Caspase-1 and Inflammasome Signaling in Spinal Cord Injury: Mechanistic Insights and Translational Gaps.

Caspase-1 is a central mediator of canonical inflammasome signaling and drives neuroinflammatory injury through interleukin-1β (IL-1β) and interleukin-18 (IL-18) maturation, gasdermin D (GSDMD)-mediated pyroptosis, and secondary inflammatory cascades. In spinal cord injury (SCI) and other neurologic disorders, experimental evidence links caspase-1 activation to blood-spinal cord barrier (BSCB) or blood-brain barrier (BBB) disruption, glial activation, neuronal and oligodendroglial injury, and worse functional outcomes. However, despite mechanistic rationale, the clinical role of caspase-1 as a

The Caspase-1-EGR4 axis regulates macrophage repolarization in acute myeloid leukemia cells.

The polarization of tumor-associated macrophages (TAMs) toward an M2-like phenotype critically promotes acute myeloid leukemia (AML) progression. Building on the clinical observation that Caspase-1 (CASP1) expression is elevated in AML and correlates with M2 macrophage abundance, we identify a novel signaling axis in AML cells, involving CASP1 and the transcription factor early growth response protein 4 (EGR4), that orchestrates macrophage polarization. Knockdown (KD) of CASP1 in human AML cells (THP-1, MOLM-13) shifted their secretome, which consequently skewed macrophage polarization away fr

STOX1 Promotes Pyroptosis of Hypoxic Trophoblast Cells Via the Nrf2/Caspase-1 Axis.

This study aimed to investigate the role of STOX1 in trophoblast pyroptosis during preeclampsia (PE) and elucidate its underlying molecular mechanisms. A trophoblast hypoxia cell model was established by exposing HTR-8/SVneo cells to hypoxic conditions. The results showed that STOX1 messenger RNA expression was significantly elevated in placental tissues from PE patients as well as in hypoxia-treated HTR-8/SVneo cells. Hypoxia markedly induced pyroptosis in HTR-8/SVneo cells, whereas STOX1 silencing enhanced cell proliferation and migration while suppressing programmed cell death. Moreover, ST

Caspase-3 regulates RNA splicing and mitochondrial dynamics in microglia during Parkinson's disease.

Caspase-3, a cysteine-aspartic protease canonically known for its role in apoptotic cell death, has been implicated in several non-apoptotic functions, particularly in microglia, the brain-resident macrophages. These novel functions appear to depend on distinct levels of caspase-3 activation; however, the role of basal caspase-3 activity remains unclear. Here, we show that basal caspase-3 regulates RNA splicing in microglia, and its deficiency in a Parkinson´s disease (PD) model leads to splicing dysregulation. Loss of basal caspase-3 activity also increases double-stranded RNA (dsRNA) accumul

An NMR sample preparation case study: Considerations for the self-destructive protease caspase-6.

Proteases represent a difficult family of proteins to purify, concentrate and store at homogeneity due to their toxicity during overexpression and their propensity to self-cleave, leading to the loss of sample stability and function. A protease of interest, caspase-6, is a member of the apoptotic family of caspases, and has been shown to be involved in human neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. Previous studies have elucidated key structural aspects and potential inhibition mechanisms of caspase-6 through various structural biology techniques such as …

Caspase 9-induced apoptosis enables efficient fetal cell ablation and disease modeling.

Fetal cell ablation models are crucial for studying congenital diseases, organ regeneration, and xenotransplantation. However, conventional knockout models offer limited control over disease severity, while conditional ablation models often require fetus-harming inducers. In the present study, we demonstrate that the inducible caspase 9 system enables precise targeting of fetal nephron progenitor cells in mice through the intrinsic apoptotic pathway. Using a safe, placenta-permeable inducer, this system facilitates specific, rapid, and efficient cell ablation. The system's temporal control all

The role of alpha-pinene on the NLRP3-Caspase 1 pathway in the ulcerative colitis model in rats.

Alpha-pinene is a natural compound that has recently attracted the attention of researchers due to its anti-inflammatory and antioxidant effects. This study aimed to investigate the impact of alpha-pinene on the NLRP3-Caspase 1 pathway activity in colon tissue in a colitis model. The colitis was induced by directly administering 4 % acetic acid (A.A) into the colon. Twenty-four hours after this treatment, the test compound alpha-pinene (at doses of 50 and 100 mg/kg), sulfasalazine (50 mg/kg), or the vehicle (liquid paraffin) was given orally. Seven days after inducing colitis, the colonic segm

Repurposed clindamycin suppresses pyroptosis in tumor-associated macrophages through Inhibition of caspase-1.

BACKGROUND: The metastatic microenvironment is often rich in tumor-associated macrophages (TAMs). In uveal melanoma (UM), high levels of TAMs positively correlate with tumor progression and poorer prognosis. We hypothesize that the immunomodulation of TAMs can remodel the UM tumor microenvironment and make it more susceptible to therapeutic interventions. METHODS: In our work, we designed a novel computational pipeline that combines single-cell transcriptomics data, network analysis, multicriteria decision techniques, and pharmacophore-based docking simulations to select molecular targets and

Overexpression of inflammatory human caspase-4 in relation to clinical severity of oral lichen planus.

