Novel proposed cutoff values for anatabine and anabasine in differentiating smokers from non-smokers.

OBJECTIVES: Anatabine and anabasine are two tobacco alkaloids used to differentiate between tobacco users and abstainers, including users of nicotine replacement therapy. Cutoff values (>2 ng/mL for both alkaloids) have not been revised since their implementation in 2002. These values may be too high, leading to increased likelihood of misclassification between smokers and abstainers. This results in major consequences, especially adverse outcomes of transplantation when smokers were incorrectly identified as being abstinent. This study proposes that a lower threshold for anatabine and anabasi

Detection and differentiation of sensorineural hearing loss in mice using auditory steady-state responses and transient auditory brainstem responses.

Sensorineural hearing loss (SNHL) comprises hearing disorders with diverse pathologies of the inner ear and the auditory nerve. To date, an unambiguous phenotypical characterization of the specific pathologies in an affected individual remains impossible. Here, we evaluated the use of scalp-recorded auditory steady-state responses (ASSR) and transient auditory brainstem responses (ABR) for differentiating the disease mechanisms underlying sensorineural hearing loss in well-characterized mouse models. We first characterized the ASSR evoked by sinusoidally amplitude-modulated tones in wild-type

Intrapharyngeal distribution of gastric acid refluxate.

OBJECTIVES/HYPOTHESIS: Determination of intrapharyngeal distribution of gastric acid refluxate is needed to better understand the pathogenesis of reflux-attributed aerodigestive tract disorders. The aims of the study were to determine intrapharyngeal distribution of gastric acid refluxate and to determine whether this distribution can better differentiate patients from control subjects compared with data obtained from a single hypopharyngeal site. METHODS: We prospectively studied 10 healthy volunteers, 12 patients with reflux laryngitis, and 15 patients with vasomotor rhinitis using a concur

Chest Radiographs for Distinguishing ADA-SCID from Other Forms of SCID.

PURPOSE: Early differentiation of adenosine deaminase deficient severe combined immunodeficiency (ADA-SCID) from other forms of SCID may initiate appropriate treatment interventions with the aim of metabolic detoxification and improved outcome. Our hypothesis was that previously described radiological features (inferior scapular angle squaring and spurring and costochondral cupping) can differentiate ADA-SCID from other forms of SCID. METHODS: Chest radiographs at clinical presentation between 2000 and 2017 of children with ADA-SCID were retrospectively included, provided that the radiologica

MicroRNA from pancreatic duct aspirate differentiates cystic lesions of the pancreas.

INTRODUCTION: Prognostication for cystic neoplasms of the pancreas continues to evolve. Beyond simple size and cystic fluid CEA determination, microRNA (miRNA) detection holds great promise as molecular diagnostics for cancer risk. In this study, we sought to identify miRNAs that could predict malignant potential of pancreatic cystic lesions. METHODS: RNA was harvested from the pancreatic duct aspirate of 72 cystic neoplasms of the pancreas. Samples with adequate RNA concentration (≥ 3 ng/μL) were selected for qRTPCR profiling using assays to 379 of the most common miRNAs. miRNA profiles were

Differential Expression Analysis of Key Inflammatory Mediators in Irreversible Pulpitis for Diagnostic Biomarkers.

INTRODUCTION AND AIMS: The diagnosis of pulpal inflammatory conditions poses significant challenges in clinical endodontics, requiring precise differentiation between reversible and irreversible states. This study investigated the differential expression patterns of 10 inflammatory genes: LCP2, PTPRC, CXCL8, TNFα, IL6, CCL2, MMP9, NOD2, ICAM1, and TLR8 in irreversible pulpitis as potential molecular diagnostic biomarkers. METHODS: In this prospective cross-sectional analytical investigation, pulp tissue samples were collected from 30 subjects with irreversible pulpitis and 10 control subjects

[Untargeted metabolomics-based analysis of metabolites influencing the efficacy of liver progenitor cell transplantation and their underlying mechanisms].

To explore the mechanisms underlying the therapeutic efficacy of cell transplantation in mouse liver, liquid chromatography-mass spectrometry (LC-MS)-based untargeted metabolomics was employed to screen the differential metabolites in the liver tissue of mice undergoing cell transplantation. We then explored the biological significance of differential metabolites through pathway analysis, correlation analysis, and cluster analysis, aiming to decipher the potential mechanisms affecting the therapeutic outcomes of cell transplantation. The augmenter of liver regeneration (ALR) has been reported

Spatially resolved proteomics provides insights into the normal-dysplasia-tumor continuum in oral squamous cell carcinoma.

BACKGROUND: Oral epithelial dysplasia is a recognized precursor lesion for oral squamous cell carcinoma (OSCC), yet the proteomic changes underlying the progression from normal epithelium to dysplasia and ultimately to OSCC remain incompletely characterized. This gap is partially due to the high interpatient heterogeneity of OSCC, the frequent reliance on cross-patient comparisons, and the omission of dysplasia as a critical intermediate stage. METHODS: A histology-guided, spatially resolved proteomics workflow was applied to formalin-fixed paraffin-embedded tissue blocks from six patients wi

An MRI-based strategy for differentiation of frontotemporal dementia and Alzheimer's disease.

