Immunome profiling in prostate cancer: a guide for clinicians.

Tumor immune microenvironment (TIME) plays a key role to understand how tumors respond to prostate cancer (PC) therapies and potential mechanisms of resistance. Previous research has suggested that specific genomic aberrations, such as microsatellite instability (MSI) or CDK12 bi-allelic loss can allow PC patients more likely to respond to immune checkpoint inhibitors (ICI) or other immune therapies. However, responses to these treatments remain highly variable even in selected patients. Thus, it is essential to obtain more information about tumor immune cells that infiltrate these tumors, and

Lipid nanoparticle-based multi-scale systemic immune programming for cancer therapy.

The clinical reach of cancer immunotherapy is currently limited by off-target toxicity, physical barriers in solid tumors, and the complex manufacturing of personalized cell therapies. This review presents multi-scale systemic immune programming as a transformative approach, in which lipid nanoparticles (LNPs) function not merely as delivery vehicles but as programmable platforms that direct immune responses across biological scales. At the molecular and cellular levels, LNP design and intracellular delivery regulate mRNA stability, translation, and antigen presentation. At the tissue level, i

Common Mechanisms of Viral Injury to the Kidney.

Viral infections in an immunocompetent host can cause both acute and chronic kidney diseases, either by direct damage to the infected kidney cells or as a consequence of systemic immune responses that impact the kidneys' function. Viruses have evolved mechanisms to hijack signaling pathways of the infected cell, including the mammalian target of rapamycin pathway to support viral replication, and to evade antiviral immune responses such as those mediated by miR-155 via microRNA mimetics expressed by the virus. At both the cellular and systemic levels, the host has also evolved mechanisms to co

Isolated glycoprotein STK11 from the nutshell of Cocos nucifera L. modulated macrophage polarization to prevent the Mycobacterium tuberculosis bacilli growth in J774A.1 macrophage.

This study examines the importance of host-directed therapy against tuberculosis by targeting macrophage polarization with Cocos nucifera L. nutshell-derived STK11. RT-PCR results revealed that M2 markers (Arg1, YM-1, FIZZ1, CD206, IL-4, IL-10) were upregulated, while M1 markers (CD40, CD68, IL-6, IL-12) were downregulated in H37Ra-infected J774A.1 macrophages. Isolated glycoprotein STK11 significantly (p < 0.05) restricted the growth of Mycobacterium tuberculosis bacilli in the Middlebrook 7H9 medium and inhibited H37Ra bacilli growth within macrophages. STK11 (10 μg/ml) was also found to dec…

rs1051838 Promotes Intracellular Survival of Mycobacterium tuberculosis H37Ra by Regulating DUSP14 Expression.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a major global health burden. Host genetic factors play a critical role in TB susceptibility, but the underlying mechanisms are not fully understood. Genome-wide association studies have identified DUSP14 as a TB susceptibility gene, and the single nucleotide polymorphism (SNP) rs1051838 in DUSP14 seems to be associated with TB risk. Using in vitro models including THP-1-derived macrophages, A549 cells, and HEK293T cells, we investigated how rs1051838 regulates DUSP14 expression and modulates macrophage responses to Mtb inf

Targeting Pseudomonas aeruginosa biofilm with an evolutionary trained bacteriophage cocktail exploiting phage resistance trade-offs.

Spread of multidrug-resistant Pseudomonas aeruginosa strains threatens to render currently available antibiotics obsolete, with limited prospects for the development of new antibiotics. Lytic bacteriophages, the viruses of bacteria, represent a path to combat this threat. In vitro-directed evolution is traditionally applied to expand the bacteriophage host range or increase bacterial suppression in planktonic cultures. However, while up to 80% of human microbial infections are biofilm-associated, research towards targeted improvement of bacteriophages' ability to combat biofilms remains scarce

Novel Dormancy Mechanism of Castration Resistance in Bone Metastatic Prostate Cancer Organoids.

Advanced prostate cancer (PCa) patients with bone metastases are treated with androgen pathway directed therapy (APDT). However, this treatment invariably fails and the cancer becomes castration resistant. To elucidate resistance mechanisms and to provide a more predictive pre-clinical research platform reflecting tumor heterogeneity, we established organoids from a patient-derived xenograft (PDX) model of bone metastatic prostate cancer, PCSD1. APDT-resistant PDX-derived organoids (PDOs) emerged when cultured without androgen or with the anti-androgen, enzalutamide. Transcriptomics revealed u

Human monocyte-derived macrophage responses to M. tuberculosis differ by the host's tuberculosis, diabetes or obesity status, and are enhanced by rapamycin.

