Insomnia treatment based on the regulation of the GABAergic system: traditional Chinese medicine perspectives and therapeutic approaches.

Insomnia is a common clinical manifestation of central nervous system (CNS) dysfunction that may be attributable to the involvement of abnormalities in multiple systems. Dysfunction of the gamma-aminobutyric acid (GABA) system is one of the key pathologic factors responsible for the increased neural excitability. GABA has been regarded as the major inhibitory neurotransmitter in the CNS, which can maintain the dynamic balance of neural networks by regulating neuronal excitability. Traditional Chinese medicine (TCM), with its multi-component, multi-target therapeutic advantages, shows unique po

Melatonin in Livestock Production: Bridging Chronobiology, Health, and Sustainable Outcomes.

Melatonin (N-acetyl-5-methoxytryptamine) is a phylogenetically ancient hormone synthesized primarily in the pineal gland in response to light-dark cycles, where it regulates circadian rhythms, but it is also produced in peripheral tissues, including the gastrointestinal tract. In livestock, melatonin extends beyond chronobiology to modulate immune function, reproduction, metabolism, and adaptation to environmental stressors such as heat, transport, and high production intensity. This review consolidates current knowledge on melatonin's application in animal nutrition, tracing from its discover

From oxygen shortage to neurocognitive challenges: behavioral patterns and imaging insights.

Environmental hypoxia, resulting from reduced oxygen supply, poses a significant risk of dysfunctioning and damaging the neurocognitive system, particularly in relation to anxiety and stress. Inadequate oxygenation can lead to acute and chronic brain damage. Scholars used behavioral, hemodynamic, and electromagnetic neurofunctional techniques to investigate the effects of normobaric and hypobaric hypoxia on neurocognitive systems. They found a correlation between hypoxia, altered psychomotor responses, and changes in EEG alpha, theta, beta, and gamma rhythms, which affect spatial attention and…

Does more sleep matter? Differential effects of NREM- and REM-dominant sleep on sleepiness and vigilance.

We investigated effects of NREM and REM predominant sleep periods on sleepiness and psychomotor performances measured with visual analog scales and the psychomotor vigilance task, respectively. After one week of stable sleep-wake rhythms, 18 healthy sleepers slept 3hours of early sleep and 3hours of late sleep, under polysomnographic control, spaced by two hours of sustained wakefulness between sleep periods in a within subjects split-night, sleep interruption protocol. Power spectra analysis was applied for sleep EEG recordings and sleep phase-relative power proportions were computed for six

Deep brain electrophysiological recordings provide clues to the pathophysiology of Tourette syndrome.

Although ample evidence suggests that high-frequency deep brain stimulation (DBS) is an effective therapy in patients with Tourette syndrome (TS), its pathophysiology and the neurophysiological mechanisms underlying these benefits remain unclear. The DBS targets mainly used to date in TS are located within the basal ganglia-thalamo-cortical circuit compromised in this syndrome: the medial and ventral thalamic nuclei, which are way stations within the circuit, the globus pallidus and the nucleus accumbens. Neuronal activity can be electrophysiologically recorded from deep brain structures durin

The C1 area of the rostral ventrolateral medulla oblongata. A critical brainstem region for control of resting and reflex integration of arterial pressure.

A population of neurons localized to a small region of the brain stem reticular formation, the rostral ventrolateral reticular nucleus (RVL), is the principal area of the brainstem regulating resting, reflex, and probably behaviorally coupled control of arterial pressure. The critical area of the RVL reticular nucleus engaged in cardiovascular control surrounds a cluster of adrenergic neurons of the C1 group, and is therefore designated the C1 area. C1 area neurons have a direct and extremely potent synaptic relationship with preganglionic sympathetic neurons of the intermediolateral (IML) nuc

Neurophysiological and neuropharmacological effects of melatonin MT2 receptors activation in MK-801-induced schizophrenia-like dysfunctions.

Schizophrenia (SCZ) is a chronic psychiatric disorder characterized by positive, negative, and cognitive symptoms that remain insufficiently controlled by current dopamine- and serotonin-based antipsychotics. Emerging evidence implicates melatonin MT2 receptors in the regulation of the sleep-wake cycle, circadian rhythms and cortical inhibition, both altered in SCZ. Here, we investigated the neuropharmacological effects of the selective MT2 partial agonist UCM924 in the MK-801 model of SCZ-like dysfunctions in male mice. UCM924 (10 mg/kg, intraperitoneally) was selected as a dose not affecting

Driving Cerebellar Theta Oscillations Interferes With Voluntary Neck Movements in Cervical Dystonia.

