The ion channel TRPA1 is required for normal mechanosensation and is modulated by algesic stimuli.

BACKGROUND & AIMS: The transient receptor potential (TRP) channel family includes transducers of mechanical and chemical stimuli for visceral sensory neurons. TRP ankyrin 1 (TRPA1) is implicated in inflammatory pain; it interacts with G-protein-coupled receptors, but little is known about its role in the gastrointestinal (GI) tract. Sensory information from the GI tract is conducted via 5 afferent subtypes along 3 pathways. METHODS: Nodose and dorsal root ganglia whose neurons innnervate 3 different regions of the GI tract were analyzed from wild-type and TRPA1(-/-) mice using quantitative re

An ion channel library for drug discovery and safety screening on automated platforms.

Ion channels represent the third largest class of targets in drug discovery after G-protein coupled receptors and kinases. In spite of this ranking, ion channels continue to be under exploited as drug targets compared with the other two groups for several reasons. First, with 400 ion channel genes and an even greater number of functional channels due to mixing and matching of individual subunits, a systematic collection of ion channel-expressing cell lines for drug discovery and safety screening has not been available. Second, the lack of high-throughput functional assays for ion channels has …

Hallmarks of ion channel gene expression in end-stage heart failure.

Electrical conductance is greatly altered in end-stage heart failure, but little is known about the underlying events. We therefore investigated the expression of genes coding for major inward and outward ion channels, calcium binding proteins, ion receptors, ion exchangers, calcium ATPases, and calcium/calmodulin-dependent protein kinases in explanted hearts (n=13) of patients diagnosed with end-stage heart failure. With the exception of Kv11.1 and Kir3.1 and when compared with healthy controls, major sodium, potassium, and calcium ion channels, ion transporters, and exchangers were significa…

Coupled movement of permeant and blocking ions in the CFTR chloride channel pore.

The cystic fibrosis transmembrane conductance regulator (CFTR) Cl- channel pore is blocked in a voltage-dependent manner by a broad range of anionic substances added to the cytoplasmic side of the membrane. Here we investigate the origin of the voltage dependence of block by intracellular Au(CN)2-, a highly permeant lyotropic anion which also acts as a high-affinity blocker of Cl- permeation. Not only the affinity, but also the voltage dependence of block by intracellular Au(CN)2- ions is strongly dependent on extracellular Cl- concentration; following replacement of most extracellular Cl- by

Several different ion channel modulators abrogate contact hypersensitivity in mice.

A major obstacle in transdermal delivery of drugs is the development of adverse skin sensitization reactions. We tested the concept that ion channel modulators as a class of agents suppress contact hypersensitivity in a mouse model. Mice were sensitized to several contact sensitizing chemicals including dinitrochlorobenzene (DNCB) and a sensitizing drug, nadolol. We report our successful use of several ion channel modulators in suppressing contact hypersensitivity, including amiloride, ethacrynic acid (ECA), nifedipine and verapamil. For this purpose, Balb/c female mice were sensitized with DN

Multiple ion channel block by the cation channel inhibitor SKF-96365 in myocytes from the rabbit atrioventricular node.

The atrioventricular node (AVN) of the cardiac conduction system coordinates atrial-ventricular excitation and can act as a subsidiary pacemaker. Recent evidence suggests that an inward background sodium current, IB,Na, carried by nonselective cation channels (NSCCs), contributes to AVN cell pacemaking. The study of the physiological contribution of IB,Na has been hampered, however, by a lack of selective pharmacological antagonists. This study investigated effects of the NSCC inhibitor SKF-96365 on spontaneous activity, IB,Na, and other ionic currents in AVN cells isolated from the rabbit. Wh…

Are B-type Ca2+ channels of cardiac myocytes akin to the passive ion channel in the plasma membrane Ca2+ pump?

The present study demonstrates that B-type Ca2+ channels observed in rat ventricular myocytes markedly reacted to agents known to affect the ion-motive plasma membrane Ca2+-ATPase (PMCA) pump. Chlorpromazine (CPZ)-activated B-type Ca2+ channels were completely blocked by internal application of PMCA pump inhibitors, namely La3+ (100 microm), eosin (10 microm) and AIF(3) (100 microm). Calmodulin (50 U/ml), the main endogenous positive regulator of PMCA, was unable to activate but significantly reduced CPZ-activated B-type channel activity. In the same manner, ATP (1 and 4 mm), the main energizi

A mechanosensitive ion channel is required for nematode predation in nematode-trapping fungi.

UNLABELLED: Mechanosensitive ion channels of small conductance-like channels (MSL) are conserved in cell-walled organisms and enable cellular responses to mechanical stimuli. Nematode-trapping fungi (NTF) rely on nematode-derived mechanical cues to form specialized predatory structures, including adhesive network and mechanically actuated constricting ring (CR) that rapidly inflate upon nematode contact. However, the sensory mechanisms underlying these processes remain unclear. Here, using the adhesive network-forming Arthrobotrys oligospora and the CR-forming Drechslerella dactyloides as repr…

Ion channel gene signature for diagnosis and antifibrotic therapy in liver fibrosis.

