Implementing reversed-phase and hydrophilic interaction liquid chromatography into the characterization of DTPA-ramucirumab conjugate before radiolabeling.

Radioimmunoconjugates represent a promising class of therapeutics and diagnostics. The characterization of intermediate chelator-antibody products, i.e., without the radionuclide, is frequently omitted, bringing significant uncertainty in the radioimmunoconjugate preparation. In the present study, we explored the utility of reversed-phase (RPLC) and hydrophilic interaction (HILIC) liquid chromatography with UV detection to characterize ramucirumab stochastically conjugated with p-SCN-Bn-CHX-A"-DTPA chelator (shortly DTPA). The conjugation was well reflected in RPLC chromatograms, while chromat…

Modified Protocol of Nivolumab in Relapsed/Refractory Hodgkin Lymphoma: A Brief Communication of Real World Data.

Immune check point inhibitors such as nivolumab are changing the treatment paradigm of relapsed/refractory Hodgkin lymphoma (r/rHL). Data from single arm studies have shown nivolumab to be an effective and safe therapy. Real world data from resource constrained settings are limited. Our study is a retrospective single center analysis of nivolumab in r/rHL from India. Data regarding baseline and pretreatment characteristics were collected for 20 patients treated with nivolumab from January 2016 to March 2021. Of 20, 15 patients received nivolumab in modified protocol, because of financial limit

TROP2 (trophoblast cell-surface antigen 2): a drug target for breast cancer.

INTRODUCTION: Breast cancer (BC) is the most common diagnosed cancer and the second leading cause of cancer-associated death in women, with the triple negative (TNBC) subtype being characterized by the poorest prognosis. New therapeutic targets are urgently needed to overcome the high metastatic potential, aggressiveness and poor survival of these tumors. Trop2 transmembrane glycoprotein, acting as an intracellular calcium signal transducer, recently emerged as a new potential target in epithelial cancers, in particular in breast cancer. AREAS COVERED: We summarize the key features of Trop2 s

Impact of belantamab mafodotin-induced ocular toxicity on outcomes of patients with advanced multiple myeloma.

Belantamab mafodotin (BLMF) is a B-cell maturation antigen-directed antibody-drug conjugate, recently approved for advanced multiple myeloma (MM). The impact of BLMF-induced ocular toxicity on patient outcomes is unknown. We studied a cohort of 38 consecutively seen patients treated with BLMF outside of trials. Of those, 75% experienced ocular toxicity, with 69% developing keratopathy. Among patients requiring ocular toxicity-related permanent BLMF discontinuation (14%) or dose reduction (11%), 70% had progression of MM within a median of 3 months (95% confidence interval: 0.2-not reached) fol

Indolent lymphomas: pushing the pace with novel agents.

Chemoimmunotherapy has been a hallmark of treatment of indolent B-cell non-Hodgkin lymphomas for the past 2 decades, with high response rates seen but relapses nearly inevitable and patients spending, on average, 20 years on and off treatment. Treatment advances, then, should be aimed at maintaining efficacy while minimizing toxicity or at achieving cure. Improved understanding of the genetic and molecular features of these diseases, as well as of the interaction between the tumor cell and its immune microenvironment, has resulted in an accelerated expansion of tolerable treatment options for

[Hodgkin lymphoma].

Hodgkin lymphoma is characterized by CD30 overexpression and downstream NF-κB pathway activation in Hodgkin/Reed-Sternberg (H/RS) cells. Accordingly, CD30 and its aberrant downstream signaling have been investigated as potential treatment targets for the disease. Given that H/RS cells are surrounded by a variety of immune cells that constitute a unique inflammatory and immunoinhibitory microenvironment, efforts have also been made to treat the disease by reversing the abnormal tumor immune milieu. In recent years, a CD30 antibody-drug conjugate and immune checkpoint inhibitors have been approv

[Cancer Stromal Targeting Therapy].

Recent advances in antibody-drug conjugate (ADC) technology have shown considerable promise in targeted cancer therapy. The ADC strategy should be confined to highly toxic anticancer agents and not to ordinary anti-cancer agents (ACAs) because the affinity of monoclonal antibodies (mAbs) diminishes if more than three ACA molecules are conjugated. According to this principle, higher amounts of ADC should be administered so that each of the ACAs is conjugated to the mAbs. Therefore for an ordinary ACA, nanoparticles should be the preferred drug delivery system (DDS). A large body of clinical evi

CBTF: new amine-to-thiol coupling reagent for preparation of antibody conjugates with increased plasma stability.

