Acne: a new model of immune-mediated chronic inflammatory skin disease.

Acne is a chronic inflammatory disease of the sebaceous-pilosebaceous unit. Interestingly, inflammation can be detected by histopathological examination and immuohistochemical analysis even in the apparently non-inflammatory acneic lesions, such as comedones. In the last years, it has been clearly demonstrated that acne development is linked to the combination of predisposing genetic factors and environmental triggers, among which a prominent role is played by the follicular colonization by Propionibacterium acnes (P. acnes). P. acnes displays several activities able to promote the development

[Proinflammatory cytokines in children with idiopathic headache].

OBJECTIVES: The pathogenesis of primary headaches is differential, with a neurogenic inflammation participation. THE AIM: The aim of this research was to establish whether inflammation participates in the pathogenesis of migraine and tension-type headaches (ETTH). In order to establish this, the levels of interleukin-6 (IL-6), interleukin-1 beta (IL-1beta), tumor necrosis factor (TNF) and soluble TNF receptor type I (sTNFRI) were detected in groups with headaches during headache-free interval and in controls and in the blood of children with idiopathic headaches to detect changes during heada…

The ambiguity of opioids revealed by immunology is changing the knowledge and the therapeutic approach in cancer and non-cancer pain: A narrative review.

Herein, we summarize the steps of a common scientific path taken by the two Guest Editors, an Anesthesiologist (EA) and an Immunologist (AS), and started 25 years ago at the National Cancer Institute in Rome. When in 1980 WHO codified the usage of opioids for cancer pain relief, it was matter of debate whether only disease progression rather than opioid tolerance were the driving force of opioid escalation. The selective intratumoral accumulation of morphine observed in an experimental xenograft model - the initial scenario of our scientific collaboration - revealed a surprising interaction be

The prevalence of IgG4-related hypophysitis in 170 consecutive patients with hypopituitarism and/or central diabetes insipidus and review of the literature.

OBJECTIVE: The prevalence and clinical characteristics of IgG4-related hypophysitis remain unclear due to the limited number of case reports. Therefore, in this study, we screened consecutive outpatients with hypopituitarism and/or diabetes insipidus (DI) to estimate its prevalence. METHODS: A total of 170 consecutive outpatients with hypopituitarism and/or central DI were screened at Kobe University Hospital for detecting IgG4-related hypophysitis by pituitary magnetic resonance imaging, measuring serum IgG4 concentrations, assessing the involvement of other organs, and carrying out an immun

Calcitonin gene-related peptide receptor expression in healthy and inflamed human pulp tissue.

AIM: To use radioreceptor analysis for comparing calcitonin gene-related peptide (CGRP) receptor expression in human pulp tissue samples collected from teeth having a clinical diagnosis of acute irreversible pulpitis, healthy pulps and teeth with induced inflammation. METHODOLOGY: Six pulp samples were obtained from teeth having a clinical diagnosis of acute irreversible pulpitis. Another eight pulp samples were obtained from healthy premolars where extraction was indicated for orthodontic purposes. In four of these premolars, inflammation was induced prior to pulp collection. All the samples

Unraveling the relationship between inflammation and cluster headache.

Cluster headache (CH) is often referred to as the 'suicide headache.' Existing research suggests that the activation of the trigeminal-vascular system, increased sensitivity of nerve fibers, and the release and interaction of various neuropeptides and inflammatory mediators may contribute to neurogenic inflammation, which serves as a crucial pathophysiological basis for the development of CH. Additionally, some neuropeptides can modulate neuronal activity related to pain transmission and may increase pain perception by sensitizing central nerves. This review discusses the neuropeptides and inf…

Interleukin 35-Producing B Cells (i35-Breg): A New Mediator of Regulatory B-Cell Functions in CNS Autoimmune Diseases.

Neuroinflammation contributes to neuronal deficits in neurodegenerative CNS (central nervous system) autoimmune diseases, such as multiple sclerosis and uveitis. The major goal of most treatment modalities for CNS autoimmune diseases is to limit inflammatory responses in the CNS; immune-suppressive drugs are the therapy of choice. However, lifelong immunosuppression increases the occurrence of infections, nephrotoxicity, malignancies, cataractogenesis, and glaucoma, which can greatly impair quality of life for the patient. Biologics that target pathogenic T cells is an alternative approach tha

Substance P and mast cells: preliminary histologic analysis of the human temporomandibular joint.

PURPOSE: Neuropeptide-containing nerves can serve as a mechanism for nervous system regulation of host defense responses. Because bacteria associated with reactive arthritis have been identified in the temporomandibular joint (TMJ), this study investigates whether the presence of substance P (SP) neuropeptide-containing nerves and mast cells can be identified in the TMJ. MATERIAL AND METHODS: Posterior bilaminar tissue removed during TMJ surgery from 9 women was evaluated for the presence of neuropeptide-containing nerves by staining with a monoclonal antibody to SP. Staining of the TMJ tissu

[Hypoxic-ischemic encephalopathy in the full-term newborn infant. Recent advances, markers of hypoxia and therapeutic options].

