Density dependence and overdispersion in the transmission of helminth parasites.

The influence of density-dependent processes on the transmission of parasitic helminths is determined by both the severity of the regulatory constraints and the degree of parasite overdispersion among the host population. We investigate how overdispersed parasite distributions among humans influence transmission levels in both directly- and indirectly-transmitted nematodes (Ascaris lumbricoides and Onchocerca volvulus). While past work has assumed, for simplicity, that density dependence acts on the average worm load, here we model density-dependence as acting on individual parasite burdens be

The distribution and ecological 'preferences' of the tick Amblyomma cajennense (Acari: Ixodidae), an ectoparasite of humans and other mammals in the Americas.

The distribution and ecological 'preferences', in the Nearctic and Neotropical regions, of the prominent, pathogen-carrying tick Amblyomma cajennense (Fabricius) were reviewed, on the basis of the published literature, satellite imagery, and the examination of several tick collections. The tick has been collected from the southern U.S.A. (Texas and Florida) to northern Argentina, but is probably absent from Uruguay and southern Brazil (the few records of the species in these areas probably represent mis-identifications). The ecological conditions prevailing in the areas where the tick has been

Surgical treatment of hepatic hydatid cyst.

Hydatid disease of the liver is caused by the tapeworm Echinococcus granulosus, and the highest incidence of human infestation occurs in sheep and cattle-raising areas. Although, still an uncommon occurrence, it is being seen with increasing frequency; hence, it is important that surgeons make themselves aware of the pathology and treatment of the disease. The liver is most commonly involved, although many other organs, including lungs and brain may also be affected. An echinococcal cyst of the liver was reported in a patient who is a resident in Northern Ireland but had originally lived and w

Marine dinoflagellates show induced life-history shifts to escape parasite infection in response to water-borne signals.

Many dinoflagellate species form dormant resting cysts as a part of their life cycle, and in some freshwater species, hatching of these cysts can be delayed by the presence of water-borne signals from grazing zooplankton. Some marine dinoflagellates can form temporary cysts, which may function to resist unfavourable short-term environmental conditions. We investigated whether the marine dinoflagellate Alexandrium ostenfeldii is able to induce an increased resistance to the parasitic flagellate Parvilucifera infectans by forming temporary cysts. We performed several laboratory experiments where

Stowaways with wings: two case reports on high-flying insects.

More people than ever before are traversing continents, either for business purposes or on holiday. Because 3-10 percent of these travelers experience skin, hair, or nail disorders related to these trips there is an increasing likelihood that Western physicians will be expected to treat exotic conditions imported from different countries. Tungiasis and furunculoid myiasis are two typical disorders of intertropical regions. They represent nuisances induced by the presence of arthropod larvae or eggs in the skin. We describe a case of tungiasis, caused by the sand flea Tunga penetrans (TP), and

Animal models for the analysis of immune responses to leishmaniasis.

This unit focuses on the murine model of cutaneous leishmaniasis and models of visceral leishmaniasis in mice and hamsters. Each basic protocol describes the methods used to inoculate parasites and to evaluate infections with regard to lesion progression and visceralization, and quantification of parasite load. Five support protocols are provided; two for the growth and isolation of metacyclic promastigotes from in vitro culture, one for isolation of tissue amastigotes from infected animals, one for cryopreservation of parasites, and one for the preparation of blood agar plates for quantitatio

[Levels of malaria transmission: methods and parameters].

Evaluation of malaria transmission levels is necessary to compare ecologically diverse areas and to assess the effectiveness of efforts to control the disease. The purpose of this report is to describe useful techniques for descriptive epidemiology and potentially pertinent indicators regarding the three links in the epidemiological chain: transmission from mosquito to man, transmission from man to mosquito, and sporogonic cycle. Standards for evaluation of transmission from mosquito to man are now well established. Techniques and resulting data, mostly entomological, have been validated in nu

The dimerisable Cre recombinase allows conditional genome editing in the mosquito stages of Plasmodium berghei.

Asexual blood stages of the malaria parasite are readily amenable to genetic modification via homologous recombination, allowing functional studies of parasite genes that are not essential in this part of the life cycle. However, conventional reverse genetics cannot be applied for the functional analysis of genes that are essential during asexual blood-stage replication. Various strategies have been developed for conditional mutagenesis of Plasmodium, including recombinase-based gene deletion, regulatable promoters, and mRNA or protein destabilization systems. Among these, the dimerisable Cre

Proteomic analysis of differentially expressed proteins in the three developmental stages of Trichinella spiralis.

