Cuticular proteins (CPs) are a large family and perform a variety of functions. However, the physiological roles of cuticle protein 21-like (Cpr21L) in the brown planthopper (Nilaparvata lugens, BPH), one of the most destructive insect pests of rice, are largely unclear. In this study, Cpr21L was revealed to be expressed in both BPH nymphs and adults, and the mRNA expression level was much higher in male adults than female adults. Spatially, the expression of Cpr21L in the testis was higher than in the ovary. The RNA interference (RNAi) of Cpr21L seriously decreased nymph survival, and no indi

Begomoviruses constitute an extremely successful group of emerging plant viruses transmitted by whiteflies of the Bemisia tabaci complex. Hosts include important vegetable, root, and fiber crops grown in the tropics and subtropics. Factors contributing to the ever-increasing diversity and success of begomoviruses include their predisposition to recombine their genomes, interaction with DNA satellites recruited throughout their evolution, presence of wild plants as a virus reservoir and a source of speciation, and extreme polyphagia and continuous movement of the insect vectors to temperate reg

BACKGROUND: Tomato chlorosis virus (ToCV) is mainly vectored by Bemisia tabaci in China, which has a worldwide distribution, and greatly reduces the yields of tomato and other vegetables. At present, control of ToCV has been focused mainly by the use of insecticides to control whitefly populations. Transcriptome sequencing showed high expression of the B. tabaci Bta11975 gene, an α-glucosidase (AGLU) during ToCV acquisition by whitefly Mediterranean (MED) species. To investigate the role of Bta11975 gene in ToCV acquisition and transmission by B. tabaci MED, we used RNA interference (RNAi) to

The fungal genus Massospora (Zoopagomycota: Entomophthorales) includes more than a dozen obligate, sexually transmissible pathogenic species that infect cicadas (Hemiptera) worldwide. At least two species are known to produce psychoactive compounds during infection, which has garnered considerable interest for this enigmatic genus. As with many Entomophthorales, the evolutionary relationships and host associations of Massospora spp. are not well understood. The acquisition of M. diceroproctae from Arizona, M. tettigatis from Chile, and M. platypediae from California and Colorado provided an op…

Zebra chip disease (ZC) in potato (Solanum tuberosum L. [Polemoniales: Solanaceae]) can produce unmarketable tubers with striped necrotic patterns. ZC is associated with the bacterium "Candidatus Liberibacter solanacearum" (Lso), which is transmitted by the potato psyllid, Bactericera cockerelli (Šulc) (Hemiptera: Triozidae). Potato psyllids are associated with numerous noncrop host plants, especially from the Solanaceae and Convolvulaceae; however, the contribution and importance of these hosts to ZC epidemiology in potato is poorly understood. To clarify seasonal phenologies on two such host…

Planthoppers are serious rice pests in Asia. Their population resurgence was first reported in the early 1960s, caused mainly by insecticides that indiscriminately killed beneficial arthropods and target pests. The subsequent resurgence involved two mechanisms, the loss of beneficial insects and insecticide-enhanced planthopper reproduction. In this review, we identify two forms of resurgence, acute and chronic. Acute resurgence is caused by traditional insecticides with rapid resurgence in the F1 generation. Chronic resurgence follows application of modern pesticides, including fungicides and

UNLABELLED: BACKGROUND Unlike synthetic pesticides, azadirachtin-based neem pesticides are environmentally friendly and are well known for their diverse pest control properties. Their use is, however, limited by the instability of azadirachtin, necessitating application at short intervals. The efficacy of relatively stable tetrahydroazadirachtin-A, therefore, needed to be established under field conditions.
RESULTS: Azadirachtin-A (Aza-A), its stable derivative tetrahydroazadirachtin-A (THA) and other neem pesticides have been evaluated for their field efficacy against major insect pests of o

Candidatus Liberibacter asiaticus (CLas) enhances the fecundity of Diaphorina citri by manipulating D. citri long noncoding RNA 5 (Dclnc5), revealing an lncRNA-mediated regulatory role in pathogen-vector interactions. We found that CLas infection significantly increased D. citri fecundity and markedly upregulated vitellogenin (Vg) at both the transcript and protein levels. Ovary lncRNA-seq comparing CLas-positive (CLas+) and CLas-negative (CLas-) identified differentially expressed lncRNAs whose predicted targets were enriched in cellular signaling, lipid metabolism, and fat digestion and abso

Grapevine red blotch virus (GRBV) reduces grapevine vigor, delays grape ripening, reduces sugar content and quality of wines in North America and other continents. To date, the three-cornered alfalfa hopper, Spissistilus festinus, has been clearly identified as a vector in the United States. Since this hopper has not been observed in some vineyards where secondary spread occurs, additional vectors of GRBV are likely. To search for vectors, we conducted a two-year survey collecting live hemipterans from infected vines and surrounding grass, trees, and shrubs. Screening a wide variety of hemipte…

Symbiotic partnerships have opened new ecological niches and contributed to the remarkable diversification of insects. The leafhopper Scaphoideus titanus, a phloem-feeding insect known to be the primary vector of Flavescence dorée phytoplasma, harbours two primary endosymbionts: the bacterium 'Candidatus Karelsulcia muelleri' and a yeast-like symbiont (YLS). While most studies on insect-associated microorganisms have focused on obligate bacterial symbionts, fungal endosymbionts, although documented for almost a century, are only now gaining renewed attention for their evolutionary and ecologic

