Gestural recovery and the role of forward and reversed syllabic repetitions as stuttering inhibitors in adults.

Instead of being the core stuttering 'problem', syllabic repetitions may be a biological mechanism, or 'solution', to the central involuntary stuttering block. Simply put, stuttering is an endogenous transitory state of 'shadowed speech', a choral speech derivative that allows for a neural release of the central block. To investigate this possibility, 14 adults who stutter read while listening to forward fluent speech, reversed fluent speech, forward stuttered speech, and reversed stuttered speech. All conditions induced significant degrees of stuttering inhibition when compared to a control c

Circadian rhythm of melatonin release in pineal gland culture: arg-vasopressin inhibits melatonin release.

The mammalian pineal gland is known to receive a noradrenergic sympathetic efferent signal from the suprachiasmatic nucleus (SCN) via the superior cervical ganglion. Arg-vasopressin (AVP) containing neurons in the SCN is one of the output paths of circadian information to the other brain areas. AVP release from the SCN is suppressed by melatonin. In turn, we determined the direct effect of AVP on melatonin release using pineal gland explant culture. AVP (1 microM) suppressed melatonin release. Noradrenaline stimulated melatonin release was attenuated by AVP. In turn, the expression of the mela

Dorsal root lesions block the expression of morphine withdrawal elicited from the rat spinal cord.

Evidence has been accumulating to indicate that the spinal cord plays an important role in the expression of several narcotic abstinence signs in the intact animal. One characteristic and reproducible withdrawal sign which can be elicited in both intact and spinal-transected dependent rats is the naloxone-induced increase in arterial blood pressure. Employing this model, this autonomic component of narcotic withdrawal was quantitated in dependent spinal (C1)-transected rats with intact dorsal roots and in those with surgical lesions of the dorsal roots from T3 to L4. The withdrawal-associated

Electroresponsive properties of dendrites in central neurons.

A brief sketch of the historic development of the concept of neuronal integration is given. Some of the properties of dendritic spikes in chromatolyzed motoneurons, alligator Purkinje cells, and fish oculomotor neurons are reviewed, as well as the various ways in which the generation of dendritic spikes has altered our basic concept of neuronal integration. The principles of dendritic inhibition are elaborated, both as a "tonic modulatory action" and as a "functional amputating system" for dendrites that generate spikes.

Determination of anodal tDCS intensity threshold for reversal of corticospinal excitability: an investigation for induction of counter-regulatory mechanisms.

Transcranial direct current stimulation is applied to modulate activity, and excitability of the brain. Basically, LTP-like plasticity is induced when anodal tDCS (a-tDCS) is applied over the primary motor cortex. However, it has been shown that specific parameters of a-tDCS can induce a plasticity reversal. We aimed to systematically assess the intensity threshold for reversal of the direction of plasticity induced by a-tDCS, monitored by corticospinal excitability (CSE), and explored mechanisms regulating this reversal. Fifteen healthy participants received a-tDCS in pseudo-random order for

Neural correlates of altered sensorimotor gating in boys with Tourette Syndrome: A combined EMG/fMRI study.

OBJECTIVES: It has been hypothesised that altered sensorimotor gating might be a core problem in Tourette Syndrome (TS). However, the underlying neurophysiological mechanisms are elusive. METHODS: We applied functional magnetic resonance imaging (fMRI) to investigate the neural correlates of altered sensorimotor gating by means of prepulse inhibition (PPI) in 22 boys with TS and 22 healthy boys using tactile PPI. The electromyography of the startle response was recorded simultaneously to the acquisition of the fMRI images. RESULTS: As expected, PPI of the startle response was reduced in boys

Semantic ambiguity and the failure of inhibition hypothesis as an explanation for reading errors in deep dyslexia.

We report evidence for dissociation between explicit and implicit access to word representations in a deep dyslexic patient (JO). JO read aloud a series of ambiguous (e.g., bank) and unambiguous (e.g., food) words and performed a lexical decision task using these same items. When required to explicitly access the items (i.e., naming), JO showed relative impairment for ambiguous compared with unambiguous words. In contrast, the same stimuli produced an advantage for ambiguous words in lexical decision. The results are discussed within a framework of deep dyslexia that considers errors in produc

Focus on the excitatory and inhibitory neurotransmission imbalance in amyotrophic lateral sclerosis: a harmful disease player or a potential therapeutic opportunity?

Amyotrophic lateral sclerosis (ALS) is a progressive and fatal neurodegenerative disease affecting both upper and lower motor neurons. Evidence indicates that ALS is a "multifactorial" and "multicellular" disease; however, the causes of ALS remain elusive, as the mechanisms underlying the disease have not yet been completely clarified. One major proposed mechanism, first described in 1990, is the glutamate excitotoxicity theory. This theory suggests that excessive glutamatergic neurotransmission, combined with impaired glutamate clearance, significantly contributes to motor neuron degeneration

Optotagging in primate cortex uncovers excitatory and inhibitory neuron specializations in form vision.

The perception of complex visual shapes is a hallmark of primates and a foundation for many of their decisions and actions. The neural mechanisms of this visual function are best studied in macaques, but progress in this animal model has been hampered by the inability to dissect the contributions of distinct neuronal cell types. Here, we developed and applied an optogenetic approach for identifying the responses of individual excitatory and inhibitory neurons in the macaque visual cortex with light. Using this optotagging approach in male macaques, we uncovered clear functional differences bet

Regulation of PV interneuron plasticity by neuropeptide-encoding genes.

