A syndromic extreme insulin resistance caused by biallelic POC1A mutations in exon 10.

POC1A encodes a protein with a role in centriole assembly and stability, and in ciliogenesis. Biallelic loss-of-function mutations affecting POC1A cause SOFT syndrome, an ultra-rare condition characterized by short stature, onychodysplasia, facial dysmorphism and hypotrichosis. Using exome sequencing, we identified a homozygous frameshift mutation (c.1047_1048dupC; p.G337Rfs*25) in a patient presenting with short stature, facial hirsutism, alopecia, dyslipidemia and extreme insulin resistance. The truncating variant affected exon 10, which is retained in only two of the three POC1A-mature RNAs

Shared genetic architecture between major depressive disorder and inflammatory bowel disease: Insights from large-scale genome-wide cross-trait analysis.

BACKGROUND: Major depressive disorder (MDD) and inflammatory bowel disease (IBD) exhibit clinical associations, yet the cross-disease genetic links and shared mechanisms between them remain unclear. This study aims to explore the genetic associations between MDD and IBD and identify the shared risk loci, potential key tissues, and related genetic mechanisms. METHODS: Leveraging large-scale genome-wide association study (GWAS) summary statistics, we employed linkage disequilibrium score regression (LDSC) and high-definition likelihood (HDL) methods to comprehensively evaluate the genetic corre

Identification of shared genetic etiology of cardiovascular and cerebrovascular diseases through common cardiometabolic risk factors.

Cardiovascular diseases (CVDs) and cerebrovascular diseases (CeVDs) are closely related vascular diseases, sharing common cardiometabolic ri … ti-omics analysis revealed methylation of two genes (CASZ1 and LRCH1) may play a causal role in the genetic pleiotropy. Notably, these pleiotropic loci are highly enriched in the targets of antihypertensive drugs, which further emphasizes the role of the blood pressure regulation pathway in the shared etiology of CVDs and CeVDs.

Genome-wide association study of primary tooth eruption identifies pleiotropic loci associated with height and craniofacial distances.

Twin and family studies indicate that the timing of primary tooth eruption is highly heritable, with estimates typically exceeding 80%. To identify variants involved in primary tooth eruption, we performed a population-based genome-wide association study of 'age at first tooth' and 'number of teeth' using 5998 and 6609 individuals, respectively, from the Avon Longitudinal Study of Parents and Children (ALSPAC) and 5403 individuals from the 1966 Northern Finland Birth Cohort (NFBC1966). We tested 2 446 724 SNPs imputed in both studies. Analyses were controlled for the effect of gestational age,

Genome-wide Pleiotropy Between Parkinson Disease and Autoimmune Diseases.

IMPORTANCE: Recent genome-wide association studies (GWAS) and pathway analyses supported long-standing observations of an association between immune-mediated diseases and Parkinson disease (PD). The post-GWAS era provides an opportunity for cross-phenotype analyses between different complex phenotypes. OBJECTIVES: To test the hypothesis that there are common genetic risk variants conveying risk of both PD and autoimmune diseases (ie, pleiotropy) and to identify new shared genetic variants and their pathways by applying a novel statistical framework in a genome-wide approach. DESIGN, SETTING,

The Master of Puppets: Pleiotropy of PDGFRB and its Relationship to Multiple Diseases.

The platelet-derived growth factor receptor beta (PDGFRB) gene is involved in proliferative and developmental processes in mammals. Variations in this gene lead to several different syndromic conditions, such as infantile myofibromatosis I, sporadic port-wine stain, primary familial brain calcification, and the Penttinen and overgrowth syndromes. Our objective was to investigate PDGFRB's genetic relationship to clinical conditions and evaluate the protein interactions using GeneNetwork, GeneMANIA, and STRING network databases. We have evidenced the gene's pleiotropy through its many connection

Genetic insights into radiomic and proteomic changes under β1-blockers treatment in hypertensive heart disease and hypertrophic cardiomyopathy.

BACKGROUND: Hypertensive heart disease (HHD) and hypertrophic cardiomyopathy (HCM) are characterized by left ventricular hypertrophy and diastolic dysfunction. Despite overlapping remodeling features, their distinct mechanisms and therapeutic responses remain unclear. This study integrated genetic, imaging, and proteomic data to identify key mediators underlying β1-adrenergic receptor blockers (β1-blockers)-related therapeutic heterogeneity between HHD and HCM. METHODS: Genetic instruments for β1-blockers were derived from two genome-wide association studies and integrated with cardiac magnet

A cross-population atlas of genetic associations for 220 human phenotypes.

Current genome-wide association studies do not yet capture sufficient diversity in populations and scope of phenotypes. To expand an atlas of genetic associations in non-European populations, we conducted 220 deep-phenotype genome-wide association studies (diseases, biomarkers and medication usage) in BioBank Japan (n = 179,000), by incorporating past medical history and text-mining of electronic medical records. Meta-analyses with the UK Biobank and FinnGen (ntotal = 628,000) identified ~5,000 new loci, which improved the resolution of the genomic map of human traits. This atlas elucidated th

Erythropoietin drug targets and cancer risk: A two-sample Mendelian randomization study.

Erythropoiesis-stimulating agents (ESAs) are widely used to treat anemia, but their on-target effects on tumor risk via erythropoietin (EPO)/erythropoietin receptor (EPOR) signaling remain debated. This study aims to evaluate the potential causal relationship between EPO signaling and the risk of 14 site-specific cancers using a drug-target Mendelian randomization (MR) approach. We proxied EPO signaling using genetic instruments for the EPOR, derived from cis-expression quantitative trait locus data from the eQTLGen Consortium. Summary-level genetic associations for 14 cancer types were source

Causal relationship between cardiac arrhythmias and sepsis: a two-sample bidirectional mendelian randomization study.

