The study investigated the applicability of three different analytical approaches for assessing post-treatment changes in liver enzyme activity following anthelmintic therapy over a 21-day period in cattle naturally infected with fasciolosis. A total of 48 animals were allocated into four groups of 12 cows each: one untreated control group and three treatment groups. Treated cattle received the following anthelmintic combinations: ivermectin plus clorsulon (IVM+CLOR), levamisole plus oxyclozanide (LEV+OXY), and albendazole (ABZ). Biochemical assessments were performed using standardized kineti

Accurate spatiotemporal single-cell quantification of intracellular enzyme activity is critical for understanding cellular heterogeneity and disease screening. However, current methods relying on single-signal modalities lack precision, and spatiotemporal multimodal strategies remain unexplored. Here, we develop a spatiotemporal sequential fluorescence-electrochemistry bimodal platform for both spatial mapping and precise quantitation of asparaginyl endopeptidase (AEP) activity in living single cells. A dual-aggregation-induced emission (AIE) probe, pyTPE-CBT, is delivered into single cells vi

Bacterial communities are vital to northern peatlands' carbon and nutrient cycling, one of the Earth's largest terrestrial carbon stores. However, their diversity and ecological roles at broad geographic scales remain partially understood, limiting our ability to predict their response to global change. Here, we combine a trans-Holarctic survey across 171 Sphagnum-dominated peatlands (SDPs) with a continental-scale reciprocal transplantation experiment to quantify how bacterial diversity shapes carbon and nutrient cycling across bioclimatic gradients. We show that bacterial diversity and compo

Deficiency of adenosine deaminase 2 (DADA2) is an autoinflammatory disease with diverse phenotypes. We describe the genetics, phenotypes, and treatment of n = 48 DADA2 patients from Germany, Austria, and Switzerland. We report a high incidence of hematological (83%) and immunological features (85%), a comparatively low anti-tumor necrosis factor full-response rate (58%), and a high decision probability (21%) for hematopoietic cell transplantation (HCT). We establish a correlation between genetic variant ADA2 activity and patient ADA2 activity. Remarkably, lower patient ADA2 activity is predict

To examine the effect of chronic alcohol exposure on the activity of CYP3A enzymes in human liver, we studied the metabolism of CYP3A-specific substrates 7-benzyloxyquinoline (7-BQ) and ivermectin in 23 preparations of human liver microsomes (HLM) obtained from donors with documented alcohol exposure, from non-drinkers to heavy alcoholics. All HLM samples were characterized for the composition of the cytochrome P450 pool by global proteomics. Our studies revealed a significant increase in the activities of CYP3A enzymes by alcohol exposure. This effect is not associated with CYP3A enzyme level

BACKGROUND: Inflammatory bowel disease (IBD) is often diagnosed during reproductive age, requiring adequate management during pregnancy. Physiological changes during pregnancy affect liver enzymes, complicating interpretation in a population prone to liver test abnormalities. This study aims to investigate liver enzyme trends throughout pregnancy in women with IBD.
METHODS: A retrospective cohort study was conducted in three Dutch university hospitals. Pregnant women with IBD were included; liver enzyme levels were analyzed throughout pregnancy using Bayesian modeling. The association between

As a core genetic biomolecule in ecosystems, the metabolic processes of DNA, particularly DNA replication and damage repair, are regulated by Flap endonuclease 1 (FEN1). Abnormal expression and dysfunction of FEN1 may lead to genomic instability, which can induce a variety of chromosome-associated disorders, including tumours. FEN1 has emerged as a prominent tumour marker. It is crucial to precisely assess FEN1 activity and identify its associated inhibitors for both experimental research and clinical applications. However, traditional detection methods are laborious and time-consuming. Despit

Most cancer patients diagnosed with late-stage head and neck squamous cell carcinoma are treated with chemoradiotherapy, which can lead to toxicity. One potential alternative is tumor-limited conversion of a prodrug into its cytotoxic form. We reason this could be achieved by transient and tumor-specific expression of purine nucleoside phosphorylase (PNP), an Escherichia coli enzyme that converts fludarabine into 2-fluoroadenine, a potent cytotoxic drug. To efficiently express bacterial PNP in tumors, we evaluate 44 chemically distinct lipid nanoparticles (LNPs) using species-agnostic DNA barc

INTRODUCTION: Congenital disorders of glycosylation (CDG) are biochemically characterized by abnormal carbohydrate assembly. These disorders can result from mutations in the genes encoding different enzymes, leading to a partial or complete reduction in catalytic activity. They can be inherited in an autosomal recessive, dominant, or X-linked manner. Approximately 130 congenital disorders are currently known, within a multisystemic clinical spectrum with high heterogeneity between family members and between different types of CDG. Deficiencies of the cytosolic enzyme phosphomannomutase 2 (PMM2

