Capture and Detection of Plasmodium vivax Lactate Dehydrogenase in a Bead-Based Multiplex Immunoassay.

Laboratory detection of malaria antigens has proved valuable for research and epidemiological purposes. We recently developed a bead-based multiplex antigen assay for pan-Plasmodium and Plasmodium falciparum targets. Here, we report integration of a Plasmodium vivax-specific target to this multiplex panel: P. vivax lactate dehydrogenase (PvLDH). Within the multiplex panel, assay signal for purified PvLDH antigen titrated into the single-digit picogram range. Against a panel of polymerase chain reaction (PCR)-confirmed samples from acute P. vivax infections (n = 36), sensitivity was 91.7% in us

Establishment and application of multiplex real-time qPCR detection for Tembusu virus and goose astrovirus genotypes 1 and 2.

The concurrent infection and epidemic of goose astrovirus type 1 (GoAstV-1), goose astrovirus type 2 (GoAstV-2), and Tembusu virus (TMUV) in geese have resulted in substantial economic losses to the goose industry. Therefore, establishing a rapid and accurate diagnostic method for these three viruses is imperative. In the present study, we designed specific primers targeting the Cap gene of GoAstV and the E gene of TMUV and developed a SYBR Green I-based multiplex real-time qPCR method for the simultaneous detection of GoAstV-1, GoAstV-2, and TMUV. The results revealed that the R2 values for t

A multiplex real-time PCR assay for detection of equid herpesvirus 1 and 4.

Equid herpesvirus (EHV) 1 and -4 are common viral pathogens of horses that can cause upper respiratory disease, neurological disease, abortion, and death. As characteristic alphaherpesviruses, both EHV-1 and EHV-4 can establish latency, resulting in a lifelong carrier state in infected animals. Here we describe the development and validation of a rapid and sensitive multiplex real-time PCR assay (EHV1-4MP) that simultaneously detects EHV-1 and EHV-4 and includes an endogenous internal control - melanocortin 1 receptor (MC1R) - targeting the equid genome. The EHV1-4MP assay analytical sensitivi

Application of Universal LNA Probe-Mediated Multiplex Real-Time PCR for the Detection of Antibiotic Resistance Genes.

The rising resistance of pathogens has increasingly complicated antibiotic treatments, leading to prolonged therapy durations and the potential for inadequate disease management. Current clinical methods (e.g., culture-based AST, conventional PCR, Sanger sequencing) for assessing microbial resistance are hampered by lengthy processing times, limited sensitivity, or high costs. In this study, we developed a multiplex quantitative polymerase chain reaction (qPCR) method using custom-designed universal locked nucleic acid (LNA) probes. This approach eliminates the need for target-specific fluores

Application of a Multiplex Quantitative PCR to Assess Prevalence and Intensity Of Intestinal Parasite Infections in a Controlled Clinical Trial.

BACKGROUND: Accurate quantitative assessment of infection with soil transmitted helminths and protozoa is key to the interpretation of epidemiologic studies of these parasites, as well as for monitoring large scale treatment efficacy and effectiveness studies. As morbidity and transmission of helminth infections are directly related to both the prevalence and intensity of infection, there is particular need for improved techniques for assessment of infection intensity for both purposes. The current study aimed to evaluate two multiplex PCR assays to determine prevalence and intensity of intest

Multiplex hydrolysis probe real-time PCR for simultaneous detection of hepatitis A virus and hepatitis E virus.

Detection of hepatitis viral infections has traditionally relied on the circulating antibody test using the enzyme-linked immunosorbent assay. However, multiplex real-time PCR has been increasingly used for a variety of viral nucleic acid detections and has proven to be superior to traditional methods. Hepatitis A virus (HAV) and hepatitis E virus (HEV) are the major causes of acute hepatitis worldwide; both HAV and HEV infection are a main public health problem. In the present study, a one-step multiplex reverse transcriptase quantitative polymerase chain reaction assay using hydrolysis probe

Development of novel DNA marker for species discrimination of Fasciola flukes based on the fatty acid binding protein type I gene.

BACKGROUND: Multiplex polymerase chain reaction (PCR) and PCR-restriction fragment length polymorphism (RFLP) for nuclear phosphoenolpyruvate carboxykinase (pepck) and polymerase delta (pold), respectively, have been used to differentiate Fasciola hepatica, F. gigantica, and hybrid Fasciola flukes. However, discrimination errors have been reported in both methods. This study aimed to develop a multiplex PCR based on a novel nuclear marker, the fatty acid binding protein type I (FABP) type I gene. METHODS: Nucleotide sequence variations of FABP type I were analyzed using DNA samples of F. hepa…

Missed Plasmodium falciparum and Plasmodium vivax Mixed Infections in Ethiopia Threaten Malaria Elimination.

Plasmodium falciparum and Plasmodium vivax are co-endemic in Ethiopia. This study investigated whether mixed infections were missed by microscopy from a 2017 therapeutic efficacy study at two health facilities in Ethiopia. All patients (N = 304) were initially classified as having single-species P. falciparum (n = 148 samples) or P. vivax infections (n = 156). Dried blood spots were tested for Plasmodium antigens by bead-based multiplex assay for pan-Plasmodium aldolase, pan-Plasmodium lactate dehydrogenase, P. vivax lactate dehydrogenase, and histidine-rich protein 2. Of 304 blood samples, 13

Effects of a diagnostic stewardship intervention to de-implement widespread use of a rapid respiratory multiplex PCR test.

UNLABELLED: Rapid respiratory diagnostic tests are widely utilized but often do not change clinical management. We evaluated the effects of an intervention to de-implement widespread use of a multiplex respiratory pathogen PCR panel (RPP) in favor of a selective use strategy. This was a retrospective, quasi-experimental study conducted in a safety-net healthcare system. We compared RPP utilization during 2 years of unrestricted, widespread use (24 July 2022-23 July 2024) and 1 year after implementation of a selective use strategy (24 July 2024-23 July 2025). The primary outcome was change in w

Development of a Serotyping Scheme for Streptococcus pasteurianus: An Underreported Zoonotic Pathogen.

Streptococcus pasteurianus is an opportunistic and underreported zoonotic pathogen capable of causing severe infections in both humans and animals, and it is also an etiological agent of swine streptococcosis. Despite its clinical and veterinary relevance, no systematic serotyping scheme has previously been established for this species. In this study, six representative strains harboring distinct cps gene clusters were selected for antiserum production, and agglutination assays identified five serotypes, designated serotypes 1a, 1b, 2, 3, and 4. Based on serotype-specific variation within the
