Testicular ACE regulates sperm metabolism and fertilization through the transcription factor PPARγ.

Testis angiotensin-converting enzyme (tACE) plays a critical role in male fertility, but the mechanism is unknown. By using ACE C-domain KO (CKO) mice which lack tACE activity, we found that ATP in CKO sperm was 9.4-fold lower than WT sperm. Similarly, an ACE inhibitor (ACEi) reduced ATP production in mouse sperm by 72%. Metabolic profiling showed that tACE inactivation severely affects oxidative metabolism with decreases in several Krebs cycle intermediates including citric acid, cis-aconitic acid, NAD, α-ketoglutaric acid, succinate, and L-malic acid. We found that sperms lacking tACE activi

Use of matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) to visualize and support interpretation of toxic effects of 4-hydroxyphenylpyruvate dioxygenase inhibitors in rat tissues.

4-Hydroxyphenylpyruvate dioxygenase inhibitors (HPPDi) are mainly used as herbicides and for therapeutic use in genetic diseases of tyrosine … s and the thyroid gland. Metabolomic profiling showed that other amino acids and metabolites of the citric acid cycle were influenced by the treatment. A metabolic adaptation was observed in the liver and kidney 28 days after the treatment, but not in other tissues analyzed. MSI of the thyroid gland seems to reveal an uneven accumulation of tyrosine in the tissue o …

Comparative analysis of myocardial enzyme activities of the energy-supplying metabolism in patients with dilative cardiomyopathies and valve diseases.

We determined representative enzyme activities of glycogenolysis (glycogen phosphorylase) glycolysis (d-glyceraldehyde-3-phosphate dehydrogenase, GAPDH), beta oxidation of free fatty acids (1-3-hydroxyacyl CoA dehydrogenase, HADH), citric acid cycle (citrate synthase, CS), lactate fermentation (lactate dehydrogenase LDH), and creatine phosphate metabolism (creatine kinase, CK) in left ventricular samples of 36 patients to investigate if the metabolic capacities of the energy-supplying pathways are differently affected in different heart diseases. There were 17 patients with mitral valve diseas…

The effect of chronic ethanol consumption on enzyme activities of the energy-supplying metabolism and the alcohol-aldehyde oxidizing system in rat hearts.

We investigated in rat hearts if chronic alcohol consumption causes an enzymatic adaption of the energy-supplying metabolism and/or of the alcohol-aldehyde metabolizing system. 16 rats were pair-fed with a liquid diet for 10 weeks. Ethanol was added to this diet to amount for 35% of calories in eight rats and was isocalorically replaced by saccharose in the control group. Selected enzyme activities of the glycolysis, the glycogenolysis, the beta-oxidation of fatty acids, the citric acid cycle and the alcohol-aldehyde oxidizing system were determined in the supernatants of the homogenized heart…

Immune suppression by human thymus-derived effector Tregs relies on glucose/lactate-fueled fatty acid synthesis.

Regulatory T cells (Tregs) suppress pro-inflammatory conventional T cell (Tconv) responses. As lipids impact cell signaling and function, we compare the lipid composition of CD4+ thymus-derived (t)Tregs and Tconvs. Lipidomics reveal constitutive enrichment of neutral lipids in Tconvs and phospholipids in tTregs. TNFR2-co-stimulated effector tTregs and Tconvs are both glycolytic, but only in tTregs are glycolysis and the tricarboxylic acid (TCA) cycle linked to a boost in fatty acid (FA) synthesis (FAS), supported by relevant gene expression. FA chains in tTregs are longer and more unsaturated

In vitro nitazoxanide exposure affects energetic metabolism of Taenia crassiceps.

Nitazoxanide (NTZ) is a broad-spectrum drug used in intestinal infections, but still poorly explored in the treatment of parasitic tissular infections. This study aimed to evaluate the in vitro responses of the energetic metabolism of T. crassiceps cysticerci induced by NTZ. The organic acids of the tricarboxylic acid cycle, products derived from fatty acids oxidation and protein catabolism were analyzed. These acids were quantified after 24 h of in vitro exposure to different NTZ concentrations. A positive control group was performed with albendazole sulfoxide (ABZSO). The significant alterat

Iron deficiency and toxicity trigger divergent metabolic responses and adaptive plasticity in Ulmus pumila: Insights from integrated transcriptomic and metabolomic analyses.

