Current Strategies for the Regeneration of Skeletal Muscle Tissue.

Traumatic injuries, tumor resections, and degenerative diseases can damage skeletal muscle and lead to functional impairment and severe disability. Skeletal muscle regeneration is a complex process that depends on various cell types, signaling molecules, architectural cues, and physicochemical properties to be successful. To promote muscle repair and regeneration, various strategies for skeletal muscle tissue engineering have been developed in the last decades. However, there is still a high demand for the development of new methods and materials that promote skeletal muscle repair and functio

The nuclear lamina and inherited disease.

Inherited disorders of the nuclear lamina present some of the most intriguing puzzles in cell biology. Mutations in lamin A and lamin C - nuclear intermediate filament proteins that are expressed in nearly all somatic cells - cause tissue-specific diseases that affect striated muscle, adipose tissue and peripheral nerve or skeletal development. Recent studies provide clues about how different mutations in these proteins cause either muscle disease or partial lipodystrophy. Although the precise pathogenic mechanisms are currently unknown, the involvement of lamins in several different disorders

Embryonic stem cell lines derived from human blastocysts.

Human blastocyst-derived, pluripotent cell lines are described that have normal karyotypes, express high levels of telomerase activity, and express cell surface markers that characterize primate embryonic stem cells but do not characterize other early lineages. After undifferentiated proliferation in vitro for 4 to 5 months, these cells still maintained the developmental potential to form trophoblast and derivatives of all three embryonic germ layers, including gut epithelium (endoderm); cartilage, bone, smooth muscle, and striated muscle (mesoderm); and neural epithelium, embryonic ganglia, a

Vibration-induced changes in vascular tone.

Longitudinal mechanical vibrations in sonic range (10-100 Hz) and of moderate amplitude (50-500) considerably reduce the contractile force of smooth muscle as well as striated muscle. Experiments elucidating the mechanism behind this effect are described. The inhibition of the contractile force is probably due to a direct interference with the action of the contractile proteins. Vibrations are supposed to increase the rate with which the interfilamentary crosslinks are broken. Thereby the number of established crosslinks is reduced and thus the force. Finally a hypothesis concerning the possib

Gross and microscopic anatomy of the major salivary glands of the ferret.

The gross and microscopic anatomy of the major salivary glands of the ferret was studied. There were 5 pairs of major salivary glands, parotid, submandibular, sublingua, molar and zygomatic. They were of compound tubuloacinar type and consisted of numerous lobes and lobules. The parotid gland was grossly similar to the parotids in other mammals but histologically it was seromucous as in the carnivores dog and cat. The parotid duct opened into the vestibule of the mouth opposite the upper third premolar tooth. The submandibular gland was single on each side, oval in shape and cervical in positi

Case report: selective occipital lobe hydrocephalus and agenesis of the left lobe of the liver in congenital myotonic dystrophy.

Myotonic dystrophy (Steinert's disease) is an uncommon familial autosomal dominant disease, characterized by progressive muscular atrophy and myotonia. Mental retardation, frontal baldness, cortical lenticular opacities and testicular atrophy are other prominent features of a disease that involves predominantly striated muscle. There is little information regarding this disease in the radiological literature. The present report introduces two additional findings involving the skull and the liver.

Suspected nutritional myopathy in two captive Bennett's wallabies (Macropus rufogriseus).

Between January and April, 1992, two captive Bennett's wallabies (Macropus rufogriseus) from a wildlife park in Iwamizawa City, Hokkaido, Japan, were diagnosed as having nutritional myopathy. One wallaby, four-year-old adult male, had mild fresh lesions, confined to the skeletal muscles, with poor cell reaction. The other wallaby, 8-month-old female joey, had marked and extensive lesions with cellular reactions and regenerative muscle fibers in the locomotor muscles. Involvement of other striated muscles such as those of the tongue, diaphragm and heart was mild with minimal cellular reaction.

Desmin related disease: a matter of cell survival failure.

Maintenance of the highly organized striated muscle tissue requires a cell-wide dynamic network that through interactions with all vital cell structures, provides an effective mechanochemical integrator of morphology and function, absolutely necessary for intra-cellular and intercellular coordination of all muscle functions. A good candidate for such a system is the desmin intermediate filament cytoskeletal network. Human desmin mutations and post-translational modifications cause disturbance of this network, thus leading to loss of function of both desmin and its binding partners, as well as

Demonstration of early lesions of muscle by infrared fluorescence microscopy.

