Transgenic mice overproducing islet amyloid polypeptide have increased insulin storage and secretion in vitro.

To determine whether chronic overproduction of islet amyloid polypeptide alters beta-cell function, we studied a line of transgenic mice which overexpress islet amyloid polypeptide in their beta-cells. At 3 months of age, these transgenic mice had greater pancreatic content of both islet amyloid polypeptide and insulin. Further, basal and glucose-stimulated secretion of both islet amyloid polypeptide and insulin were also elevated in the perfused pancreas of the transgenic animals. These findings demonstrate that chronic overproduction and secretion of islet amyloid polypeptide are associated

Decrease of stimulated amylin release precedes impairment of insulin secretion in type II diabetes.

Amylin, a 37-amino acid polypeptide, has been identified as the major protein component of pancreatic amyloid deposits in patients with non-insulin-dependent (type II) diabetes mellitus. Amylin is stored and released together with insulin and has been proposed to play a major role in the pathogenesis of type II diabetes. To compare amylin release and its proportion to insulin secretion under different metabolic conditions, oral and intravenous glucose tolerance tests (OGTT and IVGTT, respectively) were performed in healthy, lean control subjects, obese patients with normal and impaired glucose…

The role of Ca(2+)-related events in glucose-stimulated desensitization of insulin secretion.

The spontaneous decline of insulin secretion (third phase) that occurs under a variety of secretory conditions is well documented and suggests a general impairment or desensitization of the secretory process. We have examined several aspects of Ca2+ flux as well as regulators of Ca-linked second messenger events in freshly isolated rat islets chronically stimulated with glucose over 24 h, a period that encompasses initial (hour 1), peak (hour 3), and subsequent impaired or desensitized (hour 20-22) secretion. In islets incubated for these periods in HB104 medium with 22 mM glucose, 45Ca2+ upta…

Interleukin-1 inhibits glucose-modulated insulin and glucagon secretion in rat islet monolayer cultures.

Recent observations suggest a role for interleukin-1 (IL-1), a polypeptide product of macrophage/monocytic cells, in the immune-mediated destruction of pancreatic islet beta-cells observed in type 1 diabetes. In this study, we investigated the effects of IL-1 on both alpha- and beta-cell secretory functions in rat islet cell monolayer cultures. Insulin release was 97% inhibited after 6 h of incubation in RPMI-1640 medium (11 mM glucose) containing 1 U/ml IL-1 and 96% inhibited after 24 h of incubation in medium containing 0.1 U/ml IL-1. The cell content of insulin in the monolayers was decreas

Glucose utilization in islets of hyperglycemic rat models with impaired glucose-induced insulin secretion.

Under most experimental conditions islet glucose metabolism is well-correlated with short-term glucose-induced insulin secretion. Two hyperglycemic rat models (neonatal streptozotocin and glucose infusion) have been previously found to have markedly impaired insulin responses to glucose, and the glucose utilization of islets isolated from these models was therefore studied to see if reduced glucose metabolism might be related to the secretory abnormalities. It was found that glucose utilization in the islets of the two models was similar or higher than in comparable control islets. These data …

Melatonin as a daily rhythm control signal for glucose homeostasis and insulin secretion in horses.

An involvement of melatonin in the regulation of glucose homeostasis and insulin secretion in pancreatic β-cells has been proposed. To evaluate the features of the daily rhythms of plasma melatonin, blood glucose, and insulin in horses, ten clinically healthy Italian Saddle geldings, 9-10 years old with a mean body weight of 490 ± 25 kg, were enrolled. The animals were housed in individual boxes under natural environmental conditions (sunrise 06:10; sunset 18:10; 12-15°C, 77% RH). All horses were cannulated the day before the start of the study for sample collection, which was performed every …

Loss of macroautophagy results in abnormality of the endoplasmic reticulum in insulin-secreting MIN6 cells.

Macroautophagy is a highly conserved system that degrades various materials inside the cell ranging from proteins to organelles. Atg5 is a protein essential for the formation of autophagosomes, which sequester materials by double membrane and degrade them after fusion with lysosomes. MIN6 cells derived from mouse pancreatic β cells retain the ability to secrete insulin in response to a change in glucose concentration from low to high. We knocked out the Atg5 gene in MIN6 cells and identified an abnormality in the endoplasmic reticulum (ER) under normal culture conditions using electron microsc

Management of refractory hypoglycaemia in a metastatic neuroendocrine tumour co-secreting serotonin and insulin.

A 27-year-old otherwise healthy man of African descent presented to the hospital with initial symptoms of carcinoid syndrome that later evolved into symptoms of hyperinsulinemic hypoglycaemia. Investigations revealed a metastatic neuroendocrine tumour (NET), co-secreting both serotonin and insulin. Management involved a multimodal approach in an attempt to reduce tumour burden and achieve euglycaemia, which proved to be a significant challenge in the face of refractory hypoglycaemia despite the administration of multiple prohyperglycaemic agents in combination. Unfortunately, given the burden

Interplay of Ca2+ and cAMP signaling in the insulin-secreting MIN6 beta-cell line.

Ca2+ and cAMP are important second messengers that regulate multiple cellular processes. Although previous studies have suggested direct interactions between Ca2+ and cAMP signaling pathways, the underlying mechanisms remain unresolved. In particular, direct evidence for Ca2+-regulated cAMP production in living cells is incomplete. Genetically encoded fluorescence resonance energy transfer-based biosensors have made possible real-time imaging of spatial and temporal gradients of intracellular cAMP concentration in single living cells. Here, we used confocal microscopy, fluorescence resonance e

[Resistine--a new hormone secreted by adipose tissue (adipose tissue in insulin resistance)].

Obesity is an important risk factor for the development of insulin resistance and type 2 diabetes. The molecular mechanism linking obesity to insulin resistance is, however, unclear. Recently, a new circulating hormone resistine, which is expressed in adipose tissue, has been identified. Resistine has been shown to antagonize insulin action. Resistine levels are increased in diet-induced obesity as well as in genetic models of obesity and insulin resistance. Furthermore, resistine gene expression is markedly downregulated by treatment with anti-diabetic drugs called thiazolidinediones, that im

[Glucose tolerance and insulin secretion in patients with hyperthyroidism. Pathogenesis of disturbances and therapeutic consequences].

Abnormalities of carbohydrate metabolism have been frequently reported in hyperthyroid patients. Disturbed glucose tolerance might be due to increased hepatic glucose production, altered insulin metabolism, increased response to beta-adrenergic stimuli and elevated plasma FFA levels. These disturbances very often disappear when remission of thyrotoxicosis is attained during-treatment with thyrostatic drugs. Non-selective or selective beta 1-adrenoceptor blocking agents does not impair the glucose stimulated insulin secretion or the carbohydrate tolerance and impaired glucose tolerance disappea…

Lipid composition of glucose-stimulated pancreatic islets and insulin-secreting tumor cells.

The effect of glucose stimulation (25 mM for 5 min) on the phospholipid and neutral lipid composition of isolated pancreatic islets was studied to find out whether there is a change in the mass of potential lipid mediators or modulators of insulin secretion. For comparison, the lipid compositions of homogenates and subcellular fractions from RINm5F insulin-secreting tumor cells and of glucose-stimulated streptozotocin/nicotinamide-induced islet cell tumors were analyzed. After separation of the lipid extract into a neutral and an acidic fraction by anion-exchange chromatography, lipids were se…