Amine-to-thiol coupling is the most common route for the preparation of antibody-drug conjugates (ADC). It is usually achieved by using heterobifunctional reagents possessing an activated ester at one end and a maleimide group at the other. However, maleimide-based conjugates were recently revealed to have limited stability in blood circulation, which can compromise therapeutic efficacy of the conjugate. To address this issue, we have developed a heterobifunctional reagent, sodium 4-((4-(cyanoethynyl)benzoyl)oxy)-2,3,5,6-tetrafluorobenzenesulfonate (CBTF), for amine-to-thiol coupling. It compr

Novel therapies for chronic lymphocytic leukemia in the 21st century.

Chronic lymphocytic leukemia (CLL) is one of the most common types of leukemia diagnosed in the Western Hemisphere. Introduction of the purine analogs has improved outcome for patients with CLL as measured by improvements in disease-free survival and has made it possible to attain a complete remission in a minority of patients with this disease. The therapeutic success with the purine analogs in CLL has led to preclinical and early clinical investigation of a variety of other therapeutic agents. This, combined with advances in the understanding the genetic and immunobiology of CLL, leaves hope

The therapeutic potential of antibody-drug conjugates in advanced colorectal cancer: a systematic review and meta-analysis.

HER2-targeted antibody-drug conjugates (ADCs) have achieved clinically meaningful activity in several human epidermal growth factor-2 (HER2)-positive malignancies. However, the efficacy of these agents in HER2-positive colorectal cancer (CRC) remains incompletely understood. To address this gap, we conducted a systematic review and meta-analysis to compare treatment outcomes of HER2-targeted ADC therapy in patients with HER2-positive CRC versus those with HER2-positive tumors of other origins. Five prospective clinical studies evaluating HER2-targeted ADCs in HER2-positive malignancies were inc

Development of a modular click labelling method for producing radioiodine-labelled antibodies.

Direct radioiodination is a simple and convenient method for antibody labelling but suffers from reduced immunoreactivity and metabolic instability. To address these limitations, a modular click radioiodination strategy based on iodo-Azide and ADIBO-antibody conjugates was developed. The resulting [125I]iodo-Azide-Rituximab exhibited approximately 1.6-fold higher immunoreactivity, improved blood retention, and reduced thyroid radiation exposure compared to directly labelled [125I]iodo-Rituximab. This click-based approach minimizes antibody damage and enhances in vivo stability, underscoring it

uPAR-Targeting Cytotoxic Antibody-Drug Conjugates Selectively Deplete Proinflammatory Myeloid Cells for Autoimmune Indications.

Rheumatoid arthritis (RA) is an autoimmune disorder characterized by synovial inflammation and progressive joint destruction. There is no cure, and patient responses to current therapies vary, reflecting underlying pathogenic heterogeneity. Leveraging single-cell RNA sequencing (scRNA-seq) of RA synovium, we identified a PLAUR/uPAR-high myeloid subset that co-expresses pathogenic mediators, including IL1B and CXCL8. To target these cells, we developed anti-uPAR antibody-drug conjugates (ADCs) and evaluated various payloads in vitro and in vivo. ADCs bearing BCL-2 family inhibitors selectively

Biodistribution of tumor-targeted MXene-antibody conjugates.

The biodistribution profile is a critical determinant of the success of nanomaterial applications, governing both therapeutic efficacy and systemic safety in emerging photothermal therapy (PTT) agents. Despite the promise of PTT, the clinical translation of many nanomaterial-based photothermal agents is impeded by intrinsic toxicity and insufficient tumor specificity. Moreover, off-target biodistribution of photosensitizers after systemic administration can increase systemic exposure, reduce tumor delivery, and ultimately compromise therapeutic efficacy, underscoring the need for advanced mate

Cell Surface Deazaflavin-Diazirine Energy-Transfer (DarT) Photoproximity Labeling Using Antibody Conjugates.

Elucidating protein-protein interactions plays a crucial part in understanding disease mechanisms and advancing pharmacological research. Photocatalytic proximity labeling using antibody-catalyst conjugates enables the highly target-specific analysis of protein-protein interactions on the cell surface without altering the native cellular state through genetic manipulation. Here, we describe an extension of the deazaflavin-diazirine energy-transfer (DarT) labeling platform through the development and evaluation of trastuzumab-deazaflavin (Tra-dFl) conjugates for mapping the extracellular microe
