[Intracellular survival and replication of legionella pneumophila within host cells].

Legionella pneumophila is a facultative intracellular pathogen which replicates within macrophages and monocytes and finally cause a severe pneumonia known as Legionnaires' disease. An important hallmark of the pathogenesis of this bacterium is their ability to manipulate host cell processes, creating a specified replicative niche within host cells. An L. pneumophila-containing phagosome (LCP) is allowed to associate sequentially with smooth vesicles, mitochondria, and the rough endoplasmic reticulum (RER) to form a compartment called a replicative phagosome. LCPs are biologically characterize

Effect on tumor cells of blocking survival response to glucose deprivation.

BACKGROUND: Glucose deprivation, a feature of poorly vascularized solid tumors, activates the unfolded protein response (UPR), a stress-signaling pathway, in tumor cells. We recently isolated a novel macrocyclic compound, versipelostatin (VST), that inhibits transcription from the promoter of GRP78, a gene that is activated as part of the UPR. We examined the effect of VST on the UPR induced by glucose deprivation or other stressors and on tumor growth in vivo. METHODS: Human colon cancer HT-29, fibrosarcoma HT1080, and stomach cancer MKN74 cells were cultured in the absence of glucose or in

Cell aggregation: a mechanism of pathogenic Leptospira to survive in fresh water.

Transmission of leptospirosis is facilitated by the survival of pathogenic leptospires in moist environments outside their mammalian host. In the present study, the survival mechanisms of Leptospira interrogans serovar Canicola in aqueous conditions and lack of nutrients were investigated. In distilled water, leptospires were able to remain motile for 110 days (pH 7.2). However, when incubated in a semi-solid medium composed of distilled water and 0.5% purified agarose (pH 7.2), they survived 347 days. In this viscous environment, aggregates of live spirochetes were observed. Neither antibioti

Survival and axonal regeneration of retinal ganglion cells in adult cats.

Axotomized retinal ganglion cells (RGCs) in adult cats offer a good experimental model to understand mechanisms of RGC deteriorations in ophthalmic diseases such as glaucoma and optic neuritis. Alpha ganglion cells in the cat retina have higher ability to survive axotomy and regenerate their axons than beta and non-alpha or beta (NAB) ganglion cells. By contrast, beta cells suffer from rapid cell death by apoptosis between 3 and 7 days after axotomy. We introduced several methods to rescue the axotomized cat RGCs from apoptosis and regenerate their axons; transplantation of the peripheral nerv

Survival of transplanted human corneal stem cells. Case report.

PURPOSE: To report a case history involving long-term survival of transplanted human corneal stem cells. METHODS: A male patient with severe bilateral chemical burns received six corneal transplants, all of which failed. He subsequently received combined corneal transplants and stem cell transplants, which have remained clear for 3 and 4 years respectively. One of the donors was female. We studied the gender of the epithelial cells of the cheek of the patient and of the two grafts using fixation and fluorescent in situ hybridization (FISH) analyses. RESULTS: In the graft from the female dono

Leukemia inhibitory factor is an autocrine survival factor for Schwann cells.

Schwann cells play a major role in promoting nerve survival and regeneration after injury. Their activities include providing neurotrophic f … ion of the soluble LIF receptor (LIF binding protein) and anti-LIF antibodies significantly reduced cell survival, suggesting that LIF exerts autocrine effects. These results suggest that Schwann cell survival following nerve injury is potentially modulated by LIF.

Influence of aprotinin and promazine on survival of isolated pancreatic acinar cells.

Aprotinin, a protease inhibitor, and promazine, an inhibitor of phospholipase A2, were tested for possible inhibition of pancreatic acinar cell (PAC) decline induced by uncoupling of oxidative phosphorylation with 2,4-dinitrophenol (DNP) or by temporary anoxia/reoxygenation. In incubates of acinar cells isolated from rat pancreas the presence of aprotinin did not influence the survival of cells treated with these noxae. This finding excludes that extracellulary acting trypsin, possibly released from damaged cells, contributes to further cell death. While promazine at concentrations of 15 to 20

The role of membrane lipids in the survival of red cells in hereditary spherocytosis.

