Cell-mediated immune responses to chlamydial antigens in guinea pigs injected with inactivated chlamydiae.

Cell-mediated immunity (CMI) to chlamydial antigens was readily induced in guinea pigs by a single injection of Betaprone-inactivated chlamydiae in complete Freund adjuvant. The CMI was measured in vivo by delayed hypersensitivity skin tests, and in vitro by inhibition of migration of peritoneal exudate cells and by proliferation of lymph node lymphocytes. There was an overall correlation between in vivo and in vitro responses. Of the in vitro assays, migration inhibition reflected the state of sensitization, as judged by skin tests, more uniformly than lymphocyte stimulation. Extensive inter-

Defective leukotaxis in patients with lepromatous leprosy.

Serums from patients with lepromatous leprosy show a high incidence of a chemotactic inhibitor. This inhibitor acts directly on leukotactic factors (bacterial chemotactic factor, C3 fragment, and C5 fragment) to render the factors irreversibly inactive. Functionally, the inhibitor acts as a chemotactic factor inactivator. While normal serum shows no inhibitory activity under the conditions employed, inhibitory activity causing more than 30 per cent reduction of the bacterial chemotactic factor was found in the serums from 14 of 19 patients with lepromatous leprosy. Although exceptions were not

Production of MIF-like supernatants by rheumatoid arthritis lymphocytes stimulated by immunoglobulin G.

Patients with rheumatoid arthritis (RA), patients with positive tuberculin (PPD) skin-tests and controls were studied. The lymphocytes from these groups were cultured in serum-free medium to obtain cell-free supernatants. These lymphocyte cultures were pre-incubated with the appropriate antigen or reconstituted after removal of the cells. Supernatants from RA lymphocytes stimulated in vitro by IgG induced an inhibition of the leucocyte migration, as well as the supernatants from tuberculin-sensitized lymphocytes. However, supernatants from non-RA lymphocytes or tuberculin-unsensitized lymphocy

Kupffer cell reverse migration into the liver sinusoids mitigates neonatal sepsis and meningitis.

In adults, liver-resident macrophages, or Kupffer cells (KCs), reside in the sinusoids and sterilize circulating blood by capturing rapidly flowing microbes. We developed quantitative intravital imaging of 1-day-old mice combined with transcriptomics, genetic manipulation, and in vivo infection assays to interrogate increased susceptibility of newborns to bloodstream infections. Whereas 1-day-old KCs were better at catching Escherichia coli in vitro, we uncovered a critical 1-week window postpartum when KCs have limited access to blood and must translocate from liver parenchyma into the sinuso

Differentiation of mycobacteria by "in vivo" and "in vitro" delayed hypersensitivity reactions.

Guinea pigs were sensitized with heat killed M. tuberculosis, M. kansasii, M. scrofulaceum, and M. avium-intracellulare. Purified Protein Derivatives (PPDs) prepared from each of these organisms were tested for delayed type hypersensitivity by lymphocyte transformation, macrophage migration, and mantoux type skin sensitivity tests. The homologous reactions were consistently more intense than heterologous reactions in all three procedures. The skin test procedure was more sensitive than either in vitro method. Positive skin test reactions were elicited with 2 times and 50 times less antigen tha

Leucocyte migration inhibition in liver disease.

Clinical and histological features provide evidence that immunological reactions of the cell-mediated type are abnormal in some forms of chronic liver disease. By using the leucocyte migration test of Søborg and Bendixen (1967), a correlate of cellular hypersensitivity in vitro, the authors studied the production of the migration inhibitory factor by lymphocytes exposed to a fetal liver homogenate in 88 patients with acute and chronic liver disease. Abnormality of leucocyte migration was found in aggressive chronic hepatitis (inhibition in 67% of patients), cryptogenic cirrhosis (inhibition in

[Mechanism of MIF in regulating HIF-1α-mediated aerobic glycolysis in pulmonary fibrosis].

