Effects of acute adult and early-in-life bladder inflammation on bladder neuropeptides in adult female rats.

BACKGROUND: The purpose of the present study was to determine how acute adult and/or prior early-in life (EIL; P14-P16) exposure to bladder inflammation affects bladder content of calcitonin gene related peptide (CGRP) and substance P (SP). Estrous cycle influences were also studied in the adult-treatment conditions. METHODS: In Experiment 1, intravesical zymosan or isoflurane anesthesia alone was administered to adult female rats. Bladders and serum were collected 24 hours later during each phase of the estrous cycle. In Experiment 2, zymosan or anesthesia alone was administered EIL and as a

Modulation of neuronal nicotinic receptor function by the neuropeptides CGRP and substance P on autonomic nerve cells.

1. One classical example of how neuropeptides can affect the function of ligand-gated receptors is the modulation of neuronal nicotinic receptors (nAChRs) by substance P. The present review updates current understanding of this action by substance P and compares it with other neuropeptides more recently found to modulate nAChRs in the autonomic nervous system. 2. Calcitonin gene-related peptide (CGRP) and its N-terminal fragments have been shown to exert complex inhibitory as well facilitatory actions on nAChRs. Fragments such as CGRP(1-4), CGRP(1-5) and CGRP(1-6) rapidly and reversibly enhanc…

Pharmacological evidence that neuropeptides mediate part of the actions of scorpion venom on the guinea pig ileum.

Severe human scorpion envenoming is characterised by instability of several physiological systems and death. These manifestations are explai … ptors in mediating part of the contractile effects of the venom on guinea pig ileum. The release of neuropeptides may play an important role in the systemic manifestations of severe envenoming.

Neuronal overmaturation in dysraphism: ontogenic expression of neuropeptides in the fetal brain and developmental anomalies in exencephaly.

Starting from knowledge obtained in our previous studies of experimental dysraphism in chick embryos, the entity of neuronal overgrowth observed in exencephaly was further investigated. The ontogenic expression of neuron-specific enolase (NSE), vasoactive intestinal polypeptide (VIP), and somatostatin was analyzed both in chick exencephaly of the natural product and in normal chick fetuses by carrying out immunohistochemical studies. In normal fetuses, immunostained elements positive for NSE first appeared in the spinal gray matter on postincubation day 16 and increased in intensity during the

Long-term effects of alprazolam and imipramine on cerebrospinal fluid monoamine metabolites and neuropeptides in panic disorder.

Cerebrospinal fluid (CSF) homovanillic acid, 5-hydroxyindoleacetic acid, 3-methoxy-4-hydroxyphenylglycol, and somatostatin and beta-endorphin levels were measured in 12 patients with panic disorder before and after 7 months of treatment with alprazolam or imipramine. The concentrations of CSF monoamine metabolites and neuropeptides were at baseline of the same order of magnitude in panic patients and controls. Neither alprazolam nor imipramine changed metabolite or neuropeptide levels in CSF despite clinical improvement in both treatment groups. According to present CSF data, the antipanic act…

Autocrine feedback maintains homeostatic neuropeptide expression in a peptidergic hub neuron.

Neuronal circuits sustain stable function by self-regulating their gene expression and neurosecretion. Patterns for autocrine feedback by neuropeptides are widespread across nervous systems, but how they contribute to neuronal homeostasis is less well understood. Here, we identify an autocrine homeostatic mechanism in a C. elegans peptidergic hub neuron. FLP-1 neuropeptide release activates the inhibitory receptor DMSR-7 on the AVK hub neurons to self-regulate flp-1 transcription, dense-core vesicle accumulation, and peptide secretion. We show that a parallel PDF-1/PDFR-1 pathway drives flp-1 …

A neuronal GPCR modifies neuropeptide signaling to suppress proteostasis in distal tissues.

The proteostasis network (PN) safeguards the integrity of proteins by promoting various cellular activities. However, with aging, the PN's c … ns. The knockdown of gtr-1 also enhances protein degradation. These findings highlight the roles of neuropeptides as organismal coordinators of proteostasis in the face of distinct proteotoxic challenges.

Neuropeptide Y and brain ageing: an updated scoping review on the role of autophagy.

Brain ageing is a complex physiological process characterised by progressive structural and functional changes that often result in cognitive decline and reduced quality of life in the elderly. These changes are driven by multiple mechanisms, including synaptic dysfunction, oxidative stress, neuroinflammation, and impaired cellular homeostasis. Neuropeptides have emerged as important regulators of these processes. Among them, neuropeptide Y (NPY) has gained attention for its potential role in mitigating age-related brain dysfunction. However, the extent to which NPY contributes to brain ageing…

Distinct contribution of spinal neuropeptide Y and NPY1R neurons to morphine analgesia.

Opioids produce potent antinociception by activating μ-opioid receptors (MOR) in the spinal dorsal horn, where MOR is broadly expressed in both inhibitory and excitatory neurons. However, the neuron subtype-specific actions and mechanisms of spinal MOR in opioid analgesia remain elusive. We employed chemogenetic approaches and conditional gene knockout strategies across multiple pain models to assess the roles of the NPY-NPY1R pathway and MOR signalling in spinal morphine analgesia. In addition, in situ hybridization combined with electrophysiological recordings from spinal cord slices was use

Intracerebroventricular injection of neuropeptide Y suppresses luteinizing hormone pulses in mice.

