Sonodynamic therapy (SDT) is a highly promising approach for cancer therapy, but its efficacy is severely hampered by the low specificity of sonosensitizers and the unfavorable characteristics of the tumor microenvironment (TME), such as hypoxia and glutathione (GSH) overexpression. To solve these problems, in this work, we encapsulated IR780 and MnO2 in PLGA and linked Angiopep-2 (Ang) to synthesize a multifunctional nanozyme (Ang-IR780-MnO2-PLGA, AIMP) to enhance SDT. With Ang functionalization to facilitate blood-brain barrier (BBB) penetration and glioma targeting, and through the function

The alcohol dehydrogenase (ADH) activity of human short-chain l-3-hydroxyacyl-CoA dehydrogenase (SCHAD) has been characterized kinetically. The k(cat) of the purified enzyme was estimated to be 2. 2 min(-1), with apparent K(m) values of 280 mM and 22microM for 2-propanol and NAD(+), respectively. The k(cat) of the ADH activity was three orders of magnitude less than the l-3-hydroxyacyl-CoA dehydrogenase activity but was comparable with that of the enzyme's hydroxysteroid dehydrogenase (HSD) activity for oxidizing 17beta-oestradiol [He, Merz, Mehta, Schulz and Yang (1999) J. For over five decades, sodium alginate (SA) has been widely employed in pharmaceutical formulations due to its environmentally friendly, biodegradable, and biocompatible nature. Primarily sourced from brown seaweed, this natural polymer plays a crucial role in enhancing drug delivery control. It serves as a versatile excipient in numerous dosage forms such as capsules, tablets, liposomes, and microspheres. SA ionotropic gelation with calcium ions allows drug and enzyme encapsulation for controlled release. Structurally, SA consists of α-L-guluronic acid and β-D-mannuronic acid monomers linked

Mosquito-borne flaviviruses are important human pathogenic viruses and can cause an array of symptoms ranging from asymptomatic to mild illness but can progress to severe disease conditions. Prominent members of the family Flaviviridae include Dengue virus, Zika virus, and West Nile virus, whose spread has increased globally due to the expansion of their respective mosquito vectors. Despite their importance as a public health concern, there are no FDA-approved antiviral drugs targeting these viruses in humans. One of the viral proteins essential for flavivirus replication is the NS3 protein, a

Japanese encephalitis caused by Japanese encephalitis virus (JEV) infection leads to the central nervous system disorder in human and swine. Viruses utilize the host protein synthesis mechanisms to efficiently translate their RNAs. Herein, we demonstrated that the host transcription factor SOX10 downregulated an RNA-binding protein heterogeneous nuclear ribonucleoprotein H (HNRNPH1) during JEV infection. We further showed that JEV replication was promoted when HNRNPH1 inhibited RIG-I/MDA5 signal pathway to decrease interferon (IFN) expression. Remarkably, the multifunctional enzyme NS3 of JEV

Lentil fermentation has a promising potential as a strategy for development of multifunctional ingredients targeting metabolic syndrome (MetS). Response surface methodology was applied to optimize lentil fermentation and study its effects on generation of peptides, soluble phenolics and bioactivities. Fermentation using Lactobacillus plantarum and Savinase® 16 L was carried out at different pH (6.5-8.5) and times (5.5-30 h). Analysis of variance was performed to evaluate linear, quadratic and interaction effects between fermentation parameters. pH positively affected peptides, soluble phenolic

Accumulating evidence has made it clear that not only does the renin-angiotensin-aldosterone system (RAAS) exist in the circulation where it is driven by renal renin, but it is also active in many tissues-and likely within cells as well. These systems might not be completely independent of each other, but rather interact. These local RAASs affect tissue and cellular angiotensin II concentrations and appear to be associated with clinically relevant physiologic and pathophysiologic actions in the cardiovascular system and elsewhere. Evidence in support of this possibility is reviewed here. In ad

Thrombin is a multifunctional protein: in addition to its role in coagulation, thrombin has important biological effects on platelets, endothelial and smooth muscle cells, leukocytes, the heart and neurones. A detailed understanding of the structure of thrombin, of related serine proteases and of enzyme-inhibitor complexes has aided in the discovery of potent and selective new inhibitor molecules. Some of these novel thrombin inhibitors are active when administered orally and have shown remarkable efficacy as antithrombotic agents in animal models, offering a greater therapeutic potential than

Natural enzymes orchestrate complex biological processes through highly coordinated and multifunctional catalysis, yet their practical translation is limited by poor stability and environmental sensitivity. Here, we report a peptide-engineered nanozyme hydrogel that enables programmable, microenvironment-responsive catalytic switching in response to the wound microenvironment for active regulation of wound healing. Inspired by the catalytic motifs of natural hydrolases and oxidases, an amphiphilic peptide (C16-GAVFFAHHGGEE) was rationally designed to coordinate Cu2+ and self-assemble into a β-