BACKGROUND: The differential diagnosis of frontotemporal dementia (FTD) and Alzheimer's disease (AD) is difficult due to the overlaps of clinical symptoms. Structural magnetic resonance imaging (sMRI) presents distinct brain atrophy and potentially helps in their differentiation. In this study, we aim at deriving a novel integrated index by leveraging the volumetric measures in brain regions with significant difference between AD and FTD and developing an MRI-based strategy for the differentiation of FTD and AD. METHODS: In this study, the data were acquired from three different databases, in

Machine vision application in animal trajectory tracking.

This article was motivated by the doctors' demand to make a technical support in pathologies of gastrointestinal tract research [10], which would be based on machine vision tools. Proposed solution should be less expensive alternative to already existing RF (radio frequency) methods. The objective of whole experiment was to evaluate the amount of animal motion dependent on degree of pathology (gastric ulcer). In the theoretical part of the article, several methods of animal trajectory tracking are presented: two differential methods based on background subtraction, the thresholding methods bas

Sweet and bitter taste discrimination in primates: scaling effects across species.

The adaptive nature of taste discrimination has been questioned on the basis that marked differences in taste thresholds for soluble sugars across mammals may not correspond to distinct dietary tendencies and further imply inaccurate prediction of the energetic value of food. Appropriate models of the evolution of taste, however, require not only consideration of adaptations to diet but also recognition of phylogenetic and allometric effects. Here, we analyse these factors in primates exhibiting various diets and covering a wide range of body weights. Taste thresholds for sucrose and fructose

T cell responses are governed by avidity and co-stimulatory thresholds.

We analyzed the avidity and CD28-mediated co-stimulatory requirements for the activation of T cells in vivo and in vitro. The strength of the T cell/antigen-presenting cell interaction was varied by using altered peptide ligands for stimulation. Co-stimulatory requirements were studied using T cells from CD28-deficient mice. The results indicate that T cell activation is not an all-or-nothing event, but occurs in distinct steps. For each step, a certain avidity, co-stimulatory threshold or both, must be met. Depending upon the strength of the interaction between the T cell receptor and the maj

Rod-cone interactions and analysis of retinal disease.

Cone flicker threshold rises as the rods dark adapt, though the cone threshold to continuous light remains constant. The rise is normally about 1 log unit, but in certain patients who complain of night blindness it may be as great as 2.5 log units. In these persons the kinetics of the rod-cone interaction are those of the recovery of rod sensitivity. The rods impose a low-pass filter on the cones. This effect is absent in congenital nyctalopia and X-linked retinoschisis. We suggest that cone flicker is maintained through a feedback system involving horizontal cells, and when the rod dark curre

[The disinhibitory influence of the activating system when artificially excited from different points in the brain].

EEG activation can be produced by electrical stimulation of some cortical points with the same threshold current strength as by the midbrain RF and thalamic CM stimulation. Near-threshold stimulation of all these points acting simultaneously with inhibitory conditioned signals does not disturb the effector inhibition but displays an EEG difference between negative signals: the fine differentiation sound evokes considerable EEG desynchronization, while the rough one does not change the background rhythms. The same stimulation combined with a positive signal which has been made ineffective by su

Liver cystic echinococcosis: a parasitic review.

Liver cystic echinococcosis (CE), known as hydatid disease, is caused by the tapeworm Echinococcus granulosus sensu lato. Humans are accidental hosts in this zoonotic disease process, and hepatic infection accounts for over two-thirds of all cases. Since signs and symptoms are mainly non-specific, especially in early disease, clinicians should have a low threshold to include CE as a differential diagnosis in patients with positive serology and suggestive radiological findings, especially in endemic regions. The standard management for liver CE depends on the patient's symptoms, the radiologica

Cranial giant cell arteritis mimickers: A masquerade to unveil.

Giant cell arteritis (GCA) is a large-vessel vasculitis that affects cranial and extra-cranial arteries. Extra-cranial GCA presents mainly with non-specific symptoms and the differential diagnosis is very broad, while the cranial form has more typical clinical picture and physicians have a lower threshold for diagnosis and treatment. Although temporal artery biopsy (TAB) has an established role, ultrasound (US) is being increasingly used as the first-line imaging modality in suspected GCA. Vasculitides (especially ANCA-associated), hematological disorders (mainly amyloidosis), neoplasms, infec

The genetic architecture of human brainstem structures and their involvement in common brain disorders.

Brainstem regions support vital bodily functions, yet their genetic architectures and involvement in common brain disorders remain understudied. Here, using imaging-genetics data from a discovery sample of 27,034 individuals, we identify 45 brainstem-associated genetic loci, including the first linked to midbrain, pons, and medulla oblongata volumes, and map them to 305 genes. In a replication sample of 7432 participants most of the loci show the same effect direction and are significant at a nominal threshold. We detect genetic overlap between brainstem volumes and eight psychiatric and neuro

Abnormal uterine bleeding.

Disorders of the menstrual cycle are common problems in ambulatory medicine. Abnormal uterine bleeding describes bleeding that is excessive or outside the normal menstrual cycle. In the premenopausal woman, the differential diagnosis is broad, and pregnancy must always be considered. Determining whether the bleeding is ovulatory or anovulatory is a central part of the evaluation, as anovulation is one of the most common causes of abnormal uterine bleeding. In patients with anovulatory bleeding, the goal of treatment is to minimize blood loss and prevent complications from chronic unopposed est
