Melatonin, consciousness, and traumatic stress.

Descartes intuitively anticipated the so-called 'binding problem' of consciousness and thought that the pineal gland enables spatio-temporal … flected in associative sequentiality that is necessary for cognition, behavior and all processes of memory consolidation that must preserve all information in the temporal causal order and synchrony. In this context, recent findings suggest that melatonin could be a potential regulator in the processes that contribute to memory formation, long-term potentiation, and s …

Early changes in the properties of CA3 engram cells explored with a novel viral tool in mice.

Forming new memories after a one-time experience requires initial encoding then consolidation over time. During learning, multimodal information converges onto the hippocampus, activating sparse neuronal assemblies which are thought to form a memory representation through concerted activity and synaptic interconnectivity. In this work, we use a novel tool for fast fluorescent labeling of engram neurons (FLEN). FLEN is based on c-Fos activity-dependent transient expression of a destabilized fluorescent marker ZsGreen1 rapidly after one-trial learning. With FLEN, we explore the electrophysiologi

REM sleep prefrontal high-frequency oscillation chains mediate distinct cortical - hippocampal reactivation patterns compared to NREM sleep.

REM (rapid eye movement) and non-REM (NREM) sleep stages contribute to systems memory consolidation in hippocampal-cortical circuits. However, the physiological mechanisms underlying REM memory processes remain relatively unclear compared to NREM memory reactivation. Here we report, in rodents, the existence of prefrontal cortical (PFC) high-frequency oscillation (HFO) chains in REM sleep during the consolidation of recently acquired spatial memory. High-density tetrode recordings in hippocampal area CA1 and PFC reveal that REM cortical HFOs occur in characteristic chains that are phase-modula…

Cannabigerol does not affect contextual fear memory in mice but modulates nociception in a sex-dependent manner.

Conditioned fear is a response to contexts or cues previously paired with aversive stimuli. Substances from the herb Cannabis sativa (phytocannabinoids), such as cannabidiol (CBD), prevent fear responses and hold potential as new treatments for certain psychiatric disorders, such as post-traumatic stress disorder. The phytocannabinoid cannabigerol (CBG), similarly to CBD, lacks psychotomimetic effects and targets multiple receptors involved in fear-related pathways. This study tested the hypothesis that CBG inhibits memory acquisition, consolidation, and retrieval/expression in contextual fear

Effects of perinatal undernutrition on social transmission of food preference in adult male Wistar rats.

Nutrition plays a fundamental role in learning and memory, and early experimental undernutrition interferes with brain memory processes. Social transmission of food preference (STFP) is a natural olfactory paired-associate learning test that has not been used to assess the effects of early undernutrition on memory consolidation. Male Wistar rats were randomly divided into two groups: control and early undernourished. The underfed rats received different percentages of a balanced diet during gestation. After birth, pups were underfed by alternating every 12 h between two lactating dams, one wit…

An evening milk drink can affect word recall in Indonesian children with decreased sleep efficiency: A randomized controlled trial.

STUDY OBJECTIVES: Sleep is important for memory consolidation in children. This study intended to find out whether an evening milk-based drink could influence sleep efficiency and memory recall in a group of Indonesian children (5-6 years old) with sleep deprivation. METHODS: Children were randomly allocated to one of three interventions: Reference product, satiety-stimulating product, and a relaxing product. The intervention lasted for 6 weeks and children consumed two servings per day of each 200 ml, the serving in the morning being the same for all children. All measurements took place at …

A placebo-controlled study of sildenafil effects on cognition in schizophrenia.

BACKGROUND: Phosphodiesterase 5 (PDE5) inhibitors increase cyclic guanosine monophosphate (cGMP) concentrations in the intracellular pathway activated by N-methyl-D-aspartic acid receptors which is believed to mediate long-term potentiation and memory consolidation. The PDE5 inhibitor sildenafil has been shown to enhance memory in animal models. In addition, neuronal nitric oxide synthase, another component of the NMDA/nitric oxide/cGMP intracellular pathway, has been reported to be dysregulated in schizophrenia patients. MATERIALS AND METHODS: Seventeen adult schizophrenia outpatients treate…

Fast track to the neocortex: A memory engram in the posterior parietal cortex.

Models of systems memory consolidation postulate a fast-learning hippocampal store and a slowly developing, stable neocortical store. Accordingly, early neocortical contributions to memory are deemed to reflect a hippocampus-driven online reinstatement of encoding activity. In contrast, we found that learning rapidly engenders an enduring memory engram in the human posterior parietal cortex. We assessed microstructural plasticity via diffusion-weighted magnetic resonance imaging as well as functional brain activity in an object-location learning task. We detected neocortical plasticity as earl…

Modulation of cAMP and ras signaling pathways improves distinct behavioral deficits in a zebrafish model of neurofibromatosis type 1.

Neurofibromatosis type 1 (NF1) is a common autosomal-dominant disorder associated with attention deficits and learning disabilities. The primary known function of neurofibromin, encoded by the NF1 gene, is to downregulate Ras activity. We show that nf1-deficient zebrafish exhibit learning and memory deficits and that acute pharmacological inhibition of downstream targets of Ras (MAPK and PI3K) restores memory consolidation and recall but not learning. Conversely, acute pharmacological enhancement of cAMP signaling restores learning but not memory. Our data provide compelling evidence that neur…

Consolidation theory and retrograde amnesia in humans.

