This review summarizes recent progress in water-transporting mechanisms across cell membranes. Modern biophysical concepts of water transport and new measurement strategies are evaluated. A family of water-transporting proteins (water channels, aquaporins) has been identified, consisting of small hydrophobic proteins expressed widely in epithelial and nonepithelial tissues. The functional properties, genetics, and cellular distributions of these proteins are summarized. The majority of molecular-level information about water-transporting mechanisms comes from studies on CHIP28, a 28-kDa glycop

Kettin is a large modular protein associated with thin filaments in the Z-disc region of insect muscles. The sequence of a 21.3 kb contig of the Drosophila gene has been determined. The corresponding protein sequence has 35 immunoglobulin-like (Ig) domains which are separated by shorter linker sequences, except near the N and C termini of the molecule where linker sequences are short or missing. This confirms a model in which each Ig domain binds to an actin protomer. The Drosophila kettin gene is at 62C 1-3 on the third chromosome. Two P-element insertions, l(3)j1D7 and l(3)rL182 are in the k

Candida albicans is a major human pathogen whose treatment is challenging due to antifungal drug toxicity, drug resistance and paucity of antifungal agents available. Myrocin (MYR) inhibits sphingosine synthesis, a precursor of sphingolipids, an important cell membrane and signaling molecule component. MYR also has dual immune suppressive and antifungal properties, potentially modulating mammalian immunity and simultaneously reducing fungal infection risk. Wax moth (Galleria mellonella) larvae, alternatives to mice, were used to establish if MYR suppressed insect immunity and increased surviva

The cottony cushion scale, Icerya purchasi, a polyphagous pest, poses a significant threat to the global citrus industry. The hermaphroditic self-fertilization observed in I. purchasi is an exceptionally rare reproductive mode among insects. In this study, we successfully assembled a chromosome-level genome sequence for I. purchasi using PacBio long-reads and the Hi-C technique, resulting in a total size of 1,103.38 Mb and a contig N50 of 12.81 Mb. The genome comprises 14,046 predicted protein-coding genes, with 462,722,633 bp occurrence of repetitive sequences. BUSCO analysis revealed a compl

Heat shock proteins (HSPs) are essential across several insect species for adaptation to environmental stressors. Most pest insects experience fertilization concurrently with summer heat extremes, which mostly affect male testis development and spermatogenesis; although the molecular mechanisms involved are unclear. Several HSPs are exclusive to the testis in several insects, but their functional roles remain unclear. In this study, we identified a testis-specific ATP-dependent HSP, CfHSP70-2 in the spruce budworm (SBW), Choristoneura fumiferana. Reverse transcript-qualitative polymerase chain

Insulin receptors (InR) are an integral component of the insulin/insulin-like growth factor signaling pathway, which plays a vital role in insect development, lifespan, reproduction, and olfactory sensitivity. However, whether InR participate in the peripheral olfactory system of insects remains unclear. Recently, we found that 2-heptanone (2-HT) affects AcerInR expression, the gene for an InR protein, in Apis cerana cerana. We then examined the spatiotemporal expression profile of the gene in A. cerana cerana. The mRNA of AcerInR was primarily expressed in the antennae, wings, and legs of for

omega-Atracotoxin-Hv1a is an insect-specific neurotoxin whose phylogenetic specificity derives from its ability to antagonize insect, but not vertebrate, voltage-gated calcium channels. In order to help understand its mechanism of action and to enhance its utility as a lead compound for insecticide development, we used a combination of protein engineering and site-directed mutagenesis to probe the toxin for key functional regions. First, we constructed a Hairpinless mutant in which the C-terminal beta-hairpin, which is highly conserved in this family of neurotoxins, was excised without affecti

The ongoing antigenic evolution of SARS-CoV-2, particularly within the spike glycoprotein, threatens the long-term efficacy of spike-focused vaccines and serodiagnostics. While most authorized COVID-19 vaccines exclusively target the spike protein, growing evidence underscores multivalent strategies incorporating conserved viral antigens. Here, we employed a baculovirus expression vector system (BEVS) to co-express all four structural proteins, including spike, nucleocapsid, membrane, and envelope, in Sf9 insect cells. Surface-displayed antigens were used in a cell-based ELISA to profile IgG r

Invertebrates have been instrumental in understanding the mechanisms involved in infectious diseases, considering the idea to replace, reduce and refine the use of mammals as well as to understand the basic principles of immune response in insect. We evaluated the consequences of Staphylococcus aureus-induced sepsis in the last instar nymphs of Nauphoeta cinerea injected with different concentrations of bacteria preserved in two culture media. Infected groups had a decrease in hemolymph metabolites (glucose, amino acids, total proteins, and cholesterol), in contrast to the proteins in the fat

Virus-like particles (VLPs) and subviral particles (SVPs) are an alternative approach to viral vaccine design that offers the advantages of increased biosafety and stability over use of live pathogens. Non-infectious and self-assembling, VLPs are used to present structural proteins as immunogens, bypassing the need for live pathogens or recombinant viral vectors for antigen delivery. In this article, we demonstrate the different stages of VLP design and development for future applications in preclinical animal testing. The procedure includes the following stages: selection of antigen, expressi