Iron homeostasis is critical for plant growth; however, the mechanisms underlying responses to iron deficiency and toxicity remain poorly understood. We investigated the adaptive strategies of Ulmus pumila, focusing on leaf physiological, transcriptomic, and metabolomic responses to iron stresses. Both iron deficiency and toxicity impaired chlorophyll biosynthesis, PS II efficiency, and chloroplast ultrastructure, resulting in reduced photosynthetic capacity and etiolation/wilting phenotypes. Iron deficiency reduced antioxidant enzyme activity and ROS levels, while iron toxicity activated the

Regulation of PDH via cysteine oxidation on DLAT.

Pyruvate dehydrogenase (PDH) complex is a key regulator of metabolism, controlling flux from glycolysis into the tricarboxylic acid (TCA) cycle. PDH is often dysregulated in metabolic diseases and cancer. We discover here that in addition to its well characterized regulation by phosphorylation, PDH activity is also regulated via inhibitory oxidation of three cysteines of the DLAT subunit - Cys488, Cys586 and Cys618. Oxidation of these cysteines increases under oxidative stress, and is highly prevalent in mouse and human tissues. This mechanism enables cells to efficiently fight oxidative stres

Berberine Attenuates Cadmium-Induced Nephrotoxicity by Suppressing LDHA-Mediated Glycolytic Reprogramming and Restoring Mitochondrial TCA Cycle Metabolism.

Cadmium (Cd) is an environmental nephrotoxicant that preferentially accumulates in the kidney and disrupts redox and energy metabolism. However, the protective effect of berberine (Ber) against Cd-induced nephrotoxicity remains insufficiently characterized. In the present study, male C57BL/6 mice were orally exposed to CdSO4 (30 mg/kg body weight/day) for 30 days in the absence or presence of berberine (25 or 100 mg/kg/day). Renal function, histopathology, oxidative stress parameters, LC-MS/MS-based metabolomic profiling, gene and protein expression, and in silico ligand-target interactions we

Mitochondrial Energy Metabolism in Cancer, Immunoregulation and Antitumour Response.

Mitochondria are the cells' powerhouse, a dogma that has guided research for decades. While this bioenergetic view is accurate, it fails to capture the extra-metabolic signalling functions of mitochondria, as well as the complexity of mitochondrial homeostasis and its contribution to disease pathogenesis, particularly in cancer. In this chapter, we review the multifaceted role of mitochondria in cancer and immunity, starting from the bioenergetic point of view and expanding to metabolic reprogramming in cancer, their role in signalling regulation, immune cell homeostasis and promising immunoth

Harnessing the glycolysis-TCA cycle axis to boost host defense against neonatal infection.

Preterm infants are highly susceptible to infections that can lead to sepsis, yet therapies beyond antibiotics are limited. Nutrition and host energy metabolism are known as immune modulators, but how they interact to mediate newborn host infection defense remains poorly understood. Here, we identify tricarboxylic acid (TCA) cycle metabolites as key modulators of early life infection outcomes. First, in a birth cohort of 700 children, elevated plasma TCA metabolite levels were associated with reduced infection burdens and systemic inflammation. Next, in a piglet neonatal sepsis model, sustaine

Ginkgolide B drives TCA cycle to accelerate Streptococcus suis biofilm dissociation.

Streptococcus suis (S. suis) is an important zoonotic pathogen that often forms biofilm, leading to persistent clinical infections and exacerbation of antibiotic resistance. There is an urgent need to develop novel antibiofilm strategies. In this study, for the first time, lactate dehydrogenase (LDH) was targeted with the aim of reactivating the metabolic activity of bacteria embedded in biofilm using natural product molecules, thereby disrupting mature biofilm. Through virtual screening based on the TCMSP database, molecular docking, and molecular dynamics simulations, two potential molecules

Metabolic potentiation of antibiotic killing by L-arginine in drug-resistant Edwardsiella tarda.

