Hydrogen sulfide and oxygen sensing in the cardiovascular system.

Vertebrate cardiorespiratory homeostasis is inextricably dependent upon specialized cells that provide feedback on oxygen status in the tissues, blood, and on occasion, environment. These "oxygen sensing" cells include chemoreceptors and oxygen-sensitive chromaffin cells that initiate cardiorespiratory reflexes, vascular smooth muscle cells that adjust perfusion to metabolism or ventilation, and other cells that condition themselves in response to episodic hypoxia. Identification of how these cells sense oxygen and transduce this into the appropriate physiological response has enormous clinica…

Characterization of an animal model of aggressive metastatic pheochromocytoma linked to a specific gene signature.

Pheochromocytomas are chromaffin cell-derived neuroendocrine tumors. There is presently no cure for metastatic pheochromocytoma and no reliable way to distinguish malignant from benign tumors before the development of metastases. In order to successfully manage pheochromocytoma, it is necessary to better understand the biological determinants of tumor behavior. For this purpose, we have recently established a mouse model of metastatic pheochromocytoma using tail vein injection of mouse pheochromocytoma (MPC) cells. We optimized this model modifying the number of cells injected, length of tryps

Epigenetic Regulation of Carotid Body Oxygen Sensing: Clinical Implications.

Recurrent apnea with intermittent hypoxia (IH) is a major clinical problem in infants born preterm. Recent epidemiological studies showed that adults who were born preterm exhibit increased incidence of sleep-disordered breathing and hypertension. Thus, apnea of prematurity predisposes individuals to autonomic dysfunction in adulthood. Experimental studies showed that adult rats exposed to IH as neonates exhibit augmented carotid body and adrenal chromaffin cells (AMC) response to hypoxia and irregular breathing with apneas and hypertension. The enhanced hypoxic sensitivity of the carotid body…

Effect of scorpion toxin on the enterochromaffin-like cells in normal and Trypanosoma cruzi-infected rats: a morphological study.

Intravenous injection of scorpion toxin (Tityus serrulatus) in normal and Trypanosoma cruzi infected rats did not cause ultrastructural morphologic changes on enterochromaffin-like (ECL) cells of the stomach, although it induced a significant increase of the gastric secretion. Our data seem to indicate that gastric ECL cells structure is not affected by stimulation with scorpion toxin or by acute infection with T. cruzi in the rat.

[Changes in the dimensions of cells and their nuclei in the adrenal medullary substance of white rats during acute overheating].

It has been shown for the first time that the size of adrenocytes and of their nuclei does not change in the adrenal medulla on the state of motor excitation, and decreases at the heat stroke under conditions of acute overheating. According to Bennighoff (1950), the decrease of nuclear volume suggests the weakness of the adrenocyte secretory activity. But in reality the specific activity of adrenocytes may increase at hyperthermy, which contradicts to the hypothesis in question. Such a contradiction may be explained by the fact of adrenocyte dehydration under the influence of the external heat

Peripheral chemoreceptors: postnatal development and cytochemical findings in Sudden Infant Death Syndrome.

The aim of the present study is to give a review of the postnatal development of peripheral chemoreceptors - carotid body, paraganglia, and pulmonary neuroendocrine cells (PNEC) - with implications in Sudden Infant Death Syndrome (SIDS). In the postnatal period, the hypoxic chemosensitivity of the carotid body gradually develops. Changes include proliferation of type I and II cells, increased numbers of dense core vesicles and K+ channels, and modifications of neurotransmitter/neuromodulator and receptor expression. Chromaffin paraganglia show increased expression of nitric oxide synthase and

Enterochromaffin cells in macrophages in ulcerative colitis and irritable colon.

Rectal biopsies from 6 patients with ulcerative colitis, 6 patients with irritable colon and 9 control patients were studied by formaldehyde-induced fluorescence after freeze-drying and Epon embedding. The mean number of enterochromaffin cells per crypt was higher in the patients with irritable colon whereas the patients with ulcerative colitis had fewer enterochromaffin cells than the controls. There was an abundance of enterochromaffin cells in 5 of the 6 cases of irritable colon. Various types of autofluorescent granules were visible in the cytoplasm of the macrophages.

Pheochromocytoma. Preoperative approach.

Pheochromocytomas are rare neuroendocrine tumors that derive from the chromaffin cells of the adrenal medulla and secrete catecholamines. The measurement of plasma or fractionated urine metanephrines is the hormonal determination of choice for the biochemical diagnosis. Once the biochemical diagnosis is confirmed, the next step is the localization study. It is recommended to request a genetic study in all patients with pheochromocytomas since 40% of cases are hereditary. Once the diagnostic study is completed, preoperative treatment with alpha blockers should be instituted at least 7-14 days b…

Update on the diagnosis of the pheochromocytoma.