INTRODUCTION: Hypoxic-ischemic encephalopathy is the neurological consequence of a nonprogressive encephalopathic clinical picture of the hypoxic-ischemic syndrome, caused by a mixture of reduced oxygenation of the blood with increased carbon dioxide (asphyxia) and a lack of tissue perfusion (ischemia). It is the chief cause of death during the perinatal period, and of nonprogressive neurological deficits in childhood. DEVELOPMENT AND CONCLUSIONS: Regarding neurophysiological factors, we emphasise the importance of re-perfusion and reoxygenation in hypoxia-ischemia, with increased oxidative s

Environmental Noise and Childhood Asthma: A Hypothesis-Generating Review of Potential Neuro-Immune Mechanisms.

Childhood asthma is a heterogeneous prevalent chronic airway disease in paediatrics worldwide. It not only directly impairs the cardiopulmon … ly, it induces central sensitisation through the 'lung-brain axis', triggering substance P-mediated neurogenic inflammation; and it disrupts sleep rhythms, triggering airway hyperresponsiveness during the nocturnal vagal hypertonic period. Topical regimens for the management of uremic pruritus.

Uremic pruritus (UP), also known as chronic kidney disease-associated pruritus, is a distressing condition affecting patients with end-stage renal disease. Characterized by persistent itching, it significantly impacts patients' quality of life, leading to sleep disturbances, emotional distress, and decreased overall well-being. Although the exact pathophysiology remains uncertain, immune dysregulation, reduced stratum corneum hydration, and altered neurogenic inflammation contribute to UP. Consequently, treating UP poses a significant challenge. Among therapeutic regimens, topical regimens hav…

Mast cell-derived histamine mediates cystitis pain.

BACKGROUND: Mast cells trigger inflammation that is associated with local pain, but the mechanisms mediating pain are unclear. Interstitial cystitis (IC) is a bladder disease that causes debilitating pelvic pain of unknown origin and without consistent inflammation, but IC symptoms correlate with elevated bladder lamina propria mast cell counts. We hypothesized that mast cells mediate pelvic pain directly and examined pain behavior using a murine model that recapitulates key aspects of IC. METHODS AND FINDINGS: Infection of mice with pseudorabies virus (PRV) induces a neurogenic cystitis asso

Chronic stable recording of physiologically evoked neural activity in a mixed autonomic nerve in awake, freely moving normal and experimental cystitis rats.

Closed-loop control over bladder function, for conditions such as neurogenic bladder dysfunction, requires real-time monitoring of bladder state. Here, we assessed the ability of a novel recording and analysis technique to extract neural activity from awake rats as a quasi-real-time feedback biomarker of bladder fullness in normal and chronic cystitis models. Adult male rats (n = 12) were implanted with a four-electrode planar array and the bladder instrumented for continuous-flow cystometry over 2 weeks. In three rats, bladder inflammation was induced using cyclophosphamide (CYP, 75-100 mg/kg

BTX additionally modulates TRPV1-expressing afferents, which further reduces AD risk. Although hypertensive peaks improve, submaximal parameters remain unchanged, highlighting the need for additional complementary strategies. CONCLUSION: The use o …

Resveratrol mitigates activated astrocytes and microglia preventing Alzheimer's Disease (AD) progression and facilitates neuronal communication in Amyloid-β25-35 induced rat model for AD: A special emphasis on non-neuronal involvement in AD pathophysiology.

RATIONALE: Amyloid deposits initiate neuroinflammation by activating astrocytes and microglia in the hippocampus, increasing neuronal vulnerability and loss. Astrocytes, while essential for cerebral function, can contribute to neuronal dysfunction by retracting neuronal synapses, that forms a consequence of neuroinflammation, leading to cognitive deficits in Alzheimer's disease (AD). Upon Amyloid-β (Aβ) deposition, astrocytes become reactive as part of a repair mechanism, however this process can impair neurogenesis resulting in AD progression. OBJECTIVE: The current study hypothesizes that r

The innate immune system and neurogenesis as modulating mechanisms of electroconvulsive therapy in pre-clinical studies.

BACKGROUND: Electroconvulsive therapy (ECT) is a powerful and fast-acting anti-depressant strategy, often used in treatment-resistant patients. In turn, patients with treatment-resistant depression often present an increased inflammatory response. The impact of ECT on several pathophysiological mechanisms of depression has been investigated, with a focus which has largely been on cellular and synaptic plasticity. Although changes in the immune system are known to influence neurogenesis, these processes have principally been explored independently from each other in the context of ECT. OBJECTI

Sensitization of pelvic nerve afferents and mast cell infiltration in the urinary bladder following chronic colonic irritation is mediated by neuropeptides.

Irritable bowel syndrome and interstitial cystitis frequently overlap. We have shown that acute colitis sensitizes urinary bladder afferents to both mechanical and chemical stimuli and that chronic colitis similarly produces neurogenic cystitis. We hypothesize that chronic irritation of the colon releases neuropeptides from bladder afferents, leading to receptor sensitization and neurogenic inflammation. Female Sprague-Dawley rats received intrarectal trinitrobenzenesulfonic acid (TNBS) or vehicle 3 days following either systemic capsaicin (CP) pretreatment or vehicle. Ten days later, action p…