Evaluating changes in metabolic pathway activity is essential for studying disease mechanisms and developing new treatments, with significant benefits extending to human health. Here, we propose EMPathways2, a maximum likelihood pipeline that is based on the expectation-maximization algorithm, which is capable of evaluating enzyme expression and metabolic pathway activity level. We first estimate enzyme expression from RNA-seq data that is used for simultaneous estimation of pathway activity levels using enzyme participation levels in each pathway. We implement the novel pipeline to RNA-seq da

Naegleria fowleri is a ubiquitous free-living amoeba that causes primary amoebic meningoencephalitis. As a part of the innate immune response at the mucosal level, the proteins lactoferrin (Lf) and lysozyme (Lz) are secreted and eliminate various microorganisms. We demonstrate that N. fowleri survives the individual and combined effects of bovine milk Lf (bLf) and chicken egg Lz (cLz). Moreover, amoebic proliferation was not altered, even at 24 h of co-incubation with each protein. Trophozoites' ultrastructure was evaluated using transmission electron microscopy, and these proteins did not sig

Urease is a metalloenzyme including two Ni2+ ions, found in some plants, bacteria, fungi, microorganisms, invertebrate animals, and animal tissues. Urease acts as a significant virulence factor, mainly in catheter blockage and infective urolithiasis as well as in the pathogenesis of gastric infection. Therefore, studies on urease lead to novel synthetic inhibitors. In this review, the synthesis and antiurease activities of a collection of privileged synthetic heterocycles such as (thio)barbiturate, (thio)urea, dihydropyrimidine, and triazol derivatives were described and discussed according to

Protein-polymer conjugation provides an opportune means to adjust the local environment of proteins and enhance protein stability, performance, and solubility. Although much attention has been focused on tuning protein-polymer interactions, the properties of polymer-modified proteins may also be altered by polymer-polymer interactions. Herein, we sought to better understand the influence of polymer-polymer interactions on Candida rugosa lipase, which was modified with random co-polymers composed of sulfobetaine methacrylate (SBMA) and poly(ethylene glycol) methacrylate (PEGMA). Our findings su

AT-752 is a guanosine analogue prodrug active against dengue virus (DENV). In infected cells, it is metabolized into 2'-methyl-2'-fluoro guanosine 5'-triphosphate (AT-9010) which inhibits RNA synthesis in acting as a RNA chain terminator. Here we show that AT-9010 has several modes of action on DENV full-length NS5. AT-9010 does not inhibit the primer pppApG synthesis step significantly. However, AT-9010 targets two NS5-associated enzyme activities, the RNA 2'-O-MTase and the RNA-dependent RNA polymerase (RdRp) at its RNA elongation step. Crystal structure and RNA methyltransferase (MTase) act

Fluorescence (FL)-guided detection of cancer is one of the most promising approaches to achieve intraoperative assessment of surgical margins. Enzymes, such as aminopeptidase, carboxypeptidase, and glycosidase, whose activities are increased in cancer, have attracted great interest as imaging targets for rapid and sensitive visualization of cancerous tissues with fluorescent probes. Activatable probes, which are initially nonfluorescent but become strongly fluorescent upon rapid one-step cleavage of their substrate moiety by the target enzyme, are especially promising for practical clinical ap

Mitochondrial trifunctional protein (TFP) is a membrane-associated heterotetramer that catalyzes three of the four reactions needed to chain-shorten long-chain fatty acids inside the mitochondria. TFP is known to be heavily modified by acetyllysine and succinyllysine post-translational modifications (PTMs), many of which are targeted for reversal by the mitochondrial sirtuin deacylases SIRT3 and SIRT5. However, the functional significance of these PTMs is not clear, with some reports showing TFP gain-of-function and some showing loss-of-function upon increased acylation. Here, we mapped the kn

Human cytomegalovirus causes diseases in individuals with insufficient immunity. Cytomegaloviruses exploit the ubiquitin proteasome pathway to manipulate the proteome of infected cells. The proteasome degrades ubiquitinated proteins. The family of cullin RING ubiquitin ligases (CRL) regulates the stability of numerous important proteins. If the cullin within the CRL is modified with Nedd8 ("neddylated"), the CRL is enzymatically active, while CRLs lacking Nedd8 modifications are inactive. The Nedd8-activating enzyme (NAE) is indispensable for neddylation. By binding to NAE and inhibiting neddy

Ubiquitin-activating enzyme E1, UBA1, functions at the apex of the enzymatic ubiquitylation cascade, catalysing ubiquitin activation. UBA1 is thus of fundamental importance to the modulation of ubiquitin homeostasis and to all downstream ubiquitylation-dependent cellular processes, including proteolysis through the ubiquitin-proteasome system and selective autophagy. The proteasome-dependent and -independent functions of UBA1 contribute significantly to a range of processes crucial to neuronal health. The significance of UBA1 activity to neuronal health is clear in light of accumulating eviden