OBJECTIVE: This study used a bidirectional two-sample Mendelian randomization (MR) framework to investigate whether genetic liability to cardiac arrhythmias increases the risk of sepsis and whether genetic liability to sepsis increases the risk of cardiac arrhythmias. METHODS: Summary statistics were obtained from publicly available genome-wide association studies in individuals of European ancestry. Cardiac arrhythmia data were derived from FinnGen (32,416 cases and 139,739 controls; IEU ID: finn-b-CARDIAC_ARRHYTM), and sepsis data were derived from UK Biobank (11,643 cases and 474,841 contr

Genomics in Personalized Nutrition: Can You "Eat for Your Genes"?

Genome-wide single nucleotide polymorphism (SNP) data are now quickly and inexpensively acquired, raising the prospect of creating personalized dietary recommendations based on an individual's genetic variability at multiple SNPs. However, relatively little is known about most specific gene-diet interactions, and many molecular and clinical phenotypes of interest (e.g., body mass index [BMI]) involve multiple genes. In this review, we discuss direct to consumer genetic testing (DTC-GT) and the current potential for precision nutrition based on an individual's genetic data. We review important

A de novo mutation in KIT causes white spotting in a subpopulation of German Shepherd dogs.

Although variation in the KIT gene is a common cause of white spotting among domesticated animals, KIT has not been implicated in the diverse white spotting observed in the dog. Here, we show that a loss-of-function mutation in KIT recapitulates the coat color phenotypes observed in other species. A spontaneous white spotting observed in a pedigree of German Shepherd dogs was mapped by linkage analysis to a single locus on CFA13 containing KIT (pairwise LOD = 15). DNA sequence analysis identified a novel 1-bp insertion in the second exon that co-segregated with the phenotype. The expected fram

Genetic correlation and causal relationships between autoimmune thyroid disease and major depressive disorders.

BACKGROUND: Autoimmune thyroid disease (AITD) and major depressive disorder (MDD) are common genetic diseases. The comorbidity of AITD and MDD has been widely demonstrated by large amounts of epidemiological studies. However, the genetic architectures of the comorbidity remain unknown. METHODS: We use large-scale GWAS summary data and novel genetic statistical methods to assess the genetic correlation and potential causality between AITD and MDD disorders. We perform cross-trait GWAS meta-analyses to identify genetic risk variants not previously associated with the individual traits. And we u

Effects of immune cells in mediating the relationship between inflammatory bowel disease and pyoderma gangrenosum: a two-sample, two-step mendelian randomization study.

BACKGROUND: Although the precise cause of the co-occurrence of pyoderma gangrenosum (PG) and inflammatory bowel disease (IBD) is still unknown, prior research has shown that the two conditions coexist. Moreover, it is currently unknown how immune cells function in influencing the relationship between IBD and PG. METHODS: In order to choose independent single nucleotide polymorphism (SNP) as instrumental variables, we were provided with genome-wide association study (GWAS) summary data of European populations from the IEU OpenGWAS project (for IBD) and a the FinnGen database (for PG) publicall

Causal associations between gut microbiota and synovitis-tenosynovitis: a two-sample Mendelian randomization study.

BACKGROUND: Increasing evidence indicates that gut microbiota dysbiosis is related to synovitis and tenosynovitis. Nonetheless, whether these associations are causal is currently unknown. OBJECTIVES: A two-sample Mendelian randomization (MR) study was performed to reveal the causality of gut microbiota with synovitis and tenosynovitis. METHODS: The summary statistical data from a large-scale genome-wide association study (GWAS) were applied as the basis for a two-sample MR analysis. The causal effect was estimated using inverse variance weighted (IVW), weighted median, simple mode, MR-Egger,

Heritable changes in regional cortical thickness with age.

It is now well established that regional indices of brain structure such as cortical thickness, surface area or grey matter volume exhibit spatially variable patterns of heritability. However, a recent study found these patterns to change with age during development, a result supported by gene expression studies. Changes in heritability have not been investigated in adulthood so far and could have important implications in the study of heritability and genetic correlations in the brain as well as in the discovery of specific genes explaining them. Herein, we tested for genotype by age (G ×A) i

Association of Inflammatory Proteins with Neuropathic Pain: A Two-Sample Bidirectional Mendelian Randomization.

OBJECT: Previous studies have found that circulating inflammatory proteins may play an essential role in the occurrence and development of neuropathic pain. However, the majority of existing evidence is derived from observational studies, which are prone to confounding factors and reverse causality, and the inflammatory profiles associated with different types of neuropathic pain remain poorly understood. Consequently, more robust methodologies are urgently required to elucidate the causal relationships between these variables. Therefore, a bidirectional Mendelian randomization analysis was pe

Exploring the Causal Link Between Plasma Lipidome and Trigeminal Neuralgia Using Bidirectional Mendelian Randomization.

Background: Trigeminal neuralgia (TN) is a prevalent neurological disorder characterized by recurrent acute pain localized within the distribution area of the trigeminal nerve. This condition places a severe psychological and emotional burden on patients. Although lipids are associated with many diseases, their relationship with TN remains unclear. This study aims to investigate the causal association between plasma lipidome and TN using a bidirectional two-sample Mendelian randomization (MR) approach, with the ultimate goal of informing potential therapeutic strategies for TN management.
