Macrophage mitochondrial superoxides as a target for atherosclerotic disease treatment.

Cardiovascular diseases (CVD) are the leading cause of death worldwide. Although many effective therapies exist, a substantial portion of patients remain unprotected by current measures. Recent advances in the understanding of the underpinning cause of CVD, atherosclerosis, have demonstrated the important causative role of inflammation in disease progression. Monocytes are important protagonists of atherosclerosis, and they have been shown to have elevated reliance on mitochondrial metabolism producing elevated levels of superoxides as a noxious byproduct. There exists a key link between mitoc

Treatment of Heart Failure with Preserved Ejection Fraction.

Heart failure with preserved ejection fraction (HFpEF) is a growing epidemiologic problem affecting more than half of the patients with heart failure (HF). HFpEF has a significant morbidity and mortality and so far no treatment has been clearly demonstrated to improve the outcomes in HFpEF, in contrast to the efficacy of treatment in heart failure with reduced ejection fraction (HFrEF).The failure of proven beneficial drugs in HFrEF to influence the outcome of patients with HFpEF could be related to the heterogeneity of the disease, its various phenotypes and multifactorial pathophysiology, in

Emergent properties of neural repair: elemental biology to therapeutic concepts.

Stroke is the leading cause of adult disability. The past decade has seen advances in basic science research of neural repair in stroke. The brain forms new connections after stroke, which have a causal role in recovery of function. Brain progenitors, including neuronal and glial progenitors, respond to stroke and initiate a partial formation of new neurons and glial cells. The molecular systems that underlie axonal sprouting, neurogenesis, and gliogenesis after stroke have recently been identified. Importantly, tractable drug targets exist within these molecular systems that might stimulate t

Aspirin and lipid mediators in the cardiovascular system.

Aspirin is an unique compound because it bears two active moieties within one and the same molecule: a reactive acetyl group and the salicylate metabolite. Salicylate has some effects similar to aspirin, however only at higher concentrations, usually in the millimolar range, which are not obtained at conventional antiplatelet aspirin doses of 100-300 mg/day. Pharmacological actions of aspirin in the cardiovascular system at these doses are largely if not entirely due to target structure acetylation. Several classes of lipid mediators become affected: Best known is the cyclooxygenase-1 (COX-1)

ACETYLCHOLINESTERASE INHIBITION ACTIVITY OF SOME QUINOLINYL SUBSTITUTED TRIAZOLOTHIADIAZOLE DERIVATIVES.

A series of aralkanoic acids was converted into aralkanoic acid hydrazides through their esters formation. The aralkanoic acid hydrazides upon treatment with carbon disulfide and methanolic potassium hydroxide yielded potassium dithiocarbazinate salts, which on refluxing with aqueous hydrazine hydrate yielded 5-aralkyl-4-amino-3-mercapto-1,2,4-triazoles. The target compounds, 3-aralkyl-6-(substitutedquinolinyl)[1,2,4]triazolo[3,4-b][1,3,4]thiadiazoles, were synthesized by condensing various quinolinyl substituted carboxylic acids with 5-aralkyl-4-amino-3-mercapto-1,2,4-triazoles in phosphorus

Autophagy of mitochondria: a promising therapeutic target for neurodegenerative disease.

The autophagic process is the only known mechanism for mitochondrial turnover and it has been speculated that dysfunction of autophagy may result in mitochondrial error and cellular stress. Emerging investigations have provided new understanding of how autophagy of mitochondria (also known as mitophagy) is associated with cellular oxidative stress and its impact on neurodegeneration. This impaired autophagic function may be considered as a possible mechanism in the pathogenesis of several neurodegenerative disorders including Parkinson's disease, Alzheimer's disease, multiple sclerosis, amyotr

[Interleukin-17 as a therapeutic target in psoriasis].

Psoriasis is a chronic, immune-mediated inflammatory disease that affects up to 1-3% of the general population. An advanced understanding of the immune-pathogenesis of psoriasis has led to the development of new drugs that refine existing treatments or target novel molecular and immunologic pathways. IL-17 and Th17 cells play an important role in the pathogenesis of several autoimmune and immune-mediated disorders, including psoriasis. IL-17A, a pro-inflammatory cytokine, is produced by Th17 cells along with other effector cytokines, such as IL-17F an IL-22, but it is also expressed by other c

Thymic neoplasms: an update on the use of chemotherapy and new targeted therapies. A literature review.

Thymic malignancies represent a wide range of clinical, histological and molecular entities, with probably considerable heterogeneity even among tumors of the same histotype. Systemic chemotherapy with cisplatin-based regimens continues to represent the standard of care in metastatic or inoperable refractory/recurrent diseases and ADOC regimen (including cisplatin, doxorubicin, vincristine and cyclophosphamide) demonstrated the longer overall response rate and median survival in the first line setting, although no randomized trial is available; and there is still a lack of standard treatment a

Can MicroRNAs serve as reliable biomarkers and novel therapeutic targets in age-related macular degeneration? A systematic review and meta-analysis.

