction test, FECRT). Ethanolic extract of M. oleifera (MOEE) exhibited maximum inhibitory effects on egg hatching (87.77%) and larval development (86.97%) along with complete adult mortality (100% after 12 hours of incubation) @ 16.0 mg/mL and 32.0 mg/mL, respectively. However, etha

For decades, farmers relied on anthelmintic drug treatments to control gastrointestinal nematode (GIN) infections, which has led to the development of anthelmintic resistance (AR). The objective of this study was to investigate the perceptions of communal goat farmers regarding anthelmintic resistance in communal goat flocks. A structured, close-ended questionnaire was used to carry out face-to-face interviews with communal goat farmers (n = 384) across four local municipalities in the uMkhanyakude district. The results showed that elderly farmers were 1.4 times more likely to underdose compar

BACKGROUND: Infection with parasitic nematodes (helminths), particularly those of the order Strongylida (such as Haemonchus contortus), can cause significant and burdensome diseases in humans and animals. Widespread drug (anthelmintic) resistance in livestock parasites, the absence of vaccines against most of these nematodes, and a lack of new and effective chemical entities on the commercial market demands the discovery of new anthelmintics. In the present study, we searched the Global Health Priority Box (Medicines for Malaria Venture) for new candidates for anthelmintic development.

Breast cancer resistance protein/ATP-binding cassette subfamily G2 (BCRP/ABCG2) is an ATP-binding cassette efflux (ABC) transporter expressed in the apical membrane of cells in tissues, such as the liver, intestine, kidney, testis, brain, and mammary gland. It is involved in xenobiotic pharmacokinetics, potentially affecting the efficacy and toxicity of many drugs. In this study, the role of ABCG2 in parasiticide monepantel (MNP) and its primary metabolite, monepantel sulfone (MNPSO2)'s systemic distribution and excretion in milk, was tested using female and male wild-type and Abcg2-/- mice. The agropastoral farmers have employed Turraea vogelii(TVL),Senna podocarpa(SPL), and Jaundea pinnata (JPL) leaves for treating various diseases, including intestinal parasites in livestock and the human population in Nigeria. Gastrointestinal nematodes are highly significant to livestock production and people's health, and natural products are interesting as sources of new drugs. In this study, we evaluated the effectiveness of extracts derived from these plants in treating parasitic infections using third-stage infective larvae (L3) of Strongyloides venezuelensis. We obtained crude extracts
