A toxicological and dermatological assessment of aryl alkyl alcohols when used as fragrance ingredients.

The aryl alkyl alcohol (AAA) fragrance ingredients are a diverse group of chemical structures with similar metabolic and toxicity profiles. The AAA fragrances demonstrate low acute and subchronic dermal and oral toxicity. No carcinogenicity in rats or mice was observed in 2-year chronic testing of benzyl alcohol or α-methylbenzyl alcohol; the latter did induce species and gender-specific renal adenomas in male rats at the high dose. There was no to little genotoxicity, mutagenicity, or clastogenicity in the mutagenic in vitro bacterial assays, and in vitro mammalian cell assays. All in vivo mi

Safety assessment of EPA-rich triglyceride oil produced from yeast: genotoxicity and 28-day oral toxicity in rats.

The 28-day repeat-dose oral and genetic toxicity of eicosapentaenoic acid triglyceride oil (EPA oil) produced from genetically modified Yarrowia lipolytica yeast were assessed. Groups of rats received 0 (olive oil), 940, 1880, or 2820 mg EPA oil/kg/day, or fish oil (sardine/anchovy source) by oral gavage. Lower total serum cholesterol was seen in all EPA and fish oil groups. Liver weights were increased in the medium and high-dose EPA (male only), and fish oil groups but were considered non-adverse physiologically adaptive responses. Increased thyroid follicular cell hypertrophy was observed i

Genomic instability in blood cells from murine model of mucopolysaccharidosis type I.

Mucopolysaccharidosis type I (MPS I) is caused by a deficiency of alfa-iduronidase (IDUA), which leads to intralysosomal accumulation of glycosaminoglycans. Some studies have revealed that oxidative stress plays an important role in MPS I. However, the mechanisms by which these alterations occur are still not fully understood. The aim of this study was to analyze genomic instability in blood cells from murine model of MPS I by single cell gel (comet) assay and micronucleus test. The results pointed out genetic damage in blood cells as depicted by the single cell gel (comet) assay results. By c

In Vivo micronucleus assay: Increased efficiency with an automated imaging system.

The in vivo erythrocyte micronucleus (MN) test is a standard assay for detecting chromosomal damage; however, conventional manual scoring is labor-intensive and subject to inter-scorer variability. Although automated image-based scoring offers a practical alternative, its application to bone marrow analysis requires effective enrichment of erythrocytes, to reduce interference from non-target nucleated cells. We have established and validated an automated bone marrow MN scoring workflow, using the Metafer slide-scanning system and associated software platform. We have compared a conventional ce

Genotoxic effects of gadobutrol and gadoversetamide active substances used in magnetic resonance imaging in human peripheral lymphocytes in vitro.

Gadobutrol and gadoversetamide are gadolinium-based contrast agents (GBCAs) widely used during magnetic resonance imaging examination. In this study, the genotoxicity of two GBCAs, gadobutrol and gadoversetamide, was investigated by using different endpoints: chromosome aberration (CAs), sister chromatid exchange (SCEs), and micronucleus (MNi). Human peripheral lymphocytes (PBLs) were treated with five concentrations (7 000, 14 000, 28 000, 56 000, and 112 000 μg/mL) of both agents. While a few concentrations of gadobutrol significantly increased abnormal cell frequency and CA/Cell, nearly all

Genotoxic effects of exposure to waterborne uranium, dietary methylmercury and hyperoxia in zebrafish assessed by the quantitative RAPD-PCR method.

Release of chemicals and fluctuation in oxygen content in the aquatic environment represent hazards for fish health. The present study aims at assessing the genotoxic impact of low concentration exposures to waterborne uranium (U), dietary methyl mercury (MeHg) and hyperoxia in zebrafish by using the RAPD-PCR quantitative method. A significant increase of the number hybridization sites was observed in fish exposed to 30μgU/L and 100μgU/L and hyperoxia. In fish exposed to MeHg (13.5μg Hg/g, dry weight) no change in the number of hybridization sites were found, however, the frequency of PCR prod

Molecular mechanisms of micronucleus, nucleoplasmic bridge and nuclear bud formation in mammalian and human cells.

Micronuclei (MN) and other nuclear anomalies such as nucleoplasmic bridges (NPBs) and nuclear buds (NBUDs) are biomarkers of genotoxic events and chromosomal instability. These genome damage events can be measured simultaneously in the cytokinesis-block micronucleus cytome (CBMNcyt) assay. The molecular mechanisms leading to these events have been investigated over the past two decades using molecular probes and genetically engineered cells. In this brief review, we summarise the wealth of knowledge currently available that best explains the formation of these important nuclear anomalies that

The lymphocyte cytokinesis-block micronucleus cytome assay and its application in radiation biodosimetry.

The cytokinesis-block micronucleus cytome (CBMN Cyt) assay is one of the best-validated methods for measuring chromosome damage in human lymphocytes. This paper describes the methodology, biology, and mechanisms underlying the application of this technique for biodosimetry following exposure to ionizing radiation. Apart from the measurement of micronuclei, it is also possible to measure other important biomarkers within the CBMN Cyt assay that are relevant to radiation biodosimetry. These include nucleoplasmic bridges, which are an important additional measure of radiation-induced damage that

Chronic hyperhomocysteinemia alters antioxidant defenses and increases DNA damage in brain and blood of rats: protective effect of folic acid.

We have previously demonstrated that acute hyperhomocysteinemia induces oxidative stress in rat brain. In the present study, we initially investigated the effect of chronic hyperhomocysteinemia on some parameters of oxidative damage, namely total radical-trapping antioxidant potential and activities of antioxidant enzymes (superoxide dismutase, catalase and glutathione peroxidase), as well as on DNA damage in parietal cortex and blood of rats. We also evaluated the effect of folic acid on biochemical alterations elicited by hyperhomocysteinemia. Wistar rats received daily subcutaneous injectio

Chromosomal damage in urothelial cells from Egyptians with chronic Schistosoma haematobium infections.

The most common cancer in Egypt is urinary-bladder carcinoma, which is strongly associated with infection with Schistosoma haematobium. The object of this study was to determine whether infection with this parasite resulted in an increase in chromosomal breakage in the urothelium. Urine samples were collected from 37 Egyptian farmers with S. haematobium ova in their urine and from 32 non-infected males attending the same village clinic. The urine was centrifuged and the urothelial cells present in the sediment were assayed for the presence of micronuclei, a quantitative indicator of chromosoma

Genotoxicity, subchronic toxicity, and allergenicity evaluation of IgG Binding Protein CBB produced from Komagataellaphaffii.

Gut health is progressively understood to be the multifactorial outcome of genetics, diet, lifestyle, and immune function and the influence of the gut microbiota. Healthy microbial homeostasis can be disrupted by environmental factors such as diet and xenobiotics, which can lead to gut dysbiosis. Dysbiosis is frequently associated with the expansion of pathobionts such as Clostridioides difficile. IgG Binding Protein CBB facilitates the safe passage of TcdB; a toxin secreted by C. difficile, through the gastrointestinal tract helping to maintain gut homeostasis. To substantiate the safety of I