The role of metabolic state reprogramming in modulating antibiotic susceptibility has attracted growing interest as a promising strategy to combat antimicrobial resistance. Our study revealed that L-arginine potentiates chloramphenicol's bactericidal activity by at least two orders of magnitude against multidrug-resistant Edwardsiella tarda via the coordinated modulation of three interconnected metabolic pathways: the tricarboxylic acid cycle disruption, redox homeostasis alteration, and phenylalanine metabolic suppression. Mechanistically, L-arginine-mediated tricarboxylic acid cycle inhibiti

Pneumococcal H₂O₂ reshapes mitochondrial function and reprograms host cell metabolism.

Streptococcus pneumoniae (Spn), a primary cause of pneumonia, induces acute lung parenchymal damage through a unique metabolic pathway generating hydrogen peroxide (H₂O₂) as a byproduct. This study demonstrates that Spn-derived H₂O₂, primarily produced by pyruvate oxidase (SpxB), inhibits key tricarboxylic acid (TCA) cycle enzymes (aconitase, glutamate dehydrogenase, and α-ketoglutarate dehydrogenase) in lung epithelial cells, leading to citrate accumulation and diminished NADH production for oxidative phosphorylation. RNA sequencing reveals SpxB-dependent upregulation of glycolytic genes (HIF

13C-Metabolic flux analysis detected a hyperoxemia-induced reduction of tricarboxylic acid cycle metabolism in granulocytes during two models of porcine acute subdural hematoma and hemorrhagic shock.

INTRODUCTION: Supplementation with increased inspired oxygen fractions has been suggested to alleviate the harmful effects of tissue hypoxia during hemorrhagic shock (HS) and traumatic brain injury. However, the utility of therapeutic hyperoxia in critical care is disputed to this day as controversial evidence is available regarding its efficacy. Furthermore, in contrast to its hypoxic counterpart, the effect of hyperoxia on the metabolism of circulating immune cells remains ambiguous. Both stimulating and detrimental effects are possible; the former by providing necessary oxygen supply, the l

Nitric Oxide Circumvents Virus-Mediated Metabolic Regulation during Human Cytomegalovirus Infection.

Nitric oxide is a versatile and critical effector molecule that can modulate many cellular functions. Although recognized as a regulator of infections, the inhibitory mechanism of nitric oxide against human cytomegalovirus (HCMV) replication remains elusive. We demonstrate that nitric oxide attenuates viral replication by interfering with HCMV-mediated modulation of several cellular processes. Nitric oxide exposure reduced HCMV genome synthesis and infectious viral progeny with cell-type-dependent differences observed. Mitochondrial respiration was severely reduced in both uninfected and HCMV-

Distinctive Krebs cycle remodeling in iPSC-derived neural and mesenchymal stem cells.

Mitochondria play a vital role in proliferation and differentiation and their remodeling in the course of differentiation is related to the variable energy and metabolic needs of the cell. In this work, we show a distinctive mitochondrial remodeling in human induced pluripotent stem cells differentiated into neural or mesenchymal progenitors. While leading to upregulation of the citrate synthase-α-ketoglutarate dehydrogenase segment of the Krebs cycle and increased respiratory chain activities and respiration in the mesenchymal stem cells, the remodeling in the neural stem cells resulted in do

Changes in pancreatic islets in aging Wistar and Zucker rats: a histochemical and ultrastructural morphometric study.

Morphometrical and cytochemical techniques have been applied to characterize the islets of Langerhans tissue in lean and obese Zucker fa/fa and Wistar rats. The changes in cytologic composition correlated with levels of serum glucose and lipids in obese rats and progressed significantly with increasing body weight. Histochemical and enzyme abnormalities observed in Zucker fa/fa and Wistar rats reflect the degenerative and reactive processes in the pancreas: a decrease in Krebs cycle and pentose pathway and increased lysosomal acid phosphatase reflect the degeneration of the islets.
