Age-associated trajectories of hippocampus subregion volumes from childhood through later adulthood.

The hippocampus has been widely implicated in the neurobiology of neurologic and psychiatric disorders. This structure is heterogeneous, however, and comprised of multiple subregions that have different connectivity patterns and functions. Better understanding how these subregions are affected by age and their relationship with neurocognition could provide information regarding how alterations in normal trajectories play a role in disease and cognitive dysfunction. Using natural splines we modeled the trajectory of 4 hippocampus subregions (i.e., CA composite, DG composite, tail and subiculum)…

Endo-Lysosomal Network Disorder Reprograms Energy Metabolism in SorL1-Null Rat Hippocampus.

Sortilin-related receptor 1 (SorL1) deficiency is a genetic predisposition to familial Alzheimer's disease (AD), but its pathology is poorly understood. In SorL1-null rats, a disorder of the global endosome-lysosome network (ELN) is found in hippocampal neurons. Deletion of amyloid precursor protein (APP) in SorL1-null rats could not completely rescue the neuronal abnormalities in the ELN of the hippocampus and the impairment of spatial memory in SorL1-null young rats. These in vivo observations indicated that APP is one of the cargoes of SorL1 in the regulation of the ELN, which affects hippo…

Serotonin Signaling in Hippocampus during Initial Cocaine Abstinence Drives Persistent Drug Seeking.

The initiation of abstinence after chronic drug self-administration is stressful. Cocaine-seeking behavior on the first day of the absence of the expected drug (Extinction Day 1, ED1) is reduced by blocking 5-HT signaling in dorsal hippocampal cornu ammonis 1 (CA1) in both male and female rats. We hypothesized that the experience of ED1 can substantially influence later relapse behavior and that dorsal raphe (DR) serotonin (5-HT) input to CA1 may be involved. We inhibited 5-HT1A/1B receptors (WAY-100635 plus GR-127935), or DR input (chemogenetics), in CA1 on ED1 to test the role of this pathwa

Electroconvulsive therapy disrupts functional connectivity between hippocampus and posterior default mode network.

BACKGROUND: The mechanisms underlying memory deficits after electroconvulsive therapy (ECT) remain unclear but altered functional interactions between hippocampus and neocortex may play a role. OBJECTIVES: To test whether ECT reduces functional connectivity between hippocampus and posterior regions of the default mode network (DMN) and to examine whether altered hippocampal-neocortical functional connectivity correlates with memory impairment. A secondary aim was to explore if these connectivity changes are present 6 months after ECT. METHODS: In-patients with severe depression (n = 35) rece…

Adipokine ZAG Alters Depression-Like Behavior by Regulating Oxidative Stress in Hippocampus.

Zinc-α2-glycoprotein (ZAG) is an adipokine involved in body metabolism, and now it has been shown to be present in the brain and play a role in some neurological diseases such as epilepsy and Alzheimer's disease. In the present study, we employed ZAG knockout (KO) mice to investigate the effects of ZAG on behaviors after fasting and in vitro used overexpression (OV) ZAG in HT-22 cells to further clarify the possibly underlying mechanism. The results showed that ZAG exists widely in the brain tissues of mice and significantly increased during fasting. In ZAG KO group the depression-like behavio

Hippocampus-Based Mitochondrial Respiratory Function Decline Is Responsible for Perioperative Neurocognitive Disorders.

Perioperative neurocognitive disorders (PNDs) are a type of cognitive dysfunction occurring with a higher incidence in elderly patients. However, the pathological mechanism of PND and effective treatment remain elusive. We generated a PND mouse model by providing wild-type mice with surgical trauma; in our case, we used tibial fracture to investigate PND pathology. Mice aged 7-8 months were randomly divided into two groups: the surgery (tibial fracture) group and the control (sham) group. All mice were subjected to anesthesia. We examined the transcriptome-wide response in the hippocampus, a b…

Lactate induces synapse-specific potentiation on CA3 pyramidal cells of rat hippocampus.