Pheochromocytoma is a rare neuroendocrine tumour that develops from chromaffin cells in the adrenal medulla and is characterised by the excessive production of catecholamines and their metabolites. Diagnostic confirmation is performed by detecting elevated levels of catecholamines and/or their metabolites in plasma or 24-h urine. In the case of moderate elevations of normetanephrine, the clonidine suppression test may be useful to differentiate between endogenous hypersecretion and false positive results. Once the biochemical diagnosis is performed, the tumour localisation is carried out using…

Modulating vesicle priming reveals that vesicle immobilization is necessary but not sufficient for fusion-competence.

In neurons and neuroendocrine cells, docked vesicles need to undergo priming to become fusion competent. Priming is a multi-step process that was shown to be associated with vesicle immobilization. However, it is not known whether vesicle immobilization is sufficient to acquire complete fusion competence. To extend our understanding of the physical manifestation of vesicle priming, we took advantage of tomosyn, a SNARE-related protein that specifically inhibits vesicle priming, and measured its effect on vesicle dynamics in live chromaffin cells using total internal reflection fluorescence mic…

[Melanosis of the large intestine].

Studies of 100 appendices and 20 snips of the large intestine in cancer (17) and in Hirschprung's disease (3) were carried out, which showed that enterochromaffin cells containing greyish-brown pigment in cytoplasm, constituted the permanent constituent of the epithelial linings of these organs. A possibility of phagocytosis of this pigment by macrophages of the connective layer of the mucous membrane is described. Basing on the analysis of literature data and her own findings the author comes to the conclusion that the phagocytoused enterochromaffin pigment in macrophages of the mucous membra…

A Case of MEN 2A Syndrome With Bilateral Recurrent Pheochromocytoma: A Case Report and Literature Review.

Pheochromocytoma is a neuroendocrine tumor arising from chromaffin cells and producing catecholamines. Due to the excessive release of catecholamines, these tumors can present with paroxysmal episodes of high blood pressure, headache, diaphoresis, and palpitations. We present the case of a 23-year-old female patient who presented to the clinic with a history of headache, palpitations, and vomiting for two months. She was admitted for further investigations and management. Laboratory studies revealed elevated urinary and plasma catecholamine levels. Further work-up revealed bilateral adrenal ma…

New Insights on the Genetics of Pheochromocytoma and Paraganglioma and Its Clinical Implications.

Pheochromocytomas (PHEOs) and paragangliomas (PGLs) are rare neuroendocrine tumors that arise from chromaffin cells. PHEOs arise from the adrenal medulla, whereas PGLs arise from the neural crest localized outside the adrenal gland. Approximately 40% of all cases of PPGLs (pheochromocytomas/paragangliomas) are associated with germline mutations and 30-40% display somatic driver mutations. The mutations associated with PPGLs can be classified into three groups. The pseudohypoxic group or cluster I includes the following genes: SDHA, SDHB, SDHC, SDHD, SDHAF2, FH, VHL, IDH1/2, MHD2, EGLN1/2 and H…

Fast myocardial recovery ensured by the combined use of V-A ECMO and IMPELLA CP in cardiogenic shock related to a pheochromocytoma crisis.

BACKGROUND: Pheochromocytoma is a rare catecholamine-secreting tumor derived from chromaffin cells in the adrenal glands. An excessive stimulation of cardiac myocytes, when pheochromocytoma 'crisis' occurs, lead to myocardial damage with cardiogenic shock. AIM OF THE STUDY: We present the case of a A 28-year old female patient admitted with signs of severe cardiogenic shock. She was successfully supported with extracorporeal membrane oxygenation (ECMO) combined with IMPELLA CP heart pump (Abiomed Danvers, MA), for left ventricular unloading. Mechanical circulatory support (MCS) was used to fa…

Postpartum Diagnosis of Cardiac Paraganglioma: A Case Report.

BACKGROUND: Extra-adrenal pheochromocytomas, or paragangliomas, originate from neural crest chromaffin cells and can be found anywhere along the sympathetic chain from head to toe. CASE REPORT: A 34-year-old female presented 4 days postpartum with episodes of palpitations, hypertension, and shortness of breath. Two episodes in the emergency department confirmed hypertension and supraventricular tachycardia (SVT). A mediastinal mass was noted during workup for pulmonary embolus and was subsequently diagnosed as a cardiac paraganglioma. Our patient underwent surgical resection and was doing wel…

[Malignant pheochromocytoma. A case].

A case of malignant phaeochromocytoma is reported. The primary tumour was removed in 1970. Metastases were diagnosed in 1979 and management of the condition included antihypertensive treatment with alphamethylparatyrosine, antitumoral chemotherapy and surgical reduction of secreting tissue. In these rare tumours malignancy can only be confirmed by metastases, i.e. presence of chromaffin cells in loci where they are not usually found. Histology is of little value. High levels of catecholamine precursors or their metabolites in the urine may be a sign of malignancy, but some asymptomatic tumours…
