Mycolactone enhances the Ca2+ leak from endoplasmic reticulum by trapping Sec61 translocons in a Ca2+ permeable state.

The Mycobacterium ulcerans exotoxin, mycolactone, is an inhibitor of co-translational translocation via the Sec61 complex. Mycolactone has previously been shown to bind to, and alter the structure of the major translocon subunit Sec61α, and change its interaction with ribosome nascent chain complexes. In addition to its function in protein translocation into the ER, Sec61 also plays a key role in cellular Ca2+ homeostasis, acting as a leak channel between the endoplasmic reticulum (ER) and cytosol. Here, we have analysed the effect of mycolactone on cytosolic and ER Ca2+ levels using compartme

Pioneering 1,3,5-triaryl pyrazolines as promising dual inhibitors of COX-2 and 15-LOX endowed with potent anticancer activity: design, synthesis, and in silico study.

Despite selective COX-2 inhibitors existing, they do not fully meet the demand for safer anti-inflammatory drugs. COX-2 and 15-LOX dual inhibition offers a promising avenue for developing safer NSAIDs and may play a crucial role in the fight against cancer. Thus, we designed and synthesized new 1,3,5-triaryl pyrazolines as dual COX-2/15-LOX inhibitors with potent anticancer activity. The synthesized compounds were evaluated in vitro as dual COXs/15-LOX inhibitors and for anticancer activity against human colorectal cancer cell lines HT-29 and HCT116. The pyrazolines 4a, 4c, 5h, and 5i exhibite

Discovery of new imidazole[1,2-a] pyridine derivatives as CDK9 inhibitors: design, synthesis and biological evaluation.

Colorectal cancer (CRC) is a highly aggressive and extensive malignancy. Presently, targeting the transcriptional regulation of cyclin-dependent kinase 9 (CDK9) is a promising therapeutic approach. Herein, twenty-five compounds (LA-1-LA-13 and LB-1-LB-12) were designed and synthesized with AZD5438 as the lead compound using an imidazole[1,2-a] pyridine skeleton. Compound LB-1 exhibited potent CDK9 inhibition and induced apoptosis in the HCT116 cell line. Moreover, compared with AZD5438, LB-1 demonstrated highly selective CDK9 inhibitory activity, with an IC50 value of 9.22 nM. Accordingly, com

Metastatic or xenograft colorectal cancer models induce divergent anabolic deficits and expression of pro-inflammatory effectors of muscle wasting in a tumor-type-dependent manner.

We investigated the impact of tumor burden on muscle wasting in metastatic (m) and xenograft (x) models of colorectal cancer (CRC). Male Nod SCID γ and CD2F1 mice were injected subcutaneously or intrasplenically with HCT116 or C26 tumor cells, respectively. CRC tumors resulted in significant muscle wasting regardless of tumor type or model, although muscle loss was exacerbated in mHCT116 hosts. The mHCT116 model decreased ribosomal (r)RNA content and rDNA transcription, whereas the mC26 model showed no loss of rRNA and the upregulation of rDNA transcription. The xHCT116 model reduced mTOR, RPS

Poly (ADP-ribose) polymerase inhibitors sensitize cancer cells to hypofractionated radiotherapy through altered selection of DNA double-strand break repair pathways.

PURPOSE: Poly (ADP-ribose) polymerase inhibitors (PARPi) are known to induce radiosensitization. However, the exact mechanisms of radiosensitization remain unclear. We previously reported that PARPi may have a unique radiosensitizing effect to enhance β-components of the linear-quadratic model. The aim of this study was to evaluate PARPi in combination with high-dose-per-fraction radiotherapy and to elucidate the underlying mechanisms of its radiosensitization. MATERIALS AND METHODS: Radiosensitizing effects of PARPi PJ34, olaparib, and veliparib were measured using a colony-forming assay in

Identification of benzoxazine scaffold derivatives as novel CDK4 inhibitors for the treatment of cancer.

