Identification and characterization of Clostridium botulinum group III field strains by matrix-assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI-TOF MS).

Animal botulism is primarily due to botulinum neurotoxin (BoNT) types C, D or their chimeric variants C/D or D/C, produced by Clostridium botulinum group III, which appears to include the genetically indistinguishable Clostridium haemolyticum and Clostridium novyi. In the present study, we used matrix-assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI TOF MS) to identify and characterize 81 BoNT-producing Clostridia isolated in 47 episodes of animal botulism. The instrument's default database, containing no entries for Clostridium botulinum, permitted reliable identif

Is schizophrenia a dopamine supersensitivity psychotic reaction?

Adolf Meyer (1866-1950) did not see schizophrenia as a discrete disorder with a specific etiology but, rather, as a reaction to a wide variety of biopsychosocial factors. He may have been right. Today, we have evidence that gene mutations, brain injury, drug use (cocaine, amphetamine, marijuana, phencyclidine, and steroids), prenatal infection and malnutrition, social isolation and marginalization, can all result in the signs and symptoms of schizophrenia. This clinical picture is generally associated with supersensitivity to dopamine, and activates dopamine neurotransmission that is usually a

Integrative transcriptomics identifies an 18-gene candidate blood-accessible biomarker panel for longitudinal monitoring of spaceflight-induced muscle atrophy.

Skeletal muscle atrophy is a critical physiological risk during long-duration spaceflight, yet candidate blood-accessible transcriptomic biomarkers for longitudinal monitoring remain unavailable. We developed an integrative multi-mission framework combining a primary discovery transcriptome from THP-1 monocytic cells exposed to International Space Station (ISS) microgravity (GEO: Sci. Adv. 2026 (https://doi.org/10.1126/sciadv.adw7832, Data S1-S10); NASA OSDR: Al-Ahmadi et al. 2026; n = 8 spaceflight, 8 ground control) - yielding 4,522 spaceflight-differentially expressed genes (SDEGs) - with a

Epigenetics in Space: The Dynamic Cellular Response.

Space exploration is moving toward the next milestone: space colonization. The risks for human health, associated to the permanence in the space, are already object of studies by several experiments both in orbit and, under simulated space conditions, on hearth. Now, these assessments will need to be extended over the increased permanence, in view of the stable life on other planets. It is well known that the environmental conditions present in the space, primarily related to the altered gravity, introduce a physical stress and the lack of a fundamental bond that contributes to the spatial org

Biological clocks and blood clots in space: circadian disruption-induced modulation of thrombosis risk.

Long-duration space missions expose astronauts to microgravity, radiation, confinement, fluid redistribution, and circadian disruption, which together induce physiological adaptations, including cardiovascular hemodynamics. Emerging evidence suggests that spaceflight may modulate hemostasis and potentially increase thrombotic risk in the head/neck; however, the association of circadian disruption and hemostasis in space, and its ground-based analogs, has yet to be systematically evaluated. This systematic review aimed to synthesize evidence from real-spaceflight missions and ground-based analo

Domestication of radiation sensitive Caenorhabditis Elegans strains suitable for liquid culture systems in long-term space mission.

With the continuous advancement of manned deep-space exploration, the need for effective radiation-risk assessment and protection is increasingly urgent. There is a need to screen space radiation-sensitive Caenorhabditis elegans strains suitable for long-term microfluidic chip-based cultivation and observation in space on orbit. In the study, green fluorescently protein-labeled nematode strains carrying stress-related genes (involved in stress response, DNA-damage, germ-cell apoptosis) were selected. Nematode strains that significantly responded to Co60γ-ray radiation stimulation were screened

To boldly go where no microRNAs have gone before: spaceflight impact on risk for small-for-gestational-age infants.

In the era of renewed space exploration, comprehending the effects of the space environment on human health, particularly for deep space missions, is crucial. While extensive research exists on the impacts of spaceflight, there is a gap regarding female reproductive risks. We hypothesize that space stressors could have enduring effects on female health, potentially increasing risks for future pregnancies upon return to Earth, particularly related to small-for-gestational-age (SGA) fetuses. To address this, we identify a shared microRNA (miRNA) signature between SGA and the space environment, c

Exposure to multiple ion beams, broadly representative of galactic cosmic rays, causes perivascular cardiac fibrosis in mature male rats.

Long-duration space exploratory missions to the Earth's moon and the planet Mars are actively being planned. Such missions will require humans to live for prolonged periods beyond low earth orbit where astronauts will be continuously exposed to high energy galactic cosmic rays (GCRs). A major unknown is the potential impact of GCRs on the risks of developing degenerative cardiovascular disease, which is a concern to NASA. A ground-based rat model has been used to provide a detailed characterization of the risk of long-term cardiovascular disease from components of GCRs at radiation doses relev

Regulation of Bone by Mechanical Loading, Sex Hormones, and Nerves: Integration of Such Regulatory Complexity and Implications for Bone Loss during Space Flight and Post-Menopausal Osteoporosis.

During evolution, the development of bone was critical for many species to thrive and function in the boundary conditions of Earth. Furthermore, bone also became a storehouse for calcium that could be mobilized for reproductive purposes in mammals and other species. The critical nature of bone for both function and reproductive needs during evolution in the context of the boundary conditions of Earth has led to complex regulatory mechanisms that require integration for optimization of this tissue across the lifespan. Three important regulatory variables include mechanical loading, sex hormones

Angiotensin converting enzyme (ACE) inhibitors as radiation countermeasures for long-duration space flights.