BACKGROUND: Cervical dystonia (CD) is a movement disorder with a complex pathophysiology, including cerebellar abnormalities. Transcranial alternating current stimulation (tACS), a noninvasive neuromodulation technique capable of entraining brain oscillations, can transiently modulate neuronal activity and enhance resonant rhythms. OBJECTIVE: The aim of this study was to explore whether cerebellar tACS delivered at specific cerebellar-resonant frequencies modifies fast voluntary neck movements in patients with CD and whether botulinum toxin (BoNT) therapy influences tACS effects. METHODS: Ei

Impact of NMDA receptors block versus GABA-A receptors modulation on synaptic plasticity and brain electrical activity in metabolic syndrome.

PURPOSE: Metabolic syndrome (MetS) is a common disorder associated with disturbed neurotransmitter homeostasis. Memantine, an N-methyl-d-aspartate receptor (NMDAR) antagonist, was first used in Alzheimer's disease. Allopregnanolone (Allo), a potent positive allosteric modulator of the Gamma-Amino-Butyric Acid (GABA)-A receptors, decreases in neurodegenerative diseases. The study investigated the impact of Memantine versus Allo administration on the animal model of MetS to clarify whether the mechanism of abnormalities is related more to excitatory or inhibitory neurotransmitter dysfunction. Plasma metabolomics changes comparing daytime to overnight infusions of home parenteral nutrition in adult patients with short bowel syndrome: Secondary analysis of a clinical trial.

BACKGROUND: Home parenteral nutrition (HPN) is often cycled nocturnally and is expected to result in glucose intolerance and sleep disruption partly due to circadian misalignment. This study aimed to define the metabolic response when HPN is cycled during the daytime compared to overnight. METHODS: This secondary analysis leveraged samples from a clinical trial in adults with short bowel syndrome consuming HPN (ClinicalTrials.gov: NCT04743960). Enrolled patients received 1 week of HPN overnight followed by 1 week of HPN during the daytime. Fasting blood samples were collected following each s

A novel N-terminal isoform of the neuron-specific K-Cl cotransporter KCC2.

The neuronal K-Cl cotransporter KCC2 maintains the low intracellular chloride concentration required for the hyperpolarizing actions of inhibitory neurotransmitters gamma-aminobutyric acid and glycine in the central nervous system. This study shows that the mammalian KCC2 gene (alias Slc12a5) generates two neuron-specific isoforms by using alternative promoters and first exons. The novel KCC2a isoform differs from the only previously known KCC2 isoform (now termed KCC2b) by 40 unique N-terminal amino acid residues, including a putative Ste20-related proline alanine-rich kinase-binding site. DNA methylation in bipolar disorder: mechanistic insights and translational challenges.

Bipolar disorder (BD) is a severe and recurrent psychiatric disorder characterized by alternating manic, hypomanic, and depressive episodes, frequent comorbidity, high relapse rates, and an increased risk of suicide. Although the pathophysiology of BD remains incompletely understood, increasing evidence suggests that DNA methylation may represent an important epigenetic regulatory layer involved in BD-related biological heterogeneity. DNA methylation alterations have been reported in genes related to dopamine, serotonin, glutamate, and gamma-aminobutyric acid (GABA) systems, suggesting a poten

GABA: The Peacekeeper Neurotransmitter-Gut-microbiota Derived Origins and Salivary Biomarker Detection Using Elisa.

BACKGROUND: The human body is a highly integrated biological system in which the gut flora wields regulatory control beyond simple digestion. Intestinal bacteria control neuronal and behavioral processes via the gut-brain axis; microbial gamma-aminobutyric acid (GABA) synthesis seems to be an essential controller of sleep and central nervous system activity. Increasing data show that neuroactive substances produced from bacteria influence mental health results, circadian rhythm, and sleep structure. SUMMARY: This review investigates the function of gut-derived GABA in sleep control, the part

Beta cell secreted GABA sets appropriate insulin secretion by modulating islet calcium oscillations.