BACKGROUND: Liver fibrosis (LF) is a progressive pathological process that may lead to cirrhosis and liver failure. Human ion channel genes (HICGs) participate in hepatic mechanotransduction and immune regulation, but their contributions to LF remain insufficiently characterized. This study aimed to profile the expression of HICGs in LF and to identify key genes with diagnostic and therapeutic relevance. METHODS: Multiple transcriptomic datasets were integrated to identify differentially expressed HICGs in LF. Weighted gene co-expression network analysis and single-cell RNA sequencing were ap

From ion channel biology to clinical practice: suzetrigine as a precision analgesic.

Acute pain remains one of the most prevalent and burdensome clinical challenges requiring pharmacological intervention, yet opioids-with their well-documented risks of respiratory depression, addiction, and overdose-remain a cornerstone of moderate-to-severe pain management in the absence of efficacious alternatives. Suzetrigine (VX-548) is a novel preferential inhibitor of the voltage-gated sodium channel (VGSC) isoform Nav1.8, recently approved by the US Food and Drug Administration for the treatment of moderate-to-severe acute pain. Through specific inhibition of Nav1.8, which is expressed

How to Perform a Tracer Displacement BRET Assay for the TRPML1 Ion Channel.

The transient receptor mucolipin subtype 1 (TRPML1) is a ubiquitously expressed ion channel involved in lysosomal homeostasis. Recent pharmaceutical interest in developing agonist ligands has emerged due to beneficial effects in neurodegenerative diseases. The major high-throughput screening techniques to investigate this ion channel involve fluorescent calcium imaging and electrophysiology. Despite their high capacity for screening compounds, it is well known that both methods face hurdles, such as the need for expensive, specialized equipment. Here, we present a novel technique to screen for

Ion Channel-Targeting Modulators in Glioma: Pharmacological Advances and Therapeutic Perspectives.

Highly aggressive gliomas remain difficult to treat because of their marked invasiveness, frequent recurrence, and association with glioma-related epilepsy. Increasing evidence suggests that ion channels expressed in glioma cells, including voltage-gated Na+ channels, large-conductance Ca2+-activated K+ (BKCa) channels, intermediate-conductance Ca2+-activated K+ (IKCa) channels, and inwardly rectifying K+ (Kir) channels, contribute to these malignant phenotypes and represent potential therapeutic targets. This perspective summarizes current evidence for eight representative drugs and phytocons…

The structural basis of the G protein-coupled receptor and ion channel axis.

Sensory neurons play an essential role in recognizing and responding to detrimental, irritating, and inflammatory stimuli from our surroundings, such as pain, itch, cough, and neurogenic inflammation. The transduction of these physiological signals is chiefly mediated by G protein-coupled receptors (GPCRs) and ion channels. The binding of ligands to GPCRs triggers a signaling cascade, recruiting G proteins or β-arrestins, which subsequently interact with ion channels (e.g., GIRK and TRP channels). This interaction leads to the sensitization and activation of these channels, initiating the neur…

Targeted drug screening for autism based on Cav1.2 calcium ion channel.

This study presents a targeted virtual drug screening approach for autism spectrum disorder (ASD), focusing on Cav1.2 calcium ion channels as potential therapeutic targets. ASD is a complex neurodevelopmental disorder characterized by impairments in social communication and behavior, with genetic factors playing a significant role. Cav1.2 channels have been implicated in the pathophysiology of ASD due to their role in regulating neuronal excitability and synaptic transmission. We employed computational methods to virtually screen a large database of compounds for their potential to modulate Ca…

ASIC1a-Dependent Potentiation of Acid-Sensing Ion Channel Currents by Cyanide.

Cyanide (CN) is a potent, fast-acting toxicant that impacts endogenous biomolecules in the nervous system, including acid-sensing ion channels (ASICs), which play a vital role in various neurological and psychological conditions. Here, we demonstrate that CN rapidly potentiates ASIC currents in cultured mouse cortical neurons in a dose-dependent manner while causing a leftward shift in the pH dose-response curve. Notably, this potentiation was unaffected by a 30-min CN treatment or the presence of ATP in the recording pipette. Further investigations into the role of zinc revealed that TPEN, a …

A privileged ER compartment for post-translational heteromeric assembly of an ion channel.

Mechanisms underlying heterotypic subunit assembly of ion channels and other oligomeric complexes are poorly understood. In the human heart, heteromeric assembly of two isoforms encoded by the human ether-à-go-go related gene (hERG) is essential for the normal function of cardiac IKr in ventricular repolarization, with loss of hERG1b contributing to arrhythmias associated with long QT-syndrome (LQTS). While hERG1a homomers traffic efficiently to the plasma membrane, hERG1b homomers are retained in the endoplasmic reticulum (ER). When expressed together, the two subunits avidly associate during…

The TRPA1 Ion Channel Mediates Oxidative Stress-Related Migraine Pathogenesis.

Although the introduction of drugs targeting calcitonin gene-related peptide (CGRP) revolutionized migraine treatment, still a substantial proportion of migraine patients do not respond satisfactorily to such a treatment, and new therapeutic targets are needed. Therefore, molecular studies on migraine pathogenesis are justified. Oxidative stress is implicated in migraine pathogenesis, as many migraine triggers are related to the production of reactive oxygen and nitrogen species (RONS). Migraine has been proposed as a superior mechanism of the brain to face oxidative stress resulting from ener
