In 2007, 2.7 million vertebrates were used for animal experiments and other scientific purposes in Germany alone. Since 1998 there has been an increase in the number of animals used for research purposes, which is partly attributable to the growing use of transgenic animals. These animals are, for instance, used as in vivo models to mimic human diseases like diabetes, cancer or Alzheimer's disease. Here, transgenic model organisms serve as valuable tools, being instrumental in facilitating the analysis of the molecular mechanisms underlying human diseases, and might contribute to the developme

This chapter describes the erratic course of the research journey of our research group into the elucidation of the genetic basis of apomixis in the common dandelion, Taraxacum officinale s. l. Research is rarely straightforward; sometimes, it comes to a standstill, requiring a shift in approach. Our journey took years to complete, starting with the theoretical genetic models and culminating in the cloning of the parthenogenesis gene. Along the way, we employed a suite of genetic, cell biological, and molecular techniques.

Spondyloarthropathies (SpA), including ankylosing spondylitis (AS) and psoriatic arthritis (PsA), have been shown to have a substantial genetic predisposition based on heritability estimates derived from family studies and genome-wide association studies (GWAS). GWAS have uncovered numerous genetic loci associated with susceptibility to SpA, with significant associations to human leukocyte antigen (HLA) genes, which are major genetic risk factors for both AS and PsA. Specific loci differentiating PsA from cutaneous-only psoriasis have been identified, though these remain limited. Further resea

The recent explosion of hereditary breast cancer genetic research has strengthened our understanding of genetic influences on health and disease. These scientific advances have significant implications for clinical oncology nurses and advanced practice nurses. In this paper, the Human Response to Illness Model (Mitchell, Gallucci, & Fought, 1991) is utilized to gain a comprehensive understanding of breast cancer related to the human response of genetic mutations. Accordingly, the research literature related to the physiological, pathophysiological, behavioural, and experiential perspectives of…

Gene therapy has revolutionized modern medicine by offering innovative treatments for genetic disorders, cancers, and immune-related conditions through technologies such as viral vector delivery, genome editing, and genetically modified cell therapies. Despite significant advancements, the classification of gene therapy medicinal products (GTMPs) as genetically modified organisms (GMOs) under EU legislation imposes significant regulatory burdens, hindering early and timely patient access to such therapies. Current GMO regulations, originally designed for agricultural biotechnology, require env

BACKGROUND: Hereditary tumor predisposition syndromes (HTPSs) are being recognized more frequently in the etiology of pediatric cancer. Previous research indicates that disclosure of tumor susceptibility is a significant event in adolescents' lives. Insight into adolescents' adjustment to knowledge of their syndromes can guide healthcare delivery, particularly genetic counseling. This study explored the experiences of adolescents with hereditary tumor predisposition and their perceptions of living at risk.
METHODS: Seven adolescents, ages 14 to 17, representing six different childhood-onset H

Genome-wide testing platforms are increasingly used to promote "agnostic" approaches to the discovery of gene variants associated with the risk of many common diseases and quantitative traits. The early track record of genome-wide association (GWA) studies suggests that some proposed associations are replicated quite consistently with large-scale subsequent evidence from multiple studies, others have a more inconsistent replication record, some have failed to be replicated by independent investigators and many more early proposed associations await further replication. An important question is

Mapping individuals' genes could transform health care. A person susceptible to a certain disease could take corrective action years before onset. A doctor could become a lifelong lifestyle coach. Comparing genetic maps across populations could reveal factors that combine to cause diseases. Some hospitals are pushing forward on the genomics front despite daunting obstacles, including cost, fear of misuse of data by insurers, and an inability to share data.

This study employed an exploratory qualitative methodology to examine how IVF clinics in the United States develop and implement policies regarding the transfer of embryos that test positive for genetic conditions through preimplantation genetic testing for monogenic disorders (PGT-M). The study explored who is involved in creating these policies, and the key factors influencing these decisions. Semi-structured interviews were conducted with genetic counselors working in IVF clinics across the United States. Participants were asked about clinic policies regarding PGT-M positive embryos, their

Population-wide genomic screening (PWGS) offers a novel approach to identifying individuals at risk for hereditary cancer conditions such as Hereditary Breast and Ovarian Cancer (HBOC) Syndrome and Lynch Syndrome (LS). However, limited data exist on patient experiences and clinical outcomes following result disclosure. This study evaluated participant data from In Our DNA SC, a PWGS initiative offering free, CLIA-certified genomic screening and genetic counseling. We analyzed survey and electronic medical record data of participants who received a clinically actionable result for HBOC or LS an

