Phosphorus in the +5 oxidation state (i.e., phosphate) is the most abundant form of phosphorus in the global ocean. An enigmatic pool of dissolved phosphonate molecules, with phosphorus in the +3 oxidation state, is also ubiquitous; however, cycling of phosphorus between oxidation states has remained poorly constrained. Using simple incubation and chromatography approaches, we measured the rate of the chemical reduction of phosphate to P(III) compounds in the western tropical North Atlantic Ocean. Colonial nitrogen-fixing cyanobacteria in surface waters played a critical role in phosphate redu

Optimizing photosynthesis is imperative for providing energy and organics for all life on the earth. Here, carbon dots doped with pyridinic nitrogen (named lev-CDs) are synthesized by the one-pot hydrothermal method, and the structure-function relationship between functional groups on lev-CDs and photosynthesis of Chlorella pyrenoidosa (C. pyrenoidosa) is proposed. Pyridinic nitrogen plays a key role in the positive effect on photosynthesis caused by lev-CDs. In detail, lev-CDs act as electron donors to supply photo-induced electrons to P680+ and QA+ , causing electron transfer from lev-CDs to

Fatty acid esters of 3-monochloropropane 1,2-diol (3-MCPD esters) are processing-induced food toxicants, with the kidney as their major target organ. For the first time, this study treated Sprague Dawley (SD) rats with 3-MCPD 1-monooleate at 10 and 100 mg/kg BW/day and 1-monostearate at 15 and 150 mg/kg BW/day for 90 days and examined for their potential semi-long-term nephrotoxicity and the associated molecular mechanisms. No bodyweight difference was observed between groups during the study. Both 3-MCPD 1-monooleate and 1-monostearate resulted in a dose-dependent increase of serum urea creat

Interaction with ozone transfers its anomalous (non-mass-dependent) 17O enrichment to atmospheric nitrogen oxides and nitrate. The 17O anomaly (Delta17O) in nitrate can be used to identify atmospheric nitrate inputs into terrestrial and aquatic environments as well as to study the role of ozone in the atmosphere's reactive nitrogen cycle. We report here on an online method for analysis of the 17O anomaly, using a strain of denitrifiers to convert nitrate to N2O, which decomposes quantitatively to N2 and O2 in a gold furnace at 800 degrees C, followed by gas chromatographic separation and isoto…

In nine patients undergoing diagnostic bronchoscopy, intrapulmonary gas distribution was evaluated by means of nitrogen washout. The investigations were performed with the patients in the supine position, first awake during spontaneous breathing and then during general anesthesia with high-frequency positive-pressure ventilation (HFPPV) via a pneumatic valve connector. With HFPPV a ventilatory frequency (f) of 60/min and a relative insufflation time (t%) of 22% of the ventilatory cycle were used. Gas distribution in terms of lung clearance index and nitrogen washout delay improved during HFPPV

Typhoons and rainstorms (>250 mm/day) are extreme weather events changing hydrological characteristics and thus nitrogen (N) cycle in coastal waters. However, responses of N cycle to rainstorms and typhoons and their underlying mechanisms need to be elucidated. In this study, we conducted an analysis of a comparative dataset encompassing concentrations of nitrate (NO3-), ammonium (NH4+), dissolved oxygen (DO), chlorophyll a (Chl a), hydrological parameters, dual isotopic composition of NO3- (δ15N-NO3- and δ18O-NO3-) in Zhanjiang Bay during three distinct periods: the normal wet season, rainsto

The application of bio-manganese (Mn) redox cycling for continuous removal of contaminants provides promise for addressing coexisting contaminants in groundwater, however, the feasibility of constructing Mn redox cycling system (MCS) through community assembly remains to be elucidated. In this study, Mn-reducing strain MFG10 and Mn-oxidizing strain MFQ7 synergistically removed 94.67 % of 17β-estradiol (E2) within 12 h. Analysis of potential variations in Mn oxides suggested that MCS accelerated the production of reactive oxygen species (ROS) and Mn(III), which interacted to promote E2 removal.

INTRODUCTION: Urea cycle disorders are metabolic disorders of nitrogenous waste substances due to either complete or partial deficiency of enzymes. Hyperammonemia associated with urea cycle disorders should be addressed immediately in the acute setting, as it can cause irreversible neurological injury or death.
CASE PRESENTATION: We report the case of a 48-year-old woman who presented with lethargy, weakness, and altered mental status following prolonged nausea and vomiting despite an esophageal dilatation procedure 3 weeks prior. Further investigation with assistance from the genetics consul

Amino acid catabolism during fasting requires coordinated nitrogen disposal and glucose production, but the transcriptional logic linking the urea cycle to gluconeogenesis remains unclear. Forkhead box O (FoxO) transcription factors regulate fasting metabolism, and here we identify FoxOs as direct hepatic regulators of argininosuccinate synthase 1 (Ass1). Acute hepatic FoxO1/3a knockdown in fasted mice reduced Ass1 expression, lowered blood glucose, and altered urea-cycle amino acids, with decreased arginine and increased ornithine, even in Klf15-deficient livers. Ass1 silencing phenocopied th

Hyperammonemia outside the neonatal period is a time-critical emergency and may reveal a late-onset urea cycle disorder, including ornithine transcarbamylase deficiency (OTCD), which can be particularly challenging to recognize in females. We report the case of an 11-month-old female who presented with seizures and transient focal neurological deficits in the context of recurrent vomiting and acute hepatic dysfunction, with respiratory alkalosis and marked hyperammonemia accompanied by biochemical features compatible with a proximal urea cycle defect. Brain imaging showed stroke-like changes t