The recombinant form of a highly immunogenic 14.6 kDa protein in Triatoma infestans saliva (rTiSP14.6) is a potential epidemiological marker for the detection of triatomine bug populations using IgG responses in peridomestic chickens. However, the persistence of the IgG response prevents it being of value for several months in areas where triatomine control programmes have been implemented. In this investigation, IgM-antibody reactions to crude salivary antigens or rTiSP14.6 decayed rapidly after exposure of chickens and were measurable for only 18 days after a single challenge with T. infesta

Female Aedes aegypti secrete urine rapidly post-bloodmeal ingestion, with diuresis beginning immediately for removal of excess salts and water. This post-prandial diuresis includes a peak, post-peak and late phase, involving the combined actions of multiple hormones, including diuretic and anti-diuretic factors. Calcitonin-like diuretic hormone 31 (DH31) and kinin peptides stimulate diuresis through actions on their cognate receptors localized in the Malpighian 'renal' tubules (MTs). In contrast, the anti-diuretic neurohormone CAPA inhibits secretion by MTs stimulated by select diuretic hormon

BACKGROUND: Aquaporin (AQP) water channels are important for water homeostasis in all organisms. Malaria transmission is dependent on Anopheles mosquitoes. Water balance is a major factor influencing mosquito survival, which may indirectly affect pathogen transmission.
METHODOLOGY/PRINCIPAL FINDINGS: We obtained full-length mRNA sequences for Anopheles gambiae aquaporin 1 (AgAQP1) and identified two splice variants for the gene. In vitro expression analysis showed that both variants transported water and were inhibited by Hg(2+). One splice variant (AgAQP1A) was exclusively expressed in adult

BACKGROUND: Leishmaniases are a group of medically and veterinary important diseases caused by protozoan parasites of the genus Leishmania (Kinetoplastida) and transmitted by blood-feeding female sand flies (Diptera: Phlebotominae). To assess the risk of Leishmania transmission, host exposure to sand fly bites can be measured through the detection of host antibodies to vector salivary proteins. Anti-sand fly saliva antibodies are elicited by repeated exposure of the mammalian host to sand fly salivary proteins deposited into the host skin during blood feeding. These antibodies are species-spec

Bacillus thuringiensis (Bt) strains naturally produce pesticidal proteins as nanocrystalline inclusions that are extraordinarily stable in aqueous environments, but which dissolve selectively at specific pH conditions. These proteins have been used in agriculture for >50 years and are critical to global food security. The majority of previously determined Bt Cry protein structures lack the extended C-terminal "crystallization domain," which is thought to stabilize crystal packing and control selective solubility in insect targets, often via manipulation of disulfide bridges. It has also recent

Targeted protein degradation is an invaluable tool in studying the function of proteins. Such a tool was not available in Trypanosoma brucei, an evolutionarily divergent eukaryote that causes human African trypanosomiasis. Here, we have adapted deGradFP (degrade green fluorescent protein [GFP]), a protein degradation system based on the SCF E3 ubiquitin ligase complex and anti-GFP nanobody, in T. brucei. As a proof of principle, we targeted a kinetoplastid kinetochore protein (KKT3) that constitutively localizes at kinetochores in the nucleus. Induction of deGradFP in a cell line that had both

During its life cycle, Trypanosoma brucei shuttles between a mammalian host and the tsetse fly vector. In the mammalian host, immune evasion of T. brucei bloodstream form (BSF) cells relies on antigenic variation, which includes monoallelic expression and periodic switching of variant surface glycoprotein (VSG) genes. The active VSG is transcribed from only 1 of the 15 subtelomeric expression sites (ESs). During differentiation from BSF to the insect-resident procyclic form (PCF), the active ES is transcriptionally silenced. We used mass spectrometry-based interactomics to determine the compos

Insect nephrocytes provide a valuable model for kidney disease, as they are structurally and functionally homologous to mammalian kidney podocytes. They possess an exceptional macromolecular assembly, the nephrocyte diaphragm (ND), which serves as a filtration barrier and helps maintain tissue homeostasis by filtering out wastes and toxic products. However, the elements that maintain nephrocyte architecture and the ND are not understood. We show that Drosophila nephrocytes have a unique cytoplasmic cluster of F-actin, which is maintained by the microtubule cytoskeleton and Rho-GTPases. A balan

Wolbachia pipientis (Rickettsiales; Anaplasmataceae) is an obligate intracellular alpha proteobacterium that occurs in arthropods and filarial worms. Some strains of Wolbachia can be maintained as persistent infections in insect cell lines. C/wStr1 cells from the mosquito Aedes albopictus maintain a robust infection with Wolbachia strain wStr, originally isolated from the planthopper, Laodelphax striatellus. To explore possible functions of penicillin-binding proteins expressed from the wStr genome, C/wStr1 cells were exposed to ampicillin. Absolute levels of Wolbachia increased 3.5-fold in am
