Spinal interleukin-17 promotes thermal hyperalgesia and NMDA NR1 phosphorylation in an inflammatory pain rat model.

It is known that interleukin-17 (IL-17) is associated with autoimmune disorders and that peripheral IL-17 plays a role in arthritis and neuropathic pain. The present study investigated the possibility of spinal cell expression of IL-17 during inflammatory pain and possible IL-17 involvement in such pain. Hyperalgesia was induced by injecting complete Freund adjuvant (CFA, 0.08mL, 40μg Mycobacterium tuberculosis) into one hind paw of the rat. Paw withdrawal latency (PWL) was tested before (-48h) and 2 and 24h after CFA injection to assess hyperalgesia. IL-17 antibody (0.2-2μg/rat) was given int…

IL-1ra alleviates inflammatory hyperalgesia through preventing phosphorylation of NMDA receptor NR-1 subunit in rats.

Although it has been shown that pro-inflammatory cytokines such as interleukin-1beta (IL-1beta) facilitate perception of noxious inputs at the spinal level, the mechanisms have not been understood. This study determined the cell type that produces IL-1beta, the co-localization of IL-1 receptor type I (IL-1RI) and Fos and NR1 in the spinal cord, and the effects of IL-1 receptor antagonist (IL-1ra) on NR1 phosphorylation and hyperalgesia in a rat model of inflammatory pain. Phosphorylation of NR1, an essential subunit of the NMDA receptor (NMDAR), is known to modulate NMDAR activity and facilita…

NK3 receptor blockade prevents hyperalgesia and the associated spinal cord substance P release in monoarthritic rats.

Previous studies in vitro have shown that NK3 receptors exist on primary afferent terminals in rat spinal cord and mediate potentiation of the depolarisation-evoked substance P (SP) release. In the present study we have investigated the role of the NK3 receptor-mediated SP release system in a model of inflammatory pain. Monoarthritis was induced in rats by unilateral injection of complete Freund's adjuvant (CFA); withdrawal latencies to a thermal stimulus were subsequently measured at various times following CFA. The CFA-treated paw displayed hyperalgesia as early as 4 h after CFA injection an…

SR 48968 specifically depresses neurokinin A- vs. substance P-induced hyperalgesia in a nociceptive withdrawal reflex.

To determine the role of neurokinin A and tachykinin NK2 receptors in processing of nociceptive information at the spinal level, the selective NK2 receptor antagonist, SR 48968 (S)-N-methyl-N [4-(4-acetylamino-4-[phenyl piperidino)-2-(3,4-dichlorophenyl)-butyl] benzamide, was tested for its effects on the hyperalgesia produced in the tail flick reflex by intrathecal administration of neurokinin A and of substance P. SR 48968 was also tested in a model in which noxious peripheral stimulation has been shown to produce hyperalgesia via a substance P mechanism. SR 48968 given intrathecally had a d…

Repetitive Transcranial Magnetic Stimulation of the mPFC Ameliorates Stress-Induced Hyperalgesia by Alleviating Microglia Activation in Rats.

PURPOSE: Repetitive transcranial magnetic stimulation (rTMS) is a noninvasive neuromodulation technique that uses magnetic fields to stimulate or inhibit specific populations of neurons in the brain. rTMS targeting the cortex has emerged as an effective treatment for chronic pain. It is established that stress can heighten pain sensitivity, a condition termed stress-induced hyperalgesia (SIH). However, neither direct evidence for rTMS treatment of SIH nor a clear understanding of its therapeutic mechanisms has been established. METHODS: In this article, we studied the effectiveness of rTMS in…

Monocyte Transmigration Involves in Remifentanil Induced Mechanical Hyperalgesia in Animal Models and a Randomized Controlled Clinical Trial.

Remifentanil-induced hyperalgesia (RIH) complicates postoperative pain management under total intravenous anesthesia. Although monocyte transmigration contributes to neuropathic pain, its role in RIH remains undefined. Here, we investigated monocyte dynamics in RIH through preclinical and clinical studies. In male mice, intraperitoneal remifentanil infusion (4 μg/kg/min, 1 h) induced sustained mechanical hyperalgesia (reduced paw withdrawal thresholds, PWTs) over 48 hours, paralleled by a transient decline in circulating monocytes (3 h post-infusion) and increased macrophage infiltration in pa…

Minocycline inhibits somatic hyperalgesia induced by malocclusion through up-regulating spinal IL-10 expression in female rats.