OBJECTIVE: The mitochondrial serine catabolic pathway (MSCP) supports tumor proliferation and metastasis, yet no therapies target the MSCP. Because cancer cells rely on the MSCP when respiration is suppressed, we hypothesized that reactivating respiration would inhibit the MSCP.
METHODS: Mitochondrial respiration was activated in triple negative breast cancer (TNBC) cells using uncouplers [niclosamide ethanolamine (NEN) and BAM15]. Metabolic activity through the MSCP was assessed using U-13C-serine tracing and expression of key MSCP enzymes (SHMT2, MTHFD2, and MTHFD1L) were evaluated at the m

The primary role of break-induced replication (BIR) is to repair single-ended double-strand breaks (seDSBs) generated at broken replication forks and eroding telomeres. In this study, we demonstrated that when senataxin (SETX), an RNA/DNA helicase, is defective, hyper-recombination using the BIR mechanism is induced at R-loops/hybrid-accumulated double-ended DSBs (deDSBs), uncovering a role for BIR in the repair of R-loops/hybrids-associated deDSBs. Intriguingly, the loss of SETX not only triggers non-canonical hyper-end resection requiring RAD52 and XPF but also stalls Polα-primase-initiated

Variants in SETX have been implicated in recessively and dominantly inherited disorders, ataxia with oculomotor apraxia type 2 (AOA2 OMIM# 606002) and amyotrophic lateral sclerosis (ALS4, OMIM# 602433) respectively, in humans. We report two novel bi-allelic pathogenic variants in SETX in patients suffering from ataxia with oculomotor apraxia type 2, extending the allelic spectrum of the gene variants. We also discuss the pathogenicity of SETX variants in relation to the evolutionary conservation status of the affected amino acids. Our analyses suggest that variants of some amino acids which ar

AOA are autosomal recessive ataxias with a common feature of oculomotor apraxia (OA) - inability to coordinate eye movements. The group includes AOA1 (APTX gene), relatively common AOA2 (SETX gene) and AOA3 (PIK 3R5 gene) described in 2012 in a Saudi family. OA is typical also for Louis-Bar ataxia-telangiectasia and its variants. А first Russian AOA2 case confirmed by DNA test is presented. The disease in a 25-year-old male started in 18 years, in 23 years he lost independent walking due to incoordination and weakness. OA produced few symptoms and was not recorded previously. Dicer has long been viewed as the RNAse III enzyme that generates small regulatory RNAs to drive RNA interference (RNAi) machinery and posttranscriptional gene silencing. However, a growing body of evidence reveals that Dicer's functional landscape extends far beyond its canonical role in the cytoplasmic RNAi pathway. In this review, we delineate the classical, RNAse III-dependent small RNA processing activities from an expanding set of noncanonical functions in which Dicer operates independently of RNAi outputs. We highlight emerging principles that underlie these functions, including structu

Although aptamers held promise for precision cancer therapy, their physiological instability and the lack of multifunctional delivery platforms that enabled synergistic therapeutic effects remained key challenges. In this study, a nanocarrier based on protamine-modified CuAu bimetallic nanozyme was constructed to co-load AT11 aptamer and Doxorubicin (Dox), named BPAD. In this system, the AT11 aptamer was adsorbed onto the surface of the positively charged nanocarrier via electrostatic interactions, which reversed the surface charge and formed a dense targeting shell, thereby significantly enha

Hypertension, a major risk factor for cardiovascular disease, is largely regulated by the renin-angiotensin system (RAS). This study evaluates the multitarget potential of three amaranth-derived peptides -SFNLPILR, FNLPILR, and AFEDGFEWVSFK- previously identified as renin inhibitors. We assessed their ability to inhibit angiotensin-converting enzyme (ACE), modulate ACE2 activity, and their bioavailability. In vitro assays demonstrated that SFNLPILR and FNLPILR are potent ACE inhibitors (IC₅₀ = 0.075 and 0.055 mM, respectively), with selective or minimal modulation of ACE2 enzymatic activity. B

Novel peptides generated from solid-state fermented hempseed cake were characterized by peptidomics and validated for bioactivity. Fermentation with Aspergillus niger, Bacillus subtilis, and Lactobacillus rhamnosus markedly increased peptide numbers compared with the unfermented control and promoted the release of short peptides mainly from Cupin type-1 domain proteins, with leucine frequently occurring at cleavage termini. In silico prediction and molecular docking identified antioxidant, angiotensin-converting enzyme (ACE), and dipeptidyl peptidase-IV (DPP-IV) inhibitory candidates. Synthesi

β-1,4-Endoglucanase (ENG) is a canonical effector protein secreted by the esophageal glands of plant-parasitic nematodes. Homology analysis identified a β-1,4-endoglucanase homolog in Hirschmanniella mucronata, designated as HmENG1. In situ hybridization demonstrated the specific localization of Hmeng1 transcripts within the esophageal glands of H. mucronata. Recombinant HmENG1 exhibited cellulase activity in vitro. In planta RNAi experiments confirmed that silencing Hmeng1 significantly reduced the infection capability of H. mucronata. In addition to its potential role in degrading plant cell