INTRODUCTION: Age-related macular degeneration (AMD) is a leading cause of irreversible vision loss, affecting 196 million people globally with projections reaching 288 million by 2040. Its pathogenesis involves oxidative stress, chronic inflammation, angiogenesis, and retinal pigment epithelium (RPE) dysfunction. MicroRNAs (miRNAs)—highly stable, non-coding post-transcriptional regulators—have emerged as key modulators of AMD-related pathways, including nuclear factor kappa B mediated inflammation (miR-146a, miR-155), angiogenesis (miR-23a, miR-27a), and RPE homeostasis (miR-204, miR-211). Arginine vasopressin: A promising therapeutic target for metabolic syndrome.

Globally, the prevalence of metabolic syndrome (MetS) among adults is around 20%-25%. MetS is associated with an increased risk of cardiovascular morbidity and mortality. The pathogenesis of MetS is multifactorial, with no single factor being an obvious target for treatment. Rather, complex genetic and environmental factors result in visceral obesity, insulin resistance, hyperglycemia, dyslipidemia, and hypertension. A growing body of epidemiological evidence is indicative of an association between arginine vasopressin (AVP) and MetS and its components. Indeed, the clinical marker of AVP, cope

Activin A-Targeted Therapy in Cancer: An Updated Review on Challenges and Opportunities in Clinical Translation.

Activin A (ActA) is a cytokine from the TGF-β superfamily that mediates a vast number of physiological mechanisms, mainly through the SMAD signaling pathway. Growing evidence indicates that ActA overexpression is also correlated with poor prognosis in cancer patients and several tumor characteristics, including cancer proliferation, metastasis, immunosuppression, drug resistance, cachexia, and cancer-associated fibroblast activation. As such, ActA-targeted therapy has been viewed as a potential adjuvant therapy alongside other anti-cancer modalities that may result in more efficient anti-cance

Pathophysiologic mechanisms and molecular targets in preeclampsia.

Preeclampsia is a pregnancy-specific multisystem disorder. With a global incidence of 2%-8%, preeclampsia is a major contributor to maternal and neonatal morbidity and mortality. The pathogenesis of preeclampsia is commonly described by a two-stage model. The first stage involves defective placentation in early pregnancy, marked by insufficient extravillous trophoblast invasion, impaired spiral artery remodeling, dysregulated sphingolipid metabolism, and an imbalance in immune tolerance; these anomalies lead to placental ischemia and hypoxia. The second stage consists of systemic maternal resp

Serine Hydroxymethyltransferase (SHMT) in biology and disease: molecular mechanisms and therapeutic targeting.

BACKGROUND: Serine Hydroxymethyltransferase 1 and 2 (SHMT1/2), as key enzymes in folate metabolism, regulate diverse physiological processes through one-carbon metabolism, including nucleotide biosynthesis, methylation reactions, and redox homeostasis. As a form of metabolic reprogramming, dysregulated SHMT-mediated one-carbon metabolism has been increasingly reported in various human cancers and liver metabolic diseases. Therefore, a concise review of its regulatory roles and disease-associated functions is necessary to clarify its biological and translational significance. MAIN BODY: This re

Targeting the untargeted: Molecular insights and emerging therapeutic strategies for primary aldosteronism.

Primary aldosteronism (PA), the most common form of endocrine hypertension, is evolving from a simple hormonal disorder to a complex molecular syndrome. While somatic mutations in genes like KCNJ5 and CACNA1D illuminate part of the pathogenesis, the integrated role of epigenetic dysregulation remains a critical frontier. This review argues that the traditional "one-size-fits-all" approach, reliant on conventional mineralocorticoid receptor antagonists (MCRAs), is inadequate due to its off-target effects and failure to address residual cardiovascular risk. We comprehensively synthesize emerging

CD47 as a potent target in cancer immunotherapy: A review.

Cancer is the second-highest cause of death worldwide. Accordingly, finding new cancer treatments is of great interest to researchers. The current platforms to fight cancer such as chemotherapy, radiotherapy, and surgery are limited in efficacy, especially in the metastatic setting. In this war against cancer, the immune system is a powerful ally, but tumor cells often outsmart it through alternative pathways. Cluster of differentiation 47 (CD47), a protein that normally prevents healthy cells from being attacked by immune cells, is often overexpressed on cancer cells. This makes CD47 a prime

Sodium's role and therapeutic targeting in cancer.

Cancer cells exhibit unique metabolic reprogramming characterized by a significant increase in intracellular sodium ion levels. Sodium influences cancer cell metabolism, immune function, and drug resistance, and can trigger a unique cell death pathway when overloaded. Sodium-related transporters regulate cellular sodium ion levels and cancer progression. Targeting these transporters with specific inhibitors might therefore be an effective way to treat cancer. However, the precise relationship between sodium and cancer cell behavior is insufficiently studied and our understanding of the relevan

The role of Piezo1 in cardiovascular diseases: from molecular mechanisms to targeted therapeutic potential.

Piezo1 is a mechanosensitive cation channel protein that responds to mechanical stimuli such as fluid shear stress, stretch force, and extracellular matrix stiffness. It converts mechanical signals into biochemical signals, thereby regulating cellular biological behaviours including inflammation, oxidative stress, mitochondrial homeostasis, and angiogenesis. Growing evidence indicates that multiple cellular biological behaviours modulated by Piezo1 play a critical role in the physiological and pathological processes of cardiovascular diseases (CVDs). This review focuses on the regulatory facto