Previous investigations demonstrated decrease or loss of IR fluorescence in certain lesions of smooth and striated muscle. These investigations were extended to diseases of skeletal muscle. Sections were stained and photographed as described (Puchtler et al. 1980). Various diseases caused significant changes of IR fluorescence. Often, IR fluorescence demonstrated lesions in muscle fibers which appeared unremarkable in direct light and/or conventional darkfield microscopy. The patterns of IR fluorescence varied widely, even within groups of diseases, e.g. muscular dystrophies. However, owing to

Update on cellular expression and function of ANO1 channels in urethral smooth muscle.

Urinary continence is maintained by the cooperation of smooth and striated muscles where urethral smooth muscle cell (USMC) contractions contribute to resting urethral tone during bladder filling, whereas striated muscle contracts in response to sudden abdominal pressure rises. Anoctamin-1 (ANO1), a calcium-activated chloride channel (CaCC), has been identified as a potential regulator of USMC excitability. This review evaluates recent advances in understanding the cellular expression profile and functional contributions of ANO1 to USMC activity, for which there are significant species-specifi

The absence of desmin leads to cardiomyocyte hypertrophy and cardiac dilation with compromised systolic function.

Desmin is the muscle-specific member of the intermediate filament family of cytoskeletal proteins, expressed both in striated and smooth muscle tissues. In mature striated muscle fibers, the desmin filament lattice surrounds the Z-discs, interconnects them to each other and links the entire contractile apparatus to the sarcolemmal cytoskeleton, cytoplasmic organelles and the nucleus. There have been increasing reports of human cardiomyopathies associated with abnormal accumulation and aggregation of desmin filaments. Recently identified desmin mutations in humans suffering from skeletal muscle

[Morphological investigation of external sphincter muscle of the male posterior urethra].

For investigation of function of the male posterior urethra, morphological study was performed in terms of the muscle architecture including sphincter muscle of the posterior urethra. The muscle localization was demonstrated by microdissection in eight male cadavers. Histological examination was also employed with HE staining and immunohistochemical method to detect the nature of these muscles. At the dorsal side, the muscle fibers of the deep trigone were located from the inter-ureteral ridge beyond the bladder neck to the vermontanum surrounding the posterior urethra. They were regarded as t

Diagnosis and classification of granulomatous myositis.

The term granulomatous myositis is applied to a myopathic syndrome associated with non-specific epithelioid granulomas in striated muscle. This rare entity is most frequently related to sarcoidosis, but other uncommon causes have been reported, including an idiopathic form only after systemic disorders known to cause similar myopathological abnormalities have been excluded. Symmetrical proximal or distal muscle weakness is the rule in the clinical presentation, sometimes associated with dysphagia. Although the clinical profile together with electromyography (EMG) studies may be useful, definit

The function of the urethral blood flow in maintaining urinary continence. A preliminary report.

Urethral pressure is made up of three components, equal in size: the striated muscle component, the smooth muscle component, and the vascular component. Anti-embolism stockings have been helpful in bringing about significant improvement in blood flow through the lower extremities. A pilot study was undertaken with 15 women to find out whether or not the same approach led to increase in urethral pressure. Simultaneous urethrocystometry was applied to women with and without stockings. Significant rise in urethral pressure was recorded from women with urinary stress incontinence who used stocking

[The brown bowel syndrome. A case report].

We present a patient with celiac sprue in whom intestinal lymphoma was suspected. At laparotomy, a brown discolouration of small bowel loops was observed, and a transmural biopsy confirmed a brown bowel syndrome, an entity that is found in long-standing malabsorption conditions. We believe that the intestinal disorder caused by deposition of lipofuscin in the bowel non-striated muscle may cause radiologic changes similar to those seen in intestinal lymphoma.

In vitro myogenic differentiation of human bone marrow-derived mesenchymal stem cells as a potential treatment for urethral sphincter muscle repair.

To explore a new treatment strategy for urinary incontinence, human bone marrow mesenchymal stem cells (MSC) of the first in vitro passage were exposed to 5-azacytidine (AZA) to induce myogenic differentiation, and cultured for a total of six passages. Expression of stem cell surface antigens and intracellular alpha-actin was examined by flow cytometry at the end of each passage and compared to that of native MSC (not exposed to AZA) cultured in parallel. To analyze differentiation into striated muscle, expression of the transcription factor MyoD1 and myosin heavy chain (MyHC) was examined by
