Deletion of MLIP (muscle-enriched A-type lamin-interacting protein) leads to cardiac hyperactivation of Akt/mammalian target of rapamycin (mTOR) and impaired cardiac adaptation.

Aging and diseases generally result from tissue inability to maintain homeostasis through adaptation. The adult heart is particularly vulnerable to disequilibrium in homeostasis because its regenerative abilities are limited. Here, we report that MLIP (muscle enriched A-type lamin-interacting protein), a unique protein of unknown function, is required for proper cardiac adaptation. Mlip(-/-) mice exhibited normal cardiac function despite myocardial metabolic abnormalities and cardiac-specific overactivation of Akt/mTOR pathways. Cardiac-specific MLIP overexpression led to an inhibition of Akt/

Mitochondrial complex I as a therapeutic target for Alzheimer's disease.

Alzheimer's disease (AD), the most prominent form of dementia in the elderly, has no cure. Strategies focused on the reduction of amyloid beta or hyperphosphorylated Tau protein have largely failed in clinical trials. Novel therapeutic targets and strategies are urgently needed. Emerging data suggest that in response to environmental stress, mitochondria initiate an integrated stress response (ISR) shown to be beneficial for healthy aging and neuroprotection. Here, we review data that implicate mitochondrial electron transport complexes involved in oxidative phosphorylation as a hub for small …

A Comprehensive Analysis of Transcriptomics and Proteomics Elucidates the Cold-Adaptive Ovarian Development of Eriocheir sinensis Farmed in High-Altitude Karst Landform.

Background: In high-altitude regions, sporadic two-year-old immature Chinese mitten crabs (Eriocheir sinensis) would overwinter and mature in their third year, developing into three-year-old crabs (THCs) with a cold-adaptive strategy. Compared to two-year-old crabs (TWCs) from low-altitude Jiangsu, THCs from Karst landform and high-altitude Guizhou exhibit significantly larger final size but lower gonadosomatic index (GSI) (p < 0.01). Methods: To elucidate the molecular mechanisms underlying this delayed ovarian development, integrated transcriptomic and proteomic analyses were conducted. Resu

Neandertal Cold Adaptation: Technological, Anatomical, and Physiological Responses to Cold Stress in One of Our Closest Fossil Relatives.

Neandertals occupied western Eurasia for over 100 000 years, repeatedly enduring climates that ranged from seasonally cold to glacial. This paper reexamines the question of Neandertal cold adaptation using updated fossil, physiological, and archaeological evidence. While some populations lived outside glacial extremes, all faced periodic cold stress, and their survival depended on a diverse set of strategies. Technological buffers, including fire use, hide processing tools, and possible clothing and footwear, likely played a primary role in reducing cold exposure. Anatomically, Neandertals exh

Nitric oxide enhances ethanol stress tolerance in Wickerhamomyces anomalus through modulating cell membrane and cell wall homeostasis.

BACKGROUND: Wickerhamomyces anomalus has garnered significant interest for its remarkable ability to shape wine flavor profiles. However, during fermentation, yeast cells are inevitably exposed to ethanol stress. Nitric oxide (NO) is a key signaling molecule that plays diverse physiological roles in living organisms. However, its impact on cell wall and membrane homeostasis under ethanol stress remains unclear. RESULTS: In this study, we investigated the regulatory effects of NO on the cellular integrity of W. anomalus under ethanol stress by supplementing with the NO donor (SNP) or the NO sca

Eukaryotic initiation factor 2 phosphorylation and translational control in metabolism.

Regulation of mRNA translation is a rapid and effective means to couple changes in the cellular environment with global rates of protein synthesis. In response to stresses, such as nutrient deprivation and accumulation of misfolded proteins in the endoplasmic reticulum, phosphorylation of the α subunit of eukaryotic initiation factor 2 (eIF2α~P) reduces general translation initiation while facilitating the preferential translation of select transcripts, such as that encoding activating transcription factor 4 (ATF4), a transcriptional activator of genes subject to the integrated stress response…

MAPK-activated protein kinase 2-deficiency causes hyperacute tumor necrosis factor-induced inflammatory shock.

BACKGROUND: MAPK-activated protein kinase 2 (MK2) plays a pivotal role in the cell response to (inflammatory) stress. Among others, MK2 is known to be involved in the regulation of cytokine mRNA metabolism and regulation of actin cytoskeleton dynamics. Previously, MK2-deficient mice were shown to be highly resistant to LPS/d-Galactosamine-induced hepatitis. Additionally, research in various disease models has indicated the kinase as an interesting inhibitory drug target for various acute or chronic inflammatory diseases. RESULTS: We show that in striking contrast to the known resistance of MK

Polyethylene Terephthalate Microplastics Induce More Pronounced Physiological and Molecular Responses in Pepper Plants than Polyethylene: Insights from Physiological, Transcriptomic, and Metabolomic Perspectives.

Microplastics (MPs) threaten agroecosystems; however, polymer-specific phytotoxicity remains unclear. In this study, we integrated physiological, transcriptomic, and metabolomic analyses to examine the effects of polyethylene (PE) and polyethylene terephthalate (PET) on pepper (Capsicum annuum L.). MPs of both polymers inhibited growth in a dose-dependent manner, with PET inducing more severe root damage. PET upregulated photosynthesis-related genes in roots, suggesting systemic stress signaling or a compensatory response. Integrated transcriptomic and metabolomic analyses revealed that MPs in

Neuroendocrine Stress Response in Acute Stroke: Physiological and Clinical Perspectives.

Acute ischemic stroke constitutes a significant systemic stress event that initiates a complex cascade of neuroendocrine and autonomic responses. Central to this process is activation of the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic nervous system, reflecting the brain's integrated response to acute cerebral injury. These changes are accompanied by widespread alterations in physiological regulation, including endocrine signaling, autonomic balance, circadian organization, and cardiovascular control. Understanding the nature and coordination of these responses is important f

Emerging roles of silicon in plant signaling networks and microbiome dynamics under salt stress.

Silicon (Si) is widely recognized as a beneficial element that can improve plant performance under salt stress. However, a comprehensive understanding of its biological functions requires moving beyond isolated physiological responses toward an integrated view of plant signaling and rhizosphere processes. This narrative review critically synthesizes current evidence and proposes an "inside-outside" framework for Si-associated salt-stress mitigation. Internally, Si treatment has been reported to influence phytohormone homeostasis, particularly abscisic acid, jasmonic acid, and salicylic acid, w

Conservation and divergence of abiotic stress-responsive gene co-expression networks in diploid and tetraploid cotton.

Cotton (Gossypium spp.) is highly susceptible to abiotic stresses, including temperature extremes, drought, and salinity. The diploid species Gossypium arboreum retains stress adaptive traits that are poorly represented in modern tetraploid cultivars, yet the evolutionary and regulatory bases of these ploidy associated differences remain insufficiently understood. Here, we performed a comparative transcriptomic analysis of 54 seedling leaf RNA-seq libraries from Gossypium arboreum, Gossypium hirsutum, and Gossypium barbadense subjected to four abiotic stress treatments. By integrating standard
