Mechanisms of pathogenesis and the evolution of parasite virulence.

When studying how much a parasite harms its host, evolutionary biologists turn to the evolutionary theory of virulence. That theory has been successful in predicting how parasite virulence evolves in response to changes in epidemiological conditions of parasite transmission or to perturbations induced by drug treatments. The evolutionary theory of virulence is, however, nearly silent about the expected differences in virulence between different species of parasite. Why, for example, is anthrax so virulent, whereas closely related bacterial species cause little harm? The evolutionary theory mig

Deciphering Kiwifruit Bacterial Canker: Molecular Insights, Host-Pathogen Dynamics, and Innovative Management Strategies.

Kiwifruit bacterial canker, caused by Pseudomonas syringae pv. actinidiae (Psa), severely threatens global kiwifruit production. Psa uses type III secretion, effectors, biofilms, and hormone manipulation to suppress the host immunity and achieve successful colonization. In response, kiwifruit activates multilayered defense systems involving pattern-triggered immunity, effector-triggered immunity, structural barriers, reactive oxygen species signaling, and hormone-mediated defense pathways. However, these immune mechanisms remain insufficiently validated in Actinidia species, limiting the trans

The Pathology of Viral Infections.

If you were to ask, "Biologically speaking, what are humans made of?", almost everyone would reply "Human cells and molecules, of course." This seemingly logical answer, however, does not truly capture the diversity of the human organism. Over the past couple of decades scientists have discovered that humans are a collective of cohabitating human, bacteria, and fungi cells along with countless numbers of viruses. Collectively, these microorganisms are referred to as the microbiome (Lederberg and McCray, The Scientist 15:8, 2001). The most recent estimates suggest the biological material from t

Beyond toxicity: Systems-level virulence of entomopathogenic fungi in termite social immunity.

Termites live in dense colonies where frequent social contact can facilitate pathogen transmission, but coordinated social immunity can also limit disease spread. Entomopathogenic fungi (EPF) are particularly relevant to termite pathology because they infect primarily through the cuticle, using adhesion, enzymatic degradation, penetration, and internal proliferation to overcome host barriers. This review synthesizes current knowledge on EPF virulence in termites and argues that fungal pathogenicity should be understood not only as direct toxicity (lethal effects of pathogen-derived compounds)

Is a Commensal Here, a Pathogen There? Genotype Specific Responses by the Sea Anemone Nematostella vectensis to Vibrio Species.

Animals rely on diverse molecular mechanisms to maintain beneficial microbial associations while also defending against pathogens, yet the extent to which these responses vary among genotypes of a single species remains poorly understood. Using individuals from different locations of the sea anemone Nematostella vectensis, we compared transcriptional responses to a commensal (Vibrio diabolicus) and pathogen (Vibrio coralliilyticus) bacterium. We find striking genotype-specific divergence: individuals from Nova Scotia mounted a strong transcriptional response to V. diabolicus, whereas the North

Novel infection by Mucor hiemalis kills Caenorhabditis hosts through intestinal perforation.

The nematode Caenorhabditis elegans has emerged as a popular model system to investigate cell biology and host-pathogen interactions. Presently, C. elegans is studied as a natural host of intracellular pathogens such as microsporidia and Orsay virus, along with extracellular bacterial and fungal pathogens. The use of C. elegans as a model in host-pathogen research is limited by the number of naturally occurring pathogens that infect this organism. Through a sampling project to identify new pathogens of C. elegans, we identified the fungus Mucor hiemalis as a pathogen of Caenorhabditis species.…

mGem: A perfect storm in the era of global warming-the convergence between thermotolerant fungi and altered immunity.

The evidence supporting the impact of global warming on the epidemiology of infectious diseases, including fungal infections, is increasing. Fungi have a remarkable ability to adapt to heat and pollution, and to disseminate via air, water ecosystems, or wildfire smoke. Their genetic plasticity can lead to thermotolerance, the ability to find new ecological niches, and fitness gains. Natural reservoirs of the fungal biomass, which are heavily affected by global warming, may serve as the environments from where new fungal diseases originate, as illustrated by the recent emergence of Candidozyma

Substituents of the polysaccharide region of LPS in Legionella pneumophila sg1 modulate interactions with host cells.

Legionella pneumophila is a Gram-negative bacterium commonly found in natural freshwater reservoirs, where it exists as an intracellular parasite of cohabiting protozoa, primarily of the genus Acanthamoeba. Inhalation of aerosol contaminated with the bacterium leads to its proliferation within pulmonary macrophages, ultimately resulting in pneumonia known as Legionnaires' disease. The lipopolysaccharide (LPS) of L. pneumophila sg1 constitutes the most exposed component of the bacterial cell wall, playing a key role in every stage of its developmental cycle associated with host cells. The orf8

Gut-microbiota-mediated host immune modulation: mechanisms, pathological dysbiosis, and therapeutic frontiers.

The mammalian immune system has evolved in constant dialogue with its diverse microbiota, forming an ecological and molecular partnership that is fundamental to health. This review examines how microbial communities shape immunity across developmental and functional axes, the immunological consequences of dysbiosis during infection and inflammatory disease, and emerging microbiota-targeted interventions. The host-microbiota-pathogen triad offers a framework to understand how commensals and pathogens compete for ecological niches and immune recognition, and how disturbances in this balance can

Stripe Rust Virulence and Host Reaction Assessment Protocols: From Lab to Field.