Red cells in hereditary spherocytosis (HS) have a decreased ratio of membrane surface area to cell volume and therefore a spheroidal shape. This abnormality in shape predisposes them to pooling and destruction in the spleen. Although splenectomy prevents hemolysis in HS, the red cell defect, as manifested by spheroidicity, increased autohemolysis, excesive permeability to sodium, and hypermetabolism, persists. The role of membrane lipids in these manifestations in vitro and in cell survival in vivo was examined. Before splencetomy, and in spite of the presence of a young cell population, the c…

Enrichment-Free Rare Cell Profiling in Pediatric Osteosarcoma Reveals a Circulating Cell Population Associated with Relapse-Free Survival.

BACKGROUND: Outcomes for patients with osteosarcoma (OS) presenting with metastases at diagnosis or relapsed remain poor with five-year survival rate of only 20-30%. Risk stratification at diagnosis remains limited, and no validated prognostic liquid biopsy analyte currently exists for this disease. OS lacks the recurring mutational hotspots and epithelial surface markers that underpin liquid biopsy strategies developed for carcinomas, necessitating fundamentally different approaches to circulating biomarker detection. METHODS: Peripheral blood samples from 25 pediatric and young adult OS pat

Red blood cell transfusion survival in sickle cell disease is reduced by donor characteristics and recipient spleen activity.

Red blood cell (RBC) transfusion therapy in sickle cell disease (SCD) is used to treat and prevent disease complications by providing nonsickle erythrocytes that survive for weeks in circulation. The suppression of sickle hemoglobin between transfusion episodes is dependent on donor RBC survival. We present an observational clinical trial of transfusion survival in pediatric and adult patients with SCD receiving chronic transfusion therapy using biotin-labeling to measure donor RBC life span. For each transfusion episode, aliquots from 2 to 3 RBC units were separated from each unit, biotin-lab

New comorbidity index associated with survival after chimeric antigen receptor T-cell therapy for large B-cell lymphoma.

The cumulative impact of baseline comorbidities on outcomes of chimeric antigen receptor T-cell (CAR-T) therapy is not well established. Therefore, we developed and validated a Cellular Therapy Comorbidity Index (CT-CI) to predict outcomes following CD19-directed CAR-T therapy for large B-cell lymphoma (LBCL). Patients aged 18 or older receiving commercial CAR-T therapy for LBCL during 2017 to 2020 were selected from the Center for International Blood and Marrow Transplant Research registry. Patients were randomly assigned to training or validation cohorts. Comorbidities given weighted scores

Inferior survival in double refractory large B-cell lymphoma eligible for third-line CD19 CAR T-cell therapy.

Outcomes following CD19 chimeric antigen receptor (CAR) T-cell therapy in third-line treatment and beyond for patients with large B-cell lymphoma (LBCL) refractory to both an anthracycline during initial and platinum-based salvage therapy, referred to as double refractory (DR), are not well-described. It is also unclear if these patients may be less likely to proceed to CAR T-cell infusion. Our objectives were to assess third-line CAR T-cell survival outcomes in DR- and non-DR (NDR)-LBCL cohorts including the failure rates to proceed to cell infusion. Review of 199 patients with LBCL referred …

"B" symptoms and CBV conditioning negatively impact survival in mantle cell lymphoma patients undergoing autologous stem cell transplantation.

INTRODUCTION: Mantle cell lymphoma (MCL) is an aggressive B-cell non-Hodgkin lymphoma with limited curative options. Autologous stem cell transplantation (ASCT) is widely used as consolidation in younger, fit patients. This study aimed to evaluate long-term outcomes after ASCT and identify clinical factors influencing survival. METHODS: This retrospective study included 119 patients with MCL who underwent ASCT between 2007 and 2024 in first complete or partial remission. Patient characteristics, outcomes, and transplant-related complications were assessed. RESULTS: At 3 and 5 years, overall

Impact of regulatory T cell therapy on immune cell composition and fetal survival rate in abortion prone mice.