Natural killer (NK) cells exhibit impaired anti-tumor activity upon entering the tumor microenvironment (TME); however, the precise mechanism(s) remains elusive. In this study, we demonstrate that AQP5+ gastric cancer stem cells contribute to the dysfunction of NK cells by reprogramming the urea cycle (UC). Mechanistically, AQP5 competitively binds ATP-dependent RNA helicase A (DHX9) over karyopherin subunit beta 1 (KPNB1), inhibiting DHX9 nuclear translocation and transcriptionally down-regulating argininosuccinate synthase 1 (ASS1). Low-arginine condition in the TME reshaped by AQP5+ tumor c

Urea cycle disorders (UCD) are inherited defects in clearance of waste nitrogen with high morbidity and mortality. Novel and more effective therapies for UCD are needed. Studies in mice with constitutive activation of autophagy unravelled Beclin-1 as druggable candidate for therapy of hyperammonemia. Next, we investigated efficacy of cell-penetrating autophagy-inducing Tat-Beclin-1 (TB-1) peptide for therapy of the two most common UCD, namely ornithine transcarbamylase (OTC) and argininosuccinate lyase (ASL) deficiencies. TB-1 reduced urinary orotic acid and improved survival under protein-ric

Ornithine transcarbamylase (OTC) deficiency is the most common inborn error in the metabolism of the urea cycle with an incidence of 1 in 14,000 live births. Pregnancy can trigger potentially fatal hyperammonemic crises. We report a successful pregnancy in a 29-year-old primiparous patient with a known diagnosis of OTC deficiency since infancy. Hyperammonemic complications were avoided due to careful multidisciplinary management which included a detailed antenatal, intrapartum and postnatal plan. Management principles include avoidance of triggers, a low-protein diet and medications which prom

Intensive marine shrimp aquaculture discharges substantial nitrogen (N) and phosphorus (P) into coastal environments while generating substantial greenhouse gas emissions. Scalable in-situ nutrient recovery technologies that can be integrated with operational aquaculture production systems remain largely absent. This study developed and validated a floating permeable nutrient uptake system (FPNUS) containing mixotrophic Chaetoceros sp. in pilot-scale marine shrimp cultivation. Membrane screening (0.1, 0.5, and 1.0 µm) identified the 0.1 µm membrane, which sustained ammonium (NH4+-N) and phosph

Spontaneous day-time periodic breathing (sPB) constitutes a common phenomenon in systolic heart failure (HF). However, it is unclear whether PB during wakefulness could be easily induced and what are the physiological and clinical correlates of patients with HF in whom PB induction is possible. Fifty male HF patients (age 60.8 ± 9.8 years, left ventricle ejection fraction 28.0 ± 7.4%) were prospectively screened and 46 enrolled. After exclusion of patients with sPB the remaining underwent trial of PB induction using mild hypoxia (stepwise addition of nitrogen gas to breathing mixture) which re

Internalization of pathogenic bacteria by macrophages results in formation of antibiotic-tolerant persisters. These cells are maintained in a non-growing state for extended periods of time, and it is assumed that their growth resumption causes infection relapse after cessation of antibiotic treatment. Despite this clinical relevance, the signals and conditions that drive persister regrowth during infection are not yet understood. Here, we found that after persister formation in macrophages, host reactive nitrogen species (RNS) produced in response to Salmonella infection lock persisters in gro

The cell-intrinsic nature of tumor metabolism has become increasingly well characterized. The impact that tumors have on systemic metabolism, however, has received less attention. Here, we used adult zebrafish harboring BRAFV600E-driven melanoma to study the effect of cancer on distant tissues. By applying metabolomics and isotope tracing, we found that melanoma consume ~15 times more glucose than other tissues measured. Despite this burden, circulating glucose levels were maintained in disease animals by a tumor-liver alanine cycle. Excretion of glucose-derived alanine from tumors provided a

BACKGROUND: Hyperammonemic encephalopathy in newborns with urea cycle disorders and certain organic acidurias can cause severe brain injury, coma and death. Standard therapy includes protein restriction, nitrogen-scavenging drugs, prevention of catabolism and hemodialysis. Neuroprotective hypothermia as part of the treatment has been reported only 3 times. It has been suggested that mild systemic hypothermia can contribute to better neurological outcomes in hyperammonemic encephalopathy. However, the limited experience precludes accurate conclusions on safety and efficacy.
METHODS: Whole body

OBJECTIVE: In patients with severe urea cycle defects (UCD) metabolic decompensation with hyperammonaemia typically occurs during the first days of life resulting in severe neurological damage or death. Benzoate can eliminate nitrogen independent of the urea cycle. Usually, benzoate is started soon after birth, but prenatal administration might improve metabolic stability.
DESIGN: Two fetuses with a prenatal diagnosis of UCD (female: citrullinaemia; male: ornithine transcarbamylase deficiency) were loaded with benzoate prenatally via the placenta by infusing their mothers with benzoate. Benzo

Arabidopsis wildtype plants suffer symptoms of stress at a sudden increase in CO2 concentration, resulting from perturbation of photosynthetic electron transport. Defense-related gene induction includes increased methionine cycle and glucosinolates metabolism. Elevated CO2 (eCO2) increases photosynthetic performance of plants, but also leads to decreased nitrogen-to-carbon ratio and a long-term decline in photosynthetic activity, known as photosynthetic acclimation. It is unclear whether initially increased CO2 assimilation or perturbation of the physiological homeostasis triggers acclimation.