Trichinella spiralis, an intracellular parasitic nematode, can cause severe foodborne zoonosis, trichinellosis. The life cycle of T. spiralis consists of adult (Ad), muscle larvae (ML) and newborn larvae (NBL). The protein profiles in different developmental stages of the parasite remain unknown. In the present study, proteins from lysates of Ad, ML and NBL were identified by isobaric tags for relative and absolute quantitation (iTRAQ). A total of 4691 proteins were identified in all the developmental stages, of which 1067 proteins were differentially expressed. The number of up-regulated prot

Somatostatin concentrations in the pancreatic-intestinal tissues of the sea lamprey, Petromyzon marinus L., at various periods of its life cycle.

1. Somatostatin concentrations were measured in homogenates of the pancreas-intestinal tissues from each period of the life cycle of Petromyzon marinus using radioimmunoassay. 2. Levels were very low in larva (4.0 pg/mg wet weight) and in the first three stages of metamorphosis, but increased from stage 4 onwards and reached a high in upstream-migrating adults (210.0 ng/mg). 3. These data correlate well with our previous morphological and immunohistochemical observations on the morphogenesis of somatostatin-containing D-cells during the life cycle and indicate that the increased concentration

Analysing how plants in coastal wetlands respond to varying tidal regimes throughout their life cycles.

Important to conserve plant species in coastal wetlands throughout their life cycle. All life stages in these habitats are exposed to varying tidal cycles. It is necessary to investigate all life stages as to how they respond to varying tidal regimes. We examine three wetlands containing populations of an endangered halophyte species, each subjected to different tidal regimes: (1). wetlands completely closed to tidal cycles; (2). wetlands directly exposed to tidal cycles (3). wetlands exposed to a partially closed tidal regime. Our results showed that the most threatened stage varied between w

Cultivation of plasmodium spp.

Cultivation of both human and non-human species of Plasmodium spp., the causal agent of malaria, has been a major research success, leading to a greater understanding of the parasite. Efforts at cultivating the organisms in vitro are complicated by the parasites' alternating between a human host and an arthropod vector, each having its own set of physiological, metabolic, and nutritional parameters. Life cycle stages of the four species that infect humans have been established in vitro. Of these four, P. falciparum remains the only species for which all stages have been cultured in vitro; diff…

New Developments in the Treatment of Hepatitis B Virus Infection.

Chronic hepatitis B virus (HBV) infection remains a serious global public health concern and a leading cause of liver cirrhosis and hepatocellular carcinoma. Although currently available oral therapies can effectively suppress HBV DNA, there is no approved regimen that has shown to reliably achieve a functional cure. In recent years, various classes of novel therapy for HBV infection have undergone considerable research and advanced to clinical trials, targeting multiple stages of the viral life cycle and systemic immunomodulatory pathways. This article discusses the complex HBV life cycle and

Dengue Virus Life Cycle and Host Protein Interactions: Focus on RNA Binding Proteins and Therapeutic Advances.

Dengue, a widespread arthropod-borne viral disease, remains endemic in more than 100 nations, affecting over 40% of the world's population, with millions of cases reported annually, and has a major burden on global public health systems. The causative agent of this infection is the dengue virus which belongs to the Flaviviridae family of RNA viruses. The DENV infection leads to a broad spectrum of clinical symptoms, ranging from mild to severe, and even fatal in the cases of secondary infection. In the absence of promising antiviral therapies or vaccines to effectively combat the infection, un

Receptors for growth and development of Schistosoma mansoni.

The growth and development of schistosomes are tightly regulated by various receptors throughout their life cycle. Each stage of the parasite inhabits a distinct habitat and responds to different factors that drive its growth and development. With two hosts involved in its life cycle (mammalian and snail), the parasite must go through additional free-living stages to transition between them. Moreover, communication between male and female worms is essential for the maturation of females. The ability of adult schistosomes to survive in human hosts for up to thirty years demonstrates their capac

Colpodella sp. (ATCC 50594) Life Cycle: Myzocytosis and Possible Links to the Origin of Intracellular Parasitism.

Colpodella species are free living bi-flagellated protists that prey on algae and bodonids in a process known as myzocytosis. Colpodella species are phylogenetically related to Apicomplexa. We investigated the life cycle of Colpodella sp. (ATCC 50594) to understand the timing, duration and the transition stages of Colpodella sp. (ATCC 50594). Sam-Yellowe's trichrome stains for light microscopy, confocal and differential interference contrast (DIC) microscopy was performed to identify cell morphology and determine cross reactivity of Plasmodium species and Toxoplasma gondii specific antibodies

ApiAP2 Transcription Factors in Apicomplexan Parasites.

Apicomplexan parasites are protozoan organisms that are characterised by complex life cycles and they include medically important species, such as the malaria parasite Plasmodium and the causative agents of toxoplasmosis (Toxoplasma gondii) and cryptosporidiosis (Cryptosporidium spp.). Apicomplexan parasites can infect one or more hosts, in which they differentiate into several morphologically and metabolically distinct life cycle stages. These developmental transitions rely on changes in gene expression. In the last few years, the important roles of different members of the ApiAP2 transcripti…
