Method for the Destruction of Endotoxin in Synthetic Spider Silk Proteins.

Although synthetic spider silk has impressive potential as a biomaterial, endotoxin contamination of the spider silk proteins is a concern, regardless of the production method. The purpose of this research was to establish a standardized method to either remove or destroy the endotoxins present in synthetic spider silk proteins, such that the endotoxin level was consistently equal to or less than 0.25 EU/mL, the FDA limit for similar implant materials. Although dry heat is generally the preferred method for endotoxin destruction, heating the silk proteins to the necessary temperatures led to c

Modulation of Secondary Structure, Bioavailability, Immunomodulation, and Tissue Repair Outcomes Using Differential Processing of Silk Biomaterials.

The ability to modulate structure, physicochemical properties, and function makes naturally derived polypeptides attractive biomaterials for human health. Silk fibroin protein, derived from silkworm cocoons, has been explored in tissue engineering, wound healing, and drug delivery. Despite these advances, the influence of silk polypeptide processing on secondary structure, bioavailability, tissue responses, and healing outcomes is poorly understood. Here, we fabricated silk fibroin-indocyanine green (ICG) dye films and modulated their properties using different processing conditions, including

Iron-Chelated Silk Microfiber/Hydrogel Composites as Injectable, Magnetically Aligned Cell Guidance Scaffolds.

In the design of biomaterial scaffolds for traumatic spinal cord injury (SCI), anisotropic composites of magnetically aligning architectures and injectable hydrogels have garnered increasing interest for their combined neuroregenerative and neuroprotective advantages, uniquely supporting both axonal orientation cues and minimally invasive delivery. Herein, ferric iron-chelated silk fibroin microfibers (Fe3+-mSF) are explored as a magnetic nanoparticle-free alternative to existing magneto-responsive fiber architectures. Functionalization of nascent mSF via iron chelation exhibited stability at

Rapid preparation of a bioadhesive with wet adhesion based on supramolecular interactions for wound closure and healing.

Achieving wet adhesion is a fundamental yet challenging requirement for bioadhesives. Poly(α-lipoic acid) (PLA) adhesives derived from the polymerization of α-lipoic acid (LA) have attracted increasing attention in the biomedical field. However, their practical application is hindered by drawbacks such as environmental dependency and prolonged polymerization time. In this study, we report the rapid fabrication of a bulk adhesive, LAS@NP, at room temperature through the supramolecular assembly of LA, Ag+, silk fibroin (SF), and polydopamine nanoparticles containing Ag (PDA/Ag-NP), without the n

A Core-Shell Structured Microneedle Patch With Adjustable Release of Kinetically for the Treatment of Melasma.

Melasma is a facial hyperpigmentation disease that significantly impacts patients' quality of life. Clinical treatment is limited by the short half-lives and hydrophilicity of drugs, necessitating release curve optimization to maintain a stable therapeutic concentration for an extended period. This article utilizes natural biomaterials to design a core-shell structured microneedle, combining the "immediate release" and "delayed release" module to achieve programmed drug release. Silk fibroin with a crystalline structure is used as the shell to ensure the harmlessness of the accessible areas, w

Hierarchically Structured Cellulose Acetate@Silk Protein Membrane with Enhanced Mechanical and Electromagnetic Interference Shielding Performances.

Compared to conventional fibers, electrospun porous nanofibers with hierarchical structures often involve additional active sites, interfaces, and internal spaces which boost the performances of functional materials. Here in this study, coaxial composite cellulose acetate@silk fibroin (CA@SF) fibrous membranes are constructed through an electrostatic spinning technique combining solvent-induced phase separation. Hierarchical core-shell structures on the fibers are achieved, which significantly increases the surface area and benefits the mechanical property, flux, as well as the electroless dep

Ingrowth of human mesenchymal stem cells into porous silk particle reinforced silk composite scaffolds: An in vitro study.

Silk fibroin protein is biodegradable and biocompatible, exhibiting excellent mechanical properties for various biomedical applications. However, porous three-dimensional (3-D) silk fibroin scaffolds, or silk sponges, usually fall short in matching the initial mechanical requirements for bone tissue engineering. In the present study, silk sponge matrices were reinforced with silk microparticles to generate protein-protein composite scaffolds with desirable mechanical properties for in vitro osteogenic tissue formation. It was found that increasing the silk microparticle loading led to a substa

Insights into the material properties of dragline spider silk affecting Schwann cell migration.