Angiotensin converting enzyme (ACE) inhibitors are effective countermeasures to chronic radiation injuries in rodent models, and there is evidence for similar effects in humans. In rodent models ACE inhibitors are effective mitigators of radiation injury to kidney, lung, central nervous system (CNS) and skin, even when started weeks after irradiation. In humans, the best data for their efficacy as radiation countermeasures comes from retrospective studies of injuries in radiotherapy patients. We propose that ACE inhibitors, at doses approved for human use for other indications, could be used t

Effects of spaceflight and ground recovery on mesenteric artery and vein constrictor properties in mice.

Following exposure to microgravity, there is a reduced ability of astronauts to augment peripheral vascular resistance, often resulting in orthostatic hypotension. The purpose of this study was to test the hypothesis that mesenteric arteries and veins will exhibit diminished vasoconstrictor responses after spaceflight. Mesenteric arteries and veins from female mice flown on the Space Transportation System (STS)-131 (n=11), STS-133 (n=6), and STS-135 (n=3) shuttle missions and respective ground-based control mice (n=30) were isolated for in vitro experimentation. Vasoconstrictor responses were

Effect of altered gravity on the neurobiology of fish.

In vertebrates (including humans) altered gravitational environments such as weightlessness can induce malfunction of the inner ears due to a mismatch between canal and statolith afferents. This leads to an illusionary tilt because the inputs from the inner ear are not confirmed by the other sensory organs, which then results in intersensory conflict. Vertebrates in orbit therefore face severe orientation problems. In humans the intersensory conflict may additionally lead to a malaise commonly referred to as space motion sickness (SMS). After the initial days of weightlessness the orientation

Proteomics of Toxigenic Corynebacteria.

Within the genus Corynebacterium, six species are potential carriers of the tox gene, which encodes the highly potent diphtheria exotoxin: Corynebacterium diphtheriae, Corynebacterium belfantii, Corynebacterium rouxii, Corynebacterium ulcerans, Corynebacterium pseudotuberculosis and Corynebacterium silvaticum. Based on their potential to infect different host species and cause either human infections, zoonotic diseases or infections of economically important animals, these bacteria are of high scientific and economic interest and different research groups have carried out proteome analyses. Impact of inhaled nitric oxide on the sulfatide profile of neonatal rat brain studied by TOF-SIMS imaging.

Despite advances in neonatal intensive care leading to an increased survival rate in preterm infants, brain lesions and subsequent neurological handicaps following preterm birth remain a critical issue. To prevent brain injury and/or enhance repair, one of the most promising therapies investigated in preclinical models is inhaled nitric oxide (iNO). We have assessed the effect of this therapy on brain lipid content in air- and iNO-exposed rat pups by mass spectrometry imaging using a time-of-flight secondary ion mass spectrometry (TOF-SIMS) method. This technique was used to map the variations

Structure, properties and functions of the forewing-hindwing coupling of honeybees.

Worker honeybees (Apis mellifera) are morphologically four-winged, but are functionally dipterous insects. During flight, their fore- and hindwings are coupled by means of the forewing posterior rolled margin (PRM) and hindwing hamuli. Morphological analysis shows that the PRM can be connected to the hamuli, so that the fore- and hindwing are firmly hinged, and can rotate with respect to each other. In the present study, using a combination of scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM), we investigate the micromorphology and material composition of the cou

Carriage of Extended Spectrum Beta-Lactamase and Carbapenemase Genes in Food Animals and Their Human Handlers: A One Health Perspective in a Low-Resource Setting in Africa.

We investigated fecal colonization with third-generation cephalosporin-resistant (3GC-r) Enterobacterales and carbapenem-resistant Enterobacterales among food animals and their handlers in Ghana. A total of 252 fecal samples were collected from 211 animals and 41 human handlers across 20 farms between May and August 2023. Enterobacterales were isolated using standard methods and identified using Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry fingerprints and 16SrRNA sequencing. Antibiotic susceptibility testing was done using standard methods. Extended spectrum be

Environmental antibiotic and fungicide exposure differentially affects survival and carbohydrate-fueled flight performance in Apis mellifera.

Environmental contamination by antibiotics and fungicides is an emerging concern due to their widespread occurrence in agricultural ecosystems. The western honey bee, Apis mellifera, a key managed pollinator, is exposed to these contaminants through multiple pathways, including treated crops, contaminated water, and residues from apiculture, livestock waste, agricultural runoff, and wastewater discharges. The agrochemicals tetracycline and boscalid disrupt cellular energy metabolism and mitochondrial function in non-target organisms and may consequently compromise bee survival and the sustaine

Microgravity-induced muscle atrophy in rodent and human models.

Exposure to microgravity leads to severe skeletal muscle atrophy, particularly in muscles that act to maintain posture against gravity, and results in significant reductions in muscle mass, cross-sectional area, and contractile force. This review examines the physiological and cellular response of microgravity in both rodent and human models, emphasizing shifts in muscle fiber type, altered contractile properties, and molecular adaptations. Specifically, rodents exhibit rapid atrophy, predominantly in slow-twitch fibers, whereas human studies suggest greater intersubject variability, with both

Field cage studies and progressive evaluation of genetically-engineered mosquitoes.

BACKGROUND: A genetically-engineered strain of the dengue mosquito vector Aedes aegypti, designated OX3604C, was evaluated in large outdoor cage trials for its potential to improve dengue prevention efforts by inducing population suppression. OX3604C is engineered with a repressible genetic construct that causes a female-specific flightless phenotype. Wild-type females that mate with homozygous OX3604C males will not produce reproductive female offspring. Weekly introductions of OX3604C males eliminated all three targeted Ae. aegypti populations after 10-20 weeks in a previous laboratory cage
