Tunable molecular packing modes via H- or J-aggregates in the supramolecular helical nanostructures from an achiral C3 symmetric molecule.

The self-assembly of a C3-symmetric molecule benzene-1,3,5-tricarboxylate substituted with methyl cinnamate (BTECM) has been investigated by a reprecipitation method in H2O and cetyltrimethylammonium bromide (CTAB) aqueous solution, respectively. The nanostructures and characteristics of the assemblies were monitored by UV-Vis spectroscopy, fluorescence (FL) spectroscopy, circular dichroism (CD) spectroscopy, scanning electron microscopy (SEM) and transmission electron microscopy (TEM). It was found that helical nanostructures were successfully assembled from the achiral C3 molecule BTECM. Mor

Effects of novel triple-stage antimalarial ionic liquids on lipid membrane models.

Primaquine-based ionic liquids, obtained by acid-base reaction between parent primaquine and cinnamic acids, were recently found as triple-stage antimalarial hits. These ionic compounds displayed significant activity against both liver- and blood-stage Plasmodium parasites, as well as against stage V P. falciparum parasites. Remarkably, blood-stage activity of the ionic liquids against both chloroquine-sensitive (3D7) and resistant (Dd2) P. falciparum strains was clearly superior to those of the respective covalent (amide) analogues and of parent primaquine. Having hypothesized that such behav

Dietary fibers and associated phytochemicals in cereals.

Epidemiological studies have linked whole-grain (WG) cereal consumption to a reduced risk of developing several chronic diseases-coronary heart disease, arteriosclerosis, type-2 diabetes, and some form of cancers. The underlying physiological mechanisms behind the protective effects of WG are unclear, but can most likely be assigned to a concerted action of dietary fiber (DF) and a wide variety of phytochemicals. Physiologically, it is important that soluble nonstarch polysaccharides contribute to higher viscosity in the small intestine as this may influence rate and extent of digestion and ab

Efflux pump gene expression in Erwinia chrysanthemi is induced by exposure to phenolic acids.

Salicylic acid (SA) is an important signaling molecule in local and systemic plant resistance. Following infection by microbial pathogens and the initial oxidative burst in plants, SA accumulation functions in the amplification of defense gene expression. Production of pathogenesis-related proteins and toxic antimicrobial chemicals serves to protect the plant from infection. Successful microbial pathogens utilize a variety of mechanisms to rid themselves of toxic antimicrobial compounds. Important among these mechanisms are multidrug-resistance pumps that bring about the active efflux of toxic

The cell membranes undergo biochemical and biophysical changes in response to hydroxycinnamic acid derivatives: ferulic acid and caffeic acid.

Ferulic acid (FA) and caffeic acid (CA), due to their strong antioxidant properties, are widely used both in dietary supplements and as ingredients in cosmetics. However, despite the undoubtedly beneficial properties of these molecules, they can also exhibit toxic effects. This study aimed to investigate which concentrations of the compounds are toxic for different types of cells and which are beneficial. Detailed analyses were performed on human cells (U937, HL-60, SK-N-SH) and mouse cells (B16-F0), as well as on cell membrane models created using the Langmuir technique. Based on biochemical

Xanthomonas campestris Employs the RND Family Pump HepABCD for Phenolic Acid Efflux and Enhancing Viability and Antioxidant Activity.

Xanthomonas campestris pv. campestris (Xcc) is the causal pathogen of black rot in cruciferous plants. Upon infection, Xcc triggers the accumulation of some phenolic acids in the host plants. The mechanism by which Xcc copes with these defense compounds remains unclear. Here, we revealed that Xcc exports benzoic acid, cinnamic acid, and their monohydroxylated derivatives through the resistance-nodulation-division family efflux pump HepABCD. This efflux system influenced glutathione and catalase levels in the wild-type strain XC1, affecting cell viability and modulating ROS levels. We identifie

Synthesis of Melatonin Derivatives and the Neuroprotective Effects on Parkinson's Disease Models of Caenorhabditis elegans.

Melatonin (MT) is a hormone with antioxidant activity secreted by the pineal gland in the human brain, which is highly efficient in scavenging free radicals and plays an important role in the neuro-immuno-endocrine system. Emerging evidence showed that MT supplementation was a potential therapeutic strategy for Parkinson's disease (PD), which inhibits pathways associated with oxidative stress in PD. In this study, we reported a C7-selective olefination of melatonin under rhodium catalysis with the aid of PIII-directing groups and synthesized 10 new melatonin-C7-cinnamic acid derivatives (6a-6j

Contemporaneous 3D characterization of acute and chronic myocardial I/R injury and response.

Cardioprotection by salvage of the infarct-affected myocardium is an unmet yet highly desired therapeutic goal. To develop new dedicated therapies, experimental myocardial ischemia/reperfusion (I/R) injury would require methods to simultaneously characterize extent and localization of the damage and the ensuing inflammatory responses in whole hearts over time. Here we present a three-dimensional (3D), simultaneous quantitative investigation of key I/R injury-components by combining bleaching-augmented solvent-based non-toxic clearing (BALANCE) using ethyl cinnamate (ECi) with light sheet fluor

Biotransformation of Plant-Derived Phenolic Acids.

Phenolic acids are abundant biomass feedstock that can be derived from the processing of lignin or other byproducts from agro-industrial waste. Although phenolic acids such as p-hydroxybenzoic acid, p-coumaric acid, caffeic acid, vanillic acid, cinnamic acid, gallic acid, syringic acid, and ferulic acid can be used directly in various applications, their value can be significantly increased when they are further modified to high value-added compounds. This review summarizes and discusses the new advances in cell-free and whole-cell biocatalysis technologies for reactions important for conversi

Reduction of L-phenylalanine in protein hydrolysates using L-phenylalanine ammonia-lyase from Rhodosporidium toruloides.

L-Phenylalanine ammonia-lyase (PAL, EC 4.3.1.25) from Rhodosporidium toruloides was utilized to remove L-phenylalanine (L-Phe) from different commercial protein hydrolysates. A casein acid hydrolysate (CAH, L-Phe ~2.28 %) was employed as a model substrate. t-Cinnamic acid resulting from deamination of L-Phe was extracted, analyzed at λ = 290 nm, and used for PAL activity determination. Optimum reaction conditions, optimized using successive Doehlert design, were 35 mg mL(-1) of CAH and 800 mU mL(-1) of PAL, while temperature and pH were 42 °C and 8.7, respectively. Reaction kinetics of PAL wit
