Seven patients with stage IV mycosis fungoides [TNM classification] have been treated with combination chemotherapy consisting of cyclophosphamide, vincristine, methotrexate, and prednisone [COMP]. A complete response rate of 57% was produced with an overall response of 100% in patients having limited prior steroid and/or chemotherapy. Diffuse histiocytic lymphoma developed in a 48-year-old man with Wegener's granulomatosis after nine years of therapy with cyclophosphamide. He died despite aggressive surgical and medical therapy for the lymphoma. This may be the first report of diffuse histiocytic lymphoma following treatment of Wegener's granulomatosis with cyclophosphamide. Recommendations for the approach towards extended therapy of smoldering Wegener's granulomatosis are discussed.

The compensatory effect of a bacterial lysate, Broncho-Vaxom (BV) on the immunosuppressive action of cyclophosphamide (CY) was investigated. The present studies assessed the relationship between murine splenic B-lymphocyte colony-forming cells and the number of mature B lymphocytes in the spleen following administration of either cyclophosphamide or azathioprine. An 11-year-old white boy had Wegener's granulomatosis, a rare condition in the pediatric age group. The clinical course, pathological findings, and mode of treatment are outlined. The disease is in remission on a regimen of cyclophosphamide therapy as judged by both clinical and pathological criteria. This syndrome with protein manifestations should be considered in children with symptoms of repeated upper respiratory tract infections along with pulmonary and renal involvement. There were 33 patients with clinically relapsing advanced prostatic carcinoma (stages C and D) treated by 2 different combinations of chemotherapy. The 13 patients in group 1 received 5-fluorouracil and cyclophosphamide and 20 patients in group 2 received adriamycin and cyclophosphamide. The over-all response was 69.3 per cent in group 1 and 65 per cent in group 2. Our preliminary results should encourage further evaluation of these drug combinations by other investigators under carefully controlled conditions.

Delayed drug-related mortality in rats treated with a single high dose (75 mg/kg) of cyclophosphamide complicated experiments using this drug treatment. We observed that this delayed mortality was due to dental abnormalities including broken teeth, absent teeth, extra long teeth, and/or supernumerary teeth. These dental abnormalities developed about 140 days after treatment and, if left untreated, interfered with eating. Eventually, the untreated rats starved. INTRODUCTION: Women diagnosed with cancer often undergo aggressive chemotherapy that can impair fertility and lead to long-term ovarian damage, significantly affecting their quality of life. Cyclophosphamide (CPA), a chemotherapeutic agent known for its gonadotoxic effects, has been shown to reduce ovarian follicle reserves, thereby contributing to the development of primary ovarian insufficiency in both humans and animal models.
