The effect of binding of spider-derived antimicrobial peptides, oxyopinins, on lipid membranes.

Oxyopinins (Oxki1 and Oxki2) are antimicrobial peptides isolated from the crude venom of the wolf spider Oxyopes kitabensis. The effect of oxyopinins on lipid bilayers was investigated using high-sensitivity titration calorimetry and (31)P solid-state NMR spectroscopy. High-sensitivity titration calorimetry experiments showed that the binding of oxyopinins was exothermic, and the binding enthalpies (DeltaH) to 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC) small unilamellar vesicles (SUVs) were -18.1 kcal/mol and -15.0 kcal/mol for Oxki1 and Oxki2, respectively, and peptide parti…

The interaction of local anaesthetics with synthetic phospholipid bilayers.

The preferred position and orientation of two local anaesthetics in lipid bilayers has been determined using fluorescence quenching techniques. The aromatic amine of tetracaine and butesin quenches the fluorescence of a series of n-(9-anthroyloxy) fatty acids (n = 2,6,9,12,16) which place a fluorophore at a graded series of depths from the surface to the centre of a bilayer. A fluorescence method is used to resolve partition coefficients in the transverse plane of the membrane. The results show that the anaesthetics take up a distribution of positions about one or more preferred maxima. The ar…

Membrane-Modifying Effects of Perfluoroalkyl Substances in Model Bacterial Membranes.

Understanding the interactions of per- and polyfluoroalkyl substances (PFAS) with bacterial membranes is essential for evaluating their ecological and health impacts. To mimic the diverse environments found in bacterial membranes, we constructed model membranes as bilayers, liposomes, and supported bilayers using binary lipid mixtures of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) or 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), with 1,2-dioleoyl-sn-glycero-3-phospho-(1'-rac-glycerol) (sodium salt) (DOPG)all sharing the same acyl chains but differing in headgroup types and charges

Phase changes in mixed lipid/polymer membranes by multivalent nanoparticle recognition.

Selective addressing of membrane components in complex membrane mixtures is important for many biological processes. The present paper investigates the recognition between multivalent surface functionalized nanoparticles (NPs) and amphiphilic block copolymers (BCPs), which are successfully incorporated into lipid membranes. The concept involves the supramolecular recognition between hybrid membranes (composed of a mixture of a lipid (DPPC or DOPC), an amphiphilic triazine-functionalized block copolymer TRI-PEO13-b-PIB83 (BCP 2), and nonfunctionalized BCPs (PEO17-b-PIB87 BCP 1)) with multivalen

Interaction of Choline-Based Ionic Liquids with Model Lipid Membranes: Force-Field Parametrization and Membrane Partitioning.

In recent years, ionic liquid (IL)-based formulations have gained attention for their potential use in drug delivery and antibacterial and antiseptic applications. Molecular dynamics simulations can provide insights into complex interaction mechanisms, serving as valuable tools to guide experimental efforts to design novel formulations. However, to study interactions that involve micellar aggregates and membrane partitioning dynamics, simulations should be able to access time scales of several microseconds, requiring reliably parametrized coarse-grained molecular models. In this study, we inve

The amyloidogenic C-terminal region of TMEM106B modulates lipid membrane biophysical properties: Functional and pathological insights.

The lysosomal transmembrane protein 106B (TMEM106B) forms amyloid filaments in the human brain in an age-dependent manner, observed both in … lar vesicles of varying lipid composition that mimic lysosomal membranes. TST binds peripherally to lipid bilayers and remodels their fluidity and elasticity in a composition-dependent manner. 31P NMR spectra revealed reduced chemical shift anisotropy and increased asymmetry, accompanied by an isotropic component indicative of enhanced headgroup motion and local …

Effect of very long-chain lipids on the organization of biological membranes: A simulation study.

Lipids in all biological membranes are distributed heterogeneously across the bilayer. A particularly striking example of this asymmetry is the yeast plasma membrane, which exhibits a high concentration of very long-chain tail-asymmetric sphingolipids in its outer leaflet. Experimental observations indicate the existence of highly ordered gel-like plasma membrane domains that are enriched in sphingolipids but depleted of the major yeast sterol ergosterol. For a better mechanistic understanding of these unusual domains, we have performed coarse-grained molecular dynamics simulations with membra

Coarse-grained molecular dynamics simulation analysis of the effects of phospholipid and cholesterol on electroporation.

Membrane composition is a critical factor for electroporation. Although existing research focuses on pore formation driven by local phospholipid headgroup clusters, the large-scale membrane dynamics post-pore formation remain underexplored. In this study, coarse-grained (CG) molecular dynamics (MD) simulations were employed to investigate the effects of phospholipid headgroups (PC, PE and PS), tail characteristics (DLPC, DPPC, POPC and DOPC) and cholesterol (CHOL) content (0 mol%-50 mol%) on membrane electroporation. The results demonstrate that membrane structural parameters, such as area per

Biointerfacial mechanical responses of supported lipid membranes to Gramicidin D revealed by QCM-D.

Gramicidin D (GrD) is a well-established model ion channel-forming peptide whose mechanical interactions with supported lipid membrane architectures remain less well characterized. Here, we combine overtone-resolved Quartz Crystal Microbalance with Dissipation (QCM-D), fluorescence microscopy, and solution-phase assays to examine how GrD perturbs supported lipid bilayers (SLBs) and supported vesicular layers (SVLs) formed from neutral DOPC and anionic DOPC:DOPS (92:08). On neutral SLBs at low peptide levels, GrD induces mild outer-leaflet softening consistent with β6.3 channel formation, produ…

Computational structure-based approach to study chimeric antigens using a new protein scaffold displaying foreign epitopes.