Neuronal activity within the physiologic range stimulates lactate production that, via metabolic pathways or operating through an array of G-protein-coupled receptors, regulates intrinsic excitability and synaptic transmission. The recent discovery that lactate exerts a tight control of ion channels, neurotransmitter release, and synaptic plasticity-related intracellular signaling cascades opens up the possibility that lactate regulates synaptic potentiation at central synapses. Here, we demonstrate that extracellular lactate (1-2 mM) induces glutamatergic potentiation on the recurrent collate

Environmental noise exposure modifies astrocyte morphology in hippocampus of young male rats.

BACKGROUND: Chronic exposure to noise induces changes on the central nervous system of exposed animals. Those changes affect not only the auditory system but also other structures indirectly related to audition. The hippocampus of young animals represents a potential target for these effects because of its essential role in individuals' adaptation to environmental challenges. OBJECTIVE: The aim of the present study was to evaluate hippocampus vulnerability, assessing astrocytic morphology in an experimental model of environmental noise (EN) applied to rats in pre-pubescent stage. MATERIALS A…

Coxsackievirus Adenovirus Receptor Loss Impairs Adult Neurogenesis, Synapse Content, and Hippocampus Plasticity.

UNLABELLED: Although we are beginning to understand the late stage of neurodegenerative diseases, the molecular defects associated with the initiation of impaired cognition are poorly characterized. Here, we demonstrate that in the adult brain, the coxsackievirus and adenovirus receptor (CAR) is located on neuron projections, at the presynapse in mature neurons, and on the soma of immature neurons in the hippocampus. In a proinflammatory or diseased environment, CAR is lost from immature neurons in the hippocampus. Strikingly, in hippocampi of patients at early stages of late-onset Alzheimer's…

Psychological stress induced hippocampus zinc dyshomeostasis and depression-like behavior in rats.

There are strong evidences showed that psychological stress (PS) could result in depression. Recently, many attentions were paid to the roles of corticosterone (CORT) and zinc dyshomeostasis in the development of depression. In this study, we investigated the zinc level in rat hippocampus after exposure to PS and accompanied behavior change. Male SD rats were randomly divided into the control and PS groups. Each group had two subgroups: 7-d group and 14-d group. A communication box was used to produce the PS model in rats. Compared to control group, the PS-treated group showed decreased total …

Acute high-intensity sound exposure alters responses of place cells in hippocampus.

Overstimulation is known to activate neural plasticity in the auditory nervous system causing changes in function and re-organization. It has been shown earlier that overstimulation using high-intensity noise or tones can induce signs of tinnitus. Here we show in studies in rats that overstimulation causes changes in the way place cells of the hippocampus respond as rats search for rewards in a spatial maze. In familiar environments, a subset of hippocampal pyramidal neurons, known as place cells, respond when the animal moves through specific locations but are relatively silent in others. Thi…

Distributed neural codes of the 3D position in the marmoset frontal cortex and hippocampus.

The neural encoding of the animal's position is essential for spatial navigation and cognition, yet the three-dimensional (3D) positioning system in the primate brain remains poorly understood. By combining wireless multi-channel recordings from common marmosets with a free-forging task in the 3D space, we identified 42% and 24% neurons in the frontal cortex (FC) and hippocampus (HPC) as place cells, respectively. Place cells in FC and HPC encoded the 3D position with similar spatial information, sparsity and stability, and exhibited stronger functional connections than non-place cells. Notabl…

Synaptic density in the hippocampus of depressed patients: A quantitative electron microscopic study.

BACKGROUND: Synaptic dysfunction or loss of synapses is implicated in the pathophysiology of major depressive disorder (MDD). Recent in vivo functional neuroimaging studies document reduced number of synapses in the brains of MDD patients. Here, we performed a post-mortem quantitative electron microscopic study to confirm the putative synaptic loss in the hippocampus of patients with depression. METHODS: Hippocampal tissue from subjects with MDD (n = 11, aged 19-79 years) and psychiatrically healthy controls (n = 17, aged 18-77 years) were investigated. A systematic quantitative analysis was …
