Carcinogenicity and mechanistic insights on the behavior of epoxides and epoxide-forming chemicals.

Many epoxides and their precursors are high production volume chemicals that have major uses in the polymer industry and as intermediates in … animals and/or workers exposed to several of these chemicals. The classifications of these chemical carcinogens by IARC and NTP are based on animal and human data and results of mechanistic studies. Reducing occupational and environmental exposures to these chemicals will certainly reduce human cancer risks.

Deceleration of carcinogenic potential by adaptation with low dose gamma irradiation.

Animals that have been exposed to a very low dose of radiation are known to have many physiological benefits. Very low dose of ionizing radiation also induces mechanisms whereby cell or tissue become better fit to cope with subsequent exposures of high doses. This phenomenon of low dose radiation is termed 'adaptive response'. This response has been reported to be true in many biological systems and confirmed by experiments on chromosomal and chromatid aberrations, micronucleus formation, sister chromatid exchange tests, DNA mutation and cell survival study and using many other biological end

Long-term toxicity and carcinogenicity study of cyclamate in nonhuman primates.

Twenty-one monkeys (cynomolgus, rhesus, African green) were fed cyclamate (100 mg/kg and 500 mg/kg) in the diet five times per week from a few days after birth and continuing for up to 24 years. Malignant tumors were diagnosed in three 24-year-old cyclamate monkeys; these were metastatic colon carcinoma (rhesus; 500 mg/kg), metastatic hepatocellular carcinoma (cynomolgus; 500 mg/kg), and a small, well differentiated adenocarcinoma of the prostate (cynomolgus; 100 mg/kg). Benign tumors were found at necropsy in three females; these were adenoma of the thyroid gland (rhesus; 100 mg/kg) and two c

Carcinogenic aspect of xenobiotic molecules belonging to the peroxisome proliferator family.

It is known that a short-term exposure of rat, mice or incubation of hepatic cells with fibrate molecules leads to increase in peroxisome number and cell hyperplasia. Further, long-term incubation of cells (at least a year) show transformed characteristics with foci and nodules. To explain the hepatocarcinogenic effect of peroxisome proliferators in rodents we studied the effect of peroxisome proliferators on rat liver oncogenes expression. Earlier, we reported an increase in liver and kidney mRNA level of c-myc and N-myc. Since several metabolic genes are activated by PPAR (peroxisome prolife

Twenty-four-month oral carcinogenicity study of ebrotidine in rats.

Three groups of Sprague-Dawley CD rats (males and females) were initially administered p.o. with ebrotidine, a novel H2-receptor antagonist, mixed with the diet, at 50, 200, and 500 mg/kg/d, respectively. Two concurrent control groups of animals were used. After 13 months, initial 200 mg/kg was lowered to 150 mg/kg, and a new group was administered with 300 mg/kg, due to the body weight reduction observed in the top dose group. After 24 months, survivors were killed and necropsied, and a histopathological study was performed. The frequencies of the different tumour types that were found were n

Radio frequency radiation (RFR): the nature of exposure and carcinogenic potential.

Epidemiologic evidence on the relation between radio-frequency radiation (RFR) and cancer is reviewed. Radio-wave communications are used extensively in modern society; thus, we are all subject to RFR created by radio, television, wireless telephony, emergency communications, radar, etc. Interest in the health effects of RFR has been motivated by the rapid growth in wireless communications and by media reports expressing concern that specific diseases may be caused by RFR exposure, e.g., from cellular telephone handsets. Due to the ubiquitous presence of RFR, the public health implication of a

Effects of riboflavin deficiency and riboflavin administration on carcinogen-DNA binding.

A study was conducted to assess the effects of riboflavin deficiency and riboflavin supplementation on carcinogen-DNA binding. After 12 wk on a riboflavin-sufficient or a riboflavin-deficient diet male Wistar rats were administered 3H-labelled benzo[a]pyrene (BP) ip. [3H]BP was given either at a uniform dose of 450 muCi/rat irrespective of body weight or at a dose adjusted to body weight. After 17 hr the animals were killed, various organs were dissected and the level of [3H]BP bound to DNA was quantified in organs that are known to be the seats of drug metabolism (i.e. the liver, lungs and in

Species differences in the metabolism of trichloroethylene to the carcinogenic metabolites trichloroacetate and dichloroacetate.

Differing rates and extent of trichloroethylene (TCE) metabolism have been implicated as being responsible for varying sensitivities of mice and rats to the hepatocarcinogenic effects of TCE. Recent data indicate that the induction of hepatic tumors in mice may be attributed to the metabolites trichloroacetate (TCA) and/or dichloroacetate (DCA). The present study was directed at determining whether mice and rats varied in (1) the peak blood concentrations, (2) the area under the blood concentration over time curves (AUC) for TCE and metabolites in blood, and (3) the net excretion of TCE to the

Carcinogenic effects of bis(2,3-dibromopropyl)-phosphate in Wistar rats.