Puccinia striiformis, the causal agent of stripe rust disease, poses a significant threat to cereal crops worldwide. Since 2000, the emergence of heat-adapted races/lineages of the wheat-infecting form P. striiformis f. sp. tritici (Pst) has enabled the disease to spread beyond its traditional high-elevation, cool, and moist regions. It can now occur wherever wheat is cultivated, making it a pandemic concern. This chapter provides an overview of the methodologies and practices established in our lab for working with this pathogen. These methodologies include isolate collection, recovery, purif

A Novel Pathosystem With the Model Plant Arabidopsis thaliana for Defining the Molecular Basis of Taphrina Infections.

Plant-associated yeasts modulate host immunity to promote or prevent disease. Mechanisms of yeast perception by the plant innate immune system remain unknown, with progress hindered by lack of a model system with the model plant Arabidopsis thaliana (Arabidopsis). A yeast strain of Taphrina tormentillae, named M11, was previously isolated from wild Arabidopsis. Taphrina have been found on many non-host plants, and their complex ecology remains understudied. Here, the interaction of M11 with Arabidopsis was characterised. Infection of Arabidopsis with the birch pathogen T. betulina, used as a n

Chlamydia trachomatis: a model for intracellular bacterial parasitism.

Chlamydia comprises a diverse group of obligate intracellular bacteria that cause infections in animals, including humans. These organisms share fascinating biology, including distinct developmental stages, non-canonical cell surface structures, and adaptations to intracellular parasitism. Chlamydia trachomatis is of particular interest due to its significant clinical importance, causing both ocular and sexually transmitted infections. The strain L2/434/Bu, responsible for lymphogranuloma venereum, is the most common strain used to study chlamydial molecular and cell biology because it grows r

Unraveling the Intricacies of Xenophagy: Lessons from Legionella pneumophila.

Autophagy, a conserved eukaryotic process for maintaining cellular homeostasis, plays a crucial role in innate immunity by targeting intracellular pathogens through selective autophagy, known as xenophagy. While xenophagy is vital for pathogen clearance, many intracellular bacteria have evolved sophisticated strategies to avoid or subvert this process. Legionella pneumophila, a Gram-negative intracellular pathogen, manipulates host pathways by an extensive repertoire of effector proteins delivered through its type IV secretion system. This review summarizes the current understanding of xenopha

Harnessing miRNAs: A Novel Approach to Diagnosis and Treatment of Tuberculosis.

Mycobacterium tuberculosis (Mtb) complex, responsible for tuberculosis (TB) infection, continues to be a predominant global cause of mortality due to intricate host-pathogen interactions that affect disease progression. MicroRNAs (miRNAs), essential posttranscriptional regulators, have become pivotal modulators of these relationships. Recent findings indicate that miRNAs actively regulate immunological responses to Mtb complex by modulating autophagy, apoptosis, and immune cell activities. This has resulted in increased interest in miRNAs as prospective diagnostic indicators for TB, especially…

The immune microenvironment of pathogen-associated cancers and current clinical therapeutics.

Pathogen-associated cancers account roughly 15-20% of all malignancies worldwide and arise from chronic infections by oncogenic viruses, bacteria and parasites. Pathogens can alter the tumor microenvironment (TME) into an immune-suppressive niche using multiple mechanisms, including immune checkpoint manipulation, chronic inflammation, metabolic reprogramming, and direct epigenetic modifications. Immune dysfunctions related to infections linked to cancers can include M2-polarized macrophages, expansion of MDSCs, exhausted T/NK cells, and tolerogenic dendritic cells, all progressing immune evas

Convergent mechanisms of epithelial cell structure manipulation by intestinal pathogens.

The epithelial layer that lines the digestive system serves as the primary barrier to infection by intestinal pathogens. While this layer has evolved complex molecular mechanisms to identify and respond to infection, pathogens have also evolved equally complex mechanisms to subvert this response and remodel the epithelium to their benefit. The structure of the intestinal epithelial cell is a common target of this remodeling effort. This review focuses specifically on the phenotypes and mechanisms of epithelial cell structure manipulation that have convergently evolved in human intestinal patho

Pathogen-specific social immunity is associated with erosion of individual immune function in an ant.

Contagious diseases are a major threat to societies in which individuals live in close contact. Social insects have evolved collective defense behaviors, such as social care or isolation of infected workers, that prevent outbreaks of pathogens. It has thus been suggested that individual immunity is reduced in species with such 'social immunity'. However, this hypothesis has not been tested functionally. Here, we characterize the immune response of the ant Lasius niger using a combination of genomic analysis, experimental infections, gene expression quantification, behavioural observations and

Predicting phage-host interactions via feature augmentation and regional graph convolution.

Identifying phage-host interactions (PHIs) is a crucial step in developing phage therapy, which is the promising solution to addressing the issue of antibiotic resistance in superbugs. However, the lifestyle of phages, which strongly depends on their host for life activities, limits their cultivability, making the study of predicting PHIs time-consuming and labor-intensive for traditional wet lab experiments. Although many deep learning (DL) approaches have been applied to PHIs prediction, most DL methods are predominantly based on sequence information, failing to comprehensively model the int