OBJECTIVE: Involvement of non-canonical inflammasome, comprising inflammatory human caspase-4, caspase-5, and Gasdermin D, in the pathogenesis of oral lichen planus (OLP) has never been demonstrated. We aimed to determine human caspase-4, caspase-5, and Gasdermin D expressions in OLP, to correlate their expressions with OLP severity, and to measure salivary interleukin (IL)-1β levels. STUDY DESIGN: OLP and normal oral mucosal specimens (n = 42 each) were processed for immunohistochemistry or immunoblotting. The clinical score for OLP severity was assessed at the most severe site. The immunohi

Neuroprotective effects of sinomenine and metformin in diabetic stroke: Role of NLRP3/caspase-1 and mitophagy.

Type 2 diabetes mellitus (T2DM) significantly increases the risk and severity of cerebral ischaemia-reperfusion (IR) injury, yet effective neuroprotective treatments for diabetic stroke remain limited. This study explored whether a combination of sinomenine and metformin could offer enhanced neuroprotection in diabetic rats subjected to cerebral IR injury, and investigated the molecular pathways involved. Male rats were rendered diabetic using a high-fat diet and low-dose streptozotocin. After establishing diabetes, rats underwent transient middle cerebral artery occlusion followed by 24 h of

KLF7-IT1-ARPC2 axis in macrophages orchestrates caspase-4-driven hypercoagulation in sepsis.

Sepsis is characterized by dysregulated inflammation and coagulation leading to organ dysfunction, yet the molecular mechanisms linking these processes remain elusive. Through transcriptomic profiling of leukocytes from septic and non-septic patients, we identified Kruppel-like factor 7 intronic transcript 1 (KLF7-IT1) as one of the most robustly upregulated long non-coding RNAs in sepsis. Elevated KLF7-IT1 expression correlated strongly with hypercoagulability, inflammatory cytokines, and multi-organ injury, suggesting a pathogenic role in septic coagulopathy. Functional studies revealed that

Targeting the DYNLL2-PAK1 axis inhibits caspase-11-dependent pyroptosis to alleviate sepsis.

Sepsis, a life-threatening syndrome caused by dysregulated host responses to infection, lacks effective therapeutic strategies due to its complex immune pathophysiology. Here, we identify Dynein Light Chain LC8-Type 2 (DYNLL2) as a critical risk gene driving sepsis progression through bioinformatics and machine learning analysis of clinical datasets. Elevated DYNLL2 expression correlates with poor prognosis and monocyte expansion in sepsis patients. Mechanistically, DYNLL2 interacts with p21-Activated Kinase 1 (PAK1) to regulate the endocytosis of Gram-negative bacterial outer membrane vesicle

Role of RGS17 in cisplatin-induced cochlear inflammation and ototoxicity via caspase-3 activation.

Cisplatin is a chemotherapy drug used to treat different solid tumors, including ovarian, bladder, lung, and head and neck cancers. One of its significant side effects is ototoxicity, especially when high doses are required. Cisplatin-induced ototoxicity is associated with increased cochlear cell death resulting from DNA damage, caspase activation, oxidative stress, inflammation, and glutamate excitotoxicity. The regulator of G protein signaling 17 (RGS17), a member of the RGS-RZ subfamily, hastens the hydrolysis of GTP to GDP on the Gα subunit. In the current study, we demonstrate the role of

Caspase-8 drove apoptosis of BMECs to promote neutrophil infiltration and DE205B clearance in meningitis.

Avian pathogenic Escherichia coli (APEC), a significant virulence reservoir for human extraintestinal pathogenic E. coli (ExPEC), poses an escalating zoonotic risk through the food chain. Our previous study demonstrated that the poultry-derived strain DE205B shared high genetic similarity with the neonatal meningitis-associated E. coli (NMEC) strain RS218 and induced meningitis in a rat model. Here, we further demonstrated that DE205B crossed the blood-brain barrier (BBB) via a transcellular pathway without compromising barrier integrity. During this process, brain microvascular endothelial ce

Polyethylene glycol and caspase inhibitor emricasan alleviate cold injury in primary rat hepatocytes.

Current methods of storing explanted donor livers at 4 °C in University of Wisconsin (UW) solution result in loss of graft function and ultimately lead to less-than-ideal outcomes post transplantation. Our lab has previously shown that supplementing UW solution with 35-kilodalton polyethylene glycol (PEG) has membrane stabilizing effects for cold stored primary rat hepatocytes in suspension. Expanding on past studies, we here investigate if PEG has the same beneficial effects in an adherent primary rat hepatocyte cold storage model. In addition, we investigated the extent of cold-induced apopt