Four groups of 40 Wistar rats of each sex per dose level were fed diets containing 0, 80, 400 or 2000 ppm of the magnesium salt of bis(2,3-dibromopropyl)phosphate (Bis-BP) for 24 months. A high incidence of tumours was induced, in both sexes, in the digestive system. Tumours included papillomas and adenocarcinomas of the tongue, oesophagus and forestomach, adenocarcinomas of the intestine and hepatocellular adenomas (neoplastic nodules) and carcinomas. Pre-terminal mortalities were associated with an increased incidence of forestomach papillomas in both sexes, adenocarcinomas of the small inte

Species differences in carcinogenicity: the role of metabolism and pharmacokinetics in risk assessment.

Attempts to improve quantitative risk assessments inevitably lead to the use of additional biological data in the risk calculation. The need for more data increases further when differences in response between laboratory animals result in uncertainty in the choice of either the species or the tumour incidence on which to base the risk assessment. Of the many stages of carcinogenesis, the first stage, which involves the uptake and activation of the chemical, is probably the most understood and is by far the easiest to measure experimentally. A review of the use of metabolism and pharmacokinetic

Molecular basis of hexavalent chromium carcinogenicity: effect on gene expression.

The carcinogenicity of chromium(VI) compounds is explained in terms of the "uptake-reduction" model. According to this model, chromium (VI) enters cells as the anionic tetrahedral species chromate, CrO4(2-), using normal cellular anion transport systems, such as the sulfate transport system. Redox-active enzymes and small molecules reduce chromium (VI) intracellularly and produce "reactive intermediates" capable of damaging cellular constituents. DNA is presumed to be a critical target within the cell; however, chromium (VI) itself is unreactive toward DNA under physiological conditions in vit

Enhancement of DNA repair capacity of mammalian cells by carcinogen treatment.

To determine whether DNA excision repair is enhanced in mammalian cells in response to DNA damage, as it is in bacteria as part of the SOS response, we used an expression vector-host cell reactivation assay to measure cellular DNA repair capacity. When UV-damaged chloramphenicol acetyltransferase (CAT) vector DNA was introduced into monkey cells (CV-1), the level of CAT activity was inversely related to the UV fluence due to inhibition of CAT gene expression by UV photoproducts. When CV-1 cells were treated with either UV radiation or mitomycin C, 24-48 h before transfection, CAT expression fr

Biological basis for assessing carcinogenic risks of low-level radiation.

Ionizing radiation is carcinogenic to many, if not most, tissues. Its carcinogenicity varies, however, depending on the tissue exposed, conditions of exposure, genetic background, sex, age of the exposed individual, and other factors. The neoplasms induced by radiation also vary in their types and in their times of onset, depending on the age and sex of the exposed individual. The long induction period for radiation carcinogenesis and the enhancing or inhibiting effects of other agents acting after irradiation imply that the induction of cancer is a multistage process, in keeping with experime

Autoimmune haemolytic disease in mice after exposure to a methylating carcinogen.

N-Methyl-N-nitrosourea (MNU) but not methyl methanesulphonate (MMS) induced both autoimmune haemolytic anaemia and thymic lymphoma in susceptible strains of mice, particularly the C57BL/6. These effects could be positively correlated with the formation of O6-methylguanine in target DNA. All murine lymphoid cells showed lack of ability to remove O6-methylguanine from their DNA, therefore the variation of responses between different mouse strains indicated that other host factors, probably genetic, must be involved. The results do indicate however that a potent pre-mutagenic DNA base modificatio

A uridine adduct with an ester of the carcinogen 3-hydroxy-1-methylxanthine.

The structure of the uridine adduct with the acetate ester of the carcinogen 3-hydroxy-1-methylxanthine has been determined. Covalent binding is between C-8 of xanthine and O-2 of uracil. This was determined from studies of the NMR spectrum, mass spectra and solvolysis in liquid hydrogen sulfide. The nucleoside adduct, formed with uridine, is identical with the adduct with polyuridylic acid after enzymatic hydrolysis. Treatment with aqueous ammonia or pH 7 at 100 degrees C leads to the loss of ribose. Thymidine also forms an adduct with 3-acetoxy-1-methylxanthine in a similar yield. Model stud

Stimulation of drug and carcinogen metabolism by prolonged oral tobacco consumption.

Oral administration of tobacco to rats for 21 days caused remarkable stimulation of the metabolism of phenacetin, aniline and benzo[a]pyrene, a carcinogen, by hepatic microsomal mixed function oxidases (MFO). Such treatment for 6 days resulted in a small increase in the activities of phenacetin O-dealkylase and aromatic hydrocarbon hydroxylase (AHH) without affecting aniline hydroxylase activity. Nicotine given orally was found to be a relatively weak inducer of phenacetin O-dealkylase and aniline hydroxylase, and elicited a maximum increase in their activities within 6 days which remained unc