Human macrophages play a major role in controlling tuberculosis (TB), but their anti-mycobacterial mechanisms remain unclear among individuals with metabolic alterations like obesity (TB protective) or diabetes (TB risk). To help discern this, we aimed to: i) Evaluate the impact of the host's TB status or their comorbidities on the anti-mycobacterial responses of their monocyte-derived macrophages (MDMs), and ii) determine if the autophagy inducer rapamycin, can enhance these responses. We used MDMs from newly diagnosed TB patients, their close contacts and unexposed controls. The MDMs from TB

Graft-versus-Host Disease after Living-Unrelated Kidney Transplantation.

Graft-versus-host disease (GVHD) is a rare complication after solid organ transplantation and consists of a reaction of donor derived immune cells directed against host tissues. The vast majority of cases reported in the literature involve liver, small intestine and pancreas transplantation. We report a case of GVHD in a 48-year-old man after living-unrelated kidney transplantation at another center. Six months postoperatively he developed a skin rash, anorexia, and diarrhea that resulted in malnutrition and a 90 pound weight loss. At this point he was transferred to our center with a BMI of 1

Pandemic Response Box Screening Identified CRM1/XPO1 as an Anti-Mammarenavirus Druggable Target.

Mammarenaviruses (MaAv) cause persistent infection in their natural rodent hosts across the world and via zoonotic events can cause severe disease in humans. Thus, the MaAv Lassa virus (LASV) in Western Africa and Junin virus (JUNV) in the Argentinean Pampas cause hemorrhagic fever diseases with significant case fatality rates in their endemic regions. In addition, the globally distributed MaAv lymphocytic choriomeningitis virus (LCMV) is an underrecognized human pathogen of clinical significance capable of causing devastating infections in neonates and immunocompromised individuals. The best of both worlds - managing the cancer, saving the bone.

In the context of breast cancer, the importance of the skeleton in the regulation of primary tumour development and as a site for subsequent metastasis is well characterized. Our understanding of the contributions made by the host bone and bone marrow cells increasingly demonstrates the extent of the interaction between tumour cells and normal host cells. As a result, the need to develop and utilize therapies that can impede the growth and/or function of tumour cells while sparing normal host bone and bone marrow cells is immense and expanding. The need for these new treatments is, however, su

Immunomodulatory therapies for cystic fibrosis.

Progressive pulmonary disease is the primary cause of morbidity and mortality in adult patients with cystic fibrosis (CF). The decrease in lung function associated with infection with Pseudomonas aeruginosa has been related to the severity of pulmonary inflammation. Thus therapies that reduce pulmonary inflammation may prove to be clinically efficacious. Therapeutic interventions that target pulmonary inflammation may be directed either at the infecting organism, especially P aeruginosa, or at host responses. Eradication of P aeruginosa from the airways of patients with CF has not been accompl

IFNs in host defence and parasite immune evasion during Toxoplasma gondii infections.

Interferons (IFNs) are a family of cytokines with diverse functions in host resistance to pathogens and in immune regulation. Type II IFN, i.e. IFN-γ, is widely recognized as a major mediator of resistance to intracellular pathogens, including the protozoan Toxoplasma gondii. More recently, IFN-α/β, i.e. type I IFNs, and IFN-λ (type III IFN) have been identified to also play important roles during T. gondii infections. This parasite is a widespread pathogen of humans and animals, and it is a model organism to study cell-mediated immune responses to intracellular infection. Its success depends,

"Envbiotics" -- a novel framework for microbiota-targeted therapeutic strategies in type 2 diabetes mellitus.

Imbalance of the gut microbiota is an important trigger for insulin resistance in type 2 diabetes mellitus(T2DM). Microbiota-targeted therapies have gradually become an emerging research direction for treating T2DM. However, exogenous strain interventions (supplementing probiotics and fecal microbiota transplantation), endogenous optimization of the gut microbiota (supplementing prebiotics, synbiotics, and postbiotics), and phage therapy focus on "directly introducing microorganisms," "feeding microorganisms," or "directly utilizing microbial components." These approaches cannot cover active s

In Vivo Activity of Repurposed Amodiaquine as a Host-Targeting Therapy for the Treatment of Anthrax.

Anthrax is caused by Bacillus anthracis and can result in nearly 100% mortality due in part to anthrax toxin. Antimalarial amodiaquine (AQ) acts as a host-oriented inhibitor of anthrax toxin endocytosis. Here, we determined the pharmacokinetics and safety of AQ in mice, rabbits, and humans as well as the efficacy in the fly, mouse, and rabbit models of anthrax infection. In the therapeutic-intervention studies, AQ nearly doubled the survival of mice infected subcutaneously with a B. anthracis dose lethal to 60% of the animals (LD60). In rabbits challenged with 200 LD50 of aerosolized B. anthra
