A Tunisian patient with two rare syndromes: triple a syndrome and congenital hypogonadotropic hypogonadism.

BACKGROUND/AIMS: The coexistence of triple A syndrome (AAAS) and congenital hypogonadotropic hypogonadism (CHH) has so far not been reported in the literature. This study aimed to characterize at the clinical and genetic level one patient presenting an association of AAAS and CHH in order to identify causal mutations. METHODS: Clinical and endocrinal investigations were performed and followed by mutational screening of candidate genes. RESULTS: At the age of 18, the patient presented sexual infantilism, a micropenis and gynecomastia. No mutation was revealed in GnRHR, TACR3/TAC3, PROK2/PROKR

Progressive bulbospinal amyotrophy in triple A syndrome with AAAS gene mutation.

Triple A (3A) syndrome, a rare autosomal recessive disorder, is characterized by adrenocorticotropic hormone-resistant adrenal insufficiency, achalasia of the cardia, alacrima, and variable autonomic and neurologic dysfunction. The gene responsible, AAAS, recently has been identified. We describe the neurologic phenotype of the first adult case of 3A syndrome presenting bulbospinal amyotrophy as the prominent sign in association with a homozygous nonsense mutation identified in the AAAS gene.

Acetyl-CoA-dependent processes are preferentially supported by local metabolite synthesis.

The cell nucleus is an active metabolic site. Numerous enzymes best known for their roles in cytosolic or mitochondrial pathways also function in the nucleus, where they contribute to gene regulation and DNA replication and repair. Although metabolites can diffuse through nuclear pores, it remains unclear the extent to which the nucleus and cytosol operate as continuous versus distinct metabolic spaces. Both compartments require acetyl-CoA-for example, for histone acetylation and lipid synthesis-and the acetyl-CoA generating enzyme ATP-citrate lyase (ACLY) resides in both locations, but the si…

The cytoskeleton dynamics-dependent LINC complex in periodontal ligament stem cells transmits mechanical stress to the nuclear envelope and promotes YAP nuclear translocation.

BACKGROUND: Periodontal ligament stem cells (PDLSCs) are important seed cells in tissue engineering and clinical applications. They are the … ssion of the LINC complex. This further strengthened the pull on the nuclear envelope, enlarged the nuclear pore, and facilitated YAP's nuclear entry, thus enhancing the expression of proliferation-related genes. In this process, mitochondria were transported to the periphery of the nucleus along the reconstructed microtubules. They generated ATP to aid YAP's …

Multiple lines of evidence for disruption of nuclear lamina and nucleoporins in FUS amyotrophic lateral sclerosis.

Advanced pathological and genetic approaches have revealed that mutations in fused in sarcoma/translated in liposarcoma (FUS/TLS), which is pivotal for DNA repair, alternative splicing, translation and RNA transport, cause familial amyotrophic lateral sclerosis (ALS). The generation of suitable animal models for ALS is essential for understanding its pathogenesis and developing therapies. Therefore, we used CRISPR-Cas9 to generate FUS-ALS mutation in the non-classical nuclear localization signal (NLS), H517D (mouse position: H509D) and genome-edited mice. Fus WT/H509D mice showed progressive m

Highly selective adsorption and lattice process of cesium by cubic cyanide-based functional materials.

Cesium (Cs) is a byproduct of nuclear bombs, nuclear weapons testing, and nuclear fission in nuclear reactors. Cs can enter the human body through food or air and cause lasting damage. Highly efficient and selective removal of 137Cs from low-level radioactive effluents (LLREs), which contain many radionuclides and dissolved heavy metal species, is imperative for minimizing LLRE volume, and facilitating their final disposal. Prussian blue analogs (PBAs) have received much attention as materials for the removal of radioactive Cs because of their affinity for adsorbing Cs+. In this study, an inex

Biallelic NDC1 variants that interfere with ALADIN binding are associated with neuropathy and triple A-like syndrome.

Nuclear pore complexes (NPCs) regulate nucleocytoplasmic transport and are anchored in the nuclear envelope by the transmembrane nucleoporin NDC1. NDC1 is essential for post-mitotic NPC assembly and the recruitment of ALADIN to the nuclear envelope. While no human disorder has been associated to one of the three transmembrane nucleoporins, biallelic variants in AAAS, encoding ALADIN, cause triple A syndrome (Allgrove syndrome). Triple A syndrome, characterized by alacrima, achalasia, and adrenal insufficiency, often includes progressive demyelinating polyneuropathy and other neurological compl…

Identification of two-pore channel 2 as a novel regulator of osteoclastogenesis.