OBJECTIVES: Gamma-aminobutyric acid (GABA) is produced in pancreatic beta cells and is implicated in modulating islet function, yet its precise physiological role remains uncertain. This study aimed to determine the function of endogenous beta cell-derived GABA on insulin secretion and islet calcium dynamics by developing a conditional beta cell-specific knockout of GABA-synthesizing enzymes (GAD65 and GAD67). METHODS: Conditional knockout mice (GadβKO) lacking both GAD65 and GAD67 specifically in pancreatic beta cells were generated. Glucose-stimulated insulin secretion was measured in isola

Non-invasive versus invasive techniques for predicting idiopathic normal-pressure hydrocephalus in shunt-responsive patients.

The diagnosis of normal-pressure hydrocephalus (NPH) can be challenging, particularly distinguishing between shunt-responsive and non-responsive patients. Overlapping clinical symptoms with aging and neurodegenerative diseases such as Alzheimer’s and Parkinson’s may hinder its diagnosis. Thus, more effective diagnostic tools are necessary To compare non-invasive techniques, such as electroencephalography (EEG), transcranial Doppler (TCD), intracranial compliance (ICC) measurement, and optical coherence tomography (OCT), with invasive procedures, such as lumbar puncture (LP) and infusion tests

Surveillance of Fontan Associated Liver Disease in Childhood and Adolescence.

Fontan associated liver disease (FALD) has been recognized as a potentially serious sequela of the Fontan circulation. Prevalence of FALD among different age groups and risk factors for advanced changes were assessed. FALD screening included abdominal ultrasound and laboratory tests. A "liver disease score (LDS)" incorporating items from ultrasound and blood testing was calculated to grade FALD severity (5 items each, maximum score 10 points). 240 patients (male: n = 139, female: n = 101, systemic right ventricle: n = 160) underwent FALD screening 10 (IQR 7-15) years after Fontan surgery. Technical Note: Cardiac synchronized volumetric modulated arc therapy for stereotactic arrhythmia radioablation - Proof of principle.

PURPOSE: Ventricular tachycardia (VT) is a rapid, abnormal heart rhythm that can lead to sudden cardiac death. Current treatment options include antiarrhythmic drug therapy and catheter ablation, both of which have only modest efficacy and have potential complications. Cardiac radiosurgery has the potential to be a noninvasive and efficient treatment option for VT. Cardiac motion, however, must be accounted for to ensure accurate dose delivery to the target region. Cardiac synchronized volumetric modulated arc therapy (CSVMAT) aims to minimize the dose delivered to normal tissues by synchroniz

GABAA receptors mediate excitation in adult rat GnRH neurons.

Gonadotropin-releasing hormone (GnRH) neurons form the final common pathway for the central regulation of reproduction. Gamma-amino butyric acid (GABA), the main inhibitory neurotransmitter in the adult brain, has long been implicated in playing key roles in the regulation of GnRH neurons. Two groups reported recently that GABA depolarizes GnRH neurons, although one group reported a hyperpolarizing action of GABA. In this study, we investigated the GABA-induced changes in [Ca(2+)](i) of GnRH neurons from GnRH-enhanced green fluorescent protein (GnRH-EGFP) rats both to confirm the depolarizing

Seasonal testicular regression in Yangzhou geese: Transcriptomic evidence for an immune-reproductive trade-off during the transition from peak to shutdown egg production.

This study investigated developmental changes and the underlying molecular mechanisms in testicular tissue of Yangzhou geese during the transition from the peak production (PP) to egg production shutdown (PF) periods. We hypothesized that this transition is accompanied by a reciprocal shift in resource allocation-declining spermatogenic activity offset by enhanced immune function-reflecting an adaptive life-history trade-off in this seasonally breeding species. Five Yangzhou geese were randomly selected each during the PP (day 73) and PF (day 135) periods, based on the endogenous seasonal repr

The effect of bilateral high-definition γ-tACS on negative symptoms and mismatch negativity in schizophrenia.

BACKGROUND: At present, there is a lack of good treatment methods for the negative symptoms of schizophrenia. Previous studies have suggested that high-definition transcranial alternating current stimulation (HD tACS) in the dorsolateral prefrontal cortex (DLPFC) can significantly improve negative symptoms and that γ oscillation may also be effective for treating negative symptoms. The aim of our study was to investigate the therapeutic effect of HD γ-tACS on the negative symptoms of schizophrenia. Active auditory mismatch negativity (MMN) was introduced as neurobiological evidence to verify t