This paper summarizes what is known about the diagnosis, treatment, and follow-care of people with the inherited disorder multiple endocrine neoplasia type 2a (MEN2a). Several databases were searched throughout the year 2000, including MEDLINE, CINAHL, CancerLit, PsychINFO, and Dissertation Abstracts International. MEN2a is a complex disease involving cancer care and control by surgery, health maintenance, and life-long observation of gene mutation carriers. Genetic testing is the standard of care, prophylactic surgery is recommended during directive counseling, and life-long surveillance is e

Advancing genetic testing technologies mean that population-based carrier screening for multiple inherited conditions is now available. As the number of genetic conditions being screened increases, there is a need for research into how people experience these screening programs. This research aimed to explore how women experience simultaneous carrier screening for three inherited conditions: cystic fibrosis (CF), spinal muscular atrophy (SMA), and fragile X syndrome (FXS). A qualitative approach was adopted using in-depth semi-structured interviews to explore the experiences of ten female part

Tourette syndrome (TS) is a complex disorder characterized by repetitive, sudden, and involuntary movements or vocalizations, called tics. Tics usually appear in childhood, and their severity varies over time. In addition to frequent tics, people with TS are at risk for associated problems including attention deficit hyperactivity disorder (ADHD), obsessive-compulsive disorder (OCD), anxiety, depression, and problems with sleep. TS occurs in most populations and ethnic groups worldwide, and it is more common in males than in females. Previous family and twin studies have shown that the majorit

PURPOSE OF REVIEW: Prenatal genetic testing can be divided into two categories: screening and diagnostic. This article will focus on reviewing prenatal genetic screening tests.
RECENT FINDINGS: Cell-free DNA (cfDNA) is a new prenatal genetic screening test with a high degree of accuracy for identifying certain genetic conditions like trisomy 21, 18, and 13. However, cfDNA has also been applied in the screening of other genetic conditions without similar research support.
SUMMARY: Prenatal genetic screening evaluates at risk pregnancies - including both carrier screening, which can be done at

In vivo Chimeric Antigen Receptor (CAR)-T cell therapy reprograms a patient's own T cells directly inside the body, bypassing the complex and costly traditional manufacturing process. This is achieved by systemically delivering viral or non-viral vectors that genetically modify endogenous T lymphocytes to produce functional CAR-T cells de novo. By eliminating ex vivo cell processing, this strategy can simplify workflows, reduce costs, improve accessibility, and allow faster treatment. Key delivery platforms include engineered lentiviral and adeno-associated viral (AAV) vectors for lasting CAR

Inherited retinal diseases (IRDs), including retinitis pigmentosa, Leber congenital amaurosis, and Stargardt disease, represent a major cause of inherited visual impairment and blindness, particularly in working-age populations. These disorders are genetically heterogeneous and result in progressive degeneration of photoreceptors and the retinal pigment epithelium. The eye offers a uniquely favorable environment for gene therapy due to its anatomical compartmentalization, accessibility, and relative immune privilege; however, successful clinical translation remains constrained by multiple biol

The clinical success of gene therapy depends critically on the development of delivery platforms capable of achieving precise, efficient and tissue-specific delivery of genetic payloads in vivo. A diverse array of carriers, including viral, non-viral, synthetic and natural vectors, have been explored to address this challenge. Among them, adeno-associated viruses, lipid nanoparticles and extracellular vesicles have emerged as leading candidates, each offering distinct advantages and translational hurdles. Here we provide a comparative analysis of these delivery modalities, highlighting their r

Pulmonary fibrosis comprises a wide range of fibrotic lung diseases with unknown pathogenesis and poor prognosis. Familial pulmonary fibrosis (FPF) represents a unique subgroup of patients in which at least one other relative is also affected. Patients with FPF exhibit a wide range of pulmonary fibrosis phenotypes, although idiopathic pulmonary fibrosis is the most common subtype. Despite variable disease manifestations, patients with FPF experience worse survival compared with their counterparts with the sporadic disease form. Therefore, ascertaining a positive family history not only provide

Non-viral gene therapy for hereditary skin diseases is an attractive prospect. However, research efforts dedicated to this area are rare. Taking advantage of the branched structural possibilities of polymeric vectors, we have developed a gene delivery platform for the treatment of an incurable monogenic skin disease - recessive dystrophic epidermolysis bullosa (RDEB) - based on highly branched poly(β-amino ester)s (HPAEs). The screening of HPAEs and optimization of therapeutic gene constructs, together with evaluation of the combined system for gene transfection, were comprehensively reviewed.