Cyclin-dependent kinase 4, a pivotal regulator of cell cycle progression, is widely recognised as a therapeutic target for controlling tumour proliferation. Inspired by the reported CDK4 inhibitor PF-07220060, a series of novel benzoxazine derivatives were designed, synthesised, and evaluated for their biological activity. From this series, compound 8j emerged as the most potent anti-proliferative agent. Subsequent in vitro studies showed 8j effectively inhibits HCT116 cell proliferation, clonogenic survival, and migration. Mechanistic investigations revealed that 8j induces G1 phase cell cycl

Discovery of the First Highly Potent and Selective Peptide Inhibitor Targeting Microtubule-MAP7 Interaction via Structure-Based Design for Suppressing Colorectal Cancer Cell Proliferation.

Microtubules are core cytoskeletal components for cellular activities. Their interaction with microtubule-associated protein 7 (MAP7) is crucial for cell proliferation, cellular component transport, and morphological regulation. The conserved microtubule surface region binds tightly to MAP7's microtubule-binding domain (MTBD). Its function relates closely to tumorigenesis and progression. Using structure-based drug design, we first designed peptides 1-3, peptide inhibitors targeting microtubule-MAP7 interaction. These peptides disrupt microtubule-MAP7 interaction by specifically binding to mic

Fast High-Resolution Laser Ablation-Inductively Coupled Plasma Mass Spectrometry Imaging of the Distribution of Platinum-Based Anticancer Compounds in Multicellular Tumor Spheroids.

Multicellular tumor spheroid models serve as an important three-dimensional in vitro cell model system as they mimic the complex tumor microenvironment and thus have contributed to valuable assays in drug discovery studies. In this study, we present a state-of-the-art laser ablation-inductively coupled plasma mass spectrometry (LA-ICPMS) setup for high spatial resolution elemental imaging of multicellular tumor spheroids and an approach to account for variations in cell density. A low dispersion LA-ICPMS setup was employed, providing accelerated throughput and high sensitivity and permitting a

Mutated CEACAMs Disrupt Transforming Growth Factor Beta Signaling and Alter the Intestinal Microbiome to Promote Colorectal Carcinogenesis.

BACKGROUND & AIMS: We studied interactions among proteins of the carcinoembryonic antigen-related cell adhesion molecule (CEACAM) family, which interact with microbes, and transforming growth factor beta (TGFB) signaling pathway, which is often altered in colorectal cancer cells. We investigated mechanisms by which CEACAM proteins inhibit TGFB signaling and alter the intestinal microbiome to promote colorectal carcinogenesis. METHODS: We collected data on DNA sequences, messenger RNA expression levels, and patient survival times from 456 colorectal adenocarcinoma cases, and a separate set of

Association between variation of circulating 25-OH vitamin D and methylation of secreted frizzled-related protein 2 in colorectal cancer.

BACKGROUNDS: Colorectal cancer (CRC) results from the accumulation of epigenetic and genetic changes in colon cells during neoplasic transformation, which the activation of Wingless (Wnt) signaling pathway is a common mechanism for CRC initiation. The Wnt pathway is mainly regulated by Wnt antagonists, as secreted frizzled-related protein (SFRP) family. Indeed, SFRP2 is proposed as a noninvasive biomarker for CRC diagnosis. Vitamin D also antagonizes Wnt signaling in colon cancers cells. Several studies showed that vitamin D was able to alter DNA methylation, although this mechanism is not yet

Downregulation of lncRNA H19 Enhances Radiosensitivity in Colorectal Cancer by Inactivation of the ATR/CHK1 Signaling.

Radiotherapy (RT) is commonly used in the treatment of colorectal cancer (CRC), but the resistance of cancer cells to radiation remains a significant challenge in clinical practice. This study aimed to investigate whether long non-coding RNA (lncRNA) H19 could affect the sensitivity of CRC cells to radiation. The levels of H19 in CRC tissues and adjacent normal tissues, as well as in radioresistant and radiosensitive CRC tissues, were evaluated using RT-qPCR. The levels of H19 were found to be higher in CRC tissues compared to adjacent normal tissues, as well as in radioresistant CRC tissues c