Osteoclast differentiation is one of the critical steps that control bone mass levels in osteoporosis, but the molecules involved in osteoclastogenesis are still incompletely understood. Here, we show that two-pore channel 2 (TPC2) is expressed in osteoclast precursor cells, and its knockdown (TPC2-KD) in these cells suppressed RANKL-induced key events including multinucleation, enhancement of tartrate-resistant acid phosphatase (TRAP) activities, and TRAP mRNA expression levels. With respect to intracellular signaling, TPC2-KD reduced the levels of the RANKL-induced dynamic waving of Ca(2+) i

Functional consequences of deleting the two C-terminal residues of the scorpion toxin BmTX3.

We deleted the two C-terminal residues of the scorpion toxin BmTx3, a peptidyl inhibitor of a transient A-type K(+) current in striatum neurons in culture, to assess their contribution to receptor recognition. The sBmTX3-delYP analog was shown to have a native-like structure in one-dimensional 1H-nuclear magnetic resonance (NMR) spectroscopy. We found that sBmTX3-delYP bound to its receptor less efficiently than the wild-type molecule (by a factor of about 10(5)) in binding assays with rat brain membranes, and that this molecule did not block the A-type K(+) current (at a concentration of 35 m

Genetic Spectrum of Nephrotic Syndrome: Impact of Podocytopathy in Adult Life.

A substantial number of patients with focal segmental glomerulosclerosis (FSGS) have a pathogenic genetic mutation in a podocyte protein as the cause of their disease. The mutations can affect a wide range of cell functions including the actin cytoskeleton, cell adhesion and motility, mitochondrial function, and nuclear pore proteins. The likelihood of a genetic cause declines with age, from approximately 30% in children and adolescents to 10% in adulthood, and the specific proteins involved and the pattern of inheritance differ in the 2 age groups. The presence of a genetic cause for FSGS can…

Speculations based on the morphology of the Golgi systems in several types of proteinsecreting cells.

Electron microscopical observations on the relationship of the Golgi region to other intracellular organelles in certain protein-secreting cells have substantiated and extended existing hypotheses. In micrographs of several cell types, the juxtanuclear Golgi regions were observed to be closely associated with nuclear "pores." The "transition elements" of the ergastoplasmic membranes possess "blebs" which may represent a transport process facilitating the movement of intracisternal contents into the Golgi zone. A "blebbing" process of this nature may be one source of the small variety of Golgi

Selective Plugging and Flow Reallocation Characteristics of a Time-Controllable Silica-Reinforced Cationic Polymer Gel in High-Temperature, High-Salinity Heterogeneous Porous Media.

Preferential flow through high-permeability pathways remains a central challenge for polymer-based conformance control in reservoirs where high temperature, high salinity, and strong heterogeneity compromise gel stability and selectivity. Here, a delayed-gelation organic-inorganic gel was formulated from a microbranched cationic copolymer, P-(AM-co-DMDAAC), a urea-formaldehyde/phenol-formaldehyde prepolymer cross-linker, and silica nanoparticles. The precursor retained injectability and formed a grade H gel within 15 h at 90 °C in 240,000 ppm brine. Transmission electron microscopy and element

Glassy dynamics of LiCl · 6H2O solution in nanoporous media.

Understanding how nanoconfinement alters the dynamics of glass-forming aqueous electrolytes is essential for clarifying the interplay among ionic hydration, hydrogen-bond structure, and interfacial effects. Here, LiCl · 6H2O was investigated in the bulk and under confinement in SBA-15 mesoporous silica with an average pore diameter of 8 nm. Differential scanning calorimetry, Raman spectroscopy, quasielastic neutron scattering, 1H spin-lattice relaxation, and pulsed-field-gradient nuclear magnetic resonance (NMR) were combined to probe thermal behavior, hydrogen-bond structure, local mobility,

Construction of Unsaturated Coordinated Cu-Metal-Organic Frameworks for Iodine Adsorption.

Radioactive iodine from nuclear energy poses serious risks to aquatic environments, driving the need for efficient adsorbents for nuclear wastewater treatment. Metal-organic frameworks (MOFs) have shown promise in iodine capture due to their tunable pores and functional surfaces. In this work, three copper-triazolate-based MOFs with unsaturated coordination (CuCl2(TRZ) for 1, Cu4OCl3(TRZ)3 for 2, Cu3HOCl2(TRZ)3·2.4H2O for 3; TRZ = 1,2,4-triazole) were synthesized and characterized. Compound 1 exhibits a one-dimensional chain structure, compound 2 exhibits a two-dimensional layer, and compound

Colloid osmotic parameterization and measurement of subcellular crowding.

Crowding of the subcellular environment by macromolecules is thought to promote protein aggregation and phase separation. A challenge is how to parameterize the degree of crowding of the cell interior or artificial solutions that is relevant to these reactions. Here I review colloid osmotic pressure as a crowding metric. This pressure is generated by solutions of macromolecules in contact with pores that are permeable to water and ions but not macromolecules. It generates depletion forces that push macromolecules together in crowded solutions and thus promotes aggregation and phase separation.