Stress impairs reproduction, at least in part, by inhibition of Kisspeptin/Neurokinin B/Dynorphin (KNDy) cells and suppression of gonadotropin secretion. However, the mechanism for this inhibition is not clear. Neuropeptide Y (NPY) is a pleiotropic molecule that is produced in neurons throughout the brain and mediates several physiological processes including energy homeostasis and stress responses. Centrally administered NPY elicits variable effects on luteinizing hormone (LH) secretion, which vary with species, gonadal status, and the site of infusion. Therefore, our objective was to determi

Pro-neuropeptide Y as a circulating biomarker for poor prognosis in prostate cancer.

Prostate cancer (PCa) is common world-wide. Current diagnostics based on testing for circulating levels of prostate specific antigen (PSA) is unspecific, and novel prognostic markers are needed for personalized therapeutic strategies. Pro-neuropeptide Y (pro-NPY) has been reported as a tissue marker for PCa related to poor prognosis. This study explored the prognostic value of circulating pro-NPY in PCa. Plasma samples were obtained from two patient cohorts: (1) men examined due to increased PSA levels in 2003-2011 (n = 796) and (2) patients treated for PCa in 2013-2016 (n = 92). Cohort 2 also

Neuropeptide TIP39 induces autophagy in PTH2 receptor-positive myeloid neoplasms.

Neuropeptides are chemical messengers that are synthesized and released by nerve cells. Studies suggest that neuropeptides released from the nervous system in bone marrow may be involved in the regulation of hematopoiesis and survival of leukemic stem cells (LSC). Parathyroid hormone 2 receptor (PTH2R), a new LSC marker, is expressed on CD34 + leukemic cells. Its ligand, tuberoinfundibular peptide of 39 residues (TIP39), is expressed in the nervous system. However, the role of the TIP39-PTH2R axis in leukemic cells is unclear. We investigated the function of this axis in leukemic cell lines, a…

A single injection of neuropeptide QRFP in the lateral hypothalamus decreased food intake.

BACKGROUND AND AIM: Severe eating disorders, such as obesity, bulimia, and anorexia, keep increasing to epidemic proportions worldwide. Understanding of neuropeptides' role in complex hunger/satiety mechanisms may allow new prospects for treatment and prevention. Pyroglutamylated arginine-phenylalanine-amide peptides (QRFPs) are thought to enhance feeding following the central administration. METHODS: In our study, QRFP-26 was delivered into the lateral hypothalamic area of male Wistar rats by direct microinjections, as QRFP-26 expressing neurons and binding sights are densely present in this…

Rapamycin reveals neuropeptide Y as a regulator of senescence and inflammatory pathways in arthritis.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic inflammatory disease characterized by immune dysregulation and joint destruction. Cellular senescence has been implicated in the progression of RA through the senescence-associated secretory phenotype (SASP), yet its molecular links to inflammation remain unclear. Rapamycin, an mTOR inhibitor with anti-inflammatory and anti-senescence properties, provides a valuable tool for exploring these mechanisms. OBJECTIVE: To investigate the link between senescence and inflammation in a murine model of RA by comparing the transcriptome of diseased join

Exploring the Role of Neuropeptide PACAP in Cytoskeletal Function Using Spectroscopic Methods.

The behavior and presence of actin-regulating proteins are characteristic of various clinical diseases. Changes in these proteins significantly impact the cytoskeletal and regenerative processes underlying pathological changes. Pituitary adenylate cyclase-activating polypeptide (PACAP), a cytoprotective neuropeptide abundant in the nervous system and endocrine organs, plays a key role in neuron differentiation and migration by influencing actin. This study aims to elucidate the role of PACAP as an actin-regulating polypeptide, its effect on actin filament formation, and the underlying regulato

Activity of the hypothalamic neuropeptide Y increases in adult and decreases in old rats.

Middle-aged obesity and aging anorexia with muscle loss (sarcopenia) of old people present public health burden. These alterations may appear both in humans and rodents suggesting the role for regulatory alterations. Previously, we demonstrated that biphasic changes in the weight-reducing (catabolic) effects of neuropeptides of the hypothalamus-adipose tissue axis (e.g. leptin) may contribute to both trends. With regard to the anabolic effects of the hypothalamic neuropeptide Y (NPY) inhibited by leptin, we hypothesized non-linear age-related changes with shifts in the opposite directions. We …

Cross talk about the role of Neuropeptide Y in CNS disorders and diseases.

A peptide composed of a 36 amino acid called Neuropeptide Y (NPY) is employed in a variety of physiological processes to manage and treat conditions affecting the endocrine, circulatory, respiratory, digestive, and neurological systems. NPY naturally binds to G-protein coupled receptors, activating the Y-receptors (Y1-Y5 and y6). The findings on numerous therapeutic applications of NPY for CNS disease are presented in this review by the authors. New targets for treating diseases will be revealed by medication combinations that target NPY and its receptors. This review is mainly focused on diso

Neuronal diversity of neuropeptide signaling, including galanin, in the mouse locus coeruleus.

The locus coeruleus (LC) is a small nucleus in the pons from which ascending and descending projections innervate major parts of the central nervous system. Its major transmitter is norepinephrine (NE). This system is evolutionarily conserved, including in humans, and its functions are associated with wakefulness and related to disorders, such as depression. Here, we performed single-cell ribonucleic acid-sequencing (RNA-seq) to subdivide neurons in the LC (24 clusters in total) into 3 NE, 17 glutamate, and 5 γ-aminobutyric acid (GABA) subtypes, and to chart their neuropeptide, cotransmitter,