The identification and recombinant production of functional antigens and/or epitopes of pathogens represent a crucial step for the development of an effective protein-based vaccine. Many vaccine targets are outer membrane proteins anchored into the lipidic bilayer through an extended hydrophobic portion making their recombinant production challenging. Moreover, only the extracellular loops, and not the hydrophobic regions, are naturally exposed to the immune system. In this work, the Domain 3 (D3) from Group B Streptococcus (GBS) pilus 2a backbone protein has been identified and engineered to

Simulations of naïve and KLA-activated macrophage plasma membrane models.

Macrophages (MAs), which play vital roles in human immune responses and lipid metabolisms, are implicated in the development and progression of atherosclerosis, a major contributor to cardiovascular diseases. Specifically, the abnormal lipid metabolism of oxidized low-density lipids (oxLDLs) in MAs is believed to be a crucial factor. However, the precise mechanism by which the MA membrane contributes to this altered lipid metabolism remains unclear. Lipidomic studies have revealed significant differences in membrane composition between various MA phenotypes. This study serves to provide and ch

Discovery of Kinetic Trapping of Poloxamers inside Liposomes via Thermal Treatment.

Poloxamers, a class of biocompatible, commercially available amphiphilic block polymers (ABPs) comprising poly(ethylene oxide) (PEO) and poly(propylene oxide) (PPO) blocks, interact with phospholipid bilayers, resulting in altered mechanical and surface properties. These block copolymers are useful in a variety of applications including therapeutics for Duchenne muscular dystrophy, as cell membrane stabilizers, and for drug delivery, as liposome surface modifying agents. Hydrogen bonding between water and oxygen atoms in PEO and PPO units results in thermoresponsive behavior because the bound …

Development of Robust Cationic Light-Activated Thermosensitive Liposomes: Choosing the Right Lipids.

Extensive research has been conducted on cationic light-activated thermosensitive liposomes (CLTSLs) as a means for site-specific and controlled drug release; however, less attention has been given to the stability of these nanoparticles. Selecting the appropriate lipids is crucial for the development of a stable and responsive system. In this study, we investigated the impact of various lipids on the physical properties of cationic light-activated liposomes. Incorporating poly(ethylene glycol) PEG molecules resulted in uniform liposomes with low polydispersity index, while the addition of uns

Protonation and orientation: a computational approach to cocaine diffusion through a model membrane.

We study the diffusion of cocaine through a DMPC lipid bilayer as an example of a protonable, amphiphilic molecule passing a biological membrane. Using classical molecular dynamics simulations, the free energy surfaces are computed applying the umbrella sampling technique for the protonated and the neutral molecule. For the combined surface, we numerically solve the diffusion equation at constant flow and for time-dependent concentrations. We find a potential of mean force dominated by a barrier of 3.5 kcal mol-1 within the membrane, and a pH-dependent entry and exit barrier of 2.0 kcal mol-1

Self-assembly, stability and conductance of amphotericin B channels: bridging the gap between structure and function.

Amphotericin B (AmB), one of the most powerful but also toxic drugs used to treat systemic mycoses, is believed to selectively permeabilize fungal cell membranes to ions in a sterol-dependent manner. Unfortunately, the structure of the biologically active AmB channels has long eluded researchers, obstructing the design of safer alternatives. Here, we investigate the structural and thermodynamic aspects of channel formation, stability, and selective ion conduction. We combine fluorescence lifetime imaging and molecular simulations to trace the process of channel assembly until the formation of

Efficient electroformation of supergiant unilamellar vesicles containing cationic lipids on ITO-coated electrodes.

Giant unilamellar vesicles (GUVs) represent a versatile in vitro system widely used to study properties of lipid membranes and their interaction with biomacromolecules and colloids. Electroformation with indium tin oxide (ITO) coated coverslips as electrodes is a standard approach to GUV production. In the case of cationic GUVs, however, application of this approach leads to notorious difficulties. We discover that this is related to aging of ITO-coated coverslips during their repeated use, which is reflected in their surface topography on the nanoscale. We find that mild annealing of the ITO-

Partition of N-monodemethylated phenothiazine drugs to phosphatidylcholine bilayer vesicles studied by second-derivative spectrophotometry.

The partition coefficients (Kps) of three N-monodemethylated metabolites of phenothiazines (chlorpromazine (CPZ), triflupromazine, and promazine) between phosphatidylcholine (PC) vesicles (SUV) and water were determined to evaluate their affinity to biomembranes by second-derivative spectrophotometry without any separation procedures. The second derivative spectra showed distinct derivative isosbestic points confirming the entire elimination of the residual background signal effects of the SUV, which were observed in the absorption spectra. From the relationship between PC SUV concentration an

[Modification of latrotoxin channels under the effect of pronase and antibodies to latrotoxin].

The action of pronase and latrotoxin antibodies on alpha-latrotoxin channels in lipid bilayers has been investigated. Proteolysis of the protruding portions of the inserted channels changed slightly its conductivity and selectivity. Latrotoxin antibodies added to either side of the membrane decreased channel conductivity. The effect depends on the membrane potential. It is suggested that the channel conformation depends on the transmembrane potential and selective centre localized in the inner region of a bilayer is insensitive to proteolysis of protruding segments.

Bilayer Elastic Properties Modulate Interface Energetics of Transmembrane β-Barrels.

Assembly energetics of transmembrane proteins occurs through the orchestrated molecular interplay of the protein's membrane partitioning free energy and the mechanical and physicochemical properties of the bilayer. While amphipathic residues help accommodate diametrically opposite forces and the steep energy gradient at the membrane-water interface, the magnitude to which the intrinsic curvature and elastic energy of the bilayer regulates protein thermodynamics at the interface remains poorly understood. Here, using PagP as a model β-barrel, we compare the per-residue energetics of an interfac