Neuronal activity must be regulated in a narrow permissive band for the proper operation of neural networks. Changes in synaptic connectivity and network activity-for example, during learning-might disturb this balance, eliciting compensatory mechanisms to maintain network function1-3. In the neocortex, excitatory pyramidal cells and inhibitory interneurons exhibit robust forms of stabilizing plasticity. However, although neuronal plasticity has been thoroughly studied in pyramidal cells4-8, little is known about how interneurons adapt to persistent changes in their activity. Here we describe

Mutual oligosynaptic inhibition of group Ia afferents between the anterior and posterior parts of the deltoid in humans.

The anterior (DA) and posterior parts of the deltoid (DP) show alternating contraction during shoulder flexion and extension movements. It is expected that an inhibitory spinal reflex between the DA and DP exists. In this study, spinal reflexes between the DA and DP were examined in healthy human subjects using post-stimulus time histogram (PSTH) and electromyogram averaging (EMG-A). Electrical conditioning stimulation was delivered to the axillary nerve branch that innervates the DA (DA nerve) and DP (DP nerve) with the intensity below the motor threshold. In the PSTH study, the stimulation t

Brainstem inhibitory neurons enhance behavioral feature selectivity by sharpening the tuning of excitatory neurons.

The brainstem is a hub for sensorimotor integration, which mediates crucial innate behaviors. This brain region is characterized by a rich population of GABAergic inhibitory neurons, required for the proper expression of these innate behaviors. However, what roles these inhibitory neurons play in innate behaviors and how they function are still not fully understood. Here, we show that inhibitory neurons in the nucleus of the optic tract and dorsal-terminal nuclei (NOT-DTN) of the mouse can modulate the innate eye movement optokinetic reflex (OKR) by shaping the tuning properties of excitatory

Electrical field distribution of Change-N1 and its prepulse inhibition.

An abrupt change in a sound feature (Test) in a continuous sound elicits an auditory evoked potential, peaking at approx. 100-180 ms (Change-N1) after the change onset. Change-N1 is attenuated by a preceding weak change stimulus (Prepulse), in the phenomenon known as prepulse inhibition (PPI). In this electroencephalographic study, we compared these two indexes among scalp electrodes. Change-N1 was elicited by an abrupt 10-dB increase in sound pressure in repeats of a 70-dB click sound at 100 Hz and was recorded using 22 electrodes in 31 healthy subjects. The prepulse was a 10-dB decrease in t

Bittersweet: The Neuroscience of Ambivalent Affect.

Ambivalent affective states, such as bittersweetness, nostalgia, and longing, which are felt as having both positive and negative aspects, are an important component of human experience but have received little attention. The most influential theoretical frameworks in affective neuroscience focus largely on univalenced states. As a result, it is not known whether ambivalent affect corresponds to a simultaneously positive and negative valenced state or whether it results from a rapid vacillation between positive and negative states. Here we hypothesize that ambivalent affect involves both mecha

The Effect of Optogenetic Inhibition of the Anterior Cingulate Cortex in Neuropathic Pain Following Sciatic Nerve Injury.

Cortical disinhibition is the underlying pathological alteration contributing to neuropathic pain associated with peripheral nerve injury. Nerve injury resulting in disinhibition of the anterior cingulate cortex has been reported. However, the effect of optogenetic inhibition of the anterior cingulate cortex (ACC) on the sensory component of nerve injury-induced neuropathic pain has not been well studied. To investigate the feasibility of optogenetic ACC modulation, we injected an optogenetic virus or a null virus into the ACC of a nerve injury-induced neuropathic pain model. The unilateral AC

Modulating functional connectivity with non-invasive brain stimulation for the investigation and alleviation of age-associated declines in response inhibition: A narrative review.

Response inhibition, the ability to withhold a dominant and prepotent response following a change in circumstance or sensory stimuli, declines with advancing age. While non-invasive brain stimulation (NiBS) has shown promise in alleviating some cognitive and motor functions in healthy older individuals, NiBS research focusing on response inhibition has mostly been conducted on younger adults. These extant studies have primarily focused on modulating the activity of distinct neural regions known to be critical for response inhibition, including the right inferior frontal gyrus (rIFG) and the pr

EEG Correlates of Preparatory Orienting, Contextual Updating, and Inhibition of Sensory Processing in Left Spatial Neglect.

Studies with event-related potentials have highlighted deficits in the early phases of orienting to left visual targets in right-brain-damaged patients with left spatial neglect (N+). However, brain responses associated with preparatory orienting of attention, with target novelty and with the detection of a match/mismatch between expected and actual targets (contextual updating), have not been explored in N+. Here in a study in healthy humans and brain-damaged patients of both sexes we demonstrate that frontal activity that reflects supramodal mechanisms of attentional orienting (Anterior Dire

Focal inhibitory seizure with prolonged deficit in adult Sturge-Weber syndrome.

Sturge-Weber syndrome is a sporadic congenital neurocutaneous disorder often related to varying degrees of motor impairment. The phenomenon of prolonged ictal paresis is a rare seizure sign that can be due to lesions affecting the centro-parietal lobe. Focal inhibitory motor seizures can be difficult to differentiate from other clinical entities such as stroke, migraine or postictal paresis. We describe the case of a 40-year-old patient suffering from Sturge-Weber syndrome, admitted due to prolonged right-sided hemiparesis following a usual seizure. Repeated EEGs during the prolonged deficit s