Recent research on the cognitive dysfunctions experienced by human anmesic patients indicates that very long term (multidecade) changes may occur in memory. Flat retrograde amnesia (RA), consisting of a uniform memory deficit for information from all preamnesia time periods, indicates a simple, monolithic retrieval problem, whereas graded RA, with greater memory deficits for information from recent as opposed to remote time periods, suggests the presence of a gradual long-term encoding, or consolidation, process. An evaluation of 247 outcomes from 61 articles provides strong evidence of graded

Vagus nerve stimulation as a tool for enhancing extinction in exposure-based therapies.

RATIONALE: Emotionally traumatic experiences can lead to maladaptive memories that are enduring and intrusive. The goal of exposure-based therapies is to extinguish conditioned fears through repeated, unreinforced exposures to reminders of traumatic events. The extinction of conditioned fear depends upon the consolidation of new memories made during exposure to reminders. An impairment in extinction recall, observed in certain patient populations, can interfere with progress in exposure-based therapies, and the drive to avoid thoughts and reminders of the trauma can undermine compliance and in

Accelerated forgetting in association with temporal lobe epilepsy and paraneoplastic encephalitis.

The association between epilepsy and amnesia is studied in patient J.T. who presented with a very unusual pattern of memory loss with retention of information for hours to days but rapid forgetting of information that exceeded this time frame. J.T.'s unusual memory profile was studied with several tests administered over week-long intervals of time. There was evidence that his retention decreased in conjunction with increased seizures. During a trial of paraldehyde, a decrease in seizure frequency was associated with enhanced memory. J.T.'s memory problem was unlike that described in prototypi

Activation of astrocytes in hippocampus decreases fear memory through adenosine A1 receptors.

Astrocytes respond to and regulate neuronal activity, yet their role in mammalian behavior remains incompletely understood. Especially unclear is whether, and if so how, astrocyte activity regulates contextual fear memory, the dysregulation of which leads to pathological fear-related disorders. We generated GFAP-ChR2-EYFP rats to allow the specific activation of astrocytes in vivo by optogenetics. We found that after memory acquisition within a temporal window, astrocyte activation disrupted memory consolidation and persistently decreased contextual but not cued fear memory accompanied by redu…

Effects of transcranial direct current stimulation over the posterior parietal cortex on episodic memory reconsolidation.

Consolidated memories may return to labile/unstable states after their reactivation, thus requiring a restabilization process that is known as reconsolidation. During this time-limited reconsolidation window, reactivated existing memories can be strengthened, weakened or updated with new information. Previous studies have shown that non-invasive stimulation of the lateral prefrontal cortex after memory reactivation strengthened existing verbal episodic memories through reconsolidation, an effect documented by enhanced delayed memory recall (24 h post-reactivation). However, it remains unknown

Posttraining Alpha Transcranial Alternating Current Stimulation Impairs Motor Consolidation in Elderly People.

The retention of a new sequential motor skill relies on repeated practice and subsequent consolidation in the absence of active skill practice. While the early phase of skill acquisition remains relatively unaffected in older adults, posttraining consolidation appears to be selectively impaired by advancing age. Motor learning is associated with posttraining changes of oscillatory alpha and beta neuronal activities in the motor cortex. However, whether or not these oscillatory dynamics relate to posttraining consolidation and how they relate to the age-specific impairment of motor consolidatio

Memory for public events in patients with unilateral temporal lobe epilepsy.

Memory for public events (PEs) was assessed as a marker of remote declarative memory in 36 patients with temporal lobe epilepsy (TLE) and compared with that of 19 patients with extra-TLE (ETLE), 17 patients with idiopathic generalized epilepsy (IGE), and 23 healthy volunteers. Verbal IQ, inventory-based evidence of depression, handedness, onset of illness, disease duration, and medication were obtained. Memory for PEs was reduced in all patient groups (TLE, P<0.0001; ETLE, P=0.009; IGE, P=0.008). The TLE group showed reduced memory for PEs compared with the other patients with epilepsy (P=0.00

How Do Amyloid Pathology and Aberrant Neuronal Activity Disrupt Plasticity and Memory in Alzheimer's Disease?

Alzheimer's disease (AD) is increasingly understood as a disorder of network-state and plasticity-capacity, in which amyloid-β and tau pathologies disrupt the activity-dependent mechanisms that build and stabilize memory engrams. Here, I review how amyloid-β-driven neuronal hyperactivity contributes to plasticity and memory deficits in AD. I also discuss how various cellular pathologies reinforce one another, leading to a cellular environment that is impermissive to plasticity. I relate these cellular and circuit-level disturbances to failures in memory encoding, consolidation, and recall, emp

Extending possible applications of an episodic-like memory task in rats.

Recently, an object recognition episodic-like memory task was proposed in rodents. However, the short retention delay of the task narrows its applications. This study verifies whether this task can be evoked after 24h. Additionally, the effect of a classical amnestic agent (scopolamine) on episodic-like memory consolidation was investigated. Rats showed increased exploration of old over recent objects and spent more time exploring displaced over stationary object. Both old over recent and displaced over stationary objects preferences were abolished by pos-training scopolamine administration (1…