Dragline silk of Trichonephila spiders has attracted attention in various applications. One of the most fascinating uses of dragline silk is in nerve regeneration as a luminal filling for nerve guidance conduits. In fact, conduits filled with spider silk can measure up to autologous nerve transplantation, but the reasons behind the success of silk fibers are not yet understood. In this study dragline fibers of Trichonephila edulis were sterilized with ethanol, UV radiation, and autoclaving and the resulting material properties were characterized with regard to the silk's suitability for nerve

Progress in the application of spider silk protein in medicine.

Spider silk protein has attracted much attention on account of its excellent mechanical properties, biodegradability, and biocompatibility. As the main protein component of spider silk, spidroin plays important role in spider spinning under natural circumstances and biomaterial application in medicine as well. Compare to the native spidroin which has a large molecular weight (>300 kDa) with highly repeat glycine and polyalanine regions, the recombinant spidroin was maintained the core amino motifs and much easier to collect. Here, we reviewed the application of recombinant spider silk protein

Viscoelastic and antimicrobial dental care bioplastic with recyclable life cycle.

Medical plastic-appliance-based healthcare services, especially in dentistry, generate tremendous amounts of plastic waste. Given the physiological features of our mouth, it is desirable to substitute dental care plastics with viscoelastic and antimicrobial bioplastics. Herein, we develop a medical-grade and sustainable bioplastic that is viscoelastic enough to align the tooth positions, resists microbial contamination, and exhibits recyclable life cycles. In particular, we devise a molecular template involving entanglement-inducing and antimicrobial groups and prepare a silk fibroin-based den

Skin-like wound dressings with on-demand administration based on in situ peptide self-assembly for skin regeneration.

Burn injuries without the normal skin barrier usually cause skin wound infections, and wound dressings are necessary. Although various dressings with antibacterial ability have already been developed, the biosafety and administration mode are still bottleneck problems for further application. Herein, we designed skin-like wound dressings based on silk fibroin (SF), which are modified with the gelatinase-cleavable self-assembled/antibacterial peptide (GPLK) and epidermal growth factor (EGF). When a skin wound is infected, the gelatinase over-secreted by bacteria can cut the GPLK peptides, leadi

Bioprinting of anisotropic functional corneal stroma using mechanically robust multi-material bioink based on decellularized cornea matrix.

Corneal scarring is a common cause of blindness, affecting millions globally each year. A huge gap between the demand and supply of donor tissue currently limits corneal transplantation, the only definitive therapy for patients with corneal scarring. To overcome this challenge, researchers have harnessed the efficacy of 3D bioprinting to fabricate artificial corneal stromal constructs. With all the different bioinks available, the decellularized corneal matrix-based bioprinted construct can fulfill the required biological functionality but is limited by the lack of mechanical stiffness. In Situ ATR-FTIR Studies on the β-Sheet Formation of Native and Regenerated Bombyx mori Silk Material in Solution and Its Potential for Drug Releasing Coatings.

Dynamic attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy at both solutions and coatings of a semicrystalline silk material derived from Bombyx mori was applied to monitor the β-sheet conformation, which is known to correlate with silk protein crystallinity. The secondary structure-sensitive Amide I band was analyzed. Two silk protein samples were studied: native-based silk buffer fibroin (NSF) was extracted from silk glands and regenerated silk fibroin (RSF) was extracted from degummed cocoons. Solutions of both NSF and RSF at 2 mg/mL featured low initial β-sheet

Development of silk-based scaffolds for tissue engineering of bone from human adipose-derived stem cells.

Silk fibroin is a potent alternative to other biodegradable biopolymers for bone tissue engineering (TE), because of its tunable architecture and mechanical properties, and its demonstrated ability to support bone formation both in vitro and in vivo. In this study, we investigated a range of silk scaffolds for bone TE using human adipose-derived stem cells (hASCs), an attractive cell source for engineering autologous bone grafts. Our goal was to understand the effects of scaffold architecture and biomechanics and use this information to optimize silk scaffolds for bone TE applications. Silk sc

Liquid-liquid phase separation enables chromatography-free purification and high-performance spidroin-amyloid hybrid silk fibers.

Large-scale production of artificial spider silk fibers requires heterologous expression of spider silk proteins (spidroins), yet current methods remain limited by low yields and costly purification processes. To overcome these challenges, we engineered mini-spidroins in which the poly-alanine motifs of the repetitive region were replaced with the non-natural amyloidogenic β16 peptide, significantly enhancing expression yields and solubility. Furthermore, we developed a simple, chromatography-free purification method for these constructs based on NaCl-induced liquid-liquid phase separation (LL