Controlling the Asian citrus psyllid (ACP), Diaphorina citri, is key to preventing spread of huanglongbing, a disease that has decimated citrus industries worldwide. One promising control method is the Sterile Insect Technique (SIT). This involves rearing large numbers of a target pest, sterilizing them using radiation, and releasing them into the field to compete with wild conspecifics for mating, ultimately suppressing population growth. The success of SIT is largely influenced by 3 factors: (i) ensuring released insects are sufficiently sterile such that mating between wild and sterile indi

Defensive symbioses in which beneficial microbes protect hosts from natural enemies are ubiquitous across animals and plants, but the underlying mechanisms remain poorly understood. Field surveys and laboratory assays revealed that infection of the invasive whitefly Bemisia tabaci by the bacterial symbiont Rickettsia and plant begomovirus were positively correlated with each other but each negatively correlated with a parasitic fungal infection (Beauveria bassiana) in the host. We show that begomovirus conferred whitefly's resistance to the parasitic fungus by triggering the expression of chit

Most problems caused by the use of common pesticides can be avoided or reduced by the use of well-established, safe, unique, and selective organic compounds. For example, juvenile hormone analogues exhibit promise as insect growth regulators (IGRs) due to their effective mechanism of action against pests and lower toxicity to children compared to conventional pesticides. Thus, as analogues of insect growth regulators, a special class of pure 1-(1-naphthalenyloxy)-2-propanol derivatives has been created. Through elemental and modern spectroscopy investigations (infrared (IR), ultraviolet (UV),

UNLABELLED: Infection with pathogens can increase the fecundity and other fitness-related traits of insect vectors for their own advantage. Our previous research has reported the pivotal role of DcKr-h1 in the fecundity improvement of Diaphorina citri induced by the bacterium, "Candidatus Liberibacter asiaticus" (CLas), and the fungus, Cordyceps fumosorosea (Cf). However, the posttranscriptional regulation of this process remains poorly understood. Given the significance of miRNAs in gene regulation, we delved into their roles in shaping phenotypes and their underlying molecular mechanisms. Ou

UNLABELLED: Most arthropod-borne viruses produce intermittent epidemics in infected plants. However, the underlying mechanisms of these epidemics are unclear. Here, we demonstrated that rice stripe mosaic virus (RSMV), a viral pathogen, significantly increases the mortality of its overwintering vector, the leafhopper species Recilia dorsalis. Cold-stress assays indicated that RSMV reduces the cold tolerance of leafhoppers, a process associated with the downregulation of leafhopper cuticular protein genes. An RSMV-derived small RNA (vsiR-t00355379) was found to facilitate the downregulation of

In many insects the surface of the eye is nanostructured by arrays of protuberances termed ommatidial gratings which provide the cuticle with anti-reflective, anti-wetting and self-cleaning properties. The hypothesised anti-contamination role of the gratings against dust and pollen results from theoretical predictions on grating geometry and experiments on synthetic replicas of ommatidia surfaces but has not yet been proven in an animal. Whiteflies are biological test beds for anti-contamination surfaces because they deliberately distribute wax particles extruded from abdominal plates over the

Exosomes play a key role in virus exocytosis and transmission. The exportin family is usually responsible for cargo nucleocytoplasmic trafficking, and they are frequently found in exosomes. However, the function of exportins sorted in exosomes remains unknown. Here, we successfully isolated "cup holder"-like exosomes from the saliva of ∼30,000 small brown planthoppers, which are vectors of rice stripe virus (RSV). RSV virions were packed in comparatively large exosomes. Four viral genomic RNAs at a certain ratio were identified in the saliva exosomes. The virions contained in the saliva exosom

The Asian citrus psyllid, Diaphorina citri Kuwayama, is among the most important pests of citrus. It is the main vector of the Huanglongbing (HLB) pathogen Candidatus Liberibacter asiaticus (CLas), which causes severe losses in citrus crops. Control of D. citri is therefore of paramount importance to reduce the spread of HLB. In this regard, using RNA interference (RNAi) to silence target genes is a useful strategy to control psyllids. In this study, using RNAi, we examined the biological functions of the V-ATPase subunit E (V-ATP-E) gene of D. citri, including its effect on acquisition of CLa

BACKGROUND: The Asian Citrus Psyllid (ACP), Diaphorina citri, can transmit the bacterium Candidatus Liberibacter while feeding on citrus flush shoots. This bacterium causes Huanglongbing (HLB), a major disease of citrus cultivation worldwide necessitating the development of new tools for ACP surveillance and control. The olfactory system of ACP is sensitive to variety of odorants released by citrus plants and offers an opportunity to develop new attractants and repellents.
RESULTS: In this study, we performed single-unit electrophysiology to identify odorants that are strong activators, inhib

The neurons innervating muscles that deliver the enormous power enabling froghopper insects to excel at jumping were revealed by backfilling the nerves from those muscles. The huge trochanteral depressor muscle (M133) of a hind leg consists of four parts. The two largest parts (M133b,c) occupy most of the metathorax and are innervated by the same two motor neurons that have small, laterally placed somata in the metathoracic ganglion and axons in nerve N3C(2). They are also supplied by three dorsal unpaired median (DUM) neurons with the largest diameter somata in the central nervous system. A s