Objective To investigate the mechanism by which macrophage migration inhibitory factor (MIF) regulates hypoxia-inducible factor 1α (HIF-1α)-mediated aerobic glycolysis in the pathological process of pulmonary fibrosis. MethodsC57BL/6 mice were divided into four groups: control group, bleomycin (BLM) model group, BLM combined with negative control lentivirus (BLM combined with sh-NC) group, and BLM combined with MIF-shRNA lentivirus (BLM combined with sh-MIF) group. After 28 days of intratracheal administration, lung tissues, bronchoalveolar lavage fluid (BALF), and serum samples were collected

Oxidized MIF is an Alzheimer's disease drug target relaying external risk factors to tau pathology.

During deep co-evolution of viruses and host cells, viruses have selected specific host cellular proteins redirected from physiological functions to viral needs, thereby disturbing cellular proteostasis and increasing the risk of triggering protein misfolding diseases (PMDs). Identifying virus-specific, repurposed host proteins also allows the study of fundamental cellular events in "sporadic" PMDs, independent of the virus. Here, we identify a small molecule with very strong activity against neurotropic herpes simplex virus 1 (HSV-1), modulating an allosteric site of macrophage migration inhi

Mitogenic factor as an in vitro correlate of delayed hypersensitivity in the guinea pig.

The relationship of mitogenic factor (MF) to delayed hypersensitivity was examined in the guinea pig. MF was readily produced in vitro by blood leucocytes from animals sensitized with Keyhole limpet haemocyanin in Freund's complete adjuvant. Such animals exhibited delayed skin reactions and macrophage migration inhibition. Treatment with cyclophosphamide markedly reduced serum antibody but caused no diminution of MF or delayed skin reactions. A migration inhibition response was observed 10, but not 20, days after sensitization by means of incomplete adjuvant.

Experimental production of granulomatous pneumonitis. Comparison of immunological and morphological sequelae with particulate and soluble antigens administered via the respiratory route.

Rabbits were sensitized with either a soluble protein antigen (BSA) or a particulate thermophilic actinomycete antigen (Micropolyspora faeni) via the respiratory route, followed by monitoring of sequential morphologic changes and the humoral plus cellular immunologic response. Primary respiratory tract sensitization with BSA resulted in a humoral anti-BSA response, Arthus and delayed skin reactivity, and in some cases specific antigen-induced alveolar macrophage migration inhibition, all in the absence of pulmonary lesions. Lesions characterized by mild multifocal perivascular mononuclear cell

TCEB2 promotes M2 polarization of macrophages in triple negative breast cancer by mediating ubiquitination degradation of Slit2 through recruiting NEDD4.

BACKGROUND: M2-polarized tumor-associated macrophage infiltration is a key risk factor for poor prognosis in triple-negative breast cancer (TNBC). This study investigated the role of transcription elongation factor B polypeptide 2 (TCEB2) in regulating M2 macrophage polarization during TNBC development. METHODS: The expression of gene or protein was tested by qRT-PCR, western blot, and IHC. CCK8, colony formation, wound healing, and Transwell assays were used to evaluate TNBC cell malignant behaviors, including cell viability, proliferation, migration, and invasion. The secretion levels of cy

Low-dose naltrexone inhibits the epithelial-mesenchymal transition of cervical cancer cells in vitro and effects indirectly on tumor-associated macrophages in vivo.

The metastasis of cervical cancer has always been a clinical challenge. We investigated the effects of low-dose naltrexone (LDN) on the epithelial mesenchymal transition of cervical cancer cells in vitro as well as its influence on macrophage polarization and associated cytokines in vivo. The results suggested that LDN supressed the proliferation, migration and invasion abilities and promote their apoptosis in Hela cells, whereas the opioid growth factor receptor (OGFr) silenced significantly reversed these effects in vitro. Knockdown the expression of OGFr, the inhibitory of LDN on EMT was we

Evidence for cell-mediated autoimmunity in patients with pemphigus vulgaris and bullous pemphigoid.