Epidemiological studies have revealed that temporomandibular disorder (TMD) and fibromyalgia syndrome (FMS) often co-occur, significantly reducing patients' quality of life, with limited and unsatisfactory treatment options. This study aimed to investigate whether the microglial inhibitor minocycline attenuates unilateral anterior crossbite (UAC)-induced somatic hyperalgesia, examine its effects on inflammatory cytokine expression in the spinal cord, and explore the underlying mechanisms. In female rats, UAC significantly reduced mechanical withdrawal threshold and thermal withdrawal latency o…

TDP-43 mediated oxidative stress induced mitochondrial dysfunction in neurons and hyperalgesia in sciatic nerve injured mice.

Neuropathic pain (NP) is a chronic pain with a highly complex pathogenesis, in which oxidative stress and mitochondrial dysfunction play sig … mitochondrial fission. Furthermore, intrathecal injection of siRNA to knock down TDP-43 alleviated hyperalgesia and allodynia in CCI mice. These data indicate that TDP-43 in spinal neurons may contribute to NP by impairing mitochondrial function induced by oxidative stress, which may provide a new potential target for the treatment of NP.

Cannabidiol and it fluorinate analog PECS-101 reduces hyperalgesia and allodynia in trigeminal neuralgia via TRPV1 receptors.

Trigeminal neuralgia (TN) is an intense and debilitating orofacial pain. The gold standard treatment for TN is carbamazepine. This antiepileptic drug provides pain relief with limited efficacy and side effects. To study the antinociceptive potential of cannabidiol (CBD) and its fluorinated analog PECS-101 (former HUF-101), we induced unilateral chronic constriction injury of the infraorbital nerve (IoN-CCI) in male Wistar rats. Seven days of treatment with CBD (30 mg/kg), PECS-101 (3, 10, and 30 mg/kg), or carbamazepine (10 and 30 mg/kg) reduced allodynia and hyperalgesia responses. Unlike car…

Anodal Transcranial Direct Current Stimulation Reduces Secondary Hyperalgesia Induced by low Frequency Electrical Stimulation in Healthy Volunteers.

The aim of the study was to determine whether transcranial direct current stimulation (tDCS) reduced pain and signs of central sensitization … ate pain compared to sham and other conditions. Anodal tDCS of DLPFC completely abolished secondary hyperalgesia. Unexpectedly, however, concurrent anodal tDCS over M1 and DLPFC did not reduce pain or hyperalgesia more than M1 alone or DLPFC alone. Overall, these findings suggest that anodal tDCS over M1 suppresses pain, and that anodal tDCS over DLPFC modulat …

Spinal CCK contributes to somatic hyperalgesia induced by orofacial inflammation combined with stress in adult female rats.

In some chronic primary pain conditions such as temporomandibular disorder (TMD) and fibromyalgia syndrome (FMS), mild or chronic stress enhances pain. TMD and FMS often occur together, but the underlying mechanisms are unclear. The purpose of this study was to investigate the role of cholecystokinin (CCK) in the spinal cord in somatic hyperalgesia induced by orofacial inflammation combined with stress. Somatic hyperalgesia was detected by the thermal withdrawal latency and mechanical withdrawal threshold. The expression of CCK1 receptors, CCK2 receptors, ERK1/2 and p-ERK1/2 in the spinal cord…

Nerve growth factor induces facial heat hyperalgesia and plays a role in trigeminal neuropathic pain in rats.

There is preclinical evidence that nerve growth factor (NGF) contributes toward inflammatory hyperalgesia in the orofacial region, but the mechanisms underlying its hyperalgesic effect as well as its role in trigeminal neuropathic pain require further investigation. This study investigated the ability of NGF to induce facial heat hyperalgesia and the involvement of tyrosine kinase receptor A, transient receptor potential vanilloid 1, and mast cells in NGF pronociceptive effects. In addition, the role of NGF in heat hyperalgesia in a model of trigeminal neuropathic pain was evaluated. NGF injec…