CONTEXT: Implantation of fertilised eggs and survival of a semi-allogenic embryo rely on the interactions between the cells and molecules preparing the uterus. We investigated the effect of regulatory T cell (Treg) therapy on the mechanism of local immune tolerance of mice prone to spontaneous abortion. METHODS: Naive T cells were stimulated in vitro with 17β-oestradiol (E2), progesterone (P4) and TGF-β1 for 96h to generate induced Tregs (iTreg). The iTregs were injected into DBA/2-mated pregnant CBA/J female mice (abortion prone model). On day 14 of pregnancy, mice were killed and decidual a

A new survival model based on ferroptosis-related genes for prognostic prediction in clear cell renal cell carcinoma.

In this study, we analyzed the clinical significance of ferroptosis-related genes (FRGs) in 32 cancer types in the GSCA database. We detected a 2-82% mutation rate among 36 FRGs. In clear cell renal cell carcinoma (ccRCC; n=539) tissues from the The Cancer Genome Atlas database, 30 of 36 FRGs were differentially expressed (up- or down-regulated) compared to normal kidney tissues (n=72). Consensus clustering analysis identified two clusters of FRGs based on similar co-expression in ccRCC tissues. We then used LASSO regression analysis to build a new survival model based on five risk-related FRG

Kinetics of immune cell reconstitution predict survival in allogeneic bone marrow and G-CSF-mobilized stem cell transplantation.

The clinical utility of monitoring immune reconstitution after allotransplant was evaluated using data from Blood and Marrow Transplant Clinical Trials Network BMT CTN 0201 (NCT00075816), a multicenter randomized study of unrelated donor bone marrow (BM) vs granulocyte colony-stimulating factor (G-CSF)-mobilized blood stem cell (G-PB) grafts. Among 410 patients with posttransplant flow cytometry measurements of immune cell subsets, recipients of G-PB grafts had faster T-cell reconstitution than BM recipients, including more naive CD4+ T cells and T-cell receptor excision circle-positive CD4+ a

Low T-Cell Responses to Mitogen Stimulation Predicts Poor Survival in Recipients of Allogeneic Hematopoietic Stem Cell Transplantation.

BACKGROUND: Successful engraftment and reconstitution of the innate and adaptive immune system are associated with improved outcomes in recipients of allogeneic hematopoietic stem cell transplantation (HSCT). A clinically meaningful and simple biomarker of immunosuppression could potentially assist clinicians in their decision-making. We aimed to determine the relationship between T-cell production of interferon gamma (IFN-γ) in response to phytohemagglutinin (PHA) to clinical outcomes in HSCT recipients. METHODS: A prospective observational multicenter study of 73 adult allogeneic HSCT recip

Consolidative therapy with stem cell transplantation improves survival of patients with mantle cell lymphoma after any induction regimen.

Intensive induction regimen followed by high-dose chemotherapy and autologous stem cell transplantation (auto-SCT) is frequently used to improve outcomes in patients with mantle-cell lymphoma. The comparative impact of conventional vs intensive induction regimen before transplantation is unknown. Forty-eight patients with mantle-cell lymphoma receiving SCT at our institution between January 2000 and December 2010 were included in this study. At the time of initial presentation, 43 (89.5%) had stage IV disease and 18 (37.5%) received more than one chemotherapy regimen before transplantation. Fo

Phospho-proteomic analysis of mantle cell lymphoma cells suggests a pro-survival role of B-cell receptor signaling.

BACKGROUND: Mantle cell lymphoma (MCL) is currently an incurable entity, and new therapeutic approaches are needed. We have applied a high-throughput phospho-proteomic technique to MCL cell lines to identify activated pathways and we have then validated our data in both cell lines and tumor tissues. METHODS: PhosphoScan analysis was performed on MCL cell lines. Results were validated by flow cytometry and western blotting. Functional validation was performed by blocking the most active pathway in MCL cell lines. RESULTS: PhosphoScan identified more than 300 tyrosine-phosporylated proteins, a