Lymphoid cells from 4 of 5 patients diagnosed as pemphigus vulgaris (PV) and 6 of 7 patients diagnosed as bullous pemphigoid (BP) demonstrated specific cell-mediated immunity by the production of migration inhibitory factor (MIF) in the presence of autologous epidermal saline extracts. Clinical treatment of these patients with immunosuppressive agents resulted in a state of unresponsiveness of their lymphoid cells to similar concentrations of the antigen. Controls consisted of lymphoid cells from patients with bullous burns or various drug allergies which failed to show significant MIF product

Abrogation of cell-mediated immunity by a serum blocking factor isolated from patients with infectious mononucleosis.

Lymphocytes from 80% of patients with infectious mononucleosis in this study failed to produce macrophage migration-inhibition factor in response to partially purified early antigen of Epstein-Barr virus or to tetanus toxoid, whereas lymphocytes from normal subjects did produce this lymphokine. Subsequent analysis of serum from the patients with infectious mononucleosis revealed a serum factor that completely abrogated antigen-specific inhibition of migration by human leukocytes as well as lymphocyte blastogenesis. The serum blocking factor was present in sera from 11 (73%) of 15 patients with

Cepharanthine inhibits in vitro VSMC proliferation and migration and vascular inflammatory responses mediated by RAW264.7.

Pathogenesis of atherosclerosis involves vascular smooth muscle cell (VSMC) migration and proliferation followed by an inflammation mediated by activated macrophages in the tunica intima of blood vessels. Cepharanthine (CEP) belongs to bisbenzylisoquinoline alkaloids found in the plant Stephania cepharantha, which has been used for various diseases like cancer, alopecia areata, venomous snakebites, and malaria. In this study, we investigated whether CEP suppresses VSMC migration and proliferation and inhibits inflammatory mediator production in macrophage (RAW264.7). Our results showed that CE

Escherichia coli inhibits endometriosis by inducing M1 polarity of peritoneal macrophages and the IL-1 signaling pathway.

The development of endometriosis is closely linked to macrophages, and the type M1 macrophage has been hypothesized to play an inhibitory role in its progression. Escherichia coli induces macrophage polarization toward M1 in numerous diseases and differs in the reproductive tract of patients with and without endometriosis; however, its specific role in endometriosis development remains unknown. Therefore, in this study, E. coli was selected as a stimulator to induce macrophages, and its effects on the growth of endometriosis lesions in vitro and in vivo were investigated using C57BL/6N female

Cell-mediated immunity after bacterial infection of the lower respiratory tract.

Lower respiratory tract and systemic cell-mediated immunity have been studied in rabbits after infection with Listeria monocytogenes or Diplococcus pneumoniae. Respiratory tract cell-mediated immunity was evaluated by direct and indirect assays of migration inhibitory factor (MIF) production. Systemic delayed hypersensitivity was determined by means of intradermal testing with appropriate antigens. Aerosol exposure to listeria was followed by markedly increased numbers of free lower respiratory tract cells. These cells manifested antigen-stimulated inhibition of migration (mean inhibition of m

Protocols for Assessing the Nuclease Activity of MIF.

Macrophage migration inhibitory factor (MIF) exhibits nuclease activity critical in DNA damage pathways linked to neurodegenerative diseases and cancer. Here, we describe dual approaches for measuring this nuclease activity, combining biochemical and cellular analysis. Recombinant MIF is subjected to enzymatic assays with L1 DNA substrates, analyzing cleavage patterns via agarose gel electrophoresis and time-dependent kinetics over 5-30 min. SH-SY5Y cells treated with the DNA-damaging agent MNNG, with or without the MIF nuclease inhibitor PAANIB-1, undergo pulse-field gel electrophoresis to as
